1.Preparation of Salvia miltiorrhiza utility aqueous extract by hydrochloric acid method and evaluation on osteoblast

Aim To prepare and qualitatively control the water extracts bioactive compounds (Danshen Gubao Ⅱ) from Danshen(Salvia miltiorrhiza) and to evaluate the effects of Danshen Gubao Ⅱ on osteoblast in vitro.Methods Danshen Gubao Ⅱ was extracted from salvia miltiorrhiza with a diluted hydrochloric acid method. The HPLC was used to determine the amounts of the bioactive compounds.PNPP methods were developed for the estimation of pharmacodynamic action by testing osteoblast activity (ALP expression).Results Compared Danshen Gubao Ⅱ with the ordinary water extract of salvia miltiorrhiza(DUAE Ⅰ), the amount of danshensu (main compound of salvia miltiorrhiza) was elevated to 3.28 times,11.50?0.52 mg?g-1, the amount of salvianolic acid B was reduced 1.46 times,16.40?1.22 mg?g-1,and each of their content was 1%~1.5%,the amount of protocatechualdehyde was elevated to 1.46 times,0.233?0.02 mg?g-1. The activity of Danshen Gubao Ⅱ on osteoblast was stronger than that of DUAE Ⅰ, 0.25 mg?L-1 of Danshen Gubao Ⅱ elevated 52%?10% osteoblast ALP expression,1.0 mg?L-1 of DUAE I elevated 5.5%?0.3% osteoblast ALP expression, and the results indicated that Danshen Gubao Ⅱ had more capacity on the osteoblast.Conclusions Danshensu, salvianolic acid B, protocatechualdehyde were stabilized in contents from Danshen Gubao Ⅱ, which could be qualitatively controlled.Danshen Gubao Ⅱ had good effect on osteoblast, which provided new data for the research of salvia miltiorrhiza's anti-osteoporosis preparation.
2.Accurate localization of pulmonary segments on pulmonary perfusion SPECT/CT imaging

Objective To investigate the methods for localization of the lung segments on three-dimentional (3D) pulmonary perfusion SPECT/CT imaging.Methods Twelve healthy adults were recruited into this study.Lung segments were defined based on the anatomical criteria of CT on transaxial, sagittal and coronal images, including apical, posterior, anterior segments in right upper lobe ( RUL), lateral, medial segments in right middle lobe ( RML), superior, anterior basal, lateral basal, posterior basal segments in right lower lobe (RLL); apical posterior, anterior, superior lingual, inferior lingual segments of left upper lobe ( LUL), superior, medioanterior basal, lateral basal, posterior basal segments of left lower lobe (LLL).Results (1) Eleven typical sections were selected on transaxial SPECT pulmonary perfusion imaging:sternoclavicular joint, supra-arch vessels, aortic arch, azygous arch, right upper lobar bronchus, left upper lobar bronchus, middle lobar or lingual bronchus, basal trunk of bronchus, lower lobar vein, superior and inferior basal vein and basal vein.(2) Twelve typical sections were defined on sagittal imaging:left hilum, bifurcation of left principal bronchus, septa of left pulmonary artery, cardiac apex, the first and second sections left to cardiac apex; right hilum, bifurcation of intermite bronchus, interlobar artery, bifurcation of interlobar artery, the second and third sections right to right atrium.(3) Seven typical sections were defined on coronal imaging:sternoclavicular joint, ascending aorta, bifurcation of pulmonary artery, bifurcation of trachea, intermite bronchus, basal vein and thoracic aorta.Conclusion The developed method was able to localize lung segments on pulmonary perfusion SPECT imaging.

6.Comparison of survival effects between using electron and modulated X-ray beams for boosting irradiation in breast cancer patients after breast-conserving surgery and postoperative radiotherapy

Objective To compare the survival effects between using electron beams (EB) and modulated X-ray beams (XB) for boosting irradiation in breast cancer patients after breast-conserving surgery and postoperative radiotherapy.Methods This study retrospectively included 485 breast cancer patients who underwent breast-conserving surgery at Beijing Cancer Hospital.All patients underwent either EB or XB for tumor bed boost irradiation (10-16 Gy/5-8 fractions) after whole-breast irradiation of 46-50 Gy/23-25 fractions.Results Median follow-up time for the cohort was 96.04 months.Statistically significant increase of local recurrence free Survival (LRFS) was observed in XB group than in EB group.The 5-year and 10-year LRFS was both 98.4% in XB group,as well as 94.2% and 93.2% in EB group,respectively (x2 =4.190,P < 0.05).But there was not statistically significant difference in 5-year and 10-year overall survival (OS) between XB group(96.7% and 95.8%) and EB group(94.9% and 89.4%),respectively (P > 0.05).The multivariate analysis showed that LRFS was significantly correlated with age≤40,positive pathological lymph nodes and positive expression of Her-2 receptor.But boost irradiation method was not independent prognostic factor for LRFS and OS (P > 0.05).Conclusions For cancer patients treated with breast-conserving surgery and whole-breast postoperative radiation followed by a boost irradiation to tumor bed,XB irradiation was superior to EB irradiation in term of LRFS,yet no difference of OS was observed in both groups.
7.Dynamic changes of sICAM-1 in patients suffering from hemorrhage fever with renal syndrome

