Journal of Chongqing Medical University 2007;0(08):-
Objective:To investigate the expression rule of Cyclin-E in endometrium during the early pregnancy and its role in blastocyst implantation in mice.Methods:RT-PCR and immunohistochemistry techniques were applied to detect Cyclin-E gene and protein expression in endometrium from pregnant mice on days 1~7.In addition Anti-cyclinE antibody was injected into horns of uterus of day 3 pregnant mice to observe the numbers of implanted blastocysts. Results:Expression of Cyclin-E gene and protein in pregnant mice were steady increasing since day 2 and reaching maximal level on day 5 of pregnancy.Injection of Anti-cyclinE antibody could decrease the numbers of implanted blastocyst compared with that of saline-injected group.Conclusion:Cyclin-E were persistently expressed in mouse endometrium during the early pregnancy and might be concerned with proliferation differentiation of endometrium and participates in the regulation process of mouse blastodyst implantation.
2.Ezetimibe in the prevention of ischemic stroke
International Journal of Cerebrovascular Diseases 2021;29(2):142-147
Dyslipidemia is a risk factor for stroke, and reducing low-density lipoprotein cholesterol (LDL-C) levels can decrease the risk of ischemic stroke. Both statins and PCSK9 inhibitors can effectively reduce LDL-C levels, but some patients cannot tolerate statins, and PCSK9 inhibitors are expensive. Animal experiments have shown that ezetimibe can reduce oxidative stress and endoplasmic reticulum stress response, inhibit cell apoptosis, increase autophagy, and protect nerve tissue by activating AMP activated protein kinase (AMPK) signaling pathway. Clinical studies have shown that ezetimibe can further reduce LDL-C levels on the basis of statin therapy, and reduce the risk of ischemic stroke in patients with acute coronary syndromes, and there is no obvious adverse reaction. However, ezetimibe cannot reduce the incidence of stroke in elderly people who are ≥75 years old, have elevated LDL-C and do not have coronary heart disease. At present, there are few studies on the use of ezetimibe for secondary prevention of ischemic stroke, and its effect needs to be further confirmed by large randomized controlled trials.
3.Prevention and treatment of glucocorticoid-induced eye adverse reactions in children with primary nephrotic syndrome
Chinese Journal of Applied Clinical Pediatrics 2021;36(5):333-337
Primary nephrotic syndrome refers to one of the common glomerular diseases in children.Glucocorticoid is the first choice in the treatment of nephrotic syndrome, and the using cycle is long.Long term application of glucocorticoids can cause side effects, including Cushing′s disease, hypertension, decreased immune function, growth retardation and eye adverse reactions.Among eye adverse reactions, glaucoma and cataract are more common.Glucocorticoid-induced glaucoma and glucocorticoid-induced cataract are often concealed, and children often cannot accurately express their discomforts. At the same time, some clinicians lack the understanding of the adverse reaction, which is easy to cause missed diagnosis and misdiagnosis.Eventually, irreversible visual impairment occurred. Here, the etiology, pathogenesis, clinical diagnosis, and treatment of glucocorticoid-induced glaucoma and glucocorticoid-induced cataract are elaborated, thus aiming to improve clinicians′ understanding of glucocorticoid ophthalmopathy and the level of clinical diagnosis and treatment.
