1.Analysis of clinical efficacy of Shenqi Fuzheng Injection in children with sepsis

Objective To observe the clinical effects of Shenqi Fuzheng Injection in treatment of sepsis in children and its effect on the serum levels of high mobility group protein-1 and tumor necrosis factor-α, lactic acid, C reactive protein, procalcitonin levels.Methods 140 patients with sepsis were chosen, and randomly divided into two groups, the observation group and the control group.Two groups were given treatment by the national standard of sepsis, while the control group only received basic treatment, the observation group on the basis of conventional treatment combined with Shenqi Fuzheng injection.Two groups of clinical curative effect and the levels of serum HMG-1, TNF-α, LA, CRP, PCT level were observed. Results After treatment, the observation group total effective rate (95.71%) was significantly higher than control group (70.00%),χ2 =16.29, P<0.01.Two groups before treatment, TNF-αand HMG-1 had no statistically significant difference.After treatment, 2 groups of HMG -1, TNF-αsignificantly lower than before treatment (P<0.01),which HMG-1,and TNF-αreduced significantly in observation group (P<0.01).Serum HMG-1 were positively correlated with TNF-αbefore and after treatment (P<0.05).After treatment, LA, CRP and PCT in 2 groups were significantly different compared with before treatment (P<0.01), and the observation group decreased significantly than control group (P<0.01).Conclusion The effect of Shenqi Fuzheng Injection in the treatment of children with sepsis is significant, which is worthy of clinical application.
2.A Study on the Relationship between Fatty Liver and Body Mass Index (BMI), Serum Lipid Levels in Old Retired Population

Objective To explore the relationship between fatty liver and body mass index (BMI), serum lipid levels in aging population. Method 748 of retired people (≥45 years) were studied. All participants accepted liver B -mode Ultrasonography examination, height, weight and blood lipid detection. The relationship between Fatty Liver and BMI, serum lipid levels were analyzed. Results The detection rate of Fatter Liver in male and female was 61.8% and 44.6% separately. The detection rate of Fatter Liver was 53.4% in the aging group (45~59 years), while the rate was 51.0% in elderly group (≥60 years). Compared to the 23.07 kg / m2 and 1.58 mmol/L in group without fatty liver, the BMI and serum triglyceride was 26.77 kg / m2 and 2.13 mmol / L in the group suffered the Fatter Lever. Objetive:To detect the gene mutation of a patient with anhidrotic ectodermal dysplasia. Methods: Genomic DNA was extracted from the peripheral blood of the patient and all the exon fragments were obtained by PCR. These fragments were sequenced directly. Results: In EDA gene a novel base transition was detected: -48A→G. Conclusion: There is a new base transition in EDA gene in this patient

Objective:To study the expression of human Era.Methods:The encoding region of human Era gene was inserted into pUC19 plasmid and sequenced,the gene fragment was then cloned into the prokaryotic fusion expression vector pRSET B to contruct recombinant expression plasmid pRSET B hEra.The recombinant protein was expressed in E.coli BL21DE3(plys S).Results:6His hEra was expressed after induced by IPTG and took 51.2% of the total cell lysate and mainly existed in the inclusion body.Conclusion:High level of human Era was established and this result may shed light on the further study on function of human Era.
7.Expression and significance of interleukin-1β in serum of acute myeloid leukemia

Objective The interleukin-1β (IL-13) expression in serum of acute myeloid leukemia (AML) patients was evaluated. To explore the significance of IL-1β in leukostasis and tissue infiltration by leukemic cells. Methods ELISA was used to investigate the contents of IL-1β in serum of 83 newly diagnosed with AML which contains 16 hyperleukocytic AML patients, and compared the IL-1β level between the hyperleukocytic AML group and non-hyperleukocytic AML group, the infiltrated group and non-infiltrated group. Results The content of IL-1β in AML serum [(88.23±36.30) pg/ml] was higher than that of in the control group[(29.56±15.53) pg/ml], with significant difference (P <0.01). There was a higher level for IL-1β in hyperleukocytic AML group[(136.67±65.68) pg/ml] than in non-hyperleukocytic AML group [(69.85±48.35) pg/ml],and there was a significant difference. IL-13 and peripheral blood cells were in linear correlation (r=0.74, P <0.01). There was a higher level for IL-1β in infiltrated group[(111.31 ±57.35) pg/ml] than in the other group [(79.68±43.42) pg/ml], and there was a significant difference. Conclusion The IL-1β may play an important role in leukostasis and tissue infiltration by leukemic cells in AML.

