1.Protective Effects of Zhen-Gan Xi-Feng Decoction-containing Serum on 6-OHDA-induced Oxidative Stress in PC12 Cells through Nrf2-ARE Pathway Activation

This paper was aimed to study the protective effects and related mechanisms of Zhen-Gan Xi-Feng (ZGXF) decoction containing serum on 6-OHDA-induced oxidative stress in PC12 cells.The ZGXF decoction containing rat serum with low-,medium-,and high-dose (8,16,or 32 g.kg-1) or blank serum was used to preprocess PC12 cells for 1 h,and cultured together with 100 μM 6-OHDA for 24 h.And then,cells were collected.The fluorescent probe 2',7'-dichlorofluorescin diacetate (DCFH-DA) and fluorescence microplate reader were used to detect the level of reactive oxygen species (ROS).Real-time quantitative PCR was used to analyze the mRNA expressions of nuclear factor erythroid 2-related factor 2 (Nrf2),Nfe2l2,heme oxygenase-1 (HO-1),glutamate-cysteine ligase catalytic subunit (GCLc),and GCL modulatory subunit (GCLm).The luciferase report gene system was used to detect the antioxidant response element (ARE) activation.The results showed that ZGXF decoction-containing serum inhibited the 6-OHDA-induced oxidative stress,upregulated the Nfe2l2,HO-1 and GCLc mRNA expressions in cells processed with 6-OHDA.However,it has no significant effect on GCLm mRNA expression.It was concluded that ZGXF decoction-containing serum had protective effects on 6-OHDA-induced oxidative stress in PC 12 cells.Its mechanism may be correlated with the upregulation on Nfe2l2 mRNA expression,which activated ARE and further induced its downstream gene of phase Ⅱ detoxifying enzyme,as well as the HO-1 and GCLc mRNA expressions of antioxidant enzyme gene.
2.The morphologic study of coronary sinus orifice and its peripheral myocardium

Objective To observe the cellular morphologic features of coronary sinus orifice(CSO) and its peripheral myocardium and discuss its functional significance in radiofrequency ablation atrial reentrant arrhythmias and AVNRT. Methods A total of 7 out of 15 cases were observed by means of autopsy, and serial sections of 7 cases of adult hearts in sagittal plane were stained with HE and Mallory staining. The cellular features of different muscular fibers and their arrangement were observed under light microscope; one case was studied under electron microscope. Results CSO and the majority of coronary sinus were encompassed by atrial myocardium; CSO fibers toward posterior was connected with extension of terminal crest (CT); and supravalvular muscular ring of right atrium(RAMR) and that toward anterior was connected with AVN. P cells were seen in superior-anterior and inferior-anterior wall of CSO. A lot of T cells were discovered on the inferior wall of CSO. Purkinje cells were mainly found in the superior wall of CSO. The muscular fibers from CSO to AVN were composed of T cells and dissected into two parts: the first one was called right atrial nodular bundle and the second one posterior node extension which was identical with the ablation target of slow pathway. In addition, lots of nerve fibers were seen in myocardium of CSO adjacency particularly in the tissue of the posterior wall of CSO. Conclusion Myocardium of CSO adjacency may belong to slow pathway of DAVNP. The myocardium of CSO adjacency participates in the formation of circular pathway in atrial reentrant arrhythmias and AVNRT. It is possible that CSO is an important latent pacing maker.
3.Applications of two analytical specifications in the external quality assessment schemes and the external comparision of internal quality control

