Long noncoding RNAs (lncRNAs) are non-protein coding transcripts longer than 200 nucleotides. Most lncRNAs have pro-nounced oncogenic effects associated with tumorigenesis and progression, promoting the proliferation, migration, invasion, and me-tastasis of tumor cells. The specific lncRNAs expression in particular types of cancers makes them promising diagnostic and prognostic biomarkers. Currently, studies on lncRNAs expression, functions, and mechanisms have attracted considerable attention in cancer re-search. However, these studies mainly focus on epitheliogenic malignant tumors. In this review, we outline the current state of infor-mation on lncRNAs and research progress on its role in haematopoietic and lymphoid tissue tumors.

Objective The goal of this study was to explore the effect of exercise preconditioning on the expression of calcitonin gene-related peptide(CGRP)in the dorsal root ganglion of rats.Methods Fifty healthy male SD rats were randomly divided into control group (C) and exercise preconditioning group(EP).Group EP performed intervial treadmill exercise for 3 weeks for establishing exercise preconditioning animal model.The expression of CGRP mRNA in dorsal root ganglion was investigated by real-time fluorescence quantitative PCR.The immunoreaetion of CGRP in dorsal root ganglion was shown by immunohistochemistry.Results There was no significant difference in the expression of CGRP mR.NA in two groups(P>0.05).As compared with the group C,the immunoreaction of CGRP was increased in group EP,and the positive area and mean optical density of CGRP immunoreaction in group PE were significant higher than that in group C(P<0.05).Conclusion Exercise preconditioning does not change the expression of CGRP mRNA in dorsal root ganglion,but enhances the expression of CGRP in dorsal root ganglion to promote the reserve and release of CGRP in peripheral nerve endings and has the same endogenous protection as ischemie preconditioning.

Objective To evaluate the performance of aspartate aminotransferase to platelet ratio index (APRI) in the assessment of fibrosis in chronic hepatitis B virus (HBV) infection patients with transaminase level less than two times of upper limits of normal (ULN),and to analyze the clinical utility in antiviral therapy.Methods A total of 349 clinically diagnosed chronic HBV infection with alanine aminotransferase (ALT) and aspartate aminotransferase (AST) level less than 2 × ULN were enrolled.Liver biopsy and routine laboratory tests were performed to calculate the ratio of AST to platelet (PLT),namely the APRI,and to compare the relationship between liver fibrosis and APRI.Diagnostic performance of APRI model was assessed by using receiver operating characteristic (ROC) curves and area under the ROC curve (AUC) analysis.Correlation between APRI and liver fibrosis was determined by Spearman rank correlation analysis.Results The AUC of APRI≥0.273 for significant liver fibrosis was 0.641 with sensitivity of 48.3％,specificity of 75.7％ and positive predictive value of 73.9％.The AUC of APRI≥0.311 for cirrhosis was 0.771 with sensitivity of 68.6％,specificity of 76.8％,and negative predictive value of 93.5％.In patients with HBV DNA level ranging from 1 × 103 to 1 × 105 copy/mL,the optimal cut-off value of APRI was 0.179 for significant fibrosis (P＝0.00) and 0.283 for cirrhosis (P=0.00).By Spearman rank correlation analysis,APRI was positively correlated with histologic stages of fibrosis (r=0.370,P＜0.01).Conclusions APRI can be utilized to assess liver fibrosis in HBV infection with ALT and AST level less than 2 × ULN.APRI≥0.273 (pathologic stages of fibrosis ≥S2) has obvious hepatic fibrosis,which may help physician to select the optimal time for antiviral therapy.APRI may serve as a potential tool for assessing liver fibrosis according to HBV DNA levels.

To study the effect of exercise on cardiac calcitonin gene-related peptide (CGRP) using immunnohistochemistry and the technique of computer image analysis, the expression and mechanism of cardiac CGRP on the animal model trained with different exercise intensities were investigated. The result showed that long-term low intensity of exercise was not able to induce obvious change in cardiac CGRP. After long-term moderate intensity of exercise, the expression of cardiac CGRP increased so as to improve blood supply and protect myocardium. Long-term high intensity of exercise decreased expression of cardiac CGRP and weakened the protection of myocardium which could be a chief cause of myocardial ischemia.

Neoadjuvant chemoradiotherapy (nCRT)and subsequent radical resection have become the standard care of locally advanced rectal cancer (LARC).However,conventional radical surgery is associated with high intra-and postoperative morbidities,and a temporary or permanent stoma in some cases,impaired sexual and urinary function,and it eventually impair the quality of life.Rectum-preserving surgery is a novel concept in the era of nCRT,which will maximally benefit patients through the minimized injuries.This strategy has been increasingly utilized in LARC following nCRT.Nevertheless,it lacks high-level evidence of evidence-based medicine,and the long-term oncological safety remains to be determined by larger,multicenter,randomized clinical trials.The crucial aspect of rectum-preserving surgery is to accurately evaluate the status of primary tumor and metastatic lymph nodes,and thus to selective patients who could benefit from this strategy most.

