Objective To evaluate the possible auxiliary effect of sipulin in treatment of anthracycline relapsed and metastatic breast cancer. Methods Sixty-three patients were randomly divided into two groups: sipulin group:sipulin plus docetaxel + capecitabine;control group:only administered with docetaxel + capecitabine. The leukocyte, haemoglobin and platelet,toxicity of digestive tract, body weight, Karnofsky status and efficacy of those patients were evaluated respeoively before and after therapy. Results Overall response rates were 71.9% and 54.8% (P = 0.014)for sipulin group and control group, respectively. The median survival time was 15.9 months versus 11.3 months(P=0. 026)for sipulin group and control group,respectively. The 1-year survival rate for sipulin group and control group was 51.6% versus 38.7% (P = 0.035) ,respectively. The clinical efficacy and reduce of leukocyte were better in sipulin group than in control group, the life quality and clinical symptom of the patients in sipulin group were improved more significantly than those in control group (P ＜ 0. 05, P ＜ 0. 01), the value of CD3, CD4, CD3/CD4 and NK in sipulin group were significantly higher than in control group(P ＜ 0.05). Conclusion Sipulin plus docetaxel/capecitabine regimens could improve the efficacy, reduce the toxicities of digestive tract and blood, enhance the immune function and improve life quality, improve the tolerance of relapsed and metastatic breast cancer patients for chemotherapy and should be recommended.

Objective To explore the correlation and mechanism between serum adiponectin and estrogen level in female patients with gallstones.Methods A total of 62 female gallstone patients were analyzed,of which 33 cases were premenopausal and 29 cases were postmenopausal.Sixty age-matched normal physical examination cases were selected as controls.Gallbladder stones were diagnosed by B-Ultrasound.Height and weight of the patients were measured to calculate body mass index (BMI).Blood glucose was determined by Glucose Oxidase method.Lipids were determined by turbidimetry method in automatic biochemical analyzer.Serum adiponectin and estrogen were determined by radioimmunoassay.Results Compate with control group,serum adiponectin level was significantly decreased,and estrogen level was significantly ncreased in premenopausal and postmenopausal patients with gallbladder stones.There was a positive correlation between serum adiponectin and HDL-C and ApoA1 level.Conclusions Low serum adiponectin level may be a predisposing factor of gallstone that is initiated by estrogen,and independent of obesity and diabetes.

ve To establish a method of high performance liquid chromatography (HPLC) for determi-nation of nitrite in water. Methods The trace content of nitrite in water sample was determined by indirect UV-HPLC. The mixture of methanol/o-phthalic acid (pH value was adjusted to 8.6 by 0.10 mol/L NaOH solution)5: 95 was defined as mobile phase. The water sample was directly filtered by chromatographic column pack with ODP. Results Under the conditions of wave length of 270 nm and flow rate of 0.9 ml/ min, the linear range of this assay was 0.0~20.0?g/ml nitrite, the relative standard deviation was 3.9%. The average recovery rate and detection limit were 99.9% and 0.001?g/ ml respectively. Conclusion This method could be applied to the determination of trace amount of nitrite in drinking water, purified water and mineral water.

Objective To study the inhibitory effects of modification c-myc antisense oligodeoxynucleotide (ASODN) on the growth of renal carcinoma cell line (GRC-1) and cell cycle. Methods We transfected c-myc antisense oligodeoxynucleeride in renal carcinoma cell line to investigate the effect of c-myc ASODN on it. Results ① By MTT assay,we found that the c-myc ASODN significantly inhibited the growth of carcinoma cell, compared with sense and mismatch ODN. The growth-inhibition rate of 12 ?mol?L -1 c-myc ASODN group of renal carcinoma cell was 47% at 48 hour, and the inhibitory effect increased with the increase of ASODN dosage and changed with ASODN time. ② The measurements by flow cytometry proved that ASODN mainly blocked the G 1 to S of the cycling of renal carcinoma cell. Conclusion c-myc ASODN can inhibit the growth of renal carcinoma cells with sequence specificity and cycling specificity.

OBJECTIVETo optimize the hydrolysis process of linarin by response surface methodology, and to use the model of aldose reductase to study the acacetin's activity of aldose reductase inhibitory.

METHODThe model of acacetin enzyme in vitro was established by the determination of fluorescence absorption of NADPH, the inhibition rate of acacetin aldose reductase was calculated, and then the IC50 of hydrolysis was obtained. The hydrolysis process of linarin hydrolysis condition was optimized by using response surface method.

RESULTThe results indicated that the IC50 of acacetin (2.74 mg x L(-1)) was less than the IC50 of linarin (3.53 mg x L(-1)). Hydrolyzation time of 7.4 h, sulphuric acid concentration of 0.54 mol x L(-1) and the ratio of material to liquid of 3 : 1 were the optimum conditions.

CONCLUSIONHydrolyzate acacetin has preferable inhibitory activity of aldose reductase. The optimized hydrolysis condition of linarin is convenient to use with good predictability.

However, the protein homogeneity, especially the N-glycosylation limited the further research and application of these protein drugs. The analysis method for N-glycans is believed to be critical in protein drugs development. To enhance the N-glycans isolation efficiency and accelerate the pretreatment, a new strategy was built on ultrafiltration-devices. New methods increased the isolation efficiency of N-glycans containing N-acetylglucosa mine with 10%-20%. The degrading of N-glycans containing sialic acids was also minimized with this method. 20%-100% more N-glycans with sialic acids were isolated. The pretreatment was finished within 30 min. Coupled with HPLC-HRMS, an effective and reliable strategy designed for protein drugs N-glycans analysis were developed.

The pharmacodynamic material basis of Shufeng Jiedu Capsule (SJC) was identified and the mechanism was explained. Ninety-four compounds were identified in SJC and 10 compounds were determined to be the anti-inflammatory ingredients by screening experiments. The mechanism of multi-component, multi-targets, and multi-pathways of SJC which provided a theoretical and experimental basis for the standards of quality control and the clinical application was elucidated by in vivo experiments, network pharmacology, and genomics.

The idea and model of quality research on traditional Chinese medicinal materials and compound Chinese materia medica were established and this method was applied to systematical promoting the quality standard of raw herbal materials and preparation of Shufeng Jiedu Capsule (SJC). The chemical components from Polygoni Cuspidati Rhizoma et Radix, Forsythiae Fructus, Isatidis Radix, Bupleuri Radix, Patriniae cum Radice Herba, Verbenae Herba, Phragmitis Rhizoma, and Glycyrrhizae Radix et Rhizoma were identified and 184 compounds were determined. The fingerprints of the eight medicinal materials were established, 10 compounds were identified as the anti-inflammatory substance of SJC by studying the correlation between chemical components and their anti-inflammation effect. The methods of multi-component determination and fingerprint controlling have been established and the quality standard of SJC has been improved comprehensively.

Objective: To study the relationship between the patient satisfaction with complete denture and the pain perception threshold (PPT) of alveolar ridge mucosa. Methods: Forty-five edentulous patients with complete denture were investigated with the questionnaire of patient satisfaction, PPT of alveolar ridge mucosa in the patients was measured with a pressure algometer.
