1.Studies on Effective Composition of Pinecone Ⅳ.Determination of Acid Polysaccharides in Polysaccharides of Cone of Korean Pine (Pinus koraiensis)

Acid polysaccharides in pinecone were determined quantitatively by sulfuric acid-carba zole colorimetry and the influence of amount of sulfuric acid carbazole, reaction temper ature and time forcolor-reaction were investigated. The maximum abstrption was found to be at 520nm. The absorbance was a linear function of concentrations between 0 and 7 ?g/ml acid polysaccharides (r = 0.9999, n = 7). Analytical recovcry was 91.0%~99.8% . RDS was 1.3%~2.3% (n = 6 ). The minimum detectable concentration was 0.05?g/ml.
2.PTCH2 gene alterations in keratocystic odontogenic tumors associated with nevoid basal cell carcinoma syndrome

T)of PTCH2 were identified and 9 polymorphisms (3 of which were novel) were determined in the present series. Conclusion: Although not as frequent as PTCH1 mutations, PTCH2 germline mutations were detectable in a subset of NBCCS patients with KCOTs. The pathogenetic role of these PTCH2 mutations is yet to be clarified.
3.In vitro cultivation and identification of epithelial cells of keratocystic odontogenic tumor

Objective:To establish a method of in vitro cultivation of epithelial cells of keratocystic odontogenic tumors(KCOT).Methods: Tissue block and enzyme digestion techniques were used for primary cultivation of KCOT cells.The cells were grown in keratinocyte-serum free medium(K-SFM).The biological characteristics of the cultivated keratinocytes were identified by phase microscopic observation and by immunohistochemistry of cytokeratin,vimentin,cytokeratin10 and 14.Results: KCOT keratinocytes could survive for 30-50 days in K-SFM after passing 2-4 generations.Cells were polygon and showed typical slabstone-like appearance.Immunostaining showed positive staining for cytokeratin antibody,and negative for vimentin.Conclusion: KCOT epithelial cells could be serially cultured in vitro in K-SFM by techniques suggested in this study.
4.Comparison of the effect of different local anesthetic epidural anesthesia for cesarean section

Objective To investigate the clinical effect of different local anesthetic epidural anesthesia for cesarean section. Methods 60 maternal intended to be carried out cesarean section by epidural anesthesia were randomly divided into two groups. A group(n =30 cases) was administrated with 0.5% ropivacaine to anesthesia,and B group(n = 30 cases) was administrated with 0. 5% bupivacaine to anesthesia. The changes of hemodynamic were monitored,and the analgesic effect and adverse reactions were observed in the two groups. Results The differences of blood pressure, heart rate had no statistical significance between the two groups (all P>0.05);Compared to B group, the anesthesia onset time, anesthesia time between the start of fetus, analgesic effect, adverse reactions of A group were significantly different (t = 4. 347,4. 528, x2 = 3. 834, 6. 429, all P < 0. 05 ) ; Neonatal Apgar score showed no significant differences between the two groups ( P > 0. 05 ). Conclusion Anesthesia with 0.5% ropivacaine for cesarean section had satisfactory anesthesia effect,and less adverse reactions.
5.Effect of obesity on the cardiac structure and function of the young and middle-aged obese subjects

Objective To compare and discuss the changes of cardiac structure and function between young and middle-aged obese and normal healthy subjects.Methods 67 obese objects were divided by the body mass index(BMI)into 3 groups and 23 normal objects were included.Echocardiography was performed using a HDI 3000 and a 2.0~3.0 MHZ transducer.The LV end-diastolic and end-systolic dimension were measured.LV diastolic function was determined by peak E wave velocity,A wave velocity and E/A ratio,IVRT.Results LVEDD,LVESD,IVS,LVMI of obese group were significantly greater,larger than those in control group (P0.05).BMI had significant positive correlation with the LVEDD(r=0.37,P
6.Prevention of asymptomatic coronary intimal lesion by controlling the risk factors of coronary heart disease