Objective To investigate the influence of ulinastatin combined with Xue Bi Jing injection therapy on serum interleukin interleukin-8 (IL)-8 and γ-interferon (γ-IFN) of patients with severe acute pancreatitis(SAP).Methods Seventy patients with SAP were randomly divided into the control group(n=35) and research group (n =35).Patients in the control group were given ulinastatin on the foundation of traditional treatment,and in research group were given Xue Bi Jing injection on the foundation of treatment of the control group.The rehef time of abdominal distension and abdominal pain,time of serum amylase (sAMY) and white blood cell count(WBC) returned to normal were recorded.The IL-8 and γ-IFN of before and after treatment of the two groups were measured.Results The relief time of abdominal distension,abdominal pain,times of serum amylase,WBC returned to normal in treatment group were (4.1 ± 1.5) d,(4.2 ± 1.9) d,(5.2 ± 1.4) d,(6.5 ± 1.9) d respectively,significantly shorter than the control group((6.2±2.4) d,(6.5±2.2) d,(6.8±2.5) d,(8.9±2.5) d;t=5.468,5.747,4.354,4.647;P＜0.05).The IL-8 and IFN-γ at the 3rd.day and 7th day after treatment of the two groups decreased obviously,and those in the observation group were significantly lower than the control group (P ＜ 0.05).Conclusion Ulinastatin combined with Xue Bi Jing injection therapy can conspicuously reduce the IL-8 and γ-IFN levels in patients with severe acute pancreatitis and alleviate the clinical symptom.

AIM: To further investigate the role of PKARⅠ? in the growth-promoting effects of Shuanglong-Jiegu pill (SLJGP), a Chinese medicine, on cultured osteoblasts. METHODS: pcDNA-antiPKARⅠ?, a recombinant expressing the antisense sequence of PKARⅠ?, was constructed and transformed HFOB1.19 by lipofectin. MTT was undertaken to assess the cell growth with the treatment of high dosage of SLJGP containing serum. RESULTS: Antisense gene blocked the growth-promoting effects of SLJGP containing serum on HFOB1.19. CONCLUSION: The function of SLJGP is closely related to cAMP-dependent protein kinase A. [

AIM: To investigate the regulatory effects of nuclear factor-κB ( NF-κB) and activator protein-1 (AP-1) on the expression of ectopic trypsin and proinflammatory cytokines in influenza A virus (IAV)-induced myocardi-tis.METHODS:Male BALB/c mice of 8 weeks old ( n=40) were randomly divided into 4 groups:normal control group ( NC) , infection control group ( IC) , NF-κB inhibitor group ( NI) and AP-1 inhibitor group ( AI) .The mice in NC group and IC group were instilled intranasally with 15μL saline and 40 plaque forming units ( PFU) IAV, respectively.The mice in NI group and AI group were infected intranasally with 40 PFU IAV and injected intraperitoneally with 10 mg/kg NF-κB inhibitor pyrrolidine dithiocarbamate ( PDTC) or 2.5 mg/kg AP-1 inhibitor nordihydroguaiaretic acid ( NDGA) once daily. The mice were euthanized at day 9 after instillation, and the hearts were removed for pathological and biochemical analysis. RESULTS:IAV infection induced significant up-regulation of ectopic trypsin, and proinflammatory cytokines interleukin 6 (IL-6), IL-1βand tumor necrosis factor-α(TNF-α) in the myocardium, and triggered acute myocarditis.PDTC signifi-cantly inhibited NF-κB activation and up-regulation of ectopic trypsin and proinflammatory cytokines, and effectively sup-pressed IAV replication and myocardial inflammatory response (P<0.01).NDGA effectively inhibited AP-1 activity (P<0.01) and mildly suppressed up-regulation of proinflammatory cytokines ( P<0.05) , but had no effects on the expression of ectopic trypsin, IAV replication and the extent of myocarditis ( P>0.05) .CONCLUSION:IAV infection induces up-regulation of ectopic trypsin and proinflammatory cytokines in myocardium predominantly by the activation of NF-κB.AP-1 signaling pathway might be only partially involved in the regulation of proinflammatory cytokines.

Objective To perfect the purification method of recombinant fusion protein of Hespintor (rHespintor) for increasing the protein extraction efficiency,and to investigate its effects on the proliferation,migration and invasion of hepatoblastoma cell line HepG2.Methods In the recombinant protein extraction,the inclusion body washing process was added and the protein purification buffer system was changed.BAPNA was used as the substrate.The inhibitory effect tof purified rHespintor on trypsin hydrolysis was detected.The blank group served as the control group.The MTT test,cell scratch wound healing test and tumor cell invasion test were performed to detect the effect of rHespintor on growth of hepatoblastoma HepG2 cells and its effect.Results The urea gradient washing on the inclusion body protein could effectively remove the vast majority of impure proteins from the targeted protein.After one-step purification,the target protein rHespintor exhibited a high inhibition effect of trypsin hydrolysis,and the inhibitory effect was exhibited a dose-dependent manner.After acting on hepatoblastoma HepG2 cells with rHespintor,the cell proliferation ability was inhibited,the migration ability was reduced and the number of invaded cells were significantly decreased.Conclusion rHespintor can significantly inhibit the proliferation,migration and invasion of hepatoblastoma cell line HepG2 cells in vitro.
