1.NEW DISCOVERY: REPORT OF A HUMAN SCHISTOSOME

Objective To delineate the m orp hology and ecology of a new human schistosome species. Methods (1)Adult schistosomes were obtained from infected mice and rabbi ts with cercariae shedding from snails which were infected with miracidia devel oping from the eggs in feces of acute schistosomiasis patients. (2)T he rabbits were dissected during 60-100 d infection period of cecariae, and thei r bladders were taken out for detecting eggs in the urine. (3)Schistosome eggs i n the feces of patients were examined with microscopy. Results (1)There were two types of male worms: one was with an average o f size 8.76 mm long and 0 43 mm wide while the other with an average size of 2 41 mm long and 0 091 mm wide and with undeveloped gynecophoral canal which ap peared in front of ventral sucker (2)There were two types of female worms: one was with an average of size 10 73 mm long and 0 74 mm wide while the other wi th an average of size 3 43 mm long and 0 12 mm wide and was fine, semitranspar ent and tape like with undeveloped ovary and vitellaria. Less than 10 eggs or none were found in t he uterus. (3)There were a lot of degenerated and black eggs in the feces of 2 p atients. (4) Schistosome eggs were detected in the urine and miracidia were hatc hed out from 8 out of 16 infected rabbits.(5) Others: 8 worms were detected in 1 mouse out of 12 dissected mice for evaluation of infectious water with sentry m ice in spite of that no Oncomelania snails were found in the area. Cercariae stayed on water surface in lying position like a c omma. The eggs returned forwardly a fter growing behind the ovary in female worm. ConclusionAccording to the primary investigation in morphology an d ecology, the new schistosome obtained is distinct from the 6 well defined ty p es of human schistosome. It is considered that a new schistosome species should be foun d and temporarily nominated as Schistosoma nanjingi( S.n.)
2.Experimental research on antiglioma effect of sodium phenylacetate

Objective To observe the change of tumor cells on sodium phenylacetate(NaPA) treating G422 glioma mice,and explore its mechanism.Methods Two kinds of mice models(intracranial and muscular G422 glioblasloma cells) were established,and were divided into five groups,among which 3 groups for expe rimental groups,they were received NaPA(1200,800,400 mg/kg?d).In the other two groups,the positive control group was administered BCNU(20 mg/kg) only one time.The negative control group was given saline 24 h later tumor inoculation,these mice bagan to be administered.The experimental group and negative control group were injected NaPA and saline for 14 days respectively.After that,the drug toxicity,the survival period,survival rate of the mice were observed,and the pathology and ultrastructure of glioma in every group were also observed.The muscular tumor mice were sacrificed for measurement of tumor suppression rate.Results NaPA can prolong the life span of the mice with glioma;it has concentration dependent inhibition to glioma proliferation;the pathology and electron microscopy showed the decrease of glioma nuclei fission image treated by NaPA,the increase of rough endoplasmic reliculum and the cell proptosis were found,it demonstrated the tumor cells had differentiation trend.Conclusion NaPA had antitumor effect by inducing glioma cell differentiation and inhibiting the growth of tumor.
3.Experimental research of sodium phenylacetate on induced differentiation in cerebral glioma cell

Objective To observe the curative effect of sodium phenylacetate (NaPA) on induced differentiation in human glioma cells P 168 in vitro,and preliminarily probe into its mechanism.Methods MTT assay flow cytometry and cell microscope were used to test the changes of P 168 cells treated by different concentrations of NaPA in vitro, and observed ultrastructureal changes of tumor cells.Results NaPA could inhibite the growth of human glioma,show the inhibition effect of time concentration dependent,the flow cytometric was used to find S phase is 11.99% or 13.17% in the NaPA treated P 168 cells and 20.17% in the NaPA untreated P 168 cells. Electron microscope of NaPA treated P 168 cells demonstrated there were rich mitochondria, rough endoplasmic reticulum and elastic filament,but in NaPA untreated P 168 cells,there were numerous scattered polyribosomes appeared.Conclusion NaPA not only inhibit the growth of P 168 haman glioma cells, but also remarkably induce the differentiation of these cells in vitro.
4.The expression of nitric oxide synthase and its association with proliferation and apoptosis of HCC cells

Objective This study was to investigate the relation of nitric oxide synthase with cell proliferation and apoptosis in hepatocellular carcinoma (HCC). Methods The expression and localization of inducible nitric oxide synthase (iNOS) and PCNA in 21 patients with HCC were studied by immunohistochemistry. Apoptotic cell in tissue section was detected by TUNEL (terminal deoxynucleotidyl transferase [TdT] mediated desoxyuridinetriphosphate [dUTP] nick end labeling). Results The iNOS protein was markedly expressed in the liver tissue of hepatic cirrhosis or chronic liver hepatitis, which was close to tumor edge. Meanwhile it was weakly or undetectable in the liver tissue localized beyond 1.5 cm from the tumor edge. The iNOS protein was markedly expressed at the invasive tumor edge and HCC cells that invased into stroma, but it was weakly or undetectable at the most tumor core. 5.Computed tomography features of gastrointestinal stromal tumor

