1.Analysis on Professor Yan Dexin's TCM Theory of Spleen and Stomach

Objective]To investigate academic thoughts of Prof. Yan Dexin in spleen and stomach syndrome treatment.[Methods]The paper summarized the academic origin, academic thoughts as well as practical treatment cases of Prof. Yan Dexin ’s spleen and stomach syndrome treatment theory. [Results]The main points of Prof. Yan’s academic thought on spleen and stomach therapy has succeeded and developed the therapy of Menghe physician, which includes an emphasis on spleen Qi rise and stomach Yin drop, Qi of spleen and stomach regulation, special attention on the function of the stomach-yang and spleen-yin, an advocation ofspleen leading four organs, and therapy design from spleen with an ingenious use of atractylodes rhizome, Atractylodes and Zhizhu pill. [Conclusion] Professor Yan Dexin developed his unique theoretical system in spleen-stomach diseases therapy on basis of the experiences of spleen and stomach disease treatment from ancient physicians, which is instructive for later physicians.
2.Progressive resistance exercise for improving lower limb motor function in hemiplegic stroke patients

Objective To investigate the efficacy of progressive resistance exercise for improving the lower limb motor function of hemiplegic stroke patients. Methods Eighty-nine hemiplegic patients were randomly divided into a treatment group (n =46) and a control group ( n = 43 ) . Both groups received routine rehabilitation training, while the treatment group also undertook progressive resistance training. The control group received extra quadriceps femoris training. Before and after the treatment, motor function was evaluated with the Fugl-Meyer lower limb assessment, functional ambulation category (FAC) classification and the modified Barthel index (MBI). Results Before intervention, there were no significant differences between the two groups in any of the assessments. Four weeks later, all the assessment scores obtained with the Fugl-Meyer lower limb assessment, FCA classification and MBI in the treatment group were significantly better than those in the control group. Conclusion Progressive resistance training combined with routine rehabilitation treatment can improve motor function in the lower limbs, ambulation and ability in the activities of daily living of hemiplegic patients after stroke.
3.Umbilical cord blood mesenchymal stem cells traced and transfected by recombinant lentivirus vector with enhanced green fluorescent protein for treatment of ischemic necrosis of the femoral head in rabbits*

BACKGROUND: At present, studies concerning human umbilical cord blood mesenchymal stem cells (UCB-MSCs) transplantation for repair of rat spinal cord injury, brain tumor, and myocardial infarction have been reported, and studies that human UCB-MSCs were induced differentiation into osteogenic cells under certain conditions have also been reported at home and abroad. But application of UCB-MSCs transplantation in the treatment of osteonecrosis of animals has not yet been reported. OBJECTIVE: To observe the repair results of recombinant lentivirus vector tracing enhanced green fluorescent protein (EGFP)-transfected UCB-MSCs transplantation in treatment of ischemic necrosis of the femoral head in rabbits. METHODS: Bone morphogenetic protein-2 gene plasmid, recombinant lentivirus vector carrying EGFP and UCB-MSCs were co-cultured. Rabbit models of femoral head defects were made and randomly divided into 3 groups. There was no treatment in the normal group, control group with bone defects and experimental bone defects filled with UCB-MSCs tracing transfected by recombinant lentivirus vector carrying EGFP. At 4 and 8 weeks after treatment, the imaging and histological of the femoral head were observed.RESULTS AND CONCLUSION: Imaging and histology results showed that there were osteogenic response and new bone formation in the experimental group at 4 weeks, and the bone defects were basically repaired at 8 weeks after treatment. In the control group, the bone defects filled with fibrous connective tissue fiber connective tissues at 4 weeks, and the osteosclerosis could be found surrounding femoral head, bone defects filled with fibrous connective tissue fibers and bone trabecula distributed disorderly. The recombinant lentivirus vector tracing EGFP-transfected into UCB-MSCs has strong effects bone conduction and can repair ischemic necrosis of the femoral head.
4.Effects on proliferation and apoptosis of T-cell leukemia cells by silencing NRP-1

