3.Fabrication of Allogenic Cartilage in A Tube Lined with Epithelium and A Novel Scaffold

Objective To study the feasibility of engineering cartilage tissue in a tube lined with epithelium and implanting allogenic chondrocytes into a novel scaffold consisting of chitosan nonwoven cloth coated with poly (DL-lactide-co-glycolide)(PLGA).The focus of the present study is to explore a new way of repairing laryngeal and tracheal defects.Methods Allogenic chondrocytes were obtained from the auricles of 1-month-old rabbits.After being cultured in vitro for three to four passages,the cells were implanted into the scaffolds to form composite grafts and then transplanted into the rabbits.After 6,12,and 18 weeks,the general,histological characteristics were investigated.Results The cobweb-like matrix was observed approximately 1 week after the chondrocytes had been implanted into the scaffolds.At 6 weeks,the matrix was secreted,and there were immature chondrocytes in the grafts.At 12 weeks,the allogenic cartilage in the tube lined with epithelium had been created.Chondrocytes were almost mature and the lacunae had formed.At 18 weeks,the neocartilage was similar to native cartilage.Conclusion It is feasible to fabricate allogenic cartilage in a tube lined with epithelium by implanting allogenic chondrocytes into a novel scaffold made of chitosan nonwoven cloth coated with PLGA.
4.A study on expression of platelet derived growth factor a in periodontal tissue during orthodontic tooth movement with rotating pulsed magnetic field

Objective To investigate the effects of rotating pulsed magnetic field on platelet derived growth factor a (PDGF-A) expression in periodontal tissue during tooth movement. Methods 30 white rabbits were random divided into 6 groups with 5 rabbits each group,including groups of 1,3,5,7,14 and 21 days. Under anesthesia condition by 2% pentobarbital sodium,the stainless coil springs were fixed between the first maxillary molar and the incisor producing the force of 80g. The experimental group was treated by rotating pulsed magnetic field and the force, the control group was only treated by t he force. The expression of PDGF-A was half-quantitatively investigated through immunohistochemical analysis. Results The expression of PDGF-AA in the experimental group enhanced apparently compared with that in the control group. There were significant differences among the 5,7, and 14day groups (5. 28 ± 0. 14 vs 2. 03 ±0. 18,7.63±0.27 vs 2. 84 ±0. 12,3.52 ±0. 16 vs 1.65 ±0.03;8.10±0.13 vs 4. 30 ±0. 21,13. 27 ±0. 31 vs 6.47 ± 0.15,5.66 ± 0.22 vs 3. 15 ± 0. 27, P < 0. 05). The expression of PDGF-AA of 7 day group was higher than that of other groups. Conclusion Rotating pulsed magnetic field promotes the expression of PDGF-A in the periodontal tissue and alveolar bone remodeling.
5.Stage Ⅰ Central Lung Cancer and Bronchial Disseminated Pulmonary Tuberculosis: A Comparative Analysis of Tree-in-bud Sign at CT

Purpose To investigate the difference of imaging features of tree-in-bud (TIB) sign at CT between stage Ⅰ central lung cancer and bronchial disseminated pulmonary tuberculosis in order to reduce the misdiagnosis rate of central lung cancer.Materials and Methods 32 patients of stage Ⅰ central type lung cancer confirmed pathologically or clinically (lung cancer group) and 47 patients of bronchial disseminated pulmonary tuberculosis confirmed pathologically or clinically (tuberculosis group) underwent chest CT scanning and were found with TIB sign.The imaging data of all the cases were analyzed retrospectively in terms of distribution range,accompanying CT findings of TIB sign,and abnormalities of segmental and (or) larger bronchus proximal to it.Results TIB sign was mainly focal (unilobar) in lung cancer group (96.88%),while it was mainly diffuse (more than one pulmonary lobe) in tuberculosis group (80.85%).The difference was significant (P<0.01).Accompanying CT findings:① Obstructive bronchial mucoid impaction distal to the TIB was more common in lung cancer group than in tuberculosis group (100% vs 6.38%,P<0.01).② Consolidation and/or ground-glass opacities were found both in lung cancer group (34.38%) and in tuberculosis group (48.94%).The difference was not significant (P>0.05).③ Cavity was less common in lung cancer group than in tuberculosis group (0 vs 38.30%,P<0.01).④ Nodules with diameter larger than 5 mm were much fewer in lung cancer group than in tuberculosis group (0 vs 76.60%,P<0.01).TIB sign was found in 33 lobes in lung cancer group,and the abnormity was found in all these lobes (100%);while in tuberculosis group,TIB sign was found in 144 lobes and the abnormity was found in 56 lobes (38.89%).Conclusion TIB sign shows differences between stage Ⅰ central lung cancer and bronchial disseminated pulmonary tuberculosis.The TIB signs such as focal distribution,accompanied by bronchial mucoid impaction and abnormality of segmental or (and) larger bronchus may predict the high possibility of central lung cancer.
6.Effect of compressive stress on the expression of DNAX-activating protein 12 and tartrate-resistant acid phosphatase in mouse monocyte RAW264.7.