Objective To explore the prevention of asymptomatic coronary intimal lesion by controlling the risk factors of coronary heart disease(CHD).Methods The relationship between the intimal lesion(such as yellow plaque、plaque rupture and/or thrombus formation)and risk factors(such as hypertension、diabetes、high blood LDL-C、blood pressure and blood sugar in patients with high blood LDL-C)was investigated in the non-culprit vessels of 120 patients with old myocardial infarction and stable angina pectoris by applying coronary angioscopy.Results Totally 155 non-culprit vessels in 120 patients were explored and the following findings were observed:(1) There are 80 cases(66.67%)of intimal lesion,40 cases(33.33%)without intimal lesion.(2)In the group with intimal lesion,the simple yellow plaque was observed in 30 cases(25.00%)and complex plaque in 50 cases.(3) In 120 patients,hypertension was observed in 69 cases(57.50%),high blood LDL-C in 79 cases(65.83%)and type 2 diabetes in 40 cases(33.33%).(4)The complex plaques was less common in patients whose BP was less than 140/90 mm Hg than in patients more than 140/90 mm Hg(P0.05).Conclusion The plaque rupture can be well prevented which can reduce the incidence of ACS if the high blood pressure、high blood sugar、high LDL-C are controlled to an ideal level.
7.Effects of PTCH1 mutations on the epithelial proliferation derived from keratocystic odontogenic tumour

Objective: To explore the relationship between the PTCH1 mutation and the expression of bcl-2, filaggrin, and loricrin in the keratocystic odontogenic tumour (KCOT), as well as the effects of the mutated PTCH1 on the epithelial proliferation and differentiation.Methods: The samples were collected from 20 cases of KCOT with mutated PTCH1, as well as 20 cases without mutation.All the samples were analyzed with immunohistochemical staining, for the purpose of investigating the expression of bcl-2, filaggrin, and loricrin.Results: In the samples with mutated PTCH1, the epithelia of 60% (12/20) cases expressed intensively positive bcl-2 staining, 20% (4/20) expressed moderate staining, and 20% (4/20) weak staining, but no negative bcl-2 staining samples were investigated;it was significantly different from the samples without PTCH1 mutation, in which 20% (4/20) expressed intensive staining, no moderate staining, 50% (10/20) weak staining, and 30% (6/20) negative staining were investigated (U=72, P=0.001).For the expression of filaggrin, 55% (11/20) of samples with PTCH1 mutations were stained weakly and 45% (9/20) showed negative staining, while in the samples not harboring PTCH1 mutations, 30% (6/20) cases showed moderate positive staining, 40% (8/20) weak staining and 30% (6/20) negative staining, no intensive staining was investigated (U=182, P=0.48).The loricrin expressed in all the layer of the epithelia in all the samples, while the filaggrin was mainly loca-lized within 1-4 layer cells of the epithelia.The differences of the expression of filaggrin and loricrin between the samples with mutated PTCH1 and without mutated PTCH1 displayed no significance.Conclusion: In the epithelia of KCOT, the bcl-2 expression was significantly associated with the PTCH1 mutation, which suggested that the mutated PTCH1 gene perhaps promotes the proliferation of KCOT epithelium.
8.A combination of minimally invasive percutaneous nephrolithotomy and extracorporeal shockwave lithotripsy for solitary renal staghorn calculi in patients with renal insufficiency

