1.Study on radiotherapy plus neoadjuvant chemotherapy for patients with upper esophageal carcinoma

Objective:To evaluate the effect of radiotherapy combined with neoadjuvant chemotherapy on patients with upper esophageal carcinoma.Methods:Forty-five patients with upper esophageal carcinoma were divided into two groups randomly. There were 23 patients in neoadjuvant chemotherapy plus radiotherapy group(CR),and 22 patients in radiotherapy group(R).The regimen of neoadjuvant chemotherapy including cisplatin and 5-fluorouracil was administered.Two to three weeks after two cycles of chemotherapy,the patients were irradiated.Carmofur in CR group was administered in the period of late-course radiotherapy for four weeks,and the methods of radiotherapy were identical in two groups.Results:The 1,2,3-year survival rates were 73.91%,56.52% and 47.83% for CR group and 59.09%,31.82% and 18.18% for R group,and survival rates differed significantly(?2=4.8112、P =0.0283)in two groups;however,more serious side effects were in CR group,but they could be tolerated by patients.Conclusions:Neoadjuvant chemotherapy with PF regimen plus radiotherapy may improve the outcome of upper esophageal carcinoma.
2.Clinical anslysis for 38 cases with ovarian hyperstimulation syndrome

Objective To Search for the induced factors and therapeutic strategies for ovarian hyperstimulation syndrome(OHSS).Methods 38 cases with OHSS were retrospectively analyzed.Results All cases had symptoms of stomach and intestine,hcmachrosis and ascitic fluid.Human serum albumin through intravenous dripping was one of the most successful treatment as plasma expanders through restoration of effective blood volume.Paracentesis was necessary for critical OHSS patients with severe ascites and hydrothorax.Herus could decrease the cost of albumin significantly.Conclusions OHSS was the severe complication of ovarian stimulation.All patients with OHSS got completely recovered through intense care and effective treatment.
3.Research progress of TLR9 inherent signaling pathways in tumor cells

Specific activation of toll-Like Receptor 9 (TLR9) by ligand CpG DNA/ODN can initiate downstream signaling.The activation of TLR9 signaling is regulated by Granulin,high-mobility group box-1 (HMGB1) and soluble TLR9.Interestingly,TLR9 is no longer only presented in the immune cells,but also expressed in many kinds of tumor cells.TLR9 is involved inmaintaining and promoting the malignant biological behaviors of tumor cells.Elucidation of the function of TLR9 inherent signaling pathways in the tumor cells will provide a novel idea for oncotherapy.
4.Processing of Semen raphani based on glucosinolate content

AIM:To optimize the processing parameters of Semen raphani by uniform design. METHODS:In order to compare the change of Semen raphani in quality before and after simple stir-frying,allyl glucosinolate was selected as the internal standard. The relative content of glucosinolate was determined by HPLC and the processing parameters were optimized. RESULTS:The glucosinolate content gradually decreased as the processing time increased,maxima in HPLC peak area appeared at 200-250 ℃ and in the range of 1-1. 5 min and the content of glucosinolate in the processed Semen rahani was 4. 2 times as great as that in the non-stir-frying. CONCLUSION:It shows that the optimized processing of Semen raphami is beneficial for the conservation of glucosinolates.
6.Visual function of the idiopathic macular hole

The idiopathic macular hole ( lMH ) is research priority associated with the regenerate quickly of vitrectomy. The unaided visual acuity and the best corrected visual acuity is partial for the visual acuity of the patient with lMH. The mechanism and clinical significance of modern visual function measurements associated with lMH, including contrast sensitivity, visual field, multifocal electroretinogram, and stereoscopic vision, have been introduced. These measurements could be of great value in early diagnosis of lMH, assessment of surgical indication and evaluation of visual performance after vitrectomy. They would also be helpful to the analysis of postoperative impaired visual function and its management. Having an adequate understanding of the contents and significance of visual function is helpful to the improvement of lMH surgery techniques and postoperative visual acuity.
7.Pharmacokinetics and relative bioavailability of gemfibrozil capsules in 12 healthy volunteers

Aim The pharmacokinetic parameters and relative bioavailability of capsule A(Maiwei Pharmaceutical Co Ltd, Beijing, China) and capsule B (Xianjing Pharmaceu-tical, Hunan, China) were studied. Methods A single oral dose of 600 mg gemfibrozilof these two kinds of capsules was given to 12 chinese healthy male volunteers in anopen, randomized crossover study. Plasma levels were determined with HPLC-UVmethod. Results The plasma concentration-time curve was fitted to 1-compartmentopen model with a first order and lag time absorption and the major pharmacokineticparameters of capsules A and B were shown respectively as following: C max(32. 69?5. 67 )and (29. 41?2. 60) mg?L-1; Tmax (1. 01?0. 14) and (1. 13?0. 37) h, t1/2ka(0. 46 ? 0. 18) and (0. 62 ? 0. 20) h; C max (1. 11 ? 0. 32) and (1. 32 ? 0. 26) h; MRT(2. 14 ? 0.27) and (2. 37 ? 0.26) h; AUC (91.7 ? 13.2) and (82.2 ? 7. 38) mg?h? L-1. There were no significantly differences between the pharmacokinetic parame-ters of capsule A and B. The relative bioavailability of the capsule A was (110 ? 9) % ascompared to the capsule B. Conclusion The two kinds of capsules have the equivalentbiological effects.
8.Effects of low-frequency ultrasound combined with Tongluo Zhitong herbs on urokinase-type plasminogen activator system and inflammatory cytokines in knee osteoarthritis

