1.Correlation of serum cardiac troponin levels and cardiovascular events in elderly type 2 diabetes patients

Objective To study the correlation of serum cardiac troponin levels and cardiovascular events in elderly type 2 diabetes patients,and provide the basis for prevention and treatment of cardiovascular disease combined with type 2 diabetes mellitus.Methods Totally 200 elderly type 2 diabetes patients were selected in the First Affiliated Hospital of Zhengzhou University,and divided into cardiac troponin-positive and negative groups according to serum cardiac troponin levels.The incidence of cardiovascular events and mortality was compared between the two groups.Results The levels of fructosamine and hemoglobin in cardiac troponin-positive group were significantly lower than those of cardiac troponin-negative group (P<0.05).The C-reactive protein levels were increased in cardiac troponin-positive versus negative group (P <0.05).The incidences of cardiovascular events and mortality in cardiac troponin-positive group showed higher than those of cardiac troponin-negative group[63.3% (19/30)vs.4.1 % (7/170),26.7% (8/30) vs.1.2% (2/170),x2 =73.45,28.44,all P<0.05].Conclusion Lower levels of serum fructosamine and hemoglobin,higher C-reactive protein level exist in diabetes patients with high cardiac troponin levels.The serum cardiac troponin level has a certain relationship with cardiovascular events in diabetes patients.
2.Experimental study on effect of chlorogenic acid on oxidative stress of kidney in diabetic rats

Objective To investigate the effect of Chlorogenic acid on oxidative and antioxidative system of kidney in diabetic rats.Methods 36 rats were randomly divided into three groups:normal group(N group),diabetes mellitus group(DM group)and diabetes group treated with chlorogenic acid(CA group).Ten weeks later,blood glu-cose,weight of body and kidney ,the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)lipidperoxidation(MDA),urinary albumin excretory rate(UARE)in renal tissue were measured.Results The levels of blood glucose were not significantly different between DM group and CA group,but the weight of rats and kid-ney and the activity of SOD and GSH-PX in CA group were higher than DM group,and MDA and UAER of CA group were lower(P<0.05 for each).Conclusion Chlorogenic acid effective in improving the antioxidativity,inhibiting oxidative stress and decreasing urinary albumin.
3.Clinical observation of mosapride combined with acupoint massage in treating senile type 2 diabetic gastroparesis

Objective To investigate the clinical effect of mosapride combined acupoint massage on diabetic gastroparesis(DGP)in elderly patients with type 2 diabetes.Methods 150 cases of DGP in our hospital from April 2015 to May 2016 were collected and randomly divided into three groups:the conventional treatment group(n=35,in insulin treatment),the control group(n=52,in mosapride treatment),and the experimental group(n=63,in mosapride combined with acupoint massage treatment).All patients received the conventional diabetic therapy.Clinical efficacy,gastric emptying rate and the levels of serum gastrin and fasting blood glucose were observed and compared before versus after treatment among the three groups.Results After treatment,the total effective rate was 92.1% in the experimental group,versus 51.4% and 73.1% in the conventional treatment group and control group respectively,showing a statistically significant difference(P<0.05).There were significant differences in gastric emptying rate,the levels of serum gastrin and fasting blood glucose among the 3 treatment groups before versus after treatment.And gastric emptying rate,the levels of serum gastrin and fasting blood glucose were improved in the experimental groups compared with the conventional treatment and control groups(all P<0.05).The follow-up results showed that the adverse reactions and the recurrence rates were significantly lower in the experimental group than in conventional treatment group and control group(all P<0.05).Conclusions Mosapride combined with acupoint massage in the treatment of diabetic gastroparesis have significant clinical effect,low recurrence rate and high safety,and is worthy of promotion.
4.Treatment scheme selection strategy and progress of elderly femoral neck fracture of Garden type II