Objective:To investigate the effect on proliferation and apoptosis of T-cell leukemia cells by silencing NRP-1 ( Jurkat cells).Methods:The lentivirus plasmid which expresses NRP1 gene specific shRNA was constructed in our preliminary ex-perimental.We transfected the lentivirus plasmid to human T-cell Lymphoma cells.The proliferation of Jurkat cells different groups and effect on cell proliferation after chemotherapy drug EPI-treated were found by CCK-8 kit.The proliferation level and apoptosis rate of the cells were detected by flow cytometry and Annexin-V-FITC/PI method.Results:The proliferation level of NRP-1 /shRNA interference group was decreased significantly in 48 h,72 h,96 h,which was compared with the control groups.The apoptosis rate of the NRP-1/shRNA interference group was increased compared with control groups.The chemotherapy drug sensitivity of epirubicin ( EPI ) test results showed that EPI concentration was 0.025,0.05,0.1,0.2,0.4 μg/ml,the NRP-1/shRNA interference group of cell growth inhibition rate was increased,the corresponding control group difference had statistical significance(P<0.05).We choose the drug con-centration of the EPI IC50 for next experiments.NRP-1/shRNA interference group cell apoptosis rate increased significantly after induction,compared with the control groups difference was statistically significant ( P<0.05 ).Compared with control group, the expression level of Bcl-2 protein was decreased and the expression level of bax protein was increased significantly after EPI induction.The percentage of cells at G0/G1 phase increased significantly,while those at S phase decreased significantly.Conclusion:Plasmid shRNA-NRP1 inhibited the expression of NRP1 in Jurkat cells and decreased the proliferation level of Jurkat cells and promote their apoptosis and enhance their drug sensitivity;the molecular mechanism may relate to down-regulation of Bcl-2 and up-regulation of Bax.and arrested the cell cycle at G0/G1 phase.
5.Induced differentiation of human embryonic stem cells into endothelial cells

BACKGROUND:Human embryonic stem cels exhibit self-renewal and multi-differentiation potential, and can differentiate into endothelial cels under certaininduction conditions.
OBJECTIVE:To explore induced conditions of the human embryonic stem cels differentiating into endothelial cels and to investigate the effect of vascular endothelial growth factors on theendothelial differentiation of human embryonic stem cels.
METHODS:After resuscitation,passage40 human embryonic stem cel lines H9 weresubjected to suspension culture to prepare embryos, and after 5-day culture,these cels werecultured in attachment medium to differentiate into embryoid bodies,folowed by induction with50 μg/L vascular endothelial growth factors. Passage 2 and 15 embryonic stem cels after induced differentiation weretaken for Dil-Ac-LDL uptake test and immunohistochemical staining, respectively.
RESULTS AND CONCLUSION:After 1-day culture, cord-like or polygonal monolayer cels around embryoid bodies showed bud-like andradialgrowth witharelative rapid speed merging into surrounding colonies; at 2-3 days, the number of suspension cels increased further, but the smal-round cels in the center began to die; at 5 days, embryoid bodies started to passage, and aggregated cels exhibited typical paving stone-like appearance. Moreover, some human embryonic cels after induced differentiation could actively takeupfluorescent labeled LDL,andred fluorescent particlesappeared.Additionaly, passage 15 embryonic stem cels after induced differentiation could express CD31 and FLK-1.These findings suggest that human embryonic stem cels induced by vascular endothelial growth factors can differentiate into endothelial cels.
6.Evidence-based Guidelines on Medication Therapy for Purulent Meningitis in Children:A Systematic Review

OBJECTIVE:To systematically review the related evidence-based guidelines of purulent meningitis in children,and to provide evidence-based reference for clinical treatment. METHODS:Retrieved from PubMed,EMBase,CBM,Wanfang Data-base,CJFD and VIP,NGC,GIN,TRIP and websites of domestic and international medical associations and industry bodies,the treatment guidelines about purulent meningitis in children were collected. Evidence-based evaluation was performed after data extrac-tion and quality evaluation. RESULTS:Finally 3 guidelines were enrolled in total,with development time ranging from 2004 to 2012,from USA,Britain and Australia,respectively. All of the recommendations were level B,scope and purpose and clarityshowed the higher scores in AGREEⅡ,more than 70%,and applicability showed generally low scores. Penicillin and cefotaxi-me were recommended in purulent meningitis of neonates,and vancomycin combined with cefotaxime or ceftriaxone were recom-mended for infants and children(it was combined with vancomycin when Streptococcus pneumoniae infection was suspected). The dose and duration of each guideline were certain different,and the period of treatment should be longer in neonates. In addition, the glucocorticoid was recommended in all guidelines. CONCLUSIONS:The recommendations of medicines for the treatment of pu-rulent meningitis are basically unanimous,with no regional difference,but there are some differences about dose and the course of treatment. In addition,the classification criteria of the levels of evidence and recommendation are still suboptimal,which needs fur-ther improvement. And guidelines on purulent meningitis should be improved inrigourandapplicabilityin future.
7.Specific labeling method in improving mixed placing of medical waste in neonatal intensive care unit

