1.Study on the Expression of Endothelin 1 mRNA in Keratinocytes in Patients with Vitiligo

Objective To study the expression of endothelin 1 (ET 1) mRNA in keratinocytes in patients with vitiligo. Methods Fourteen patients with progressive vitiligo were measured by hybridization in situ. Results Our results showed that the expression of ET 1 mRNA was markedly decreased. Conclusion The results revealed that decreased ET 1 expression may be associated with the pathogenesis of vitiligo. The cause and role of decreasing of ET 1 expression of keratinocyte in vitiligo lesion need to be studied further.
2.Experimental study on myocardial injury and protective effect of ulinastatin after bile duct obstruction

Objective To investigate the myocardial injury induced by bile duct obstruction and the protective effects ulinastatin(UTI). Methods Dynamic observation of of the levels of malondialdyhyde(MDA) and superoxide dismutase(SOD) of myocardial tissues, and the levels of serum TBil, alkaline phosphatase(ALP), MB isoenzyme of creatine kinase(CK MB), endotoxin (ET), and tumor necrosis factor(TNF ?) in bile duct obstruction(BLD) rats and UIT treatment rats. The left ventricles of the rats were obtained for light and electronic microscopic observation . Immunohistochemical staining method of ABC used was to locate the expression and distribution of TNF ? in myocardial tissues. Results After ligation of the common bile duct, serum TBIL, ALP, CK MB, ET, TNF ? levels and myocardium MDA gradully increased, while SOD levels gradually decreased, and the expression of TNF ? in myocardium increased. As compared with BDL group at the same phase, in UIT group, serum TBIL, ALP, CK MB, ET, TNF? levels and myocardium MDA in UTI treated groups decreased, while myocardium SOD increased, and the expression of TNF ? in myocardium decreased.Myocardial injuries of bile duct obstruction were aggravated as time progressed, and there were less myocardial injuries in UTI treated groups than in BDL groups at the same stage as shown with light and electronic microscopic observation.Conclusions UTI can effectively protect the myocardium from ET,TNF ? and free radical injury in bile duct obstucion rats.
3.Preparation of Zhitong sustained-release tablet and its release property in vitro

Objective To prepare sustained-release tablet contained paeoniflorin and evaluate its drug release mechanism.Methods Orthogonal design was used to obtain the best formula.Hydroxypropykmethyl cellulose(HPMC K4M)and ethylcellulose(EC 20CPS)were used as hydrophilic matrix material,and PVP which dissolved in ethanol was used as adhesive material,then the sustained-release tablet was prepared by the wet granule compression technique.The best formula was determined by powder technology,including compression ratio,angle of repose,flow rate,and angle of contact as indexes.The optimum formula was obtained by comprehensive scoring method.The drug release in vitro was determined,and the release formula was made to investigate the release mechanism.Results The optimized prescription was with HPMC 20%,EC 20%,PVP 0%,and ethanol concentration 80%.The dissolution curves in vitro showed that the drug release could be best described by the Higuchi equation(Q=0.367 5 t1/2-0.165 8,r=0.993 2)and Peppas equation(lnQ=0.72 lnt-1.608 6,r=0.991 8).It showed that the release mechanism was Fick diffusion and backbone corrosion.The release rate could meet the requirement of quality control on sustained-release tablet in Chinese Pharmacopoeia(2005).Conclusion The method of content determination is simple and sustained-releasing rate is significant.The release mechanism follows the kinetic model of sustained-release tablet.It is worth doing further research for the clinical use.
5.HPLC Fingerprint and chemical pattern recognition of Semen Cassiae

Objective To establish a method for the quality control of Semen Cassiae.Methods A high performance liquid chromatographic method was developed to establish the fingerprint of Semen Cassiae,and 32 samples from various batches of Semen Cassiae were analyzed.Cluster analysis and principal component analysis were applied to study HPLC fingerprint and chemical pattern recognition.Results The fingerprint of Semen Cassiae was set up.Conclusion The method could be used for the quality control and comprehensive evaluation of Semen Cassiae.
6.HPLC Fingerprint and chemical pattern recognition method of Radix Paeoniae Alba

Objective To establish a method for quality control of Radix Paeoniae Alba.Methods A high performance liquid chromatographic method was developed to establish the fingerprint of Radix Paeoniae Alba and 47 samples from various batches were analyzed.Cluster analysis and principal component analysis(PCA) were applied to study on HPLC fingerprint and chemical pattern recognition method. Results The result of PCA and cluster analysis showed that the samples were divided into two types.The HPLC fingerprinting of Radix Paeoniae Alba,showing 10 characteristic peaks,was established from 47 batches of Radix Paeoniae Alba.Conclusion The method provides an academic reference for the quality control of Radix Paeoniae Alba.

8.Clinical analysis of complex femoral and tibial fractures treated with Grosse-Kempf intramedullary interlocking nails

Objective To investigate the clinical results of the treatment of complex femoral and tibial fractures using Grosse Kempf intramedullary interlocking nails. Methods Grosse Kempf intramedullary interlocking nails were used to treat 38 cases of complex femoral and tibial fractures. 12 cases of them were opened fractures, and 26 cases were closed. There were 24 cases of femoral fractures and 14 cases of tibial fractures. The average age was 33.6 years. Most of them were reduced under x ray monitoring. CPM rehabilitation started after the operation to gain the funtional recovey. After two weeks walking with crutches began. Results After the operation, all the wounds healed by first intention. The average union time of the fractures was 10.5 weeks (8~12 weeks). The average follow up period was 14.5 months. With all the cases rated as good or excellent, the clinical results were satisfactory. Conclusion Grosse-Kempf intramedullary interlocking nails are an effective, strong, convenient and secure implant to treat complex femoral and tibial fractures.
9.Spermatic vein detection by color doppler ultrasound for minimally trauma varicocelectomy

Objective In order to choose the best surgical approach for minimally trauma varicocelectomy color Doppler ultrasound(CDU) was used to detect the anatomic relationships of spermatic vein in groin. Methods Sixty varicocele patients were randomly selected. Their spermatic veins were examined by CDU which beginning from superficial inguinal ring,passing the crossing point of spermatic vein and femoral artery ,and ending at the 3cm above the deep inguinal ring. The depths from skin to spennatic vein were measured and the relationships between spermatic vein and femoral artery were recorded. Results The average length of incision is 2.1cm and the average duration of operation is 22 minutes. The average depth from skin to spermatic vein was 1.1cm,1.55cm and 3.56cm respectively at the site of the superficial inguinal ring,the crossing point of spermatic vein and femoral artery,and the deep ingui-nal ring. Conclusion The best approach for minimally trauma varicocelectomy is at the crossing point of spermatic vein and femoral artery because here the spermatic vein is relative superficial and has merged into two or three vessels and the femoral artery can be easily touched by fingers.
10.HPLC Fingerprint and chemical pattern recognition method of polar components in Qingyan Dropping Pill

Objective To establish a method for the quality control of Qingyan Dropping Pill.MethodsAn HPLC method was developed to establish the fingerprint of polar components in Qingyan Dropping Pill,and 12 samples from various batches were analyzed.Furthermore,principal component analysis(PCA) was used to differentiate and evaluate the whole fingerprints.The relationship between Qingyan Dropping Pill and their original herbs were investigated with HPLC-DAD method.Results The result showed that liquiritin was the key component of quality control.Among the chromatographic peaks,there were six chromatographic peaks coming from Glycyrrhizae Radix et Rhizoma;Five chromatographic peaks coming from Chebulae Fructus.Conclusion This method can be used as quality control for Qingyan Dropping Pill.
