The effects of aminophylline on xenogenic graft versus host reaction and the survival of heart allograft in mice were investigated. Human T lymphocytes could be separated into two subsets when they were incubated with theophylline in vitro. One subset lost their ability to form E rosette with sheep red blood cells and was called theophylline sensitive T cell (TS). The other one maintained their ability to form E rosette was called thyeophlline resistant T cell (TR). It was shown that TS inhibited and TR enhanced GVHR induced by total T lymphocytes in cyclophosphamide treated rats. Spleen lymphocytes from mice treated with aminophyline showed significant weaker GVHR than that treated with normal saline, suggesting that aminophylline might induce suppressor cells in vivo. The effect of aminophylline on the survival of heart heterotopic allograft was then investigated and the result showed that aminophylline prolonged the survival of transplant significantly.

Objective Stromal and glandular cells of human endometrium were successfully isolated and cultured in this experiment. Methods These two types of cells were observed by light microscopy and verified by immunocytochemical staining. Results The result showed that some of stromal cells became adherent after.0 5h of culture.Two shapes of stromal cells were found.One was spindle and the another was polygonal.Immunocytochemical staining method showed that both shapes of stromal cells were positive for vimentin,with 95% of positive rate,and negative for cytokeratin 1,indicating that these cells were endometrial stromal cells.Most of glandular cells became adherent after 24h of culture and formed tightly packed whorls after 4d.Individual cells were polygonal and had a large and round nucleus.Immunocytochemical staining method showed that glandular epithelial cells were positive for cytokeratin 1,with 90% of positive rate,suggesting their nature of epithelial cells. Conclusion Stromal and glandular cells of human endometrium were successfully isolated by using two series of filter and cultured in this experiment. [

Objective This experiment was to identify the influence of recombinant human endostatin (rhES) combined with 5-FU on the growth of a xenografted gastric cancer in nude mouse. Methods After the establishment of the model, mice were divided into 4 groups (six in each) receiving respectively, for consecutive 10 days: NS, 5-FU (10 mg/kg) intraperitoneally; rhES (2 mg/kg); and 5-FU + rhES. On 15th day tumor volume was measured. The expression of VEGF, bFGF, VEGF-C, PCNA, bcl-2 and apoptosis index (AI) were examined. The tumor inhibition rate(TIR) and tumor shrinking rate(TSR) were calculated. Objective To investigate the time, location and relationship to clinical manifestation of abnormal expression of tyrosine hydroxylase(TH) in the cerebellum of Niemann\|Pick type C disease (NPC). Methods Immunohistochemical staining was applied, by using antibodies to TH, to brain sections from NPC and normal mice of various ages. Some adjacent sections were stained for calbindin D28k. Results There was no marked decrease in number of cerebellar Purkinje cells(PC), which were negatively stained for TH, in the NPC mice aged 1\|3 weeks. The PC were decreased in number starting from week 5. During postnatal weeks 8\|11, PC were significantly lost, but considerable number of PC in the nodulus and uvula vermis survived. Some survived PC and their dendritic trees were TH immunoreactive after week 8. These PC showed axonal spheroids and irregular dendrites that were bent, broken, locally enlarged or atrophied. Conclusion Mutation of NPC1 gene induces severe loss of cerebellar PC and survived PC have been damaged morphologically and show abnormal gene activity. These may be the pathogenic basis of movement disorders of NPC.

Objective To investigate the significance of overexpression of P53 and bcl-2 protein in carcinogenesis of cervix. Methods 10 cases of cervical intraepithelial neoplasis(CIN) and 57 cases of invasive cancer were investigated with immunohistochemistry technique. Results The overexpresion of P53 protein in CIN and cervical cancer was significantly higher than that of control, respectively (P＜0.01). But there was no significant difference between CIN and cervical cancer(P＞0. 05). The immunoreactivity of bcl-2 in CIN was much more higher than that of control (P＜0.05). The positive rate and immunoreactivity of bcl-2 in cervical carcinoma were both remarkably higher than those of control (P＜0. 01) ,but there was no significant difference between CIN and cervical carcinoma (P＞0. 05). It was also found that there was a remarkably positive correlation between the overexpression of bcl-2 and P53 (P＜0.01). Conclusion Because of the loss of wtP53 function,the expression of bcl-2 can not be down-reguated,which is associated with the pathogenesis and development of cervical carcinoma.

