1.Separation, Purification and Analysis of the Components of Earthworm Fibrinoly tic Enzymes

The earthworm fibrinolytic enzymes (EFE) were separated by affinity ch romatography using soybean trypsin inhibitor as a matrix. The enzymes were furth er separated and purified into 12 components after DEAE-32 chromatography and p reparative electrophoresis. The pI of these components gradually decreased f rom pH 4.0 according to electrophoresis mobility from higher to lower on PAGE. The molecular weights were in the range of 22~34 ku. 6.5 and 7 w ere glycoproteins proved by staining with the shiff reagent and thymol/sulfuric acid. The fibrinolytic activity of 7 was highest as determined usi ng chromzym UK and chromzym PL as specific substrates.
2.Research Progress in TCM Non-medicine Therapies for Depression Syndrome

Depression in modern medicine belongs to the depression syndrome in TCM. As early as the pre-Qin period, the ancient doctors recognized depression syndrome as a kind of disease associated with the function of visceral organs or emotional stimulation, which showed mood disorders and a variety of physical symptoms. The doctors in the past dynasties had explored different kinds of non-medicine therapies except for general medicine therapies, such as acupuncture and moxibustion, tuina, music and emotional therapy etc., which had obtained definite therapeutic effects. This article reviewed the non-medicine therapies for depression syndrome in ancient literatures and modern clinical researches, in order to provide some references for the treatment of depression.
3.Correlation of Blood Glucose Variability and TCM Syndromes in Type 2 Diabetic Patients

Objective To explore the correlation of blood glucose variability and TCM syndromes in type 2 diabetic patients. Methods A total of 64 type 2 diabetic patients from China-Japan Friendship Hospital during Jan. 2010 to Dec. 2013 with complete clinical data were measured by continuous glucose monitoring system (CGMs) for 3 days. Patients were divided into empirical group and deficiency group. The empirical group included heat excess and fluid deficiency syndrome, phlegm-dampness stagnation syndrome and blood stasis syndrome. The deficiency group included qi-yin deficiency syndrome and yin-yang deficiency syndrome. The mean blood glucose (MBG), and mean amplitude of glycemic excursions (MAGE) were compared between the groups and types in each group, and the correlation with TCM syndrome types were analyzed. Results There was no significant difference in MBG between two groups (P>0.05). MAGE in deficiency group was higher than that in empirical group (P<0.05). No difference of MAGE existed among TCM syndrome types in empirical group (P>0.05), but in deficiency group, MAGE in yin-yang deficiency syndrome was higher than that in qi-yin deficiency syndrome (P<0.05). Conclusion Blood glucose variability has no correlation with MBG in type 2 diabetic patients. Glucose showed significant variability in deficiency group, and the most serious glucose variability was found in type of yin-yang deficiency syndrome.
4.Development of autologous peripheral blood stem cells in the fibrous liver of rats after recombinant human granulocyte colony stimulating factor mobilization

AIM:To study the development of autologous peripheral blood stem cells in the fibrous liver after mobilizing recombinant human granulocyte colony stimulating factor and the therapeutical effect of autologous peripheral blood stem cell transplantation.METHODS:Experiments were performed at the Central Laboratory of First Affiliated Hospital,China-Japan Clinical Pathological Center and Central Laboratory of Dalian Medical University from April 2005 to April 2006.①Rat models of hepatic fibrosis were made by injecting carbon tetrachloride subcutaneously.Thirty-two successfully established rat models of hepatic fibrosis were divided into group A,group B,group C and group D with 8 in each group,male and female(half and half).②Rat models of group A were only administered with recombinant human granulocyte colony stimulating factor subcutaneously.Rat models of group B were administered with recombinant human granulocyte colony stimulating factor subcutaneously.Peripheral blood mononuclear cells containing peripheral blood stem cells were collected by density gradient centrifugation.These peripheral blood mononuclear cells were grafted in liver after labeling with PKH26-GL.Rat models of group C did not undergo any management.Serum and hepatic tissues were collected from rat models of group A,group B and group C six weeks later.Successful rat models of liver fibrosis of group D were sacrificed instantly after the last time of carbon tetrachloride administration.Serum and hepatic tissues were collected for measurement.③The alanine aminotransferase,aspartate aminotransferase,albumin and total bilirubin levels and hepatic pathological changes of rat models of all the four groups were measured.Immunofluorescent method was employed to observe the development of autologous peripheral blood stem cells in the liver.RESULTS:A total of 32 rats were involved in the result analysis.①Total bilirubin levels were lower in group A and group B than that of group C(P 0.05).②Hepatic fibrosis was significantly improved in the group A and group B as compared with the group C(P
5.Effect of miR-133b on radiosensitivity of colon cancer SW620 cells by targeting HER-2