Objective To study the dynamic changes of soluble intracellular adhesion molecule-1 (sICAM-1), and to assess its role in the pathogenesis of hemorrhagic fever(HFRS) with renal syndrome. Methods A total of 30 patients with HFRS(case group) and 20 healthy subjects (control group) from Worker Hospital of Finance & Commerce of Qiqihar city were included in the study during 2006 - 2007. Double-antibody sandwich ELISA method was used to measure the level of sICAM-1 in serum samples. Results The serum levels of sICAM-1 in patients with HFRS in early stage, critical stage, convalescent stage were(47.56±6.51), (94.23±15.36), (54.19±8.42)ng/L,respectively. The serum level of sICAM-1 in control group was (22.63±3.40)ng/L. The above values were compared between any two groups, differences were statistically significant (all P < 0.01). The serum level of urea nitrogen in patients with HFRS in early stage, critical stage, convalescent stage were (5.16±0.12), (33.84±9.24),(8.20±1.30)mmol/L, respectively. The serum level of urea nitrogen in control group was (4.20±0.56)mmol/L.The above value were compared between any two groups, the differences were statistically significant (all P< 0.01). Conclusions sICAM-1 is involved in the pathogenesis of HFRS as protective factors. Cellular immune playes an important role in the pathogenesis of HFRS.
8.Study on the differentiation of human umbilical cord perivascular cell into cardiac-like cells in vitro.

Objective To investigate the capability of cardiomyoganic differentiation of human umbilical cord perivascu- lar cells induced by 5-azacitidine in vitro.Methods Human umbilical cord perivascular colls were harvested from the umbilical cords of consenting full-term caesarian section patients,which were digested by collagenase,and were purified, expanded through repeating passaging in time in vitro.The HUCPV cell of passage 3 were treated with 5-azacytidine(5 -aza,5?10~(-6)nmol/L) for 24 hours and continued to be cultured under the previous condition for 4 weeks.The prolifera- tion and differentiated phenotype of HUCPV cells were observed.Results The isolation method of the UC vasculature and enzymatic digestion of its perivascular tissue to rapidly harvest a highly proliferative HUCPV cells.After treated with 5 -aza for 4 weeks,the passage 3 HUCPV cells formed the spindle-shaped cells and rate of nuclear and cytoplasm de- creased.Myotube-like structures were found in some cytoplasm.Immunohistochemistry results showed that the HUCPV cells were similar to MSC by stained positively for CD44 and negtively for CD34.The HUCPV cells induced by 5-aza were stained positively a-cardiac action and cardiac Troponin by immunohistochemistry.Furthermore,the expression of a -cardiac action and cardiac treponin gene as detected positively by RT-PCR after HUCPV cells were induced by 5- aza.Conclusion The HUCPV cell was a valuable material for cell-based therapies and can be induced into cardiac- like cells by treatment with 5-aza.
9.Analysis of 52 eases of aged patients with acute cholecystitis treated with laparoscopie eholecystectomy

Objective To explore the clinical features and surgical management of aged patients with acute cholecystitis,and try to command the opportunity and procedure of laparoseopic cholecystectomy(LC)better.Meth- ods Clinical data of 52 aged cases with acute cholecystits undergone LC were analyzed retrospectively.Results All of the patients(within 48h of the acute attack)were successfully recovered without serious operative complications. Conclusion Aged acute cholecystitis progressed rapidly and its operative difficulty and risk were higher;only if more attention was paid to perioperative managements and operative time and technical skill were mastered,early LC for the patients was safe and feasible.Therefore.it should be recommended in the great majority of cases except the des- perate patients whose general condition was too poor to operate.
10.Effects of Pulmicort Respules on airway resisitance and airway remodeling in rats with asthma

Objective To investigate the effects of Pulmicort Respules on the airway resistance and airway remodeling in rats with allergic asthma. Methods Thirty-two SD rats were randomly divided into blank control group,asthma model group,Pulmicort Respules group and normal saline control group,with 8 rats in each group.On the 14th day of allergization,rats were challenged by ovalbumin inhalation and were treated correspondingly.On the 28th day,airway resistance was determined,HE staining was conducted in lung tissues of each group,and airway remodeling related parameters were also measured. Results The airway resistance of Pulmicort Respules group was significant lower than that of blank control group,asthma model group and normal saline control group(P
