Objective To find the features of cryo-injured infarcted model and the therapeutic effects of skeletal satellites cells(SCs) on myocardial infarction(MI).Methods SCs were implanted into the center of cryo-injured myocardium.Immunohistochemistry,confocal immunofluorescence double-labelling staining and transmission electron microscopy were used to study the state of SCs.Results After cryo-injured frontal wall of the left ventricle(LV) for approximately(15 s) by aluminium rod with(5 mm) in diameter,it made 43% weight of LV ischemia and 17% weight of LV infarcted.At 2nd week,injured area occured as transmural infarct.After transplantation,the implanted cells survived in large numbers.The transplanted cells proliferated remarkably at early phase and differentiated into many multinuclear myotubes after 2 weeks.Some of them developed into muscle fibers with better sarcomeres,but the gap junction was not formed between implanted cells and cardiac myocytes.Conclusions Cryo-injured infarcted model is a proper model to study myocardial infarction.Skelatal satellite cell transplantation will repair morphological structure partly of myocardial infarction area.

Objective To explore the optimal condition for the differentiation of rat bone marrow mesenchymal stem cells(MSCs) into cardiomyocyte-like cells in vitro by using DMSO.Methods SD rat MSCs were isolated from rat bone marrow and cultured,then induced by 1.0% DMSO for 24 h.The cultured cells were observed through phase-contrast microscopy.Immunohistochemical technique and laser scanning confocal microscopy(LSCM) were used for detecting the expression of desmin,?-sarcomeric actin and C-TnT.The ultrastructures were examined with transmission electron microscopy.Results MSCs induced by DMSO were identified by the positive staining for desmin,?-sarcomeric actin and C-TnT.Transmission electron microscopy showed that paralleled myofilaments and a lot of rough endoplasmic reticulum were formed.Conclusion MSCs can be induced to differentiate into cardiomyocyte-like cells by DMSO.

Results　The shape and ultrastructure of myocytes became complicated gradually with development. Conclusion　All the changes are related to function of blood supply of human fetal heart.

Objective To investigate the relationship between RAD51 135G ＞ C polymorphism and prognosis in triple-negative breast cancer patients by retrospective analysis.Methods The clinical data of 62 triplenegative breast cancer patients were collected.The 62 cases underwent standard chemotherapy and radiotherapy after tumor resection from Jan.2004 to Dec.2010 in Affiliated Cancer Hospital of Zhengzhou University.RAD51 135G ＞ C polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RELP) technology.The survival curve about progress free and overall survival time were then made.Results The median progress free and overall survival time in triple-negative breast cancer patients with or with-out RAD51 135G ＞ C polymorphism were(77.00 ±5.55)and(89.00 ± 10.40) months vs(99.00 ±4.26)and (103.00 ±4.30) months.The difference had statistical significance(P =0.039 and 0.015 respectively).Conclusion RAD51 135G ＞ C polymorphism is related with prognosis of triple-negative breast cancer patients,which might be a prognostic factor for breast cancer.

Objective To identify the rale of NKX2-5 gene in cardiomyocyte differentiation and its mechanism.Methods P19 cells were divided into transfected and non-transfected groups.In the transfected group,P19 cells were with stable expression of NKX2-5 gene.The P19 cells were cultured in suspension for 4 days,and the formed aggregates were transferred to Petri dish for adherent culture.On days 4,8,12,and 16 of the adherent culture,the expressions of ct-saicomeric actin(α-SA)and cardiac troponin T(cTnT)were detected with double-labeling immunofluorescence and Western blot.The ultrastruetural changes were observed on day 16.Results In the transfected group,no expression of α-SA and cTnT was found on day 4,and the expression of these 2 proteins or co-expression existed on days 8,12,and 16.There were early cell junction and myofilament-like structure in the cytoplasm of some cells in the transfected group.In the non-transfected group,these 2 proteins were negative,and no differentiated cell was found.Conclusion Stable expression of NKX2-5 gene can induce cardiomyocyte differentiation from P19 cells,but the P19 cells with stable expression of JVKX2-5 gene is not suitable to be an in vitro model of cardiac development.

OBJECTIVETo observe the effect difference between acupuncture of "five needles therapy" and conventional acupuncture for asthma of latent cold phlegm-fluid in the lung.

METHODSTwo hundred and ten cases were randomly assigned into an observation group and a control group, 105 cases in each one. Finally 7 cases were dropped out in the observation group; 6 cases in the control group. Feishu (BL 13), Dazhui (GV 14), Fengmen (BL 12) were used in the observation group; conventional acupuncture was used in the control group, and the main acupoints were Feishu (BL 13), Zhongfu (LU 1), Tiantu (CV 22), Danzhong (CV 17), Kongzui (LU 6), Dingchuan (EX-B 1), Fenglong (ST 40), Fengmen (BL 12), Taiyuan (LU 9). The needles were retained for 30 min each time, once a day for continuous 12 days. The scores of the individual symptoms and signs were observed before treatment and on the 3rd, 6th, 9th, 12th days, including pant, cough, cough up phlegm, fullness and oppression in the chest and diaphragm, wheezing rale and shortness of breath. The clinical effects were compared between the two groups.

RESULTSThe scores of six individual symptoms and signs on the 3rd, 6th, 9th, and 12th days in the two groups were lower than those before treatment (all<0.05), except the score of wheezing rale in the control group on the 3rd day (>0.05). The scores of pant, wheezing rale, cough on the 3rd, 6th, 9th, and 12th days in the observation group were lower than those in the control group (all<0.05), except the score of wheezing rale score on the 3rd day (>0.05). There were no significant difference between the two groups about the scores of cough up phlegm, fullness and oppression in the chest and diaphragm and shortness of breath on the 3rd, 6th, 9th, and 12th days (all>0.05), except the score of fullness and oppression in the chest and diaphragm in the observation group was lower than that in the control group on the 12th day (<0.05). 46 cases were clinical cured, 39 cases were markedly effective, 10 cases were effective and 3 cases were ineffective in the observation group with the total effective rate of 96.9%. 23 cases were clinical cured, 43 cases were markedly effective, 24 cases were effective and 9 cases were ineffective in the control group with the total effective rate of 90.9%. The difference was statistical (<0.05).

CONCLUSION"Five needles therapy" has significant therapeutic effect for asthma of latent cold phlegm-fluid in the lung, which is better than conventional acupuncture.