Gastrointestinal stromal tumors (GISTs) are common mesenchymal neoplasms in the gastrointestinal tract.The results of computed tomography play important roles in the diagnosis,treatment planning and follow up of GISTs. From August 2007 to January 2012,28 patients with GISTs were admitted to the Henan Provincial People's Hospital. Sixteen tumors showed extra-luminal growth,4 showed intra-luminal growth,7 involved both intra- and extra-luminal growth,and 1 showed extra-gastrointestinal growth.After administration of contrast media,9 tumors demonstrated homogeneous enhancement,18 showed heterogeneous enhancement and 1 showed non-obvious enhancement.The enhancement of tumors in arterial and venous phase was 20 Hu higher than that in plain scan.Angiogenesis was displayed in 6 tumors,and feeding arteries were observed in 3 tumors.
6.Investigation of the apoptosis mechanism induced by lactacystin in prostate cancer cell

Objective To investigate the relationship between nuclear factor(NF)-κB activity and lactacystin induced prostate cancer cell apoptosis.Methods Two prostate cancer cell were divided into two groups:blank control group treated with culture solution,lactacystin group treated with different concentration of lactacystin(0.5,1.0,2.0,4.0 μ mol/L),the action time were 8,16 and 24 hours.The cell survival rate was measured by MTT assay.NF-κB DNA binding activity was measured by enzyme-linked immunosorbent assay,the expression of NF-κB P65 nuclear protein was detected by Western blot assay,and caspase-3 activity was analyzed by enzyme analysis assay.Results On basal condition,the NF-κ B DNA binding activity was much higher in DU145 cell than that in LNCaP cell(t=4.728,P=0.001).Compared with blank control group,different concentration of lactacystin groups'NF-κ B DNA binding activity in both the LNCaP and DU145 cell were reduced.The expression of NF-κB p65 nuclear protein decreased along with raising of lactacystin concentration in LNCaP cell,but it did not change in DU145 cell.On basal condition,caspase-3activity in DU145 cell was higher than that in LNCaP cell(t=4.519,P=0.001).After lactacystin acting of 24 hours,caspase-3 activity increased along with raising of lactacystin concentration in both the LNCaP and DU145 cell(2.0 μmol/L lactacystin group compared with 1.0 μmol/L lactacystin group,DU145 cell P=0.000,LNCaP cell P=0.000).Conclusions Lactacystin has different killing effects on prostate cancer cell.The mechanism may be related to inducing the apoptosis by down-regulation of NF-κB activity.There may be additional cell survival/death pathway in androgen-independent prostate cancer cell.
7.Microsurgical treatment of temporal lobe epilepsy

Objective To sum up clinical experiences of intractable temporal lobe epilepsy by microsurgery. Methods A retrospective review was conducted in 50 cases with epilepsy,and temporal lobe was resected by microsurgery from June 1996 to Oct. 2003. Results Follow-up patients from six months to five years.The follow up showed healing outcome in 25 cases,good results in 13 cases,unsatisfactory outcome in 9 cases,no improvement in 3 cases.Slight paresis 6 cases,divagation 8 cases,psychosis 5 cases and no death case. Conclusions Application of microsurgery can obvously improve the clinical result of patients with temporal lobe epilepsy and decrease the complication after operation.
8.Expression of glial fibrillary acidic protein in developing rat brain after intrauterine infection

9.Study on the effect of vinpocetine on the restenosis of venous bypass grafts in diabetic rats

Objective To expose the effect and its potential mechanism of vinpocetine (Vinp) on the restenosis of dia?betic grafted veins. Methods Thirty-six Sprague-Dawley rats were randomized into saline control group and Vinp treat?ment group. The autologous jugular vein to carotid artery transplantation was performed in diabetic model rats. Normal sa?line or Vinp were intraperitoneally injected. The rats were sacrificed at 0, 2 or 4 weeks after surgery, then the grafted veins were harvested. The pathological sections were used to detect the effect of Vinp on intimal hyperplasia. The protein expres?sion of proliferating cell nuclear antigen (PCNA) was detected by immunohistochemical method, and which was described by cell proliferation index. The phosphorylation of NF-κB was detected by Western blot assay. Results The treatment of Vinp on intimal hyperplasia in vivo was significant at two weeks after surgery (17.06±5.10)μm versus control group (39.79±7.84μm, P<0.01), (30.94±5.18)μm versus (63.67±18.09)μm at four weeks after surgery (P<0.01). Vinp treatment effectively reduced the protein expression of PCNA [2 weeks:(21.07±1.38)%vs. (28.13±1.35)%,P<0.01;4 weeks:(31.73±1.38)%vs. (63.67 ± 18.09)%, P<0.01]. The treatment of Vinp inhibited phosphorylation of NF-κB at two weeks (1.08 ± 0.42 vs. 0.84 ± 0.12, P < 0.01). Conclusion Vinpocetine can effectively attenuate intimal hyperplasia in diabetic grafted veins, which might be related to its effect on inhibiting phosphorylation of NF-κB as well as inflammation.
