1.Research on Relationship Between Neonatal Health and TCM Constitution Types of Early to ;Mid-Pregnancy Expectant Parents

Objective To investigate the relationship between neonatal health and TCM constitution types of early to mid-pregnancy expectant parents;To provide basis for the targeted interventions for couples of childbearing age before pregnancy-periconceptional TCM constitution. Methods Compiling scales for pregnant couple’ TCM constitution type assessment and postpartum visits to newborns, the research team got access to the information of neonatal physical problems, constitutions and changes of 675 babies’ father and mother who had established maternal manual from April, 2012 to March, 2013 at the Sanlin Community Health Center of Pudong District of Shanghai. Results Early to mid-pregnancy expectant parents mainly had peace constitution type. In detail, 55% of pregnant women had gentle constitution type, while others mainly had qi deficiency, yang deficiency and damp-heat constitution types;76% expectant fathers got peace constitution, while others mainly got damp-heat, yin deficiency and qi deficiency constitution types. Marital pregnancy physique did not change significantly. Compared with the peace constitution type, newborns, whose fathers got damp-heat constitution type, had higher risk of jaundice, and whose mothers got yin deficiency, the babies had higher risk of spits (P<0.05). The constitution types of expectant parents did not have obvious combined action on neonatal health. Conclusion There is a certain relationship between neonatal health and TCM constitution types of early to mid-pregnancy expectant parents. It is necessary to distinguish constitution types of prospective parents, and thus to take targeted health interventions.
2.Clinical significance of IgG determination in children with Henoch-Schonlein purpura

Objective To investigate the role of IgG determination in diagnosis of early renal damage in children with Henoch-schonlein purpura(HSP). Methods Urine IgG was determined in 188 children with HSP (HSP group), 99 patients with Henoch-schonlein purpura nephritis (HSPN group) and 26 healthy controls (healthy control group). Simultaneously, qualitative detection of urine protein was carried out in HSP group. Results The abnormality rate of urine IgG was significantly higher than that of urine protein in HSP group (P<0. 05). The level of urine IgG was significantly higher in HSP group than that in healthy control group (P<0. 05), while the level of urine IgG in group than that in HSP group (P<0. 05). Conclusion Urine IgG determination is simple and nonin-vasive, which if high clinical value in diagnosis of early renal damage in children with Henoch-schonlein purpura(HSP).
3.Study of the concentrations of interleukin\|5 in tissue and secretion of nasal polyps

Objective: To explore the IL\|5 values in the tissue and secretion of nasal polyp. Methods: The concentrations of IL\|5, either in tissue homogenate or in nasal secretion, were measured by ELISA. The values of total protein in tissue homogenate were measured by Bradford colorimetry. In this study, 31 cases with nasal polyp, 8 patients with perennial allergic rhinitis (PAR), 11 cases with chronic sinusitis (CS) and 6 control cases were chosen. Results: ① In tissue homogenate, the IL\|5 concentrations of nasal polyp were higher than that in CS or control group (P<0.001). ② In nasal secretion, IL\|5 values of both nasal polyp and PAR group were significantly higher than that in CS or control group (P<0.05). ③ When all cases or only patients with nasal polyp were taken as the subjects, the Pearson correlation coefficient were 0.70, 0.66 respectively. Conclusions: IL\|5 values, both in tissue homogenate and nasal secretion, are significantly higher, which suggested that there were a close relationship between IL\|5 and nasal polyp, and IL\|5 play a key role in the formation of nasal polyp.

