1.Study on porphyrin and related metal-porphyrin recognition of carbohydrate

Objective To study the molecular recognition of porphyrin compounds with carbohydrate molecular. Methods Fluorescent titration method was developed to explore the porphyrin and its metal porphyrin compounds interaction with carbohydrate compounds. Results The binding constants indicated that porphyrin and its metal compounds involved in this experiment could recognize carbohydrate compounds respectively, and interaction with lactose is stronger than glucose; Comparison with free porphyrin, metal ion Zn~(2+) and Fe~(3+) in the core of porphyrin compounds could enhance the binding capability with carbohydrate. Conclusion Porphyrin compounds has selective molecular recognition abilityto sugar molecular.
2.Detection of urogenital chlamydia and mycoplasma and analysis of drug resistance in patients with non-gonococcal urethritis ( a report of 3,280 cases )

Objective To investigate the infection of Chlamydia (CT) Untie urea mycoplasma (UU) and person type mycoplasma (MH) in urology outpatients clinic and sexually transmitted diseases clinic and drug resist-ance. Methods A British UNIPATH Clean View Chlamydia rapid immunoassay kits and Tianyang Zhongshan City electronic biosensor Limited production of mycoplasma culture and identification of susceptibility kit were used for the detection of secretion from 3,280 cases of non-gonococcal urethritis (GNU). Results Of 3,280 cases of GNU, only 241 cases were detected with positive CT, accounting for 7.35%, 1163 cases with simply UU positive, ac-counting for 35.46%, only 14 cases with MH positive, accounting for 0.43% ,. Overall detection rate of UU and CT was 42.77% and 11.59% respectively. 122 cases were detected with positive UU + MH, accounting for 3.72%. Female infection rate was higher than that of males (P < 0.01 ) ; UU, MH, MH + UU were more sensitive to azithremycin, Josamycin, doxycycline, roxithromycin, Minocycline. Conclusion The infection rate of urogenital mycoplasma and ehlamydia is higher and drug resistance rate is different. Azithromycin and Josamycin are the best to treat mycoplasma infection, followed by doxycycline, Pyronine Doxycycline, Minocycline.
3.Application of BODIPY for tooth whitening as a photosensitive drug

Objective To study the tooth whitening effect of boron-dipyrromethene (BODIPY) as a photosensitive drug combining photodynamic therapy (PDT) and the influence on the surface of the enamel.Methods After oxidation of hydrogen peroxide or BODIPY by photodynamic therapy,the extracted teeth were evaluated by Vita shade guides matching.The change of surface enamel on the crystals and microstructure was observed by scanning electron microscopy (SEM).Results The teeth treated with BODIPY by PDT were smooth and whitened with the remineralization of teeth surface in morphology.While the teeth treated with hydrogen peroxide were whitened obviously but the surface morphology of the teeth was demineralized seriously.Conclusion The results suggest that PDT is effective for teeth whitening and can promote the remineralization of teeth surface enamel with BODIPY as a photosensitive drug.PDT is expected to become a new method for tooth whitening.
4.Preparation and drug-loading property of phosphatidyl polyethyleneglycol monomethyl ether liposome

Objective To develop liposome loaded with paclitaxel for the treatment of lung cancer,novel phosphatidyl polyethyleneglycol monomethyl ether liposomes were prepared and studied.Methods A series of phosphatidyl polyethyleneglycol monomethyl ether liposomes were synthesized.Liposomes with or without drug were prepared by ultrasound.The entrapment efficiency and size of drug loaded liposomes were evaluated.The influence of liposomes to A549 lung cancer cell was investigated by MTT method.The cell uptake of liposome was observed by laser confocal scan microscope.Results The liposome loaded with paclitaxel had an entrapment efficiency of 83% and size of 100~200 nm.The MTT results showed phosphatidyl polyethyleneglycol monomethyl ether liposomes loaded paclitaxel have inhibition effect to A549 cells with long circulation,while liposomes without paclitaxel have not inhibition effect.Conclusion Drug loaded polyethyleneglycol monomethyl ether liposomes have sustained delivery to paclitaxel.They have potential application in the therapy of cancer.
5.Interaction of PLGA nanoparticles with HL60 cells by fluorescence tracking

