BACKGROUND:Studies have confirmed that nicotine affects the activity of osteoblasts, osteoclasts, fibroblasts and erythrocytes.
OBJECTIVE:To study the nicotine effects on osseointegration and the expression of osteoprotegerin and bone morphogenetic protein 2 after implantation of dental implants with surface treatment by sandblasting or acid etching.
METHODS:Twenty-four Sprague-Dawley rats were randomly assigned to two groups and received daily injections for 2 weeks as folows: Nicotine 2 mg/kg twice for experimental group, saline solution for control group. Then the titanium implants with surface sandblasting or acid etching were implanted into the tibiae folowed by continuous nicotine or normal saline injection. At weeks 2 and 4 after implantation, the implants and surrounding bone tissue were prepared for CT, X-ray and hematoxylin-eosin staining examinations to evaluate bone healing and expression levels of bone-related genes were measured by quantitative RT-PCR.
RESULTS AND CONCLUSION: Compared with the control groups, the degree of osseointegration and the expression of osteoprotegerin and bone morphogenetic protein 2 in the experimental groups were decreased significantly (P < 0.05), except that the expression of bone morphogenetic protein 2 in the experimental group with acid etching was not significantly reduced. In addition, the expression of bone morphogenetic protein 2 in the experimental group with acid etching was higher than that in the experimental group with sandblasting at 2 weeks after implantation (P < 0.05). The X-ray and CT show that the quantities of new generation bone and the degree of bone mineralization of the sandblasting group were significant lower than those of the acid etching group under the intervention of nicotine. Hematoxylin-eosin staining showed that the activity and quantity of osteoblasts around the implants down-regulated significantly, but acid etching-treated implants showed better outcomes than sandblasting-treated implants.

Journal of Chinese Physician 2015;17(8):1131-1134
Lynch syndrome.(LS) is an autosomal dominant condition caused by a mutation in the mismatch repair genes (MMR).Endometrial cancer (EC) is the most common extra-colonic cancers of LS Ⅱ type.Among the female members of LS family with the MMR gene mutation,EC has an overall lifetime risk more than 35%.LS-associated EC is more likely present as the clinical pathological characteristics of early age of onset, endometrioid adenocarcinoma, lower grade, lower uterine segment involvement, and better prognosis.LS patients with family history and early age of onset should be performed with a combined screening and genetic testing, while high-risk patients should be taken clinical monitoring methods as endometrial biopsy.Oral contraceptives mav be reasonable chemopreventive agents in the patients with LS.Although prophylactic hysterectomy with bilateral salpingo-oophorectomy is an effective strategy to prevent EC in women with LS,preoperative counseling should address the trade-offs between the reduction in the risk of cancer, and the risks and side effects of surgery. The patients need hormone replacement therapy after surgery.

AIM: To discuss the influence of high glucose on the proliferation and ICAM-1 expression of human umbilical vein endothelial cells(HUVEC) within 48 h and to observe the altered effect of high glucose when high glucose and inflammatory stimulator exist at the same time.METHODS: HUVEC were cultured with different concentrations of glucose. The proliferation of HUVEC induced by high glucose was detected by MTT assay. The ICAM-1expression of HUVEC induced by high glucose was assayed by flow cytometry and cell ELISA.The difference of ICAM-1 expression between induced by LPS and by LPS together with high glucose was compared.RESULTS: 25、(45 mmol?L~(-1)) glucose did not inhibit the proliferation of HUVEC within 48 h,and they also had no effect on the expression of ICAM-1.However,when stimulated HUVEC with LPS, the proliferation of HUVEC was extremely inhibited in high glucose groups, and the expression of ICAM-1 increased significantly in those groups.CONCLUSION: In short time, high glucose has no effect on the proliferation and ICAM-1 expression of HUVEC,but when inflammatory stimulation exists, high glucose can inhibit the proliferation and increase the expression of adhesion molecules.

0.70,cluster in two pairs of medicines are similar. Conclusion:It's reliable to cluster analysis the TCM by regarding the element contents as the character of these medicines. The larger the correlation coefficient is,the more similar the nature, flavor and efficacy of the medicines are, which suggested that trace element contents were closely related to(have much to do with) the flavor, nature and efficacy. The study provides method for the cluster relationship among TCM.

Background N-ethyl-N-nitrosourea (ENU)-induced mouse mutagenesis is a powerful approach for the study of gene function and the generation of human disease models. Objective This study was to create the corneal morphologic change and map the mutant gene of a kind of corneal opacity in ENU mutagenesis in mouse. Methods ENU was intraperitoneally injected in forty C57BL/ 6J (B6) male mice aged 8-10 weeks old.The male mice were mated with the same strain female mice.Their progenies were screened for visible eye mutation, and the mutant mice were mated with the same strain mice to confirm the heredity of mutation phenotypes.Hematoxylin & eosin staining was used to examine the histopathological change of cornea in one mouse with ENU-induced corneal opacity. To map the mutant gene,[(B6×D2)F1 ×B6] N2 mutant mice were bred, and the genome of the N2 mice was scanned by microsatellite markers distributed equally on the mouse chromosome. The microsatellite linked to the mutant gene was determined by the log odds score. This experimental procedure was approved by Ethic Committee about Experimental Animal Care and Use of Yangzhou University. Results The founder mouse, which was the progeny of an ENU-treated B6 male mouse and an untreated B6 female mouse, had a corneal opacity phenotype. After mating the mutant with B6 mice,19 of 59 descendants appeared corneal opacity phenotype.Thickening of corneal stroma, neoangiogenesis, infiltration of inflammatory cells and proliferation of fibroblasts were exhibited in cloudy cornea in ENU-induced mutated mice under the optical microscope. After linkage analysis between microsatellite markers and the mutant gene, the mutant gene was linked to D2Mi307,which was located at 63.42 cM.Three cases of 26 N2 mice underwent recombination with the LOD 3.79.The mutant gene associated with the cornea phenotype was located on chromosome 2.Conclusions This study map the mutant gene associated with the cornea phenotype on chromosome 2.The strain might be used as a mouse model for heritable human corneal opacity.