Objective To study the effect of IL-8 on cell proliferation and invasion,and to analyze the correlations between chemokine and epidermal growth factor receptor(EGFR)signaling pathways in breast carcinoma cells.Methods IL-8 secretion responded to treatment with rhEGF and anti-EGFR and expression of its receptors CXCR1,CXCR2 in MDA-231 cells were measured by ELISA and immunocytochemistry,respectively.Effect of rhIL-8 and neutralizing antibody on cell proliferation and invasion were analyzed by using MTT and matrigel invasion assay.EGFR transactivation stimulated with rhIL-8 and neutralizing an tibody was assessed by Western blot using anti-phosphotyrosine antibody.Results MDA-231 cells released hish level of IL-8 and two receptom of IL-8(CXCR1 and CXCR2)both expressed on cell membrane.Exogenous IL-8 and its neutralizing antibody did not significantly influence the proliferation of breast carcinoma cells,but rhIL-8 stimulated invasive activity in MDA-231 cells and its neutralizing antibody inhibited the in vasive activity(P<0.05).EGF and anti-EGFR both inhibited the secretion of IL-8 in breast carcinoma cells,and IL-8 had no effect on EGFR phosphorylation,but anti-IL-8 induced transactivation of EGFR after 24h.Conclusion IL-8 contributes to tumor progression in breast carcinoma through its enhancement of in vasive activitv but not act as an autocrine growth factor.The correlation of competitive inhibition rather than cross-talk is found between G protein coupled receptor(GPCR)-mediated IL-8 signaling pathway and EGFR pathway in breast carcinoma.
5.Study on the association between interleukin-5 and eosinophil in nasal polyp

Objective:To explore the expression of IL-5 and its role in the formation and development ofnasal polyp. Method: 31 patients with nasal polyp, 11 patients with chronic sinusitis (CS) and 6 control cases werechosen . Their IL-5 concentration in tissue homogenate were measured by ELISA. All patients with chronic si-nusitis,control cases and 15 patients with nasal polyp were chosen to be counted eosinophils in their HE slice.Result :①The values of IL-5(pg/ml) in nasal polyp,CS and control group separately were .. 23.44± 6.68,16.41±3.09,12.86±4.17. IL-5 concentration in nasal polyp group was higher than that in the other two groups( P <0. 001). ②The numbers of eosinophils in nasal polyp ,CS and control group were 7.42±2.33,1.30± 0. 59,1.07±0.70 separately,the numbers of eosinophils in nasal polyp group were higher than that in CR or control group(P < 0.05). Conclusion: Nasal polyp is a disease characterized by eosinophilia, and IL-5 which activateseosinophil plays part role in the formation of nasal polyp.
6.Effect of overexpression of p16 on anion exchange function of anion exchanger 1 at HeLa cell lines

AIM: To study the effect of the overexpression of p16 on an anion exchange function of band 3 in HeLa cells. METHODS: The expression of p16 and band 3 in HeLa cells was detected by immunohistochemistry (IHC). The p16 cDNA was subcloned to plasmids pEGFP-C1 by PCR and identified by restriction enzyme digestion and sequencing, and then, the recombinant pEGFP-C1-p16 plasmids were transiently transfected into HeLa cells. The expression of fusion protein in HeLa cells was detected by fluorescence microscope. 6-methoxy-N-(3-sulfopropyl)-quinolinium(SPQ)fluorescent probes were used to detect the anion exchange function of band 3. RESULTS: P16 and band 3 were expressed in HeLa cells. The amplificated p16 cDNA sequence was the same as the report sequence. The transfective efficacy of pEGFP-C1-p16 was above 60%. The anion exchange function increased after the transfection of pEGFP-C1-p16 plasmids. CONCLUSION: p16 facilitates the anion exchange function of band 3 in HeLa cells.
7.Comparison of peroral endoscopic full-thickness myotomy and circular myotomy for severe achalasia

