1. Role of MAPK signaling on human mast cell activation induced by Trichomonas vaginalis

Trichomonas vaginalis is a flagellated protozoan parasite that causes vaginitis and cervicitis in women and asymptomatic urethritis and prostatitis in men. Mast cells have been reported to be predominant in the vaginal smears and vaginal walls of patients infected with T. vaginalis. Mitogen activated protein kinase (MAPK) activated by various stimuli also regulate the transcriptional activity of various cytokine genes in the mast cells. Because of their essential role in intracellular signaling network, also appropriate targets for pharmacological treatment of inflammatory disorders.Cultivation of T.vaginalis and HMC-1 line, preparation of TvSP, to check intracellular ROS level and degranulation by FACS, to determine phosphorylation of MAPK and p47phox by immunobloting.In this study, we investigated whether MAPK were involved ROS generation and exocytotic degranulation in HMC-1 induced by T. vaginalis-derived secretory products (TvSP). We first examined that TvSP could induce activation of MAPK and NADPH oxidase in HMC-1 cells. Stimulation with TvSP induced phosphorylation of MAPK and p47phox in HMC-1 cells. Phosphorylation of p47phox is main source of ROS generation. Next, to determine involvement activation of MAPK in ROS generation and degranulation in HMC-1 cells induced by TvSP. ROS generation is required for exocytotic degranulation of mast cells induced by TvSP. Stimulation with TvSP induced phosphorylation of p47phox, ROSgeneration, and surface up-regulation of CD63 in human mast cells. CD63 is a marker for exocytosis. Pretreatment with MAPK inhibitors strongly inhibited TvSP-induced ROS generation and exocytotic degranulation.Our results suggest that TvSP could induce intracellular ROS generation and exocytotic degranulation in HMC-1 via MAPK signaling pathway.
2.The detection of trichomonas vaginalis by cytological stained tests

Background: One of the confronted problems of doctors and medical personnels is sexually transmitted disease has not been decreased in our country up to now. By last 5 years propagation of trichomoniasis was 16.7-9.5% per 10000 population and infected T.vaginalis. In the practice of parasitology of our country metilen is revealed by gram method hasn’t been introduced which is used in up to date histological analyse. This became the background of our research work. Aim of our research is to diagnose trichomoniasis which infects sexually by cytological analyse and to define its specific and sensibility.Materials and Methods: A total 99 smears of 33 females aged 19-39, used cross sectional descriptive method. Finding T.vaginalis on specimens (1) Vaginal wet mount, (2) Gram staining and (3) Pap stain. Result: In our research T.vaginalis leaked out in wet mount smear was 33%, in Gram stain was (21.2%). The sensibility quality of T.vaginalis on Gram stain is 63%, specific quality is 90%, value of kappa coefficient (К=0.58 Р<0.002). In Pap stain T.vaginalis diagnosed 27,2% and sensibility quality of T.vaginalis is 81%, specific quality is 100%, value of kappa coefficient (К=0.87 Р<0.005). In the case when 3 analysis were positive case was 5 or 15.2%, in the case where 2 analysis were positive case was 6 or 18.2%. Conclusions: The Pap stain sensibility quality of T.vaginalis is 81%, specific quality is 100%, value of kappa coefficient (К=0.87 Р<0.005), that shows should be to give effect on diagnosing of sexually transmitted infectious disease.

BACKGROUND: Microtia is often associated with hearing loss and patients typically
require treatment for hearing impairment and surgical ear reconstruction. The
occurrence of microtia is of public health importance in part due to the psychosocial
sequelae, including the stigma associated with malformations of the ear and the burden
of undergoing multiple surgeries
In addition, greater than 90% of individuals with microtia experience conductive hearing
loss on the affected side. Although children with microtia-anotia are at a greater risk of
delayed language development and attention deficit disorders.
METHODS: Our study has a 173 childrens from the UB city and countryside. 23 children
has a bilateral microtia and anotia. 6 patient wearing BAHA soft band from 6 months
age. 150 patients have an unilateral microtia. Microtia was found more commonly in
males, unilateral and right sided. Even children with bilateral microtia may have wearing
BAHA soft band before 6 months age.
RESULTS: External ear malformations are more commonly found in males. Sex ratio 2:1.
From other studies right ear malformations are more common in male children. In our
study children who have bilateral ear abnormalities wearing BAHA softband and
language therapy from 6 months age to able to have normal language development.
Children who had surgical treatment and using hearing aid can have normal social life
and increased quality of life.
CONCLUSION: Early screening in children who have external ear malformation,
monitoring language development, wearing BAHA softband from 2 months age who
have bilateral ear malformation and one ear affected other one is normal children
wear BAHA softband increases children speech and language development.
4.Effects of flavanone on cancer cells viability

Background:
In recent years, scientists have found that certain natural compounds have significant potential in cancer
prevention and early-stage cancer treatment. Flavanones, a class of polyphenolic compounds found in plants, vegetables,
seeds, fruit peels, and flowers, have been identified to possess anticancer, antioxidant, anti- inflammatory, and antibac
terial bioactivities. Cancer has become a major global challenge in terms of both economic and public health concerns.
Global statistics indicate that 22.8% of deaths are attributed to non-communicable diseases, and 16.8% are caused by
cancer, accounting for one in four and one in six deaths, respectively.
Aim :
To investigate anticancer effects of Iris Tenuifolia-derived flavanone on cancer cell lines.
Materials and Methods :
The study was conducted at the Bio-Medical Research Institute of the Mongolian National Uni
versity of Medical Sciences, investigating the effect of flavanones on cancer cell viability under in vitro conditions using
the MTT assay. In the study, colon, liver, and lung cancer cells were cultured, stabilized, and used for the experiments.
Colorectal cancer cells (MC38), liver cancer cells (HepG2), and lung cancer cells (A549) were revived, cultured, and
stabilized for use in the experimental procedures. Statistical analysis of the results was performed using Microsoft Excel
2010, and graphs were generated using GraphPad Prism 8. Differences between groups were analyzed using Student’s
t-test, and a p-value of <0.05 was considered statistically significant.
Results :
We treated MC38, HepG2, and A549 cancer cells with different concentrations of flavanone (2.5 µM, 5 µM, and
10 µM) for 24 to 48 hours to evaluate cell viability. Flavanone inhibited A549 cell viability by 2.5 μM-10%, 5 μM-25%,
and 10 μM-38%, respectively. For HepG2 cells, flavanone treatment at concentrations of 5-10 µM reduced cell viability
by 28–58%. No statistically significant effect on the viability of MC38 cells was observed following treatment with fla
vanone at concentrations ranging from 2.5 to 10 µM. Additionally, although MC38 inhibited cell viability in a dose-de
pendent manner in cell cultures, it had a statistically significant effect at higher concentrations of 30-200 μM (p<0.01).
Conclusion
Flavanone inhibits the cancer cell viability in a dose and time dependent manner
