3.Effects of E 4031 on isolated cardiac function and resting Ca2+ level of myocardial cells from rats with chronic heart failure.
Chinese Journal of Applied Physiology 2014;30(4):364-367
OBJECTIVETo investigate the effects of E 4031, a sodium calcium exchanger (NCX) agonist, on the isolated cardiac function and resting Ca2+ level in myocardial cells from rats with chronic heart failure.
METHODSRats chronic heart failure model was established by abdominal aorta coarctation with; Isolated heart perfusion by Langendorff apparatus was used to detect heart function and the effects of E 4031 on haemodynamic indexes; Myocardial cells of rats in the model group were extracted quickly and co-incubated with calcium fluorescent indicator fluo3/AM and the impact of E 4031 on the fluorescence intensity in myocardial cells were evaluated by confocal microscopy.
RESULTSHeart function of rats in the model group detected by Langendorff perfusion was significantly reduced after 12 weeks, E 4031 at 10 micromol/L could improve their left ventricular developed pressure(LVDP) and systolic / diastolic maximum rate (+/- dp/dtmax). Compared with the control and sham operation groups, the resting Ca2+ fluorescence intensity of the myocardial cells of rats in model group was at a higher level and went through a process of transient rise and drop, then stably remaining at a low level after co-incubated with 10 micromol/L E 4031.
CONCLUSIONE 4031 can improve the isolated heart function of rats with chronic heart failure, which may be associated with its enhancing the activity of NCX in the myocardial cell membrane and stabilizing intracellular Ca2+ level.

Objective To investigate the role of intracellular protein tyrosine kinase (PTK) activation in vascular smooth muscle cells (VSMCs) migration mediated by angiotensin Ⅱ (AngⅡ). Methods Intracellular PTK activities were investigated by means of ?- 32 P incorperation in cultured rat VSMCs stimulated by AngⅡ. The migrated VSMCs were enumerated in a modified Boyden Chamber. Results Without any specific stimulus, only a few migrated cells were detected. However, the number of migrated cells were increased, under the condition of AngⅡ at the concentration of 1?10 -10 , 1?10 -9 , 1?10 -8 , 1?10 -7 and 1?10 -6 mol/L, by 3.7, 4.6, 5.9, 6.6 and 6.3 times respectively, compared with the control group. Intracellular PTK activities were also increased by 1.0,1.7,2.4,3.2 and 2.4 times compared with the control group, under the condition of AngⅡ at the concentration of 1?10 -10 , 1?10 -9 , 1?10 -8 , 1?10 -7 and 1?10 -6 mol/L, respectively. Statistical analysis showed that there was a significant positive correlation between AngⅡ-induced VSMCs migration and intracellular PTK activation. Correspondingly, PTK inhibitor Genistein significantly reduced AngⅡ-induced VSMCs movement ( P

Liver is the main target organ of the metastasis of colorectal cancer.More and more researches demonstrate that including micro-environment of cancer, signal transduction pathway, abnormality of chromosomes and the gene which control cell proliferation and apoptosis are the most important mechanisms in liver metastasis of colorectal cancer. The metastasis of colorectal cancer is the result of all these factors. To study the mechanism can shed some light on prevention and therapy in liver metastasis of colorectal cancer.

Objective To investigate the interaction between HBx protein and ?-catenin for exploring molecular mechanism in HBV-associated hepatocellular carcinoma. Methods Iimmunocytochemistry and Luciferase Assay were used to detect the endocellular location and transcriptional activity of ?-catenin in normal liver cell line L02 infected by Ad-HBx.Results Immunocytochemistry: there was markedly increased expression of ?-catenin in cytoplasm and nucleus.Luciferase Assay: Ad-HBx led to a significant increase of TCF-4-dependent transcription, the TOP count of Ad-GFP and Ad-HBx-GFP was 1.4 fold and 11 fold as compared to the negative control respectively(P
9.The correlation of serum ghrelin level with the total BMC,LM and body weight in premenopausal women with different thyroid functional status
Chinese Journal of Pathophysiology 2000;0(07):-
AIM:To investigate the relationship between serum ghrelin level and body composition including bone mineral content (BMC),fat mass(FM) & lean mass(LM) in premenopausal women with different thyroid functional status.METHODS:We measured the serum ghrelin levels by radioimmunoassay (RIA),the serum levels of free triiodothyronine (FT3),free thyroxine (FT4) and sensitive thyroid-stimulating hormone (sTSH) by chemiluminescene immune assay. The total body composition including total BMC,FM and LM was measured by dual-energy X-ray absorptiometry (DXA) in 71 premenopausal women with different thyroid functional status (33 hyperthyroidism,18 hypothyroidism and 20 normal subjects).RESULTS:(1) The level of serum ghrelin in patients with hyperthyroidism was significantly lower than that in patients with hypothyroidism (P0.05).The serum ghrelin level was correlated negatively with FT3(r=-0.318,P0.05).CONCLUSION:The serum ghrelin level in the premenopausal women with different thyroid functional status may correlate with the total BMC,LM and body weight.

Objective To explore the water quality of the product water from the centralized water supply system in Hunan, 2006. Methods The sanitary quality of the product water samples collected from 28 centralized water supply system in cities and 39 centralized water supply system in counties in Hunan, 2006, which distributed among the area of Xiangjiang River, Zishui River, Yuanjiang River and Lishui River, were determined and evaluated according to the Standards for Drinking Water Quality (GB5749—2006). Results The eligible rate of the water sample was 70.1%. Among 74 water quality indexes, 12 indexes (such as antimony, aluminum,pH value, manganese and so on) exceeded the standard limits in degrees in some centralized water supply systems. Most of the routine indices of water quality were unqualified and the reference indices were qualified. The quality of some water samples presented statistical difference in some indices (such as aluminum, sulphate, arsenic, et al) among 4 different river areas in Hunan province (P