BACKGROUND: There are various treatment methods for femoral neck fracture in the elderly, and they have advantages and disadvantages, especially for the elderly type of stable femoral neck fractures Garden type II of treatment options. OBJECTIVE: To summarize the current main clinical methods of senile femoral neck fractures of Garden type II, compare the advantages and disadvantages of internal fixation and joint replacement, and provide the best treatment for orthopedic surgeons. METHODS: We retrieved PubMed database. Key words were elderly femoral neck fracture, Garden type II, selection strategy and progress. Combining with the clinical cases encountered in the comparative analysis, the relevant discussion was analyzed. RESULTS AND CONCLUSION: Although the risk of surgery and the pain of non-surgical patients in early stage, the effects are not ideal. Internal fixation of Garden type II femoral neck fracture and joint replacement in elderly patients is controversial. For the elderly and Garden type II femoral neck fractures, the rate of revision after cannulated screw fixation was significantly higher than that of hip replacement. Both cement-type and biological-type implant prosthesis can improve hip function largely, and reduce non-surgical complications, and finally get a satisfactory clinical effect. The clinician should make the best treatment plan according to the patient’s age, bone condition, fracture type and physical condition.
5.Information Management System for Rescuing Batches of the Sick and Wounded

Objective To investigate the information management method for improving the efficiency of rescuing batches of the sick and wounded.Methods A portable palm computer were used to collect the information of the sick and wounded and the collected data were transmitted in real time to the server of the higher rescuing institution in the hospital via wired and wireless networks after being counted up,analyzed and summarized.Results The higher rescuing institution took the real-time information as the decision-making base at any time to give orders and provide assistance that fed back to the rescuing site at any moment.Conclusions The system combines medical rescuing with modern computerized information system,realizing the informatization,digitalization and automation of direction and management models in rescuing batches of the sick and wounded and greatly improving the efficiency for rescuing the sick and wounded.
6.Biocompatibility of chitosan-Mg membranes with PC12 cells in vitro

Objective To prepare the chitosan-Mg membranes and to explore the biocompatibility of membranes with PC12 cells and its usage as biomaterials in tissue engineering.Methods The appearance of the chitosan-Mg membranes (CM) was observed under scanning electromicroscope (SEM) and the element of the membranes was analyzed by x-ray energy spectromete. PC12 cells were co-cultured with CM in vitro,the morphological changes of PC12 cells on membranes were observed under scanning electron microscope (SEM) and light microscope (LM),the cell vitality was detected by MTT assay. Results The surface of chitosan membranes (CS) was smoother than that of CM;but the CM groups were full of pore observed under SEM;The content of Mg element was related with the dosage of MgSO4 added into chitosan solution. Morphology observation showed that PC12 cells grew well on CM as compared with CS group. The cells were rich of microvilli and long processes on 7th day,and synapses-like structure was formed in many PC12 cells. 7.Effects of miR-206/miR-1 on Breast Cancer Stem Cell Proliferation and the Mechanism

Objective To investigate the effects of up?regulated miR?206/miR?1 on the proliferation of breast cancer stem cells and the effect mech?anism. Methods Breast cancer stem cells(BCSCs)were isolated from breast cancer cell line MCF?7 by fluorescence?activated cell sorting. Cells in the experiment were divided into the blank control group,the negative control group,the miR?206 group and the miR?1 group. The BCSCs were transfected by negative control mimic,hsa?miR?206mimic and hsa?miR?1mimic in all groups except the blank control group. MiR?206and miR?1 expression levels as well as the transcription factor EVI?1 gene were detected by real time PCR. The expression levels of the transcription factor EVI?1 protein were detected by Western blot. MTT method was used to detect the effects of miR?206 and miR?1 on the proliferation of BCSCs. Results The BCSCs(CD44+/CD24-/low cells)isolated from MCF?7 cell lines were successfully cultured in serum?free medium for subsequent studies. After transfection of hsa?miR?206mimic and hsa?miR?1mimic for 48 hours,miR?206and miR?1relative expression levels increased. EVI?1mRNA ex?pression levels significantly decreased. The results of Western blot and MTT showed that up?regulated expression levels of miR?206 and miR?1 could significantly reduce the expression of EVI?1 protein and inhibited the proliferation of BCSCs. The differences in levels of miR?206,miR?1 and EVI?1 protein were statistically significant(P<0.05). Conclusion Up?regulated miR?206 and miR?1 expression can inhibit the proliferation ability of BCSCs,which may be related to the down?regulation of EVI?1.
8.Analysis on the myocardial enzyme spectrum of mountain yaks, alien mountain yellow cattle and low altitude yellow cattle in Qinghai Plateau