Objective To study the applicability of a new analytical specification defined in WS/T 403-2012 in the external quality assessment schemes and the external comparision of internal quality control .Methods It was a quality management method study.Total error allowable criterions listed in WS/T 403-2012 and GB/T 20470-2006 were selected to assess the results of 23 analytes in the 1st challenge of 2013 routine chemistry external quality assessment.The acceptable rate of 23 analytes were calculated with the two specifications.Criterions of imprecision derived from the two standards were applied to assess the coefficient of variation with internal quality control data.Results With the specification based on WS/T 403-2012, the ratio of laboratories that all five samples were passed in the 1st challenge for 23 analytes ranged from 55.5%to 94.7%.The ratio of laboratories with 80%or more samples passed in 2013 EQA ranged from 73.9%to 98.5%.While ratios of two kinds described above evaluated based on GB /T 20470-2006 ranged from 63.0%to 99.2%, and from 90.0% to 99.7%, respectively.The acceptable rate of CV according to the two criterions ranged from 55.5% to 94.7% and 63.0% to 99.2%, respectively.Conclusions As evaluation criterions of external quality assessment allowable total error and internal quality control imprecision in routine chemistry , the specification in WS/T 403-2012 can be used to assess the analytical performance of clinical laboratory more objectively and comprehensively.It can help laboratories to identify the latent problems for further quality improvement.
4.Trueness verification and traceability assessment of results from a routine chemistry system for measurement of urea and creatinine in serum

Objective To verify the trueness and assess the traceability of results from a routine chemistry system procedure for measurement of urea and ereatinine in serun.Methods Series of fresh frozen patieot sera,whose values of urea or creatinine were assigned by isotope dilution gas chromatography mass spectrometry (ID-GC/MS) or isotope dilution liquid chromatography tandem mass spectrometry (ID-LC/MS/MS),were chosen to be analyzed by a routine chemistry system.The measurement results of urea and creatinine by the routine chemistry system were used for linear regression analysis against the assigned values bv the ID-MS method to calculate the percentage deviation and assess the expected bias.Results For urea and creatinine,the linear regression equations between the routine chemistry system and ID-MS methods were Y =0.9890X + 0.0192 (R2 =0.9990) and Y =0.9815X-6.4794 (R2 =0.9989),and the average percentage bias were-0.41% (P >0.05) and-4.20% (P < 0.05),respectively.The expected percentage bias at three medical decision levels were-0.46%,-0.83% and-0.96% for urea and -15.90%,-5.87% and-2.95% for creatinine.Conclusions The results of urea analyzed by the routine chemistry system were consistent with the ID-MS method,which suggested that the results of the routine system procedure could be traced to ID-GC/MS method.For creatinine,the bias between the results of routine procedures and the assigned values met the minimum acceptance criteria' derived from biologic deviations,which would be better if its specificity improved.
5.Data analysis of 2013 national trueness verification project of HbA1c measurement

Objective To assess system deviation of HbA1c measurement in clinical laboratories in China by the national trueness verification project.Methods Bias assessing research.Two lots samples of human whole blood pools with different HbA1c concentration levels were prepared and sent to laboratories by dry ice package.Laboratories were asked to measure these samples in 5 repeats per set in three consecutive Wednesday separately,results were reported through Web-based software.Meanwhile the IFCC reference measurement procedure was applied to assign HbA1c reference values for the two lots samples.The following information or data were analyzed:measurement systems,intra-lab CVs and inter-lab robust CVs of all laboratories,inter-lab robust CVs and bias based on peer groups,et.al.The criterion of bias was set at ± 4.5%.Results 106 of 120 laboratories submitted results,including 88 using high performance liquid chromatography method,13 using immune turbidimetry method and 5 using enzymatic methods the intra-lab CVs of lot 201311 ranged from 0 to 4.6%,with median of 1.1%,while for lot 201312 the intra-lab precision ranged from CV0 to CV4.5%,with median of CV0.9%.The inter-lab robust CVs of 201311 and 201312 with single determinations were 5.6% and 6.1% and inter-lab robust CVs of 201311 and 201312 of each lab's average results were 5.9% and 5.6% respectively.The inter-lab CVs of group BIO-RAD,TOSOH,ARKRAY and PRIMUS at two level were less than 5%.For all laboratories,the percents of pass of 201311 and 201312 were 61/106(57.5%) and 56/106(52.8%) respectively.The pass ratio of each group on two lots were as follows:of group BIO-RAD were both 19/45 (42.2%),of group TOSOH were 85% (17/20) and 75% (15/20),of group ARKRAY were 71.4% (10/14) and 50% (7/14),of group PRIMUS were 6/8,5/8; of group immune turbidimetric method were both 46.2% (6/13) and of group enzymatic were both 3/5.Conclusions There were improvement for the performance of trueness of HbA1c measurement in domestic laboratories,while some of them should be addressed.Academic,research institutions,EQA organizer,manufacturers and clinical laboratories should work together to achieve the standardization of HbA1c measurement.
6.Comparative Study on Protection Effect of Puerarin, Soybean Element, Quercetin and Rutin of CCl4-induced Acute Hepatic Injury Mice