Objective To compare the efficacy and safety of full-thickness peroral endoscopic myotomy (POEM) and circular myotomy for patients with severe achalasia.Methods The clinical data of 123 patients with severe achalasia who were admitted to the Second Xiangya Hospital of Central South University from August 2011 to May 2013 were retrospectively analyzed.Seventy patients who received full-thickness POEM were in the full-thickness myotomy group,and the other 53 patieuts who received circular myotomy were in the circular myotomy group.The clinical efficacies and incidences of complications of the 2 groups were compared.Patients in the 2 groups were followed up at the out-patient department till May 2014.The consecutive measurement data were presented by (x) ± s and analyzed using thc t test; the non-consecutive data were presented by M (range) and analyzed using the Wilcoxon rank test.Data before and after operation were compared using the repeated measure of analysis of variance.The count data were analyzed using the chi-square test.Results All the patients successfully received POEM.The operation time of the full-thickness myotomy group and the circular myotomy group were (57 ± 8)minutes and (63 ± 12)minutes,with significant difference between the 2 groups (t =3.421,P <0.05).The incidences of complications of the full-thickness myotomy group and the circular myotomy group were 14.3% (10/70) and 11.3% (6/53),with no significant difference between the 2 groups (x2=0.234,P >0.05).Atotal of 119 patients were followed up,with the median time of 18 months (range,12-24 months).The Eckardt scores at postoperative month 6 and 12 were 0 (range,0-3) and 0 (range,0-3) in the full-thickness myotomy group,and 0 (range,0-2) and 0 (range,0-3) in the circular myotomy group,with no significant difference between the 2 groups (Z =0.525,1.476,P > 0.05).The sussess rates of the full-thickness myotomy group and the circular myotomy group were 98.6% (69/70) and 98.1% (52/53),with no significant difference between the 2 groups (x2=0.040,P > 0.05).The diameters of the esophagus at postoperative month 6 of the full-thickness myotomy group and the circular myotomy group were (3.2 ± 0.3) cm and (3.4 ± 0.4) cm,with no significant difference between the 2 groups (t =1.927,P > 0.05).The diameters of the esophagus at postoperative month 6 and 12 were significantly lesser than (5.9 ± 1.0) cm and (5.9 ± 1.0) cm before operation (F =780.923,493.018,P < 0.05).No recurrence was detected in the 2 groups during the follow-up.Conclusion The short-term efficacy and incidence of complications of full-thickness myotomy and circular myotomy are comparable,while the operation time of patients who received full-thickness myotomy is shorter.
8.The Prediction model of herpangina epidemic trend based on Baidu index and hand, foot and mouth disease

Objective:
To establish a prediction model of herpangina epidemic trend based on Baidu index and hand, foot and mouth disease, so as to provide insights into analyses of communicable disease epidemics with limited or missing surveillance data.
Methods:
The incidence of hand, foot and mouth disease in Zhejiang Province during the period from the first week of 2015 through the 39th week of 2021 was retrieved from the China Information System for Disease Control and Prevention, and the Baidu index of hand, foot and mouth disease and herpangina was collected via the Baidu search engine during the same period. The correlation between the Baidu index and time series of hand, foot and mouth disease was examined using wavelet analysis. In addition, a random forest training model was created based on the Baidu index and incidence of hand, foot and mouth disease, and the fitting effectiveness was evaluated using the mean percentage error, while the Baidu index of herpangina was included in the model to predict the epidemic trend of herpangina during the study period.
Results:
The Baidu index of herpangina and hand, foot and mouth disease, and the Baidu index and incidence of hand, foot and mouth disease all appeared two peaks at the 26th and 52th week. The phase difference was less than 0.1 week between the Baidu index and time series of hand, foot and mouth disease, and the mean percentage error of the training model was 13.07%, with high concordance between the predicted number and actual report number of cases with hand, foot and mouth disease. The numbers of herpangina cases were predicted to be 28 822, 27 341, 28 422, 51 782, 52 457 and 5 691 from 2015 to 2020, and there were totally 48 702 herpangina cases reported until the 39th week of 2021. Like hand, foot and mouth disease, the incidence of herpangina peaked between May and July.
Conclusion
The random forest training model based on the Baidu index and incidence of hand, foot and mouth disease is feasible to predict the epidemic trend of herpangina.
9.Epidemiological characteristics of notifiable infectious diseasesin Zhejiang Province, 2021

