1.The therapeutic efficacy of dihydroergotoxine for headache attributed to ischemic stroke

Objective This clinical study was designed to explore the therapeutic efficacy of dihydroergotoxine for headache at-tributed to ischemic stroke .Methods Seventy eight patients fulfilling the diagnostic criteria of headache attributed to ischemic stroke were enrolled and randomly divided into two treatment group(n=42) and control group(n=36) who took dihydroergotoxine (2 mg twice daily) and placebo respectively .One week for a course of treatment ,had many courses of treatment .Efficacy was as-sessed using the patient′s diary .Results The gender ,age ,stroke aetiology ,admission NIHSS score ,quality and location of headache didn′t differ significantly between the two groups .Compared with the control group ,the duration of headache were shorter in treat-ment group(3 .78 ± 1 .44 vs .4 .88 ± 1 .45 ,P=0 .002) .The severity of headache were lower in treatment group since the third day post ischemic stroke(P<0 .05) .There were no significant side effects during treatment .Conclusion Dihydroergotoxine in treat-ment of headache attributed to ischemic stroke is beneficial to shorten the duration of headache as well as decrease the severity of headache ,and thereby improve the quality of life .

Objective To study the longitudinal change of Migraine patient′s condition ,quality of life and disability ;analyze the evolution of the relationship between headache frequency ,pain intensity ,medication and treatment efficacy .Methods Continuous registration of migraine patients ,using WHO-DAS Ⅱ to assess the disability ,using MSQ2 .1 to assess the health-related quality of life ,using headache diary to record the patient′s condition .Results Compared the conditions of before and after treatment ,the aver-age level of pain ,acute drug using and headache frequency ,were significantly improved .The three dimensions′scores of MSQ2 .1 were decreased .Conclusion In this study ,significant improvement were found in some of the disability and health-related quality of life .Continuity of treatment has an important role to improve health-related quality of life of patients and reduce disability .
5.Quantitative Proteomic Analysis of Bromotetrandrine and Tetrandrine in K562 Cell Line Using 18O-labeling Method

ObjectiveTo compare quantitative proteomic analysis of bromotetrandrine (W198) which was a Class Ⅰ new antitumor drug in China and tetrandrine (Tet) in K562 cell line using 18O-labeling method.MethodsTo illustrate its mechanism,a shotgun quantitative proteomic strategy employing 2D LC-MS-MS and trypsin catalyzed 18O-labeling quantification was carried out in this study.Compared to normal chronic leukemia cell line K562 and K562 induced by Tet,the proteomic changes of K562 induced by W198 were investigated.In order to validate the quantitation by the 18O-labeling,the analysis was done on an equivalent sample composed of the same amount of labeled and unlabeled proteins from normally cultured cells to act as a reference to the comparative sample.ResultsA threshold of ± 2-fold change for deciding whether a protein concentration was changed was settled for the following experiments.Comparing the 105 identified soluble proteins' expression levels of the apoptosis starting up K562 cells after W198 induction with the normally cultured cells,16 proteins were found with significantly altered expression levels after W198 treatment.Eight proteins were up-expressed including HMGB2,peroxiredoxin-2,and eIF4A-I,etc.Eight proteins were down-expressed including TCP-1,GRP94,GST-π,and SFGHs,etc.Compared to K562 induced by Tet,eight proteins of K562 were found with significantly altered expression levels after W198 treatment.Five proteins were up-expressed including HSP 90-β and 40S ribosomal protein S15a,etc.Three proteins were down-expressed including phosphoglycerate kinase 1,isoform 5 of interleukin enhancer-binding factor 3,etc.ConclusionThe 18O-labeling MS-MS-based method is ideal as a discovery tool,but it is not suitable for validation using a large number of samples.Other more effective methods,such as Western blotting should be used for further validation of candidate cancer proteins discovered from 18O-labeling samples.In total,105 soluble proteins were discovered,and 16 proteins were found with significantly altered expression levels after W198 treatment.These repressed or activated proteins are the potential drug targets of W198,which may provide novel targets for future development ofbiomarkers for cancer therapy.
6.Effect of Toutongning capsule for treatment of headache with stroke-associated risk factors

Objective To investigate the effect of Toutongning capsule for treatment of headache with stroke-associated risk fac-tors .Methods 60 cases of headache patients with stroke-associated risk factors were selected and divided into the treatment group and the control group ,30 cases in each group .The treatment group were given conventional therapy plus Toutongning capsule while the control group were given conventional therapy only .Results After 4 weeks ,the remission rate of treatment group was 93% and that of the control group was 63% .The attack frequency ,duration and degree between the two groups showed statistically signifi-cant difference(P<0 .05) .Conclusion Additional Toutongning capsule has a better effect for treatment of headache with stroke-as-sociated risk factors.
7.A proteomic approach to study cerebrospinal fluid from migraine patients

