1.Stability and reproducibility study in serum proteomics with matrix assisted Iaser desorption ionisation time-of-flight mass spectrometry

Objective To study the stability and reproducibility in serum proteome by matrix assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS).Methods Bruker Daltonics MALDI-TOF-MS was adopted under the linear mode, with mass ranging from 800 to 10 000 m/z.MB-WCX pre-extraction kit and Clinprotools software was used to analyze data.Results The reproducibility study was carried out under the standardized conditions.Data analysis showed that the m/z ratio was in the acceptable range, and the coefficients of variation(CV) was 12.7% ,13.1% and 18.8% ,respectively in the parallel experiment by taking 2 cancer and 1 healthy control serum samples and could identify the diseased samples.Then 30 healthy control samples were used to validate this result, all of which showed very good mass profile. The average CV was 25.5%, which could meet the requirements of clinical laboratory.Conclusion With the help of optimized experimental conditions and MALDI-TOF-MS, stable and reproducible mass spectrum can be required which is helpful for the early diagnosis of diseases.

3.Functional connectivity of EEG's delta-band in epilepsy interictal seizures based on Granger causality analysis

ObjectiveThe purpose of this research was to investigate functional connectivity of 16-channel electroencephalograph(EEG) in δ frequency band based on Granger causality analysis.MethodsThe experimental data was recorded at a sampling rate of 200 Hz from temporal lobe epilepsy (TLE) patients(6 left and 3 right TLE,and 9 normals as control group.Ten of EEG segments of 20 s length for three different states:epileptiform discharges (ED) state in interictal durations,non-ED state for TLE patients,and control state for the normal.The δ band components (1~4 Hz) were filtered from EEGs.The functional connection values Iδ between two EEG δ components were calculated separately by Granger causality analysis.The two EEG components were from inferoposterior temporal lobe (left:T5,right:T6) to frontal lobe (Fp1,Fp2,F3,F4,and parietal lobe (C3,C4) for three states.ResultsThe Iδ values for ED state was 0.1323±0.0329~0.1670±0.028 9,which was significantly higher than that of non-ED state (0.0300±0.0130~0.0420±0.0072) (P<0.05).The Iδ values for the control group (0.0153±0.0028~0.0193±0.0057) was much lower than that of ED state (P<0.01),and no obvious distinctions were observed compared with non-ED state at P=0.05 level.ConclusionThere is a stronger connection of EEG' s δ bands from the inferoposterior temporal lobe to frontal and parietal lobe for the ED state,and the over-discharges transmission is from inferoposterior temporal lobe to other brain regions.There is a weaker connection from the top temporal lobe to frontal and parietal lobe for non-ED state and control group,and the onset zones is not inferoposterior temporal lobe.
4.Distribution of Diatoms in Chuanyang River of Pudong New Area of Shanghai and Its Forensic Application

Objective To investigate the quantity and species distribution of diatoms in Chuanyang River of Pudong new area of Shanghai and provide references for the invesitigation of water body in forensic practice. Methods The water samples collected from 15 areas in Chuanyang River of Pudong new area in September 2012 were examined by microscope to identify the species of diatoms. Results Cyclotella and Pinnularia were found to be the dominant species within the 12 species of diatoms in Chuanyang River, which showed differences in species among the sections of Huangpu River, the center and the East China Sea. Conclusion The differences in subsectional distribution of diatom species in Chuanyang River may provide a new foundation for forensic identification in drowning cases especially in the deter-mination of falling location.
5.An analysis of the organic extracts from source water and drinking water in city C and the effects on DNA damage in primary hepatocytes in rats