Pancreatic cancer is characterized by high invasiveness,high malignancy and poor prognosis. Therefore,it is urgent to find biomarkers for early diagnosis and to develop targeting drugs for treatment of pancreatic cancer. MicroRNAs (miRNAs)are single-stranded,non-coding small RNA molecules with a length of 18-25 nucleotides and are highly expressed in pancreatic cancer. MiRNAs play important roles in cell proliferation,differentiation,apoptosis,invasion and metastasis,and are associated with adverse prognosis and resistance to chemotherapy in pancreatic cancer. As a new target for diagnosis and treatment of pancreatic cancer,miRNA-21 has become the hot spot of clinical research. This article reviewed the advances in studies on miRNA-21 in the pathogenesis,diagnosis and treatment of pancreatic cancer.
9.System Review of Acupotomy Treatment for Stenosing Tenovaginitis of Flexor Digitorum

Objective To systematically evaluate the efficacy of acupotomy treatment for the stenosing tenovaginitis of flexor digitorum.Methods Articles about RCTs of acupotomy treatment and local blocking for the treatment of stenosing tenovaginitis of flexor digitorum were retrieved from CBM, CNKI, VIP, Wanfang database, PubMed, Embase, and Cochrane library. And then according to the requirement of Cochrane Handbook for Systemetic Reviews 5.1.0, two reviewers independently chose tests, extracted data, had a cross check, accessed methodological qualities and finally used RevMan 5.3 software for Meta analysis. The total effective rate, cure rate and improvement rate of acupotomy treatment and local blocking in treating stenosing tenovaginitis of flexor digitorum were assessed by using odds ratio (OR).Results Totally 10 articles involving 1426 cases were included. The cure rate in acupotomy treatment group was higher than that of local blocking group [OR=13.11 (95% CI: 8.23–20.89),Z=10.83 (P<0.000 01)], funnel plot Begg’s testP=0.858, Egger’s testP=0.579; the improvement rate of local blocking rate was higher than that of acupotomy treatment group [OR=0.14 (95% CI: 0.10–0.19),Z=11.60 (P<0.000 01)], funnel plot Begg’s test P=1.000, Egger’s testP=0.926; the total effective rate of acupotomy treatment group was higher than that of local blocking group [OR=18.26 (95% CI: 9.95–33.50),Z=9.38 (P<0.000 01)], funnel plot Begg’s testP=0.592, Egger’s testP=0.936.Conclusion Acupotomy treatment for stenosing tenovaginitis of flexor digitorum is superior to local blocking injection. However, the quality of the RCTs is low, so more large-sample, multi-center, and high-quality clinical RCTs are required for further verification.

Objective To study the effects of 4 1BBL on antitumor immunity induced in vivo by murine 4 1BBL gene transfected hepatocellular carcinoma cell line Hepa1 6. Methods Eukaryotic expression vector pcDNA3 1(+) m4 1BBL was transfered into murine hepatocellular carcinoma cell line Hepa1 6 via lipofectamine method. The transfected cells were selected in RPMI1640 containing G418(400~800 ?g/ml) and termed as Hepa1 6 m4 1BBL.The TCV m4 1BBL was obtained by treating with mitomycin (MMC). Three models (immunological model, early model, and later model) were established for the study of anticancer effects of TCV m4 1BBL. Results The syngeneic mice were completely protected and could survive free from tumor for a long period by inoculation with TCV 4 1BBL. In early models, TCV 4 1BBL showed strong immunotherapy effects( P 0 05). Conclusion The antitumor effect against syngeneic murine hepatocellular carcinoma cell line in vivo is significantly enhanced by the treatment with TCV 4-1BBL.
