Objective To explore the expression of survivin gene in gastric cancerous tissue and its relationship with gastric cancer prognosis. Methods All the samples were obtained from patients with gastric cancer who underwent surgery in Zhongshan Hospital from July 1995 to June 1996.“ Envision two steps”of immunochemistry was used to detect survivin expression in the resected gastric cancerous tissue.TNM stages were determined by pathological diagnosis and clinical status. All the cases were followed up at least 5 year or until death. The discrepancy of survivin expression was compared in different pathological types and TNM stages.Survivial curves were compared in the two groups with or without survivin expression. Results Totally 96 cases(male/female:59/37;age:29-84 years old, mean age 56) were recruited and survivin expression rate was 70.8%(68/96).Among pathological types,78 cases were adenocarcinoma,18 were non-adenocarcinoma.Survivin expression was similar in different pathological types (69.7% in adenocarcinoma vs.60.0% in non-adenocarcinoma,P=0.369).Among adenocarcinoma, survivin expression rate was higher in poorly differentiated group than that in well differentiated one(82.1% vs. 62.5%,P=0.053).As the infiltration denoted in adenocarcinoma, the expression rates of survivin were 33.3% in cases limited to mucosa and submucosa,81.3% in cases limited to muscularis layer and 75.9% in cases infiltrated to whole layers (P= 0.020).Expression rate of survivin was not related with lymph node metastasis(68.1% vs.70.8%,P= 0.771).Five-year survival rate in 68 survivin positive cases was 42.93%,lower than 52.93% of 28 survivin negative cases, but no statistical difference was observed. Conclusions Survivin is highly expressed in gastric cancer.Survivin expression is closely related with differentiation status of adenocarcinoma and degree of infiltration.Further studies are needed to evaluate its role in the prognosis of gastric cancer.

Objective To observe the attenuated and synergistic effect of meteorism tablets in treating HBeAg-positive chronic hepatitis B patients. Methods Eighty patients of HBeAg-positive chronic hepatitis B were randomly divided into treatment group (meteorism tablets combined with telbivudine) and control group (telbivudine), and treated for one year. The changes of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), the negative rate of HBV-DNA, the negative rate of HBeAg, HBeAg/HbeAb serological conversion rate, creatine kinase (CK) before and after treatment, and safety were observed. Results After treatment, biochemical response, virological response and HBeAg serological response was more significant in the treatment group than that in the control group (P<0.05). The percetage of patients with elevated CK in the treatment group was significantly lower than that in the control group (32.5% vs 60%, P<0.05). Conclusion Meteorism tablets combined with telbivudine has attenuated and synergistic effect in treating HBeAg-positive chronic hepatitis B patients.

Chinese Journal of Tissue Engineering Research 2013;(27):4988-4991
BACKGROUND: Intramyocardial transplantation of autologous umbilical cord-derived mesenchymal stem cells for repair of myocardial tissue damage is paid increasing attention in the cardiovascular field. OBJECTIVE: Human umbilical cord mesenchymal stem cells were isolated and cultured. Passage 2 human umbilical cord mesenchymal stem cells were treated with various concentratins of 5-azacytidine (2.5, 5, 10, 20, 40, 80 μmol/L) for 24 hours . After removal of 5-azacytidine, cells were cultured for another 4 weeks. RESULTS AND CONCLUSION: Before 5-azacytidine treatment, filament-like structures or particles were not observed in the cells, but the amount of cytoplasm was less and uniform, nuclear/cytoplasm ratio was high, cells exhibited typical fusiform appearance and grew in a swirl-like manner, and nucleolus was obvious. After treatment with 5-azacytidine for 24 hours, some cells died in each group, and typical fusiform appearance turned into stick-like or column-like appearance, especial y in the 40 and 80 μmol/L 5-azacytidine groups. Reverse transcription-PCR results showed that atrial natriuretic peptide and α-skeletal actin gene expression levels were detected on human umbilical cord mesenchymal stem cells after treatment with 2.5 or 40 μmol/L 5-azacytidine for 4 weeks or with 5, 10, 20 μmol/L 5-azacytidine for 1, 2, 3 and 4 weeks. These findings suggest that 5-azacytidine-induced human umbilical cord mesenchymal stem cells express the specific gene of myocardiocytes.

Objective To explore the relationship between plasma interleukin-11(sIL-11) level and platelet count post-chemotherapy with hematological malignancy patients and analyse a sIL-11 level which may maintain a safe platelet count so as to guide the treatment. Methods Blood samples were collected from the patients with hematological malignancy at certain time point of pre-and post-chemotherapy, and serum level of sIL-11 and platelet count were determined separately. Different statistical methods were applied to test the relationship between sIL-11 level and platelet changes. Results 99 cases finished this study. The findings are: the sIL-11 level went up and reached the peak on day 6 post-chemotherapy, while the platelet count kept dropping to the lowest on day 10, the sIL-11 peak occurred before the lowest platelet count, patients with faster sIL-11 increase may maintain a comparatively higher plateled count. 99 eases were grouped according to the lowest platelet count and compared: the group with higher platelet count tend to have higher peak sIL-11, more cases with higher peak sIL-11, with faster daily average sIL-11 increase, the lowest platelet count occurred later. Logistic regression analysis showed the factors contributed to lower platelet includes slower daily average sIL-11 increase and sIL-11 level less than 2000 pg/ml on Day4 post-chemotherapy. Conclusion There were correlation between the serum sIL-11 level and platelet counts, the platelet count change may be predicted by determining the plasma sIL-11 level post-chemotherapy. Patients with sIL-11 level less than 2000 pg/ml on Day4 post-chemotherapy may be endangered with severe thrombocytopenia, rhIL-11 or platelet transfusion treatment should be considered.

Background and purpose: Cetuximab-containing regimen has been increasingly applied in the treatment of patients with metastatic colorectal cancer. Simultaneously, the predictive factors of outcome for cetuximab have been thoroughly researched. The aim of this study was to evaluate the relationship between K-ras status and efficacy of cetuximab containing regimen in the treatment of patients with metastatic eolorectal cancer. Methods: Between March 2006 and June 2008, twenty-seven patients with metastatic colorectal cancer were treated with cetuximab in combination with chemotherapy. The K-ras mutation status [wild-type (wt) or mutation (nat)] was examined by polymerase chain reaction (PCR) and direct sequencing. The influence of K-ras mutation status on efficacy of cetuximab-containing regimen was analyzed. Results: For 27 patiants in this cohort, K-ras wt was detected in 55.6% (15/27) cases and K-ras mt in 44.4% (12/27) cases. Statistically significant differences were found between the patients with K-ras wt and K-ras mt in terms of overall response rate (ORR) (66.7% vs 25.0%,P=0.035) and progression-free survival (PFS) (8 months vs 4 months, P=0.0028). However, there was no significant difference in overall survival (OS) (19 months vs 12 months, P>0.05). The most common treatment-related adverse effect was skin reaction, with incidence rate of 80.0% and 66.7% (P>0.05), respectively. No treatment related death was observed. Conclusion: K-ras mutation status is a predictive factor of good efficacy of cetuximab-containing regimen in the treatment of patients with metastatic colorectal cancer. Patients with K-ras wt could benefit from cetuximab in combination with chemotherapy.
