1.Roles of the liver regeneration phosphatase-3 and its inhibitors in tumor
Journal of International Oncology 2012;39(6):415-417
Phosphatase of regenerating liver-3 (PRL-3) is a novel small molecule protein-tyrosine-phosphatase,which plays an important role in the oncogenesis and deveopment of tumors.Studies show that PRL-3 regulates neoplasm progress through participating in multiple signal pathways. Its high expression can significantly promote neoplasm metastasis in cancer tissue. However, there is no expression of PRL-3 in most normal tissue.PRL-3 is expected to be a potential tumor maker of diagnosis and a new target for the therapy of cancer.

Aim To investigate the inhibitory effect and its posssible mechanism of 5F from Pteris semipinnata L on the metastasis-associated ability of human highly metastatic ovarian carcinoma HO-8910PM cells in vitro. Methods MTT assay was used to examine the effect of 5F on proliferation of HO-8910PM cells after 24 hours treatment; Transwell Chamber assay was performed to determine the effect of 5F on invasion and migratory capacity of the cells; Effect on adhesion potential of HO-8910PM cells was tested by cell-Matrigel adhesion assay; The expression levels of NF-?B(P65) and VEGF proteins were assessed by Western blot analysis. Results 5F significantly inhibited invasion, migration, and adhesion capacity of HO-8910PM cells in vitro, their inhibitory rates after treatment with 50 ?mol?L -1 5F for 6 h were (37.57?0.62)%, (28.42?0.67)%, (46.07?4.49)%. 5F d istinctly down-regulated the expressions of NF-?B(P65) and VEGF protein. Conclusion 5F can inhibit the migration, invasion and adhesion of HO-8910PM cells in vitro,Its possible mechanism may be involved in the reduction of the expression level of NF-?B(P65) and VEGF. 5F might be potential drugs to inhibit tumor metastasis.

Aim To study the effect of resveratrol on the expression of FAK and the level of phosphorylated FAK, and to investigate its possible mechanism of resveratrol on the metastasis-associated ability of human highly metastatic ovarian carcinoma HO-8910PM cells line. Methods MTT assay was used to examine cytotoxicity of resveratrol on HO-8910PM cells after 24 h treatment; The expression of FAK protein and the level of phosphorylated FAK were assessed by Western blotting analysis. Results MTT assay showed that resveratrol had weak cytotoxicity on HO-8910PM cells after 24 h treatment, its IC50 value was 163.40?2.48 ?mol?L-1. The expression of FAK and the level of phosphorylated FAK decreased obviously in HO-8910PM cells treated with 25~100 ?mol?L-1 resveratrol for 24 h. Conclusion The mechanism of resveratrol inhibiting the metastasis-associated ability of human highly metastatic ovarian carcinoma HO-8910PM cells line involves the expression of FAK and the level of phosphorylated FAK.

Journal of Practical Radiology 2017;33(4):550-553
Objective To explore the clinical application of material decomposition technique on spectral CT imaging and evaluate hemodynamic changes in different liver lobes with liver cirrhosis. Methods 30 patients with liver cirrhosis diagnosed clinically in our hospital were collected and underwent enhanced scanning of abdomen with spectral CT protocol. The monochromatic energy images and iodine-based material decomposition (MD) images were reconstructed after scanning. The iodine concentration (IC) was measured in five liver lobes (the caudate, left lateral, left inner, right anterior and right posterior lobes) and the abdominal aorta of the same axial slice in both the arterial phase (AP) and portal venous phase (VP) on the iodine-water based material decomposition images. The arterial iodine fraction (AIF) and the portal venous iodine concentration (PVIC) as well as the normalized iodine concentration (NIC) during the AP and VP were calculated. The differences of IC,the NIC,the AIF,and the PVIC in five liver lobes in AP and VP were compared by using single factor analysis of variance. Results The IC,the NIC in both AP and VP and the AIF of the caudate liver lobe were higher than those of other four liver lobes, with statistically significant difference (P<0.05), while these values in the other four lobes showed no statistically significant difference (P>0.05).The PVIC of the caudate liver lobe was slightly lower than that of the other four liver lobes, however, the difference was not statistically significant (P=0.929).Conclusion The quantitative iodine concentration measurement of liver lobes on spectral CT material decomposition technique can evaluate the hemodynamic changes in liver lobes with liver cirrhosis, and provide more information about the change of blood flow in liver cirrhosis.

Objective To study the effect of a diterpenoid 5F (ent-11?-hydroxy-15-oxo-kaur-16-en-19-oic acid) isolated from Pteris semipinnata on the expressions of NF-?B (P65), Smad3 protein and NF-?B (P65), ETS-1 mRNA, and to investigate the antitumor mechanisms of 5F. Methods MTT assay was used to examine the effect of 5F on proliferation of HO-8910PM cells. The expressions levels of NF-?B (P65) and Smad3 proteins were assessed by Western blot analysis. RT-PCR assay was used to assess the expression levels of NF-?B (P65) and ETS-1 mRNA. Results The proliferation of HO-8910PM cells can be inhibited by 5F in a dose-dependent manner. The expression levels of NF-?B (P65) and Smad3 proteins decreased obviously in HO-8910PM cells after treatment with 25—100 ?mol/L 5F for 24 h, and the effects appeared in a dose-dependent manner. 5F down-regulated significantly the expression of NF-?B (P65) mRNA, down-regulated gently the expression of ETS-1 mRNA. Conclusion The antitumor activity of 5F is involved in the expression of NF-?B (P65), Smad3 protein and NF-?B (P65), ETS-1 mRNA.

As an important member of tool enzymes, exonuclease is a kind of hydrolytic enzymes without strict base sequence dependent. In recent years, by taking advantage of different hydrolysis ways of exonuclease and nanotechnology, cycle effect of enzyme digestion, aptamer, non Watson-Crick base pairing system by metal ions, fluorescent nucleic acid probes, electrochemical methods etc. , a series of exonuclease-assisted signal amplification strategies have been developed, which played a very key role in improving the sensitivity of detection methods. Therefore, exonuclease has been widely used in high sensitive detection of nucleic acids, proteins, ions, small molecules and so on. To understand it better and apply it well in the future, the application progress of exonuclease-assisted signal amplification strategies in biochemical analysis has been summarized in this review.
