1.The Toxic Effect of Bilirubin on Different Types of Neurons of Rat in Primary Culture

Objective To investigate cytotoxicity and its mechanism of bilirubin on different types of neurons of rat in primary culture. Methods Toxic effect of bilirubin on primary cultured neurons (at cerebellar oranule hippocampal and cortical neurons) was observed. Ncuron survival was detected by fluorescei diacetate (FDA) staining. Measurement of fluorescent intensity of intracellular dilirudin were performed by microspectroflurimetry imaging system. Results Biliruin induced cytotoxicity to different type neurons in a dose-dependent manner. Exposed at 8d to different concentrations of bilirubin (4.3 ?mol/L、8.6 ?mol/L and 17.1 ?mol/L); cerebellar gtanule neuron survival werc (50.1?3.1)%,(18.5?2.3)%, (2.0?2.4)%, cortical neuron survival were (90.7?7.8)%, (85.3?5.3)%,(74.5?2.4)%, and hippocampal neuron survival were (75?6)%, (62?5)%, (49?5)% respectively. The toxic effect of bilirubin on ccrebellar granule neuron was significantly higher than that on hippocampal and cortical neurons. The fluorescent intensity of bilirubin in gra- nule, hippocampal and cortical neurons were 33.4?2.3、10.7?3.0、21.9?4.3 respectively three hours after addition of bilirubin to the medium. 2.Changes of bone mineral density in type 2 diabetes mellitus with peripheral vessel disease

Objective To investigate the changes between type 2 diabetes mellitus(T2DM) with peripheral vessel disease and osteoporosis. Methods A total of 100 cases T2DM patients were divided into non-peripheral vessel disease (A group) and peripheral vessel disease (B group). B-mode ultrasonography to measure the intima-media thickness(IMT) of carotid artery,dual energy X-ray absorptiometry (DEXA) to measured bone mineral density(BMD) and VS-1000 arteriosclerosis diagnometer to measure brachi-ankle pulse wave velocity (baPWV) were used in 100 T2DM patients. The clinical data and BMD were compared between two groups. Results Compared with normal group,the BMD in T2DM patients significantly decreased.Compared with T2DM patients without peripheral vessel disease,the BMD decreased in T2DM patients with peripheral vessel disease. Conclusions Atherosclerosis in T2DM with peripheral vessel disease is probably related to osteoporosis, and the relationship between them is worth further investigation.
3.Adhesion of bone marrow stromal cell in children with acute lymphoblastic leukemia

AIM and METHOD: The adhesion behavior of bone marrow stromal cells(BMSC) in children with acute lymphoblastic leukemia(ALL) to bone marrow mononuclear cells(BMMC) and Raji cells were investigated by MTT method. The expression of adhesion molecules ICAM-1 and VCAM-1 were detected by flow cytometry. RESULTS: The adhesion ability of BMSC in acute period of ALL to BMMC was lower than that of control group. The adhesion ability of BMSC in long term remission period of ALL to Raji cells higher than that of control group. The expression of ICAM-1 on the surface of BMSC in acute period of ALL is much lower than that of control group. CONCLUSIONS: The adhesion of BMSC to BMMC or tumor cells in children with ALL was abnormal. The abnormal adhesion behavior might be partially due to the changed expression of adhesion molecules on BMSC.
