1.Photodynamic efficiency of new porphyrin-typed drug on HGC27 and MGC803 cells

Objective To explore photodynamic therapeutic efficiency and related mechanisms of a new porphyrin-typed photodynamic therapy (PDT) drug synthesized in our lab on HGC27 and MGC803 cells.Methods Two different kinds of gastric cancer cells, HGC27 and MGC803, were chosen as cell lines to evaluate the PDT efficiency of new porphyrin-typed PDT drug based upon four group experiments: control (no drug, no light) group, drug treated group (only drug, no light), light treated group (no drug, only light) and experiment group (with drug and light at the same time). Bleaching method was used to evaluate the photostability of new porphyrin-typed PDT drug upon repititive illumination. MTT assay was carried out to test the survival rate of tumor cells after PDT. Cofocal laser scanning microscope (CLSM) was applied to observe subcellular localization of drug in HGC27 and MGC803 cells (mitochondria and lysosome). Results New porphyrin-typed PDT drug has good stability to light, no toxicity on HGC27 and MGC803 cells without light (P>0.05), while intense lethal effect was observed upon light illumination (P<0.05). The peak efficacy appeared when concentration is 3.12 μmol/L for both HGC27 and MGC803 cells and the new porphyrin-typed PDT drug localizes in lysosome other than traditional mitochondrion. Conclusion New porphyrin-typed PDT drug is a good photodynamic therapeutic sensitizer for HGC27 and MGC803 tumor cells.
2.The correlative relationship between AEEG and MRI in the diagnosis of epileptic patients

Objective To analyse the role of AEEG and MRI in diagnosis of epilepsy on its primary epileptic focus and pathologic lesions. Methods We reviewed the results of sixty epileptic patients on routine EEG, twenty-four hour active EEG(AEEG),CT scanning and MRI examinations. The relationship between epileptic discharges of AEEG and lesions of MRI was analyzed by using ? 2 test. Results AEEG showed a higher sensitivity in finding epileptic focus in 80% patients, as compared with the rate of 32% in routine EEG. The sensitivity on finding the cerebral lesion related to the epileptic focus by using MRI was 60%, as compared with the rate of 33% in CT scanning. 3.Clinical Study on Combined Needle-embedding and Medication for Depressive Sleep Disorder

Objective: To observe the clinical efficacy of combined needle-embedding and SSRIs for depressive sleep disorder. Method: Upon admission, 71 depression cases were randomized into needle-embedding group of 24 cases, using combined needle-embedding and SSRIs, acupuncture group of 21 cases, using combined acupuncture and SSRIs, and medication group of 26 cases, using SSRIs alone. After 1, 2, 4 and 6 weeks of treatment, the HAMD sleep scores were compared. Result: The HAMD sleep scores in the needle-embedding and acupuncture group were remarkably superior to the medication group and there was significant statistical difference after 1, 2 and 4 weeks of treatment (P<0.01). However, there was no statistical difference after 6 weeks of treatment. Conclusion: SSRIs can improve HAMD sleep scores. The combined needle-embedding or acupuncture with SSRIs can work faster and better than medication alone. There was no statistical difference between needle-embedding and acupuncture group, however, needle-embedding can reduce the treatment frequencies and therefore has more active clinical meaning.
4.Objective and Quantitative Evaluation for Traditional Chinese Medicine Placebo

Traditional Chinese medicine placebo has been put more and more attention to. However, there is no ac-cepted quality evaluation method for TCM placebo. How to evaluate TCM placebo objectively and quantitatively is a common problem in the industry of Chinese medicine. New technologies such as the Intelligent Sensory Technique have been used to establish the placebo evaluation methods which are suitable for TCM characteristics. This article provided the basis for establishing scientific, rational and objective evaluation guiding principles for TCM.
5.Effect of donepezil hydrochloride on the Calpain Ⅰ-Cdk5 pathway expression in the hippocampus of mice with vascular dementia