Objective:
To investigate the epidemiological features of notifiable infectious diseases in Zhejiang Province in 2021, so as to provide the evidence for formulating infectious disease control measures.
Methods:
The data pertaining to notifiable infectious diseases reported in Zhejiang Province in 2021 were retrieved from the Infectious Disease Surveillance System of China Information System for Disease Control and Prevention. The incidence and mortality of notifiable infectious diseases in Zhejiang Province in 2021 were descriptively analyzed.
Results:
Totally 29 types of notifiable infectious diseases with 396 623 cases and 391 deaths were reported in Zhejiang Province in 2021, with a reported incidence of 614.28/105 and a reported mortality of 0.605 6/105, respectively. There were 93 204 cases with class B notifiable infectious diseases, with a reported incidence rate of 144.35/105 and 303 419 cases with class C notifiable infectious diseases, with a reported incidence rate of 469.92/105; while no cases with class A notifiable infectious diseases were reported. Syphilis (39.45/105), tuberculosis (37.12/105), viral hepatitis (31.90/105) and gonorrhea (26.34/105) were the four most common class B notifiable infectious diseases, and AIDS (0.477 0/105) and pulmonary tuberculosis (0.116 2/105) were the two most deadly class B notifiable infectious diseases, while hand, foot and mouth disease (192.00/105), other infectious diarrhea (184.24/105) and influenza (86.45/105) were the three most common class C notifiable infectious diseases. According to the transmission route, intestinal and respiratory infectious diseases were the two most common infectious diseases, with reported incidence rates of 384.10/105 and 133.73/105, respectively; and according to the reported region, the highest incidence of class B notifiable infectious diseases was reported in Zhoushan and Ningbo cities, and the highest incidence of class C notifiable infectious diseases was reported in Ningbo City. Totally 1 101 COVID-19 cases were reported in Zhejiang Province in 2021, including 712 confirmed cases and 389 asymptomatic cases, and no deaths occurred.
Conclusions
The reported incidence of notifiable infectious diseases declined in Zhejiang Province in 2021 as compared to that prior to COVID-19 epidemics, with remarkable reductions in the incidence of respiratory and intestinal infectious diseases. The management of pulmonary tuberculosis, viral hepatitis and AIDS requires to be reinforced during the containment of COVID-19, to prevent the seasonable epidemic of influenza, hand, foot and mouth disease and other infectious diarrhea in Zhejiang Province.
10.Epidemiological characteristics of overseas imported COVID-19 casesin Zhejiang Province

Objective:
To analyze the epidemiological characteristics of overseas imported cases with coronavirus disease 2019 (COVID-19) in Zhejiang Province, so as to provide insights into containment of overseas imported COVID-19.
Methods:
The pertaining to overseas imported COVID-19 cases in Zhejiang Province during the period between January 1, 2020 and May 31, 2022 were captured from the Chinese Disease Prevention and Control Information System. The temporal, spatial and population distributions, the duration from entry to the first time of positive severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) nucleic acid test, and COVID-19 vaccination of overseas imported COVID-19 cases were analyzed using a descriptive epidemiological method.
Results:
A total of 1 535 overseas imported COVID-19 cases were reported in Zhejiang Province during the period between January 1, 2020 and May 31, 2022, including 596 confirmed cases and 939 asymptomatic infections, and all reported cases were imported from 102 countries and territories. Overseas imported COVID-19 cases were reported in each month during the period between March 2020 and May 2022 except May 2020, and the mean monthly number of reported overseas imported COVID-19 cases was 125 during the period between December 2021 and May 2022. Overseas imported COVID-19 cases were reported across 11 cities of Zhejiang Province, with the largest numbers reported in Hangzhou (978 cases) and Jiaxing cities (177 cases), and imported cases were reported in 76.09% of counties (districts) in Zhejiang Province. The overseas imported COVID-19 cases were predominantly identified among individuals with Chinese nationality (88.79%), at ages of 20 to 39 years (59.09%), and students (21.82%), workers (17.20%) and business servants (17.00%) were predominant occupations among the overseas imported COVID-19 cases. The median duration from entry to the first time of positive SARS-CoV-2 nucleic acid test was 3.98 (7.06) d during the period between January 1, 2020 and May 31, 2022, and was 3.23 (4.97) d during the period between December 1, 2021 and May 31, 2022, which was significantly shorter than that during the period between January 1, 2020 and November 30, 2021 (P<0.001). The proportion of normal and more severe types of COVID-19 was 15.69% among cases without COVID-19 vaccination, which was significantly higher than that (7.77%) among those receiving booster vaccination (χ2=5.345, P=0.021), but was not significantly different from that (12.65%) among those receiving full-dose vaccination (χ2=0.971, P=0.324).
Conclusions
Students with Chinese nationality, workers and business servants were predominant among overseas imported COVID-19 cases in Zhejiang Province until May 31, 2022. The duration from entry after December 1, 2021 to identification of overseas imported COVID-19 cases shortened, and booster COVID-19 vaccination facilitated the alleviation of severity of clinical symptoms. There is a long-term risk of overseas importation of COVID-19 in Zhejiang Province, and the containment of imported COVID-19 requires to be sustainably implemented among entry personnel.