OBJECTIVETo investigate the expression of DNAX-activating protein 12 (DAP12) and tartrate-resistant acid phosphatase (TRAP) in mouse monocyte RAW264.7 subjected to compressive stress.
METHODSMouse monocyte RAW264.7 was subjected to four-point bending system. The expression of DAP12 and TRAP mRNA were examined by reverse transcription-polymerase chain reaction (RT-PCR) and the expression of DAP12 protein by Western Blotting after 0, 3, 6 and 12h's compressive stress.
RESULTSAfter RAW264.7 cells were cultured by osteoclast cell culture fluid, the amount of the cells increased, and the volume enlarged, and the number of nuclei per osteoclast increased in vitro. The expression of DAP12 and TRAP mRNA and DAP12 protein of RAW264.7 cells subjected to compressive stress increased along with the time (P < 0.05).
CONCLUSIONThe mouse monocyte RAW264.7 can differentiate into osteoclast in vitro, and high expression of DAP12 exists in the process of osteoclast differentiation.

7.Research Progress on Immune Mechanism of Immune Thrombocytopenia--Review.

Abstract Immune thrombocytopenia (ITP) is an acquired autoimmune hemorrhagic disease, although the ITP pathogenesis is completely unknown, but in terms of the current view, the immune tolerance is main reason for the onset of ITP. In recent years, more and more immune cell subsets, cytokines and the new approacher were found to be closely related with the ITP, such as saliva acid, B cell activating factor, dysfunction of regulatory B cells and Th1/Th2 balance drift, CD4 CD25 T cell function defect, IL-23/Th17 pathway regulation, etc., In this paper, the latest research progress on the immune pathogenesis of ITP are reviewed, so as to provide theoretical basis and research direction for further understanding the pathogenesis of ITP.

8.Value of triglyceride glucose-body mass index in evaluating the severity of hyperlipidemic acute pancreatitis

ObjectiveTo investigate the value of triglyceride glucose-body mass index (TyG-BMI) index in predicting the severity of hyperlipidemic acute pancreatitis (HLAP). MethodsA retrospective analysis was performed for the clinical data of 185 patients with HLAP who were admitted to Tianyou Hospital Affiliated to Wuhan University of Science and Technology from January 2021 to December 2023, and according to the revised Atlanta classification criteria for acute pancreatitis, they were divided into mild group with 95 patients and moderate or severe group with 90 patients. Clinical features were compared between the two groups to analyze the correlation between TyG-BMI and the severity of HLAP, and the efficacy of TyG-BMI in predicting the severity of HLAP was analyzed. The independent-samples t test or the Mann-Whitney U test was used for comparison of continuous data between two groups, and the chi-square test was used for comparison of categorical data between two groups. Univariate and multivariate Logistic regression analyses were used to investigate the influencing factors for the severity of HLAP. A Spearman correlation analysis was used to investigate the correlation between TyG-BMI and the severity of HLAP, and the receiver operating characteristic (ROC) curve was used to analyze the efficacy of TyG-BMI in predicting the severity of HLAP. ResultsThere were significant differences between the mild group and the moderate or severe group in creatinine, blood glucose (GLU), triglyceride, TyG-BMI, and Bedside Index for Severity in Acute Pancreatitis (BISAP) score (Z=-2.059,-7.217,-7.799,-9.566 and-11.386,all P<0.05). The multivariate Logistic regression analysis showed that BISAP score (odds ratio [OR]=4.221, 95% confidence interval [CI]: 1.421 — 12.538, P=0.001), TyG-BMI (OR=1.262, 95%CI: 1.140 — 1.396, P=0.010), and GLU (OR=1.316, 95%CI: 1.040 — 1.666, P=0.022) were independent risk factors for the severity of HLAP and were positively correlated with the severity of HLAP (r=0.839, 0.705, and 0.532, all P<0.05). In the comparison of the efficacy of these indicators in predicting the severity of HLAP, TyG-BMI had a slightly lower efficacy than BISAP score (Z=-4.368, P<0.001) and a significantly better efficacy than GLU (Z=2.155, P<0.001), with an area under the ROC curve of 0.891, a sensitivity of 91.10%, and a specificity of 96.80%. ConclusionTyG-BMI index has a certain value in predicting the severity of HLAP and can be used in clinical comprehensive assessment of HLAP.
9.Advances in the Action Mechanism of Classical Pathways IKKα and IKKβ in Hematological Tumors and Drug Therapy Blocking Their Effect--Review.