Objective To investigate the effect of donepezil hydrochloride on the expression of Calpain Ⅰ-Cdk5/p25 pathway in the hippocampal CA1 area by cerebral ischemia and reperfusion in mice.Methods Mice were divided into model group,sham-operated group and donepezil-treated group.The expression of Calpain Ⅰ in hippocampal CA1 area was measured by immunohistochemistry staining respectively at 4,6 and 8 weeks post cerebral ischemia and reperfusion.Western blot was used to evaluate Cdk5 and p25 protein expression.Results The abilities of learning and memory performance was damaged significantly at 4,6 and 8 weeks after surgery compared to sham-operated group (P< 0.05).The expression of Calpain Ⅰ of model group were (0.098 ± 0.009),(0.129 ±0.01),(0.116 ± 0.01),which were higher than that of sham-operated group (0.03 ± 0.003),(0.031 ± 0.003),(0.029 ±0.003) and there was significant difference (P < 0.05).The expression of Cdk5 in model group was (0.54 ± 0.05),(0.73 ± 0.07),(0.7 ± 0.06),which were higher than that of sham-operated group (0.23 ±0.02),(0.31 ± 0.02),(0.33 ± 0.02) and there was significant difference (P < 0.05).The expression of p25 in model group was (0.44 ± 0.04),(0.51 ± 0.04),(0.55 ± 0.06),which were higher than that of sham-operated group(0.19 ± 0.02),(0.24 ± 0.02),(0.2 ± 0.02) and there was significant difference (P < 0.05).The expression of Calpain Ⅰ of donepezil-treated group was (0.041 ± 0.004),(0.054 ± 0.004),(0.046 ± 0.003),which were lower than that of model group.The expression of Cdk5 was (0.28 ± 0.02),(0.33 ± 0.03),(0.38 ± 0.02),and expression of p25 was (0.26 ± 0.02),(0.25 ± 0.03),(0.21 ± 0.02),which were lower than that of model group respectively(P < 0.05).Conclusion Donepezil hydrochloride probably improve the learning and memory abilities by reducing the expression of Calpain Ⅰ and Cdk5/p25.
6.Comparative study of cerebrospinal fluid flow in the cerebral aqueduct between empty sella syndrome and normal volunteer

Objective To compar the cerebrospinal fluid (CSF) flow between empty sella syndrome (ESS) and normal volunteer in the cerebral aqueduct with MRI in phase contrast cine mode. Methods Thirty-eight ESS patients (ESS group) and 38 normal volunteers (control group ) were involved in this study.The aqueduct CSF flow image was positioned perpendicularly to the midbrain aqueduct at the middle sagittal T1WI or T2WI image. The waveforms were analyzed for the flow direction, flow rate, flow volume rate and cardiac cycle. Results The CSF flow of the aqueduct in control group and ESS group had two directions which was downward flow during the systolic period and upward flow during the diastolic period of the cardiac cycle. The.systolic period downward peak flow rate, diastolic period upward peak flow rate, mean downward flow rate, mean upward flow rate and mean flow rate were (5.231 ± 0.262), (4.902 ± 0.281 ),(3.083 ± 0.191 ), (3.032 ± 0.151 ), (3.151 ± 0.162) cm/s in control group, and (6.244 ± 0.356), (6.091 ±0.430), (3.916 ± 0.196), (3.812 ± 0.273 ), (3.690 ± 0.291 ) cm/s in ESS group respectively,and there was no significant difference between the two groups ( P > 0.05 ). The systolic period downward peak flow volume rate, diastolic period upward peak flow volume rate, mean downward flow volume rate,mean upward flow volume rate and mean flow volume rate were (0.050 ± 0.003 ), (0.050 ± 0.004), (0.030± 0.002), (0.031 ±0.002), (0.030 ± 0.003 ), ( 0.004 ± 0.001 )ml/s in control group, and (0.058 ± 0.003 ), (0.063 ± 0.005),(0.039 ±0.002), (0.038 ±0.003), (0.038 ±0.003), (0.004 ±0.001) ml/s in ESS group respectively,and there was no significant difference between the two groups(P > 0.05 ). The correspond cardiac cycle of systolic period downward peak flow rate, correspond cardiac cycle of diastolic period upward peak flow rate, mean cardiac cycle were (40.890 ± 37.096), (501.026 ± 19.374), (719.511 ± 14.946) ms in control group,and (35.921 ±6.218), (531.553 ± 16.764), (770.700 ±21.579) ms in ESS group,and there was no significant difference between the two groups (P > 0.05 ). Conclusion Part of CSF flows into the area of saddle in ESS patients, but it has no effect on CSF indexes in area of cerebral aqueduct.
7.Study on quality standard for Wuziyanzong Pills