This study was aimed to compare the antioxidant activity of puerarin and 3 other flavonoid compounds, and to investigate their structure-activity relationship. The intragastric administration of 4 kinds of typical flavonoids compounds (soybean element, puerarin, quercetin and rutin) were given to mice, respectively. The model mice of acute hepatic injury were established with intraperitoneal injection of 0.1% carbon tetrachloride (CCl4) after 7 days. After 18 h fasting, liver tissues were removed. The histomorphology was observed after paraffin sectioning and hematoxylin-eosin staining. The activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma-glutamyl transpeptidase (GGT) and alkaline phosphatase (ALP) in serum were detected with automatic biochemical analyzer. The content of malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in liver tissues were detected with homogenization. The pathological results of liver tissues showed that hepatic damages were decreased in all 4 medicine treatment groups compared with the model group, but there were no significant differences among these treatment groups. The results of blood serum bio-chemical analysis showed that compared with the model group, puerarin and quercetin could decrease the activities of ALT, AST and GGT in serum significantly (P < 0.05, orP < 0.01). There were no content changes of ALP. In the soybean element group, only the activities of ALT and AST decreased obviously (P < 0.05, orP < 0.01). There was no obvious change in the serum of mice in the rutin treatment group. The homogenate detection results of liver tissues showed that compared with the model group, quercetin and rutin significantly lowered MDA (P < 0.05), increased SOD and GSH-Px (P < 0.05, orP < 0.01); while soybean element and puerarin only improved GSH-Px levels (P < 0.05, orP < 0.01). It was concluded that the antioxidant capacity of quercetin was better than that of soybean element, puerarin and rutin, which may be related to its structure. Compared with 3 other chemical compounds, quercetin had more polyhydroxies and its polyhydroxies were not glycosylated, which suggested that the structure of quercetin may be closely related to its antioxidation activity.
7.The role of 3 external quality assessment programs in assessment of analytical quality on determination of serum creatinine and urea

Objective To describe and compare the roles of 3 external quality assessment programs in assessment of analytical quality of serum creatinine and urea.Methods Research in quality management methods.Sixty-five laboratories those enrolled in the Natonal Center for Clinical Laboratories′programs of routine chemistry external quality assessment ( EQA ) , trueness verification ( TV ) for small molecular metabolites and external comparison of internal quality control ( IQC) simultaneously in 2013 were selected , the performances of those laboratories of serum creatinine (crea) and urea in terms of total errors(TE), bias and CV were obtained by using the above 3 programs, and these performance were assessed against the criterion listed in the analytical quality specifications for routine analysis in clinical biochemistry ( WS/T 403-2012).The failure ratio of 65 laboratories on each performance was calculated , the sensitivity of 3 external quality assessment programs in detection of analytical quality deficiency among clinical laboratories were compared.Results Only 1 laboratory failed in the 1st routine chemistry EQA in terms of TE of creatinine , failure ratio is 1.5%(1/65).Three laboratories failed in the 2nd EQA and caused a failure ratio of 4.6%(3/65).For serum urea, 3 laboratories failed in the 1st routine chemistry EQA with a failure ratio of 4.6%(3/65).Two laboratories failed in the 2nd EQA with a failure ratio of 3.1%(2/65).The failure ratios of creatinine determination in two samples in TV were 41.5%(24/65) and 21.5%(14/65) respectively, and the failure ratio of urea determination were 53.8%( 36/65 ) and 32.3%( 21/65 ) respectively.In the program of external comparison of IQC , the CVs of creatinine and urea determination ranged from 0.7% to 6.2%and from 1.0%to 7.2%respectively, their respective failure ratio range were 15.4%(10/65) and 40.0%(26/65).The failure ratio in routine EQA were much less than those in the other two programs , the laboratories failed in routine EQA program were all failed in trueness verification or /and the comparison of IQC programs, but not vice versa.Conclusions By participating in the programs of routine EQA , TV and comparison of IQC laboratories could assess the performances of inaccuracy , bias and imprecision.Laboratories should participate in different external quality assurance programs to detect their quality issues and get improved.
8.Impact of life meaning and school adjustment on mobile phone addiction of female college students