Objective To investigate if vascular endothelial growth factor-D(VEGF-D) and endostatin(ES) promote angiogenesis in colorectal cancer(CRC). Methods One hundred and five patients undergoing CRC surgery were enrolled in this study. CRC and normal tissue microarray were produced and measured for VEGF-D and ES by immunohistochemmical staining.Results VEGF-D and ES expression were stronger in CRC tissues than in control normal mucosa. MVD was higher in cancer tissues than in normal ones. ES down-regulated expression was associated with less invasiveness, positive lymph node metastasis, distant metastasis and in cases of early Dukes′ stage, while the expression of VEGF-D in corresponding tissues was on the opposite to ES. MVD increased along with tumor invasion, lymph involvement, distant metastasis and late Dukes′ stage. Conclusions VEGF-D in accord with ES promotes the angiogenesis and progression of CRC.

Objective To investigate whether the new mechanically-activated (MA) cation channel Piezo 1 protein can cause the apoptosis of human chondrocytes under compressive loading,using a Flexercell unit by activating MAPK/ERK5 signal pathway.Methods Primary human chondrocytes were isolated,cultured,and then subjected to the static compressive loading for 2 h,12 h,24.h and 48 h,respectively.The GsMTx4,which is the special inhibitor of the Piezo1 protein and the BIX02188,which is the inhibitor of the ERK5 served as the inhibitor group.The immunofluorescence was used to locate the expression of the Piezo 1 protein.The expressions of Piezo1 and ERK5 were assessed by reverse transcription-polymerase chain reaction (RT-PCR),as well as the apoptosis gene B cell lymphoma/leukemia-2 (Bcl-2),Bcl-associated X protein (Bax) and Bcl-associated death promoter (BAD).In addition,Piezo1 inhibitor,GsMTx4,was used to block the MA cation channel Piezo1,served as the inhibitor group.AVPI was used to detect the apoptosis of the OA chondrocytes.Results The location of the Piezo1 was expressed in nucleus and cytoplasm of chondrocytes.The expression of the Piezo1,ERK5,BAD,and Bax mRNA in the OA chondrocytes is weak.The 12 h stretch force group was significant increased,and the 24 h stretch force group was the highest expression.However,the expression of the 48 h group was decreasing.The expression of the Bcl-2 in the 12 h group was decreasing,and the 12 h stretch force group was the lowest expression while the 24 h stretch force group was increasing.In the GsMTx4 group,the expression of the Piezo1,ERK5,BAD,Bax was decreasing while the Bcl-2 was increasing.In the BIX02188 group,the expression of the ERKS,BAD,Bax was increasing while the Bcl-2 was decreasing,while the expression of the Piezo1 was not change.The result of AV-PI shown that the 2 h stretch force group increased early stage of apoptosis.The 12 h stretch force group increased late stage of apoptosis,and the 24 h stretch force group's apoptotic rate was the highest.However,the apoptotic rate of the 48 h group was lower than the 24 h stretch force group.The GsMTx4 could inhibit the late stage of apoptosis.Conclusion Piezo 1 plays an important role in the apoptosis of human chondrocyte through the MAPK/ERK5 signal pathway.

Objective To investigate the effect of recombinant human endostatin (rhES) on the growth of transplanted gastric cancer in nude mices. Methods At the 20 day after tumor transplantation, twelve Balb/c-nu/nu mice were randomly divided into two groups:(1)Control group: 0.9% NS 0.1ml was injected around the tumor,once a day for 10 days; (2)experimental group: rhES 2mg/kg was inpected around the tumor once a day for 10 days. The tumors bulk were measured every five days until five days after injection. VEGF, bFGF,PDGF,VEGF-c, VEGFR-3, FⅧAg, PCNA, bcl-2 and apoptosis index were examined. The tumor inhibition rate(TIR) and tumor condensing rate (TCR) were calculated. The difference of AI , tumor bulk and MVD between the 2 groups were assessed. Results The difference of the tumor bulk between the two group was no statistical significance before injection, but at the time of 15 days after begining of the injection, the difference was significant(P =0.0001); and in the experimental group the tumor bulk was also decreased significantly after treatment(P=0.0015). MVD, VEGF, bFGF, PDGF, VEGF-c,VEGFR-3 and bcl-2 expressed lower in experimental group than those in control group; PCNA expression was the same in the 2 groups. AI increased significant in experimental group compared with in control group (P =0.0000). In experimental group,the TIR was 91.2%,TCR 53.5%,and HE staining presented more necrotic areas.Conclusions rhES can inhibit angiogenesis of the tumor and promote tumor cell apoptosis, so that can inhibit the growth of tumor.

[Objective] To investigate the therapeutic effect of Phyllanthus compound (PC, mainly composed of Herba Phyllanthus, Radix Astragali, Radix Notoginseng, Radix Glycyrrhizae, etc.) and lamivudine on chronic hepatitis B. [ Methods ] Sixty cases of chronic hepatitis B were randomized to two groups: group A (the combination of PC and lamivudine) and group B (lamivudine 100mg/d). After a six-month treatment, the therapeutic effect was evaluated. [Results] After treatment, symptoms and signs were improved in group A.