Objective:To evaluate the effect of miR-133b on the apoptosis and radiosensitivity of colon cancer cell line (SW620 cells), and to explore its mechanism.Methods:SW620 cells were transfected with miR-con (miR-con group), miR-133b mimics (miR-133b group), si-con (si-con group) and si-HER-2(si-HER-2 group) by the liposome method, and then irradiated with 0, 2, 4, 6, 8 Gy. The miR-133b protein expression, HER-2 protein expression, apoptosis, cell survival fraction and cytofluoroactivity in each group were evaluated by qRT-PCR, Western blot, flow cytometry, colony formation assay and dual luciferase reporter gene assay, respectively.Results:Compared with the pre-irradiation group, the expression level of miR-133b was significantly down-regulated ( P<0.05), whereas that of HER-2 was significantly up-regulated in SW620 cells after irradiation ( P<0.05). Overexpression of miR-133b and knockdown of HER-2 remarkably reduced the survival fraction (both P<0.05), and significantly promoted the apoptosis of SW620 cells ( P<0.05). miR-133b could considerably inhibit the fluorescent activity of wild-type HER-2 cells ( P<0.05) and negatively regulate the expression of HER-2 protein. Conclusion:miR-133b can inhibit the survival of colon cancer cells, promote the apoptosis and enhance the sensitivity of radiotherapy probably via the mechanism of targeting HER-2.

Stomatology is a subject emphasizing on practical and innovation ability, so paying attention to the cultivation of students' application ability and innovation ability in the undergraduate stage, combining theoretical study and research innovation, can lay a good foundation for the study and work. In recent years, the College of Stomatology of Chongqing Medical University has created the scientific research atmosphere, established the platform of scientific research, attempted the undergraduate tutorial system, expanded the construction of teaching resource, and involved undergraduate to take part in the scientific research practice. The college has taken many active measures to train the scientific innovation ability of undergraduates, and has obtained good results. Some undergraduates have already possessed a certain ability to find and solve problems as well as independently think. They have also enhanced their awareness of reading literature and critically accepting knowledge and improved their comprehensive quality.
7.Effect of topical bevacizumab on experimental corneal neovascularization in mouse

Objective The inhibitory effects of avastin on new blood vessels in nonproliferation diabetic retinopathy, age-related macular degeneration and neovascular glaucoma have been demonstrated. But only seldom report of avastin on corneal neovascularization(CNV) was seen. Present study was to evaluate the effect of topical bevacizumab (avastin) on experimental corneal neovascularization in mice. Methods Thirty eyes of 30 Balb/c mice were chemically cauterized by applying a 2 mm-diameter filter paper soaked 1 mol/L NaOH solution at the central cornea for 40 s. All animals were randomly assigned to five groups, including 1 mg/mL, 3 mg/mL and 5 mg/mL bevacizumab eye drops group respectively, 1 mg/mL dexamethasone sodium phosphate eye drops group (positive control) and normal saline solution group (negative control) . The drug was topically utilized twice per day. CNV was examined under the slim lamp on the 3rd, 7th and 14th day after alkali burn. Animals were killed on the 14th day after alkali burn. Area of CNV was calculated in terms of pixels on digital photographs. The use of animals followed the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission. Results No significant difference was found in the grade of corneal injury among five groups (F = 0. 201, P = 0. 935). The area of neovascularization at the cornea surface was (37.11 ±3.17)% in 1 mg/mL bevacizumab group, (29.75 ±3.56)% in 3 mg/mL bevacizumab group, (18. 76 ± 2. 55) % in 5 mg/mL bevacizumab group, (20. 91 ± 2. 75) % in dexamethasone group and (41. 65 ±2. 11)% in normal saline group, showing a significant difference among groups(F = 71. 687, P =0. 000) with the further comparative decline in 5 mg/mL bevacizumab group compared with other groups (P < 0. 01) . Conclusion The topical use of bevacizumab (avastin) inhibits alkali burn-induced CNV in mice.
8.Apoptosis effect of Siraitia grosvenorii extracts on lung cancer cells A549 and its mechanisms

