China Pharmacy 2015;(34):4818-4820
OBJECTIVE:To establish a simple and feasible method for rapid analysis of drug consumption patterns, and to im-prove the level of hospital pharmacy management. METHODS:Taking Motilium tablet as an example, the actual order amount da-ta of 24 months were analyzed by time series seasonal decomposition method;several key components of change trend were extract-ed by SPSS software,i.g. smoothed trend-cycle(STC),seasonal factors(SAF)and seasonally adjusted series(SAS). The varia-tion of components data series was observed to obtain the conclusion, and then compared with direct-vision method. RESULTS:The seasonal decomposition method had high prediction accuracy, the mean absolute percent error (MARE)(0.17) was less than that of error value by direct-vision method (0.24) significantly. CONCLUSIONS:This method can found drug consumption pat-terns quickly and accurately with the help of software. Based on this aspect, stock adjustment strategy can combine daily manage-ment and long-term dynamic management conveniently so as to improve the level of drug management.

3. AKT2 reverses chemoresistance of ovarian cancer to cisplatin by regulating p27
Tumor 2008;28(6):459-462
Objective: To explore the role of v-akt murine thymoma viral oncogene homolog 2 (AKT2) and p27 in the chemoresistance of multicellular spheroids (MCS) of ovarian cancer to cisplatin. Methods: MCS of A2780 were formed by three-dimensional cell culture. Western blotting was applied to detect the expressions of AKT2 and p27. Then the interfering plasmid of AKT2 was constructed and transfected into the MCS of A2780 cells. The cell apoptosis after treatment with different concentrations of cisplatin was detected by flow cytometry (FCM) and the sensitivity of cells to cisplatin was evaluated by MTT assay. Results: Western blotting indicated that the expressions of AKT2 and p27 were higher in MCS than that in monolayer cells. The expressions of AKT2 and p27 in AKT2 shRNA-transfected MCS were significantly lower than that in MCS without transfection or transfected with empty vector. FACS analysis indicated that after treatment with various concentrations of cisplatin the apoptotic ratio of MCS was significantly lower than that in monolayer cells. AK72 shRNA transfection significantly increased the apoptotic ratio of MCS compared with those without transfection or transfected with empty vector. MTT assay showed that the sensitivity of MCS to cisplatin was increased after transfection with AK'12 shRNA. Conclusion: Interference with AKT2 shRNA down-regulated p27 and consequently reversed the drug resistance of ovarian cancer to cisplatin.
4. Expressions of calcineurin A and B in breast carcinoma tissues
Tumor 2008;28(2):146-149
Objective: To investigate the expression of calcineurin (CaN) in breast carcinoma and to analyze the relationship between the expression of the member of CaN family and clinicopathological features. Methods: The expressions of CaN Aα, CaN Aβ and CaN B in 55 cases of breast carcinoma tissues and adjacent normal tissues were measured by Western blotting. The intracellular distribution of CaN proteins were detected by immunohistochemical method. Results: The expression levels of CaN Aα and CaN Aβ and CaN B proteins were markedly increased in breast carcinoma tissues compared with adjacent normal tissues. The relative intensities were 6.45-,4.67-, and 2.31-fold of the control group, respectively (P < 0.05). The expression levels of CaN Aα and CaN Aβ were related with TNM staging (III/IV) of breast carcinoma (P = 0.05). Conclusion: The continuous activation of CaN signaling pathway maybe play an important role in tumorigenesis of breast carcinoma.

Objective To analyze diagnosis and management of traumatic atlantal fractures complicated with disruption of the transverse atlantal ligament. Methods Twenty-six patients with traumatic atlantal fractures complicated with disruption of the transverse atlantal ligament, 20 acute ones and six old ones, were managed in our department from March 1995 to March 2005. All the patients had the symptom of neck pain and a certain extent of neurological deficits. The radiographic examination showed that there were fractures in the anterior arch or/and lateral mass of the atlas and that the atlanto-dental interval (ADI) in these patients was 4 to 7 mm. Twenty-one cases were operated on with occipitocervical or atlantoaxial fusion immediately after injury. Five cases received conservative treatment. Results All were followed up for 15 (from 6 to 24) months on average. The 21 cases treated operatively reported relieved symptoms. Four of the five conservative treatments succeeded, but one of them underwent a successful atlantoaxial fusion because the original conservative treatment had failed. Conclusions The severity of clinical symptoms mostly depends on the degrees of atlantal displacement and cord compression. Diagnosis should be made on the basis of the ADI change as well as clinical presentations. For patients who have potential risk of atlantoaxial instability, occipitocervical or atlantoaxial, usion should be performed early.