Objective To compare the myocardial enzyme spectrum of native mountain yaks ( Bos grunniens) , alien mountain yellow cattle ( Bos taurus domestica) and low altitude yellow cattle breeding in Qinghai plateau, explove the mechanism that the native mountain yaks adapt to the plateau environment.Methods The samples were treated by cervical bleeding to death, and whole blood was collected.Then serum were prepared from whole blood.The myocardial enzymes including aspartate amino transferase, creatine kinase, creatinine kinase-MB isoenzyme, lactate dehydrogenase, α-hydroxybutyrate dehydrogenase and.Results In the native mountain yaks and in the alien mountain yellow cattle, levels of AST, CK, CK-MB were significantly higher than those in the low altitude yellow cattle (P<0.05);compared with the alien mountain yellow cattle, levels of AST,CK were significantly higher than those in the native mountain yaks (P<0.05),CK-MB,LDH and HBDH were lower(P<0.05).The results suggested that under the high altitude and hypoxia environment, myocardial cell injury occurred in the alien mountain yellow cattle, thus various enzymes penetrated through the cells into the blood circulation, lead to the higher serum enzymatic levels.Conclusion The phenomenon indicates that the alien mountain yellow cattle is in a state of high altitude acclimatization.But the native mountain yaks are well adapte to the high altitude and hypoxia environment.
9.Bufalin inhibits proliferation and downregulates expression of WT1 in K562 cells in vivo and vitro

Aim To investigate the effect of bufalin on proliferation and expression of WT1 in K562 cells. Methods The colony number of K562 cell was detec-ted with semi-solid culture assay. The cell cycle was measured by flowcytometry, and the expression of WT1 was observed with immunocytochemistry. Subcutaneous tumor models established by K562 cells in BALB/C nu/nu mice were divided into three groups, including model group, bufalin group and positive control group. After 21 days, the subcutaneous tumors were removed for calculating the inhibitory rate of tumor growth. HE staining and immunohistochemistry were used to ob-serve the morphological changes and the expression of WT1 . Results ① Bufalin could significantly decrease the colony number of K562 cell, arrest it at G0/G1 phase and down-regulate its expression of WT1 in a dose-dependent manner. ② Compared with the model group, the tumor inhibitory rate was much higher, while the volume and the weight were obviously lower in the other two groups. ③Bufalin could induce apop-tosis, necrosis, hemorrhage and fibrosis with HE stai-ning, and down-regulate the expression of WT1. Con-clusion Bufalin could inhibit the proliferation, arrest the cell cycle at G0/G1 phase and down-regulate the expression of WT1 in vitro. Bufalin could inhibit the tumor inhibitory rate, the volume and the weight of the subcutaneous tumors, induce apoptosis, necrosis, hemorrhage and fibrosis with HE staining and down-regulate the expression of WT1 .
10.Changes and significance of transient receptor potential melastatin 7 after myocardial infarction in the mouse

Objective To investigate the changes of TRPM 7-like current in mouse cardiac fibroblast(CFs) after myocardial infarction and the effect of myocardial ischemia on the TRPM 7 expression and current. Methods (1) The model of myocardial infarction was made and CFs were isolated;(2) CFs were cultured and infected by TRPM 7 siRNA;(3) The effects of myocardial ischemia on TRPM 7 current were recorded by whole cell patch clamp technique;(4) The influences of myocardial isehemia on Ca2+ influx in CFs were recorded by Ca2+ fluorescence imaging. (5) The effects of ischemia on total collagen content of CFs were studied. Results (1) Ca2+ inward current of CFs was increased after myocardial infarction [(16.2±1.7) vs. (7.4±0.7) pA/pF, P<0.053];(2) TRPM 7 current was reduced by 90% after siRNA infection;(3) The total collagen content of CFs after ischemia was approximately 2.3-fold higher than basic value. Conclusions Ca2+ influx in CFs plays an important role in the pathophysiological process of myocardial fibrosis.