BACKGROUND: Low-frequency ultrasound combined with Tongluo Zhitong herbs can effectively relieve pain and improve joint function of patients with osteoarthritis, but the underlying mechanisms are still unclear.OBJECTIVE: To investigate the effects of low-frequency ultrasound combined with Tongluo Zhitong herbs on urokinase-type plasminogen activator (uPA) and inflammatory cytokines of osteoarthritis rabbits and the related mechanisms.METHODS: Forty rabbits were randomly divided into four groups: normal, model, control, and experimental groups. The normal group was treated with normal feeding, not for any processing. For the other three groups, papain was injected into the rabbit knee articular cavity to establish osteoarthritis models. One week later, the experimental group was treated with low frequency ultrasound combined with Tongluo Zhitong herbs for 3 weeks, the control group treated with the same volume of normal saline, and the model group received no intervention. The levels of inflammatory cytokines in the serum and synovial fluid were measured, the histological changes of synovial tissues and articular cartilage were evaluated using Mankin's score, and the expression levels of uPA and its receptor were determined by western blot assay.RESULTS AND CONCLUSION: The levels of inflammatory cytokines in the model group were significantly higher than those in the normal and experimental groups (P < 0.05). The Mankin's scores of synovial tissue in the experimental group were significantly lower than those in the model and control groups (P < 0.05), while Mankin's scores of the cartilage tissue had no significant difference between the experimental and model groups (P > 0.05). Compared with the model group, in the experimental group, the expression levels of uPA and its receptor were significantly downregulated,and the expression of uPA inhibitor was notably upregulated. These data show that low-frequency ultrasound combined with Tongluo Zhitong herbs attenuates inflammatory reaction in the synovial tissues, by inhibiting the formation of joint fluid and production of inflammatory cytokines, which is an effective treatment for knee osteoarthritis.
9.Comparison of 2 decellularizing methods to prepare rat acelluar allograft spinal cord scaffold

Objective To compare the effect of 2 decellularizing methods,sodium deoxycholate plus Triton X-100 or plus DNase and Rnase,in the preparation of acelluar allograft spinal cord scaffold in order to provide an ideal natural spinal cord scaffold to bridge the nerve gap.Methods Spinal cord was removed from health rats,and then decellularized by the method of freeze thawing(immersing in 3% sodium deoxycholate followed by the mixture of 1?103 U/ml DNase and RNase),or by chemical extraction(immersing in 1% Triton X-100 and then 1% sodium deoxycholate).HE staining,myelin staining and scanning electron microscopy(SEM) were employed to evaluate the spinal cord scaffold after the 2 methods of decellularization.Results Both cells and myelin were completely decellularized with the 2 methods.In cross section,network of the extracellular matrix was presented without axon,sheath and cells nucleus being seen in the scaffold.Typical network of empty tubes were viewed in longitudinal sections.Conclusion An ideal spinal cord scaffold can be produced with these 2 decellularizing methods in tissue engineering.The scaffold made by the 2 methods have similar three dimensional structure with normal spinal cord,so can be used as a graft to bridge the nerve gap directly or as a scaffold to implant the seeding cells in spinal cord tissue engineering.
10.Preparation of acellular scaffold of natural spinal cord and observation of morphology

[Objective]Using a procedure of chemical agent to remove the cells and myethin in spinal cord of rat and to prepare the scaffold of extracellular matrix,so as to obtain an ideal natural spinal cord scaffold to bridge the nerve gap.[Method]Rat spinal cord was cut and treated using the method of freeze thawing and chemical extraction(3%sodiumdeoxycholate and 1KU/ml DNaseI,RNaseA).Histology was exploited to evaluate the degree of acellular and the structure of the spinal cord scaffold.[Result]In cross section,network of the extracellular matrix was presented in the scaffold.The cells,myethin and axons disappeared after the spinal cord was treated with sodium deoxycholate and DNaseI,RNaseA.Typical network of empty tubes were viewed in longitudinal sections.[Conclusion]An ideal spinal cord scaffold can be produced with the method designed in authors experiment.This scaffold has similar three dimensional structure with normal spinal cord,which can be used as a graft to bridge the nerve gap directly or as a scaffold to implant the seeding cells in spinal cord tissue engineering.The experiment indicates that cells and myethin can be removed and the three dimensional structure be reserved by chemical extraction with 3% sodium deoxycholate and 1KU/ml DNaseI,RNaseA.Chemical extraction is an ideal method to prepare tissue engineer scaffold of spinal cord.