Objective Nanoparticles are widely investigated and applied in clinical diagnosis and treatment as drug carrier,and their transmembrane process is related to their biological effects.The aim of this study was to investigate the interaction of fluorescein-labeled PLGA nanoparticles and HL60 cells via fluorescence tracking.Methods The transmembrane process of nanoparticles was quantitatively analyzed by laser scanning confocal microscopy.Results The analyzing results showed that the interaction of fluorescein-labeled PLGA nanoparticles and HL60 cells was strongly temperature-dependent.The receptor-mediated endocytosis mechanism played an important role in the transmembrane process for cellular uptake of nanopaticles.Conclusions This study provides a theoretical basis for design and application of nano-medicines.
6.Comparative study of cerebrospinal fluid flow in the cerebral aqueduct between empty sella syndrome and normal volunteer

Objective To compar the cerebrospinal fluid (CSF) flow between empty sella syndrome (ESS) and normal volunteer in the cerebral aqueduct with MRI in phase contrast cine mode. Methods Thirty-eight ESS patients (ESS group) and 38 normal volunteers (control group ) were involved in this study.The aqueduct CSF flow image was positioned perpendicularly to the midbrain aqueduct at the middle sagittal T1WI or T2WI image. The waveforms were analyzed for the flow direction, flow rate, flow volume rate and cardiac cycle. Results The CSF flow of the aqueduct in control group and ESS group had two directions which was downward flow during the systolic period and upward flow during the diastolic period of the cardiac cycle. The.systolic period downward peak flow rate, diastolic period upward peak flow rate, mean downward flow rate, mean upward flow rate and mean flow rate were (5.231 ± 0.262), (4.902 ± 0.281 ),(3.083 ± 0.191 ), (3.032 ± 0.151 ), (3.151 ± 0.162) cm/s in control group, and (6.244 ± 0.356), (6.091 ±0.430), (3.916 ± 0.196), (3.812 ± 0.273 ), (3.690 ± 0.291 ) cm/s in ESS group respectively,and there was no significant difference between the two groups ( P > 0.05 ). The systolic period downward peak flow volume rate, diastolic period upward peak flow volume rate, mean downward flow volume rate,mean upward flow volume rate and mean flow volume rate were (0.050 ± 0.003 ), (0.050 ± 0.004), (0.030± 0.002), (0.031 ±0.002), (0.030 ± 0.003 ), ( 0.004 ± 0.001 )ml/s in control group, and (0.058 ± 0.003 ), (0.063 ± 0.005),(0.039 ±0.002), (0.038 ±0.003), (0.038 ±0.003), (0.004 ±0.001) ml/s in ESS group respectively,and there was no significant difference between the two groups(P > 0.05 ). The correspond cardiac cycle of systolic period downward peak flow rate, correspond cardiac cycle of diastolic period upward peak flow rate, mean cardiac cycle were (40.890 ± 37.096), (501.026 ± 19.374), (719.511 ± 14.946) ms in control group,and (35.921 ±6.218), (531.553 ± 16.764), (770.700 ±21.579) ms in ESS group,and there was no significant difference between the two groups (P > 0.05 ). Conclusion Part of CSF flows into the area of saddle in ESS patients, but it has no effect on CSF indexes in area of cerebral aqueduct.
7.Clinical significance of the relationship between chlamydia pneumoniae infection and coronary heart disease

Objective To assess the relationship between the Chlamydia pneumonia(CPn)infection and coronary heart disease(CHD).Methods Blood samples from 200 CHD patients and 100 heathy controls were col-lected,and Cpn IgM and Cpn IgG were tested by ELISA.Results The Cpn IgM were found in 113 cases (56.5%) and in 24 controls(24.0%).The Cpn IgG positive cases were in 145(72.5%) patients and in 43 controls (43.0%).The positive rate of Cpn IgM and Cpn IgG in CHD group Was higher than that in control group(P<0.05).Conclusion:CPn positive rate is higher in CHD group than that in control group.Cpn is closely related to the pathogenesis of CHD.
8.Combined effects of cytarabine and PDT treatment on leukemia cell HL-60