Objective To explore the curative effect of combined use of minimally invasive percutaneous nephrolithotomy (MPCNL) and extracorporeal shockwave lithotripsy (ESWL) for the treatment of solitary renal staghorn calculi in patients with renal insufficiency. Methods Eight cases of solitary renal staghorn calculi associated with renal insufficiency were treated by a combination of MPCNL and ESWL. Results Stones were completely cleared away in 5 cases, while residual stones were found in 3 cases. No blood transfusion was required and no severe surgery-related complications were encountered. After surgery variable degrees of improvement in renal functions was observed. The serum creatinine (Cr) decreased from 289?166 ?mol/L pre-operation to 155?33 ?mol/L post-operation (t=4.69, P=0.004), and the blood urea nitrogen (BUN) decreased from 15.1?7.9 mmol/L to 8.3?1.9 mmol/L (t=4.00, P=0.005). The emission computed tomography (ECT) examinations showed the glomerular filtration rate (GFR) was elevated from 48.8?12.4 ml/s before operation to 63.0?8.4 ml/s after operation (t=4.68, P=0.003). Post-renal obstruction disappeared after operation. Follow-up for 0.5~4.5 years (mean, 2.8 years) in the 8 cases revealed no obvious changes in renal functions. Conclusions Combination of MPCNL and ESWL for solitary renal staghorn calculi associated with renal insufficiency is safe and effective.
9.Four-step Method for Transurethral Plasmakinetic Resection of the Prostate:a Report of 120 Cases

Objective To explore the efficacy of four-step method for transurethral enucleation of the prostate . Methods A retrospective analysis was conducted on 120 cases of benign prostate hyperplasia ( BPH) treated with four-step method ( traction, enucleation, resection, and repair ) for transurethral plasmakinetic resection of the prostate in our hospital from January 2012 to February 2014. Results The average operation time was (91.4 ±35.3) min.The intraoperative blood loss was 78.8 ±34.1 ml. The resection of the prostate tissue weight was (32.1 ±15.7) g.The postoperative bladder infusion time was (1.5 ±0.5) days.The postoperative indwelling catheter time was 3.5 ±0.8 days.The postoperative hospital stay was (5.5 ±0.5) days.No death, blood transfusion, or TURS happened.Follow-up reviews at 3 months after surgery found postoperative international prostatic symptom score (IPSS) was significantly less than the preoperative level [4.5 ±2.2 vs.24.5 ±5.1, t=39.445, P=0.000], postoperative Qmax was significantly higher than preoperative level [(21.5 ±10.5) ml/s vs.(6.0 ±1.9) ml/s, t =15.912, P =0.000], and postoperative residual urine was significantly less than the preoperative level [(12.0 ±2.0) ml vs.(145.0 ±33.0) ml;t=44.069, P =0.000].No retrograde ejaculation, sexual dysfunction, or dysuria happened. Conclusion The four-step method for transurethral enucleation of the prostate is radical , safe, and simple.
10.Research on protein expression of CD55 in triple-negative breast cancer and its effect on prognosis

Objective:To investigate the expression of CD55 in triple-negative breast cancer (TNBC) and its correlations with prognosis.Methods: The expressions of CD55 was detected by immunohistochemistry Streptavidin-Peroxidase method in 100 cases of triple-negative breast cancer and 50 cases of adjacent-carcinoma tissues, with clinical and pathological features and follow-up data, the Cox model analysis was established and to analyze the prognostic factors. Results: The positive expression of CD55 was 56.0% in the tissues from triple-negative breast cancer, whereas its positive rate was 8.0% in the corresponding adjacent breast tissues, with statistical significance (x2=32.0,P<0.05). The expression of CD55 was significantly related with tumor size, clinical stage and axillary lymph node metastases (x2=7.404,x2=8.688,x2=4.802;P<0.05); the expression of CD55 did not show correlation to age, and histologic grade. Multivariate analysis showed that the age, clinical stage, CD55 protein expression and axillary lymph node metastasis were the independent risk factors affecting the 5-year disease-free rate of patients and that clinical stage and axillary lymph node metastasis were the independent risk factors affecting the 5-year overall rate of patients.Conclusion: The expression of CD55 might be related to the development and prognosis of triple-negative breast cancer. The expression of CD55 may be used as one of the indicators in determining the progress and prognosis of TNBC.