Objective To explore the effect of specific labeling method in improving the mixed placing of medical waste in neonatal intensive care unit(NICU).Methods Medical waste classification of 34 trash cans in the NICU of a hospital between July and December 2014 were investigated,July-September was pre-implementation phase of spe-cific labeling,October-December was post-implementation phase,mixed placing of medical waste between pre-and post-implementation phase was compared.Results A total of 504 cases of medical waste classification in NICU were investigated,252 cases respectively in pre-and post-implementation phase,74 cases of mixed placing were found. Mixed placing rates before implementing specific labeling was higher than after implementing (25.40%[64/252]vs 3.97%[10/252],χ2 =46.187,P <0.001 );before implementing specific labeling,57 cases of infectious waste and non-infectious waste were mixed placing,after implementing specific labeling,only 8 cases of infectious waste and non-infectious waste were mixed placing.Mixed placing were mainly performed by trainees for in-service training and interns,accounting for 39.06% before implementing and 50.00% after implementing.Conclusion The specific labeling for standardizing and managing of medical waste can improve the classification of medical waste in NICU, significantly improve the compliance of all kinds of health care workers to the standard handling of medical waste.
8.Therapeutic effect of progesterone combined with psychological intervention on threatened abortion

Objective To investigate the effect of progesterone combined with psychological intervention on threatened abortion. Methods According to the different treatment and nursing intervention mode selected from be January 2015 to January 2017 in our hospital 60 cases of patients with threatened abortion group: the control group used dydrogesterone + routine nursing intervention, the observation group with psychological nursing mode + progesterone intervention; the two groups of patients with clinical symptoms, the incidence of adverse reactions and nursing satisfaction were recorded in detail, and the relevant data for comparative analysis. Results The psychological nursing mode of progesterone +(observation group) intervention is better than the clinical effect of patients with threatened abortion and dydrogestrone + routine nursing (control group) the clinical intervention effect, clinical symptoms improved better than the control group, the incidence rate of adverse reaction was lower than the control group, nursing satisfaction was higher than the control group, the difference was statistically significant (P<0.05). Conclusion Threatened abortion patients choose progesterone + psychological intervention effect significantly, can effectively improve the clinical symptoms and adverse reactions and nursing satisfaction, is widely used in clinical intervention of threatened abortion patients.
9.Clinical study on the therapeutic effect of pullerolol, magnesium sulfate and psychological intervention on hypertensive rats during pregnancy

Objective To investigate the effects of labetalol, Magnesium Sulfate, psychological intervention combined with the clinical effect of treatment of gestational hypertension. Methods According to the different treatment and nursing intervention mode of 120 cases of hypertension patients in January 2015 to 2017 year in January in our hospital for diagnosis and treatment of pregnancy groups: control group with routine nursing intervention in the observation group, Magnesium Sulfate +, Magnesium Sulfate + + mode labetalol psychological nursing intervention; the detailed records of the two groups of patients with clinical curative effect, complications, etc. the situation, and the related data for comparative analysis of labetalol, Magnesium Sulfate, psychological intervention combined with the clinical effect of treatment of gestational hypertension. Results The psychological nursing mode of labetalol + Magnesium Sulfate (observation group) intervention effect in patients with hypertension during pregnancy is better than the Magnesium Sulfate + routine nursing (control group) the clinical intervention effect, clinical symptoms improved better than the control group, the complication rate was lower than the control group, nursing satisfaction was higher than the control group, the difference was statistically significant (P<0.05). Conclusion Patients with gestational hypertension selection effect of labetalol + Magnesium Sulfate +psychological nursing intervention significantly, can effectively improve the clinical symptoms and reduce the incidence of complications.
10.Protective mechanism of rebamipide on non-steroid anti-inflammatory drug related small intestinal mucosal injury