Objective:
The research was to investigate the intermediary effect of school adjustment between famale college students’ life meaning and mobile phone addiction and to provide reference for mobile phone addiction prevention.
Methods:
Totally 1 355 female college students in Jinan City were investigated with Life Meaning Scale, School Adjustment Scale and Mobile Phone Addiction Scale.
Results:
Score of freshmen’ mobile phone addiction (34.02±7.87) was significantly lower than that of sophomores’ and junior’(36.67±8.03, 37.19±10.40)(F=4.58, P<0.05). Female college students’ life meaning and school adjustment were negatively correlated with mobile phone addiction(r=-0.50, -0.58, P<0.01). Female college students’ life meaning was positively correlated with school adjustment(r=0.51, P<0.01). Having a sense of life meaning and seeking a sense of life meaning could jointly explain 24% of the variation in mobile phone addiction. School adjustment played a complete mediating role between life meaning and mobile phone addiction in female college students.
Conclusion
Female college students’ life meaning can affect mobile phone addiction through school adjustment.
9.Effect of transcranial direct current stimulation on post-stroke cognitive impairment

Post-stroke cognitive impairment (PSCI) is a series of syndromes caused by stroke, involving impairment of one or more cognitive functions, such as attention, language function, executive function, visuospatial cognition, episodic memory and working memory, etc. The traditional treatment methods of PSCI include drug therapy and cognitive training. The treatment modalities are limited and the maintenance effect is not good. Therefore, an auxiliary treatment method is urgently needed to improve its therapeutic effect. Transcranial direct current stimulation (tDCS) is a safe and mature non-invasive brain stimulation technique, which generates weak direct current (1-2 mA) through electrodes placed on the scalp to change the resting membrane potential of neurons, regulate the excitability of the cerebral cortex, so as to achieve the purpose of treatment. This article reviews the effect of tDCS on PSCI, and hopes to provide reference and guidance for its rehabilitation treatment.
10.Epidemiological study of antibody to pertussis toxin IgG in newborns in Shunyi District of Beijing in 2016

Objective To investigate the levels of antibody to pertussis toxin (PT) IgG in newborns in Shunyi Women and Children's Hospital of Beijing Children's Hospital in 2016.Methods A total of 419 newborns were enrolled in this study.Umbilical cord blood sample was collected from each subject and detected by enzyme-linked immunosorbent assay to measure the concentration of PT-IgG.Besides,all newborns were followed up to January 31,2017.Chi-square test was used for statistical analysis.Results The detectable rate of umbilical cord blood samples for PT-IgG accounted for 30.1% (126/419).The median antibody level was < 5 U/ml,and the 90th and the 95th percentile were 14.3 and 24.0 U/ml,respectively.No cases of pertussis occurred at the end of follow-up.Conclusions The newborns born in Shunyi Women and Children's Hospital of Beijing Children's Hospital are generally lack of protective PT antibody.