Objective: To establish the quality standard for Wuziyanzong Pills (Fructus Lycii, Semen Cuscutae, Fructus Rubi, Fructus Schisandrae, Semen Plantaginis). Methods: Betaine, the index component of Wuziyangzong Pills was determined by dual wavelength TLC-scaning, ? s=514nm, ? R=468nm. Semen Cuscutae and Fructus Schisandrae were identified by TLC. Results: The average recovery was 99.28% and RSD was 2.66%(n=5), respectively. Conclusion: The method is reliable, accurate and specific. It can be used for the quality control of Wuziyanzong Pills.
8.A Meta-analysis of the Therapeutic Effect of Nape Acupuncture on Post-stroke dysphagia in Domestic Literature

ObjectiveTo investigate the clinical effect of nape acupuncture on post-stroke dysphagia by a meta-analysis.Method Domestic literature on clinical randomized controlled trials of nape acupuncture treatment of post-stroke dysphagia published from Jun. 2004 to Jun. 2014 were obtained by a computer search ofChina National Knowledge Internet(CNKI), Wanfang Data medical information system (WF), VIP information resource system (VIP) and Chinese Biomedical Literature Database (CBM) combined with a manual search of Jinan University library journal database. Literature inclusion and exclusion criteria were established to extract data and the qualities of the included studies were assessed using a Jadad rating scale. A meta-analysis was made using the software ReviewManager 5.2.ResultA total of 17 articles were included with 1158 patients. The results of meta-analysis showed that there were statistically significant differences in the total efficacy rate [OR=3.99, 95%CI(2.83, 5.63),P<0.00001] and the cure rate[OR=2.67, 95%CI(2.03, 3.53),P<0.00001] between the nape acupuncture and control groups.ConclusionNape acupuncture has clinically a good therapeutic effect on post-stroke dysphagia. Multi-center, large-sample and high-quality studies are still needed for validation because the qualities of the included articles are lower, which is one of the factors influencing the assessment.
9.Study on HPLC fingerprint chromatogram of Flos lonicerae

0.05 ). CONCLUSION: The qualities of Flos lonicerae from the four different areas were almost the same as that of the control. This method can be used to judge the appraisal of the Flos lonicerae and to distinguish between genuine and counterfeit Flos lonicerae.
10.Evaluation of white matter lesions in patients with mild and moderate Alzheimer's disease using proton magnetic resonance spectroscopy and diffusion tensor imaging

Objective To investigate the pattern of the cerebral white matter lesions in patients with mild and moderate Alzheimer's disease(AD)and healthy controls using proton magnetic resonance spectroscopy(~1H-MRS)and diffusion tensor imaging(DTI).Methods Twenty AD patients and Twenty healthy controls were recrnited.All subjects underwent clinical examination,neuropsychological assessment.The quantitative analysis of N-acetylaspartate(NAA),myoinositol(mI),Chotine(Cho)and Creatine(Cr)resonance signals in region of interests(ROIs)located in the paraventricular white matter region bilaterally were measured.Ratios of NAA/Cr,mI/Cr and Cho/Cr were calculated in two groups.In addition,conventional MRI and DTI scanning were received,fractional anisotropy(FA)and mean diffusivity(MD)values of white matter in the same regions were measured respectively.Results No significant difference between two groups were observed in NAA/Cr ratio(P>0.05).A significantly increased mI/Cr and Cho/Cr were found in AD patients than in controls(P<0.05).FA and MD values in AD patients were 0.470±0.082 and 0.771±0.099,and in controls were 0.539±0.068 and 0.691±0.064,respectively.FA value decreased significantly in AD patients(P<0.05),M D value increased significantly in AD patients(P<0.05).After controlling for age-related,partial correlation analysis revealed a negtive correlation between mI/Cr and FA value in the patients with AD(P<0.05).No correlation between mI/Cr and MD was found(P>0.05).Conclusion The results suggest that not only the gray matter is injured,but also the white matter is abnormal in AD patients.Combining ~1H-MRS with DTI alterations could provide the valuable informations about white matter lesions in AD patients.
