2.Clinical Observation on Effect ofSai-ErFormula in Treatment of Benign Paroxysmal Positional Vertigo

This study was aimed to summarize the effect ofSai-Er Formula in treatment of benign paroxysmal positional vertigo (BPPV). A total of 62 BPPV outpatients were randomly divided into two groups, which were the group of Epley’s maneuver combined withSai-Er Formula and the group of Epley’s maneuver. Observation was made on the treatment efficacy and vertigo integral 10 days, 1 month and 3 months after treatment. The results showed that after 10-day treatment, the effective rate of the group of Epley’s maneuver combined withSai-Er Formula was significantly higher than that of the control group (P = 0.045). And after 10-day, 1-month, 3-month treatment, the improving degree of vertigo integral was better than that of the control group (P < 0.005). It was concluded thatSai-Er Formula can relieve the vertigo symptom of BPPV, and reduce its recurrence rate.

5.Study on the balance rebuilding of regulatory T cells/Th17 cells by 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin for the treatment of rheumatoid arthritis

Objective To investigate the regulatory T cells (Treg) induced by 2, 3, 7, 8-tetrachloro-dibenzo-p-dioxin (TCDD) on the aryl hydrocarbon receptor activator from na?ve T cells, in collagen-induced arthritis (CIA)-rheumatoid arthritis (RA) mouse in the correction of rheumatoid arthritis pathological process on Th17/Treg cell imbalance. Methods ① Rat CIA model was established by bovine Ⅱ collagen injection. The rats were divided into 3 groups: the normal group (N), the CIA model group (C), and the TCDD group (T).② The percentage of Th17 and Treg from peripheral blood mononuclear cells (PBMCs) were analyzed with flow cytometry (FCM). Th17 and Treg cells related cytokine including interleukin (IL)-1β, IL-6, IL-17A, IL-23, tumor necrosis factor (TNF)-α and transforming growth factor (TGF)-β were measured with enzyme linked immunosorbent assay (ELISA). ③ The changes of the arthritis parameters, and radiological of T/NT before and after treatment were observed. ④ After two weeks of treatment, the rats were killed. The swollen joints were used for pathological assessment and arthritic pathology injury score was evaluated. ⑤ All data were analyzed by one-way analysis of variance (ANOVA) test and t test. Results In group C:When the second immunization was compared with the first, the percentage of Th17 cells was significantly increased [(2.99 ±0.16)%, (4.02 ± 0.33)%, t=-6.82, P<0.01], while Treg cells were significantly reduced [(2.67 ±0.57)%, (1.79 ±0.39)%, t=3.11, P=0.000], Th17/Treg was significantly increased (1.14 ±0.14, 2.27 ±0.39, t=-4.53, P=0.039). In group T, the percentage of Th17 cells were significantly reduced [(2.69±0.24)%, (1.21±0.25)%, t=5.12, P<0.01] before and after treatment, while Treg cells were significantly increased [(2.76 ±0.36)%, (3.78 ±0.22)%, t=-6.24, P<0.01], Th17/Treg was significantly reduced (1.08±0.07, 0.21±0.05, t=-11.92, P=0.027). In group C:When the second immunization was compared with the first, the Th17 cells related cytokines were significantly increased inclu-ding IL-1 [(96±12) pg/ml, (153±18) pg/ml, t=-13.24, P<0.01], IL-6 [(246±14) pg/ml, (295±15) pg/ml, t=-9.41, P=0.000], IL-17 [(76 ±14) pg/ml, (99 ±16) pg/ml, t=-5.78, P<0.01], IL-23 [(85 ±9) pg/ml, (102 ±11 ( pg/ml, t=-11.71, P<0.01], TNF-α [(303 ±12) pg/ml, (345 ±17) pg/ml, t=-15.56, P=0.007], while Treg cells related cytokines TGF-β was significantly reduced [(42 ±7) ng/ml, (35 ±8) ng/ml, t=7.52, P=0.012]. Th17 cells related cytokines were significantly reduced including IL-1 [(93 ±10) pg/ml, (79 ±8) pg/ml, t=10.52, P<0.01], IL-6 [(245 ±11) pg/ml, (151 ±8) pg/ml, t=19.23, P<0.01], IL-17 [(76 ±10) pg/ml, (62 ±9) pg/ml, t=7.37, P=0.005], IL-23 [(84 ±11) pg/ml, (38 ±6) pg/ml, t=14.48, P=0.000], TNF-α [(294 ±8) ng/ml, (195 ±9) ng/ml, t=23.35, P<0.01], while Treg cells related cytokine TGF-β was significantly increased (41 ±8, 61 ±10, t=-10.61, P=0.000. Conclusion TCDD can increase Treg cells, reduce Th17 cells, so the Th17/Treg cell in RA patho-genesis could be re-balanced.
6.Ingenane diterpene ester constituents from Tibetan medicine Euphorbia wallichii