Objective:To explore the protective effect and related mechanism of rebamipide on non-steroid anti-inflammatory drug (NSAID) related small intestinal mucosal injury.Methods:A total of 21 C57BL/6 mice were selected and by random number table method, they were divided into negative control group (0.9% NaCl gavage for four days), indomethacin modeling group (20 mg/kg indomethacin gavage for four days) and rebamipide intervention group (20 mg/kg indomethacin gavage for four hours and then 320 mg·kg -1·d -1 rebamipide gavage for four days), seven mice in each group. After modeling, the injury of mice intestinal mucosa of indomethacin modeling group and rebamipide intervention group was evaluated by gross observation as well as pathological analysis. The serum levels of interleukin (IL)-6, IL-10, trefoil factor 3 (TFF3), prostaglandin E2 (PGE2) and epidermal growth factor (EGF) in mice were detected by enzyme-linked immunosorbent assay (ELISA). The expression of IL-6, IL-10, TFF3, cyclooxygenase 2( COX2) and EGF at mRNA level of mice small intestinal tissues were examined by real-time quantitative polymerase chain reaction (qRT-PCR). And the relative expression of TFF3, COX2 and EGF at protein level of mice small intestinal tissues were determined by Western blotting. Levene test and independent sample t test were used for statistical analysis. Results:The scores of gross observation and histopathology of mice small intestinal mucosa injury of rebamipide intervention group were both lower than those of indomethacin modeling group (2.80±0.45 vs. 4.60±1.14, 1.67±0.52 vs. 3.00±0.71), and the differences were statistically significant ( t=2.667 and 3.618, P=0.029 and 0.006). The mouse serum level of IL-6 and the expression of IL-6 at mRNA level in intestinal tissues of indomethacin modeling group were both higher than those of the negative control group, however the serum level of IL-10 was lower than that of the negative control group ((48.83±5.40) ng/L vs. (40.96±5.92) ng/L, 5.23±2.36 vs. 1.12±0.56, (168.50±10.57) ng/L vs. (186.30±7.77) ng/L), and the differences were statistically significant ( t=2.307, 3.372 and 3.366; P=0.047, 0.007 and 0.012). The expression of IL-6 at mRNA level in mice small intestinal tissues of rebamipide intervention group was lower than that of indomethacin modeling group (1.74±0.82 vs. 5.23±2.36), however, the expression of IL-10 at mRNA level was higher than that of indomethacin modeling group (6.44±3.46 vs. 1.22±0.83), and the differences were statistically significant ( t=3.409 and 3.025, P=0.008 and 0.014). The serum levels of TFF3, PGE2 and EGF, the expression of TFF3 at mRNA level of small intestinal tissues, the relative expression of COX2 and EGF at protein level of small intestinal tissues of indomethacin modeling group were all lower than those of the negative control group ((131.20±16.37) ng/L vs. (150.30±9.66) ng/L, (32.68±6.88) ng/L vs. (41.51±3.20) ng/L, (112.70±17.17) ng/L vs. (138.20±10.10) ng/L, 0.43±0.22 vs. 1.20±0.50, 0.33±0.25 vs. 1.30±0.43, 0.28±0.19 vs. 1.15±0.10), and the differences were statistically significant ( t=2.290, 2.645, 2.867, 3.097, 3.405 and 7.106; P=0.048, 0.021, 0.025, 0.017, 0.027 and 0.002). The mice serum levels of PGE2 and EGF, expression of TFF3, COX2 and EGF at mRNA level of small intestinal tissues, as well as the expression of TFF3 and EGF at protein level of small intestinal tissues of rebamipide intervention group were all higher than those of indomethacin modeling group ((43.55±5.28) ng/L vs. (32.68±6.88) ng/L, (153.30±15.66) ng/L vs. (112.70±17.17) ng/L, 2.48±1.70 vs. 0.43±0.22, 2.95±1.56 vs. 0.88±0.45, 3.97±2.54 vs. 0.98±0.76, 1.47±0.26 vs. 0.72±0.35, 1.08±0.36 vs. 0.28±0.19), and the differences were statistically significant ( t= 2.711, 3.658, 2.656, 2.856, 2.524, 3.013 and 3.435; P=0.024, 0.008, 0.026, 0.019, 0.033, 0.039 and 0.026). Conclusions:Rebamipide alleviates small intestinal mucosal injury induced by indomethacin by inhibiting the expression of inflammatory factors and promoting the expression of intestinal mucosal protective factors suggesting that rebamipide plays a protective role in NSAID related small intestinal injury by maintaining the chemical barrier of the intestinal mucosal.