Objective To investigate the effects of the polluted organic substances in urban water environment and genetoxicity of the organic pollutants in drinking water on DNA damage in primary cultured hepatocytes in rats. Methods Qualitative analysis of the organic extracts from source water and drinking water in five waterworks in city C with GC/MS was conducted and the effects of organic extracts on DNA damage in primary cultured hepatocytes in rats were tested with single cell gel electrophoresis assay. Comet length means and percentage of tail cell were observed. Results A total of 98 organic pollutants were detected in all samples including 60 in source water and 58 in drinking water. Objective:To explore the possible role of ? endorp hi n in effect of melatonin on morphine withdrawal symptoms in rats.Method:Acute model of morphine dependent rat was estalished by gradually increasing the dose of morphine through subcutaneous injections.CSF(cerebrospinal fluid)w a s got by push-pull perfusion and intracerebroventricular injection(icv)in a fixe d tuber.Concentration of ? endorphin in CSF was determined by radioimmunoassa y (RIA).Results:(1)The symptoms of natural morphine withdrawal were r e lieved significantly by icv ? Endorphin with a dose dependent manner.(2)The s y mptoms of natural morphine withdrawal were also relieved by ip melatonin.(3)The CSF ? endorphin level was significantly lower in morphine dependent rats than normal control,but that was higher in natural morphine withdrawal rats.That was also much more increased by ip melatonin in the withdrawal rats than in morphine dependent rats or normal control.Conclusion:The withdrawal symptom s could be relieved by icv ? endorphin or by ip melatonin in morphine dependetn rats.The mechanism may be partly due to the elevation of endogenous ? endorp hin level in brain of rats in withdrawal
7.Effect of ischemia on donor coronary arteries after heart transplantation

Objective To study the relationship between Cardiac allograft vasculopathy (CAV) and donor heart ischemia time. Methods The rat model of retroperitioneal heterotopic heart transplantation was used. Seventy Wistar rats were divided into 4 groups: group 1 receiving CsA after sham operation; group 2 receiving transplantation directly; group 3 having 3 h-ischemia before transplantation; group 4 having 6?h ischemia before transplantation. After transplantation, all rats in each group were subjected to the gastric lavage of CsA for 20?days. At 20?days after transplantation, the donor heart was removed for observation of changes in pathological lesions and ultrastructues of coronary artery. Results With ischemia time being prolonged, cardiac capillary endothelial cell swelling and coronary intimal proliferation were observed which were similar to the CAV histopathological changes. Conclusion The prolongation of ischemia time can lead to intimal proliferation and hyperplasia which can promote CAV development. So ischemia time is a vital reason for the CAV.
8.Doppler Blood Flow of Uterine Artery and Umbilical Artery in Healthy Pregnant Women

In this study the umbilical artery and uterine artery blood flow was measured by Acuson 128 color computer ultrasound in 90 healthy pregnant women, and the reference ranges and mathematical models of its indices were established. The variation of observers and measurement time and the effect of different areas of the same vessel on measured blood flow values were also studied. The results show reducing of resistance and increasing of blood flow of the umbilical and uterine arteries in healthy pregnancy with increased gestational age. Doppler blood flow measurement has a good repeatability and is worth while to be used universally. The Doppler signals shound be recorded from near placenta part of umbilical artery and upper branch of uterine artery in a side near to placenta.
9.Study progress of TCM syndrome of type 2 diabetes mellitus

We summarized the researches of syndrome differentiation for type 2 diabetes mellitus (T2DM) in this article. According to the constitution feature of obesity and non-obesity and analysis of researches of different medical scholars, we put forward that the easy-suffering constitutions were the basis of the evolution and development of syndromes of T2DM, and the different inducements were the conditions of that courses. The persons whose constitutions were of qi-deficiency of spleen and kidney and excessive stomach often appeared a obese physique, and the people whose constitutions were of yin-deficiency of both the liver and kidney usually showed a non-obese shape. Affected chronically by the inducements, the evolution and development of the syndromes had its own rules respectively corresponding to the different constitutions as the diseases go on. Therefore, to integrate the constitution, disease stage and pathological change with specific zang-fu organs can better grasp its rules of syndrome differentiation.
10.Effect of Activities of Daily Living Training on Hemiplegia after Stroke

Objective To explore the effect of activities of daily living (ADL) training on hemiplegia after stroke. Methods 50 stroke patients with hemiplegia were divided into treatment group (n=27), who received ADL training and physical therapy, and control group (n= 23), who only received physical therapy. They were evaluated with modified Barthel index (MBI) and the Brunnstrom classification. Results After 6 months of treatment, the score of MBI improved more in the treatment group than in the control group (P<0.05), but there was no significant difference in Brunnstrom stage (P>0.05). Conclusion ADL training can improve the recovery of ADL in hemiplegics after stroke.。