Aim To study the apoptosis effect of Sir-aitia grosvenorii extract on human lung cancer cells A549 and its mechanisms.Methods MTT assay was applied to determine A549 cell proliferation.Hoechst 33258 staining was applied to investigate morphological changes in A549 cells.To find out the cause of cell growth inhibition,several experiments on cell cycle distribution and apoptosis were performed by flow cy-tometry analysis.The expression of p21 and Bcl-2 was determined by Western blot.Results Flow cytometry analysis showed that treatment with mogrol arrested A549 cells in the G0 /G1 phase and induced apoptosis. After treatment with Siraitia grosvenorii extract,West-ern blot experiment showed cell cycle regulator p21 was up-regulaed,while the apoptosis inhibitor Bcl-2 was down-regulated.Conclusion Treatment with Siraitia grosvenorii extract arrests the A549 cells at G0 /G1 phase and induces apoptosis that may contribute to the anti-proliferation activity of mogrol through the regula-tion of p21 and Bcl-2 expression.
9.Effect of Sciatic Nerve Compression with Different Pressures and Sites on Neural Blood Flow in Rats

Objective To observe the changes of neural blood flow when oppressed with different pressures and at different sites in rat's sciatic nerve.MethodsThe sciatic nerve compression model of rat with pressure and time controlled simultaneously was established.The changes of neural blood flow were observed by oppressing the sciatic nerve with five grades pressures using Gasbag.The rats were randomly divided into the distal compression group and proximal compression group,and the changes of neural blood flow were observed in each group.ResultsThe neural blood flow changed significantly(P<0.05) when the sciatic nerve was oppressed with different pressures and it had the decreasing tendency with the pressures increasing.The neural blood flow of each group decreased obviously(P<0.01).The effect of oppressing distal nerve on neural blood flow was more obvious than that of oppressing proximal nerve(P<0.01).ConclusionMechanical compression can influence the neural blood flow obviously.The distal vessels of sciatic nerve are the major sources of nerve blood supply.
10.Study on intervention effect of mentalization based treatment on anxiety, depression and chronic craving among methamphetamine addicts

Objective To investigate the effect of mentalization based treatment on the negative emotion such as anxiety,depression and chronic drug craving among methamphetamine addicts.Methods Totally 612 methamphetamine addicts in 6 compulsory isolated detoxification center of Liaoning served as the study subjects and randomly divided into the experimental group (mentalization based treatment group) and control group,306 cases in each group.The Beck Anxiety Inventory (BAD,Beck Depression Inventory (BDI) and the Drug Craving Inventory were used to evaluate the anxiety,depression and chronic craving in the two groups.Results The scores of BAI,BDI and Drug Craving Inventory after mentalization base treatment in the experimental group were decreased compared with those of before intervention (P<0.05).The declining trend of the scores of BAI,BDI and Drug Craving Inventory after intervention in the experimental group were more significant than that in the control group (P<0.05).Conclusion The mentalization based treatment can improve the negative emotions such as anxiety and depression to some extent,and reduces the chronic drug craving among methamphetamine drug addicts.