Objective To study the chemical constituents from the acetone extract in the root of Tibetan medicine Euphorbia wallichii. Methods The constituents were separated by column chromatography with silica gel and purified by Sephadex LH-20. Their structures were identified on the basis of spectral analysis such as IR, HRESIMS, HRSIMS, and 1D-and 2D-NMR techniques ( 1H- 1H COSY , HMQC, HMBC). Results Six compounds were isolated from the acetone extract in the root of E. wallichii. Their structures were identified as lanosterol (Ⅰ), ingenol-20-myristinate (Ⅱ), ingenol-3-myristinate (Ⅲ), acid (Ⅳ), 1-O-?-L-arabinofuranosyl-(1→6)-?-D-glucopyranosyl-3, 7-dimethyl-oct-2-en-7-ol (Ⅴ), 1-O-galloyl-?-D-glucose (Ⅵ) . Conclusion Ingenane diterpene esters ingenol-20-myristinate (Ⅱ) and ingenol-3-myristinate (Ⅲ) are new compounds and other compounds are found from this plant for the first time. It is the first time that monoterpene disaccharide glycoside, compound Ⅴ, is isolated from the plants of Euphorbia L.
7.Intensive care after transplantation

BACKGROUND:It is an important and complex issue for the incidence of bacterial infection and complications after transplantation. The monitoring and care after transplantation can improve the success rate of transplantation. OBJECTIVE:To search the database of China National Knowledge Infrastructure, North American Clinical Trial Register and Thomson Reuters Web of Science database, and to perform literature metrological analysis and clinical trials registration project analysis on the published literatures of the monitoring and care after transplantation. METHODS:A total of 138 literatures were searched with the key words of“intensive care, transplantation”in the database of China National Knowledge Infrastructure on the intensive care after transplantation. 23 literatures were used for further analysis by reading titles and abstracts and 115 papers were excluded; the Thomson Reuters Web of Science database was searched with the key words of“intensive care, transplantation”for the literatures on the intensive care after transplantation published from 2008 to 2013;the North American Clinical Trial Register was searched with the key words of“intensive care, transplantation”for the related clinical trials, and a total of 50 registered projects were obtained, only 10 interventional studies. RESUTLS AND CONCLUSION:In recent years, the literatures on the intensive care after transplantation show a gradual increase trend. Compared with international research, fewer researches in this field emerge in China, literature quantity and quality need to be improved. There have been 1 693 papers published in the Thomson Reuters Web of Science database regarding the intensive care after transplantation. United States published the most literatures than other countries, total 532 papers, accounting for the largest proportion, 31.424%of total literatures. Transplantation Proceeding published the most literatures, total 144 papers, accounting for 8.506%of total literatures. There are 50 clinical trial registration projects related to the intensive care after transplantation in the North American Clinical Trial Register, 10 of them were interventional study, accounting for the majority, fol owed by observational study. Diagnostic study had no related registration projects. The surgery of transplantation was complex with trauma. The early monitoring and care post-transplantation are directly related to the success of surgery. Strengthening the monitoring and care of patients’ vital signs, rejection, bacterial infections and other aspects can reduce the complications and improve patients survival rate and quality of life.
8.Reasonable option of digestive tract reconstruction after proctocolectomy

Digestive tract reconstruction after proctocolectomy is a critical step of colorectal surgery, proper reconstruction can promote the recovery of digestive tract function and improve the quality of life.At present, there are some candidate reconstruction methods after proctocolectomy, but standardized and proper one is not still established.Since there are differences of the reconstruction methods after proctocolectomy at the different parts of colorectum, the best reconstruction methords should be selected according to the condition and intraoperative situation of patients.
9.Experimental Study on the Effect of Fuzhengqingluo(FZQL) Capsule Combined with CTX Against S180 Tu-mor

OBJECTIVE: To observe the antitumor effect of FZQL combined with CTX on S180 tumor and the effect of FZQL on reducing CTX s toxieity .METHODS: The models of S180 tumor bering mice were established .The animals were divided into CTX therapy group, CTX and FZQL combined therapy group, FZQL therapy group and control group.The results were compared among these groups.RESULTS: The weight of tumor in FZQL therapy group was less than that in control group.All of the experimental markers in combined iherapy group were better than those in CTX therapy group.CONCLUSION: FZQL has a marked inhibiting effect on tumor(S180), and it can increase the CTX's inhibiting effect on tumor and reduce the side effect of CTX and increasing the body immunity.
10.The study of immobilization of glucose oxidase on hydrophilic-hydropholic silica gel

Objective To evaluate the performances of glucose oxidase immobilized on hydrophilic-hydrophobic silica gel, which may be employed to prepare glucose sensor for the determination of glucose in body fluids. Methods The silica gel was prepared from precursors ?-aminopropyltrimethoxysilane (APTMOS) and methyltrimethoxysilane (MTMOS) by sol-gel technique. Glucose oxidase (GOD) was covalently attached to the silica gel via carbodiimide coupling reaction between a carboxylic acid group on enzyme and an amine group of the silica gel under the participation of the linking reagents 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride and N-hydroxysuccinimide. The performances of immobilized GOD were explored. Results The optimum conditions were obtained as follows: volume fraction of APTMOS 70%, enzyme content given 16 800 U, the temperature of 35 ℃ and buffer pH 5.5. The decrement in the activity of immobilized GOD for the first 2 weeks was less than 10% of its original activity, and the activity of immobilized GOD retained more than 75% of its original activity after 1 month of testing. Six independently prepared immobilized GOD on the silica gel resulted in an average bioactivity of 1 290.9 ?mol?min -1?g -1 with an R.S.D. of 3.4%. The Michaelis constant (K m) of immobilized GOD was 9.1 mmol?L -1. Conclusion Immobilizing GOD on the silica gel via the formation of peptide bonds is an outstanding enzyme immobilization method.