In recent years, it is found that the classical IKKα and IKKβ pathway were closely relates with hematological tumors, except the classical pathogenesis, moreover the classical IKKβ pathway is deeply studied. The studies indicated that the IKKβis activated to phosphorylate the NF-κB through multiple cascades under the effect of extracellular IL-6, TNF-α and other stimulating factors. At the cellular level, the classical IKKβcan promote the tumor cell survival and proliferation, reduce the cell apoptosis, and promote the angiogenesis and cell transfer. Although the classical IKKα plays a role in regulating IKKβ activity, but its role in non-classical pathway is more prominent. This review briefly summarizes the latest advance of researches on the pathogenesis of hematological malignancies in term of IKKα and IKKβpathway, so as to provide the theoretic basis for deeply understanding and studying the pathogenesis of hematologic tumors. At present, blocking the classical IKKα and IKKβ pathway has become a new target for treatment of hematological tumors, moreover, some specific inhibitor for IKKα and IKKβpathway have been developed, for example, LY2409881, BMS 345541 and so on. Most of these drugs are in clinical trials and display some good anti-tumor effects.

10.Analysis of Blood Concentration Monitoring Results of Sodium Valproate in 856 Patients of Our Hospital from 2016 to 2018

OBJECTIVE:To reference for the rational use of sodium va lproate in clinic. METHODS :By retrospective analysis,blood concentration monitoring results of sodium valproate and medical record data in 856 patients were collected from the Affiliated Tianyou Hospital of Wuhan University of Science and Technology during Jan. 2016-Dec. 2018. The dosage form of sodium valproate ,monitoring times of therapeutic drugs ,monitoring results of steady-state blood concentration of sodium valproate up to the standard ,dosage adjustment and the combination with carbamazepin ,fluconazol and carbapenem drugs were analyzed. Fisher exact test was used to analyze the factors influencing the steady-state blood concentration of sodium valproate up to the standard. RESULTS :A total of 1 270 cases of sodium valproate were monitored in 856 patients,involving 407 males and 449 females,with age of (38.2±13.8)years and body mass of (52.3±10.0)kg. Among 1 270 cases of monitoring ,steady-state blood concentration of sodium valproate in 554 cases were in the range of 50-100 µg/mL,and 43.6% of which reached the standard. The rate of reaching the standard in patients with multiple monitoring was higher than patients with single monitoring ;the dosage of patients with last monitoring reaching the standard was higher than that of patients with the first monitoring reaching the standard. The rate of reaching the standard in Sodium valproate sustained-release tablet group was higher than general Sodium valproate tablet group;the carbamazepin/fluconazol free group was higher than the carbamazepin combination group and fluconazol combination group;the carbapenem free group was higher than the carbapenem combination group (all P<0.05). CONCLUSIONS :Clinical pharmacists should pay attention to the monitoring of sodium valproate treatment drugs , strengthen the publicity and 3551851542@qq.com education of patients and their families ,and try to use Sodium valproate sustained-release tablets. When patients additionally receive carbapenem drugs like carbamazepin or fluconazol , the standard level of sodium valproate will be reduced ,then the dosage of sodium valproate should be adjusted.