Objective To identify the biomarkers associated closely with migraine by proteomics analysis of cerebrospinal fluid (CSF).MethodsForty-five patients were involved in the present study and they were divided into the following two groups:25 with migraine (5 with aura and 20 without aura) were assigned to migraine group,and 20 with giddiness or peripheral neuropathy were assigned to control group.The patients' CSF was collected,and the protein of CSF was extracted by acetone precipitation.The two-dimensional electrophoresis (2D-PAGE) with immobilized pH gradient (IPG) was performed to display the differently expressed protein spots.These spots were then identified by two-dimensional liquid chromatography and tandem mass spectrometry (2D-LC/MS-MS),and semi-quantitatively analyzed by Western blotting.ResultsDifferences were found in the results of 2D-PAGE between migraine group and control group.Ten spots of 9 kinds of proteins were successfully identified,of which 7 spots were down regulated,including transthyretin (TTR),CBX6 53 kDa protein (CBX6),agrin precursor (AGRN),FAM3C precursor (FAM3C),neuronal pentraxin receptor (NPR),dermcidin-isform2 (DCD) and albumin (ALB);whereas junction plakoglobin (JUP) and H2A histone family,member J (H2AFJ) were up-regulated.The results of Western blotting revealed that the net protein retention (NPR) in migraineurs group (0.3351?0.0275) was obviously decreased compared with that of control group (0.8854?0.0957,P

9.Therapeutic Observation of Moxibustion plus Anus-lifting Exercise in Preventing Hemorrhoids Relapse

Objective To observe the clinical efficacy of moxibustion plus anus-lifting exercise in preventing hemorrhoids relapse.Method Seventy eligible patients with mixed hemorrhoids whose symptoms had been released by conservative treatment were randomized into a treatment group and a control group, 35 cases in each group. The treatment group was intervened by moxibustion plus anus-lifting exercise, while the control group only practiced anus-lifting exercise. The intervention was conducted successively for 4 weeks. Symptoms including hematochezia, bearing-down pain, prolapse, and size of hemorrhoid were scored in the 1st, 2nd, 3rd, and 4th treatment weeks as well as the 4th week after the intervention, meanwhile, the relapse rates of the above symptoms were observed.Result In the 3rd treatment week, there were significant differences in comparing the scores of prolapse, bearing-down pain, and hemorrhoid size between the two groups (allP<0.05); in the 4th treatment week and the 4th week after the intervention, there were significant differences in comparing the scores of hematochezia, prolapse, bearing-down pain and hemorrhoid size between the two groups (allP<0.05); in the 4th week after the intervention, there were significant differences in comparing the relapse rates of hematochezia, bearing-down pain, prolapse and hemorrhoid size between the two groups (P<0.05).Conclusion Moxibustion plus anus-lifting exercise can produce a significant efficacy in preventing the hemorrhoids relapse.
10.P21-activated kinase 1 protein expression and its significance in colorectal carcinoma and colorectal dysplastic adenoma

Objective To investigate the role of p21-activated kinase 1 (PAK1) in colorectal mucosal carcinogenesis and the relationship between PAK1 expression and biological behavior of colorectal carcinoma. Method PAK1 was detected with immunohistochemical method in 10 normal colorectal mucosas,40 colorectal villous or tubular adenomas and 60 colorectal carcinomas. Results The positive rate for PAK1 was 10.0% in normal colorectal mucesas,and 25.0%, 33.3% and 33.3% in slight, moderate and severe dys-plastic adenomas, respectively, 65.0% was found in colorectul carcinomas. The positive rate for PAK1 in col-orectal carcinomas was higher than that in normal colorectal mucosas(P<0.01), colorectal villous or tubular adenomas(P<0.01), slight dysplastic adenomas(P<0.01) and moderate dysplastic adenomas (P<0.05).The positive rate for PAK1 of poor differentiated colorectal carcinomas was higher than that of high differentiated ones (P<0.05), and the positive rate for PAK1 of patients with lymph node metastasis was higher than that of patients without lymph node metastasis (P<0.05). In clinical stages, the positive rate for PAK1 in Dukes C and D stages patients was higher than that in Dukes A stage patients, respectively (P<0.05). Conclusion PAK1 maybe play some role in the process of carcinogenesis of colorectal mucesa, and be used as an useful marker for assessment of the biological behavior and prognosis of colorectal carcinoma.
