BACKGROUND:Growth differentiation factor 5 (GDF5) has been shown to play a crucial role in the development of chondrocytes via different regulatory mechanisms.
OBJECTIVE:To explore the effect of GDF5 on the chondrogenic differentiation of adipose-derived stem cel s (ADSCs).
METHODS:ADSCs in passage 4 isolated from Japanese White rabbits were cultured in the medium containing 0 (blank control group), 10, 50, 100, 150 and 200μg/L, respectively. The morphological changes were observed, and the optimal concentration of GDF-5 was screened, and its round coverslips at 1, 2 and 3 weeks of culture were selected to undergo immunocytochemistry, toluidine blue staining and immunofluorescent staining.
RESULTS AND CONCLUSION:The growth of ADSCs was stable in the medium containing 10 and 50μg/L GDF5, while apoptotic cel s appeared after induction with 200μg/L GDF5. In the medium containing 100 and 150μg/L GDF5, some spindle-shaped cel s changed into irregular shape, and became round or oval after 7-10 days of culture. The optimal concentration of GDF5 was 100μg/L. The cel s transfected by 100μg/L GDF5 were positive for type II col agen obviously and with blue-stained nucleus. The transfected cel s were positive for toluidine blue, and metachromatic granules were visible in the cytoplasm. The proteoglycan mRNA expression of the transfected cel s was significantly increased, and reached the highest at 3 weeks. These results suggest that GDF5 promotes the chondrocytic differentiation of ADSCs.

A simple, rapid and sensitive colorimetric reverse-transcription loop-mediated isothermal amplification (RT-LAMP) was developed for rapid detection of Middle East respiratory syndrome coronavirus (MERS-CoV). The method employed six primers that recognized sequences of a nucleocapsid gene for amplification of nucleic acids under isothermal conditions at 63 degrees C for 60 min. Products were detected through a LA-320c Loopamp Turbidimeter (real-time RT-LAMP) or visual inspection of color change by pre-addition of Hydroxynaphthol Blue dye (visual RT-LAMP). Specificity of RT-LAMP was validated by detection of several human coronaviruses and common respiratory viruses. MERS-CoV real-time RT-LAMP had a linear correlation (R2) of 0.995 at 10(3)-10(6) copies. The limit of detection of real-time RT-LAMP, visual RT-LAMP and quantitative real-time PCR was 500, 1000 and 100 copies/reaction, respectively. The established RT-LAMP assay was demonstrated to be a rapid screening tool for MERS-CoV infection, and could be suitable in resource-limited clinical sites and for field studies.

OBJECTIVETo observe the efficacy of Shenmai Injection (SI) in preventing and treating adverse reation of chemotherapy on advanced non-small cell lung cancer (NSCLC).
METHODSForty-five patients with NSCLC in III b-IV stage were randomly divided into two groups, the control group treated with chemotherapy alone and the treated group with chemotherapy and SI. The therapeutic efficacy were evaluated after 3 treatment cycles.
RESULTSThere was no significant difference in short-term effect between the two groups (P >0.05), while it showed remarkable difference in Karnofsky scoring and body weight (P < 0.05). The treated group showed a better effect than the control group in reducing adverse reaction, such as decrease of leucocyte and hemoglobin (P < 0.05), while in the aspects of improving thrombocytopenia and the reducing occurrence of nausea/vomitting and alleviating injury of liver and kidney function, it only showed the lower value in the treated group, but with no significant difference as compared with the control group (P >0.05).
CONCLUSIONSI could not raise the efficacy of chemotherapy on NSCLC, but improve the quality of life, raise the body weight of patients and alleviate adverse reaction of chemotherapy as myelosuppression so as to improve the tolerance of organism to chemotherapy.

Objective To identify the clinical and demographical features that may impact the duration of mechanical ventilation(DMV) in the patients with post-thymectomy myasthenic crisis(PTMC).Methods Reviewed the patients who had PTMC from June 2008 to November 2015.Cox proportional hazard regression analysis was used to identify potential prognostic factors that may impact DMV and long-term survival,respectively.One-way ANOVA analysis was used to compare the four groups with continuous variates.Statistical powers were calculated by using XLSTAT.Results In total, seventy patients with PTMC were enrolled.Alcoholic abuse, high MGFA classification and Clavien-Dindo classification were the critical factors that remarkably delayed early extubation.Postoperative lung infection(PLI) as sole complication did not prolong DMV as compared to those without any complication, however,PLI with other more severe complications requiring at least pharmacological treatment seemed to remarkably prolong DMV,as compared to those without any complication. Conclusion Preoperative abstinence, proper treatment of MG and postoperative complications can decrease the incidence of PTMC,shorten the use of ventilator time and improve the prognosis of patients.