Objective To evaluate the combined effects of photodynamic therapy (PDT) with PSD-007 and cytarabine (Ara-c) on human acute promyelocyte leukemia cell HL-60.Methods The experiments were divided into four groups:control group,PDT-only groups (PDT 1-4 groups:the combination of PSD-007 concentrations (5 μg/ml and 7.5 μg/ml) and the energy density of laser (EDL) (1.2 J/cm2 and 2.4 J/cm2),Ara-c-only groups (Ara-c A group:0.3μg/ml,Ara-c B group:1.2μg/ml) and combination groups (the pair-wise combinations of the PDT doses and Ara-c doses above).All combination groups were treated with three treating methods,including P24A (the cells were treated with PDT for 24 h,and then cocultured with Ara-c for another 24 h),A24P (treated with Ara-c for 24 h before PDT,and then cocultured with PDT for 24 h),and PA24 (treated with the Ara-c and PDT for 24 h).CCK-8 method was used to test the cell viability,and the combined effect was analyzed using King formula.The changes of cell cycles were analyzed using flow cytometry.Results In the case of low-dose PDT,the combination groups showed coordinated effect with all three methods,whereas in the case of high-dose PDT,they showed additive or coordinated effect in P24A and A24P,but it showed antagonistic effect in the schedule of PA24.Cell cycles were inhibited to G0/G1 phase by both PDT and Ara-c.Conclusions Coordinated effects could be found when HL-60 cells were treated with the combination of Ara-c and PDT.The effects depended on the dose of Ara-c and PDT and the operation schedules.The effects at low dose were more obvious than that of high dose,and allowing a 24 h period in between the addition of PDT and Ara-c also promoted the effects.
9.Acceptability of Taxol-poly (alkyl-cyanoacrylates) micell material

BACKGROUND: Currently used poly (alkyl-cyanoacrylates) (PACA) produces aldehyde compound in its degradation, which is easily results in toxicity and stimulation to the body. Here, a novel TaxoI-PACA micell material was synthesized, which has broad application as a kind of liposolubility drug delivery carrier. OBJECTIVE: To verify the therapeutic efficacy of Paclitaxel-PECA micells for mouse breast cancer.METHODS: Paclitaxel drug delivery micells were prepared by a multi-emulsification technique and were characterized for size, drug loading capacity, and in vitro release. Bablc breast cancer model mice were randomly divided into the physiological saline, vacant control, paclitaxel positive control, and Paclitaxel-PECA micells with low-dose, medium-dose, and high-dose groups. Paclitaxel and PaclitaxeI-PECA micells were injected into the location of mouse breast cancer, and then the tumor inhibit rates were detected. RESULTS AND CONCLUSION: The mean diameter of Paclitaxel-PECA micells was 70 nm, with 19.89% loading amount of Paclitaxel. In vitro, micells maintained sustained release of Paclitaxel for 2 weeks. Compared with the physiological saline group, the PaclitaxeI-PECA micells group exhibited superior tumor inhibit effects with doses of 30, 60, and 90 mg/kg (P < 0.001 ), which was 68.49%, 77.03% and 81.87%, respectively. The results suggested that Paclitaxel-PECA micells material has excellent acceptability as sustained-release preparation for treating mouse breast cancer.

With more and more non-affiliated hospitals of universities joining the clinical teaching of infectious diseases,there have been some dilemmas on clinical teaching,such as the shortage of staff,teaching experience,poor lecture art and teaching enthusiasm,textbook lag,the shortage of case teaching abd students'fear of infectious disease etc. Countermeasures such as perfecting the organizational structure,formulating preferential policy,clearing historical mission,increasing students' interest in learning,adding new academic progress,enriching teaching methods,setting up experts'supervision will ensure the effective teaching quality.
