1.Gene expression of 5?-reductase (type 2) in reproductive gland of pubertal diabetic rats

AIM: To study the molecular mechanism of reproduction dysfunction in pubertal diabetic rat, the level of 5?-reductase (type 2) mRNA in testis, epididymis and prostate in diabetic rat was detected. METHODS: The gene expression of 5?-reductase (type 2) was detected by Northern blot. RESULTS: In the caput of the epididymis, the expression of 5?-reductase (type 2) mRNA of D and ID groups was less than that of C group. CONCLUSION: The decreased expression of 5?-reductase (type 2) results in the decreased production of dihydrotestosterone, which influences the development and function of reproduction system in pubertal rats. 2.Auditory Brainstem Responses with Forward -Masking in Newborns

Objective To study the characteristics of auditory brainstem responses with different forward - masking intervals in newborns with normal hearing ,and to find out the most efficient interval .Methods Auditory evoked brainstem responses were recorded from 30 newborns(60 ears) ,with forward - masking intervals of 4 ms ,8 ms ,16 ms ,32 ms ,and unmasked as control .The ABR data were statistically analyzed with SPSS 13 .0 .Results With forward - masking intervals ,all waves Ⅰ ,Ⅲ ,Ⅴ latencies and Ⅰ - Ⅴ ,Ⅰ - Ⅲ ,Ⅲ - Ⅴ interpeak latencies were significantly (P< 0 .01) prolonged compared with those of in the unmasked ,except wave Ⅲ and Ⅰ - Ⅲ interpeak latencies with 4 ms forward - masking intervals .The waves from 8 ms interval were the most prolonged .Conclusion Forward masking increases human newborn ABR latencies and interpeak latencies .The interval time influenced the effect ,and 8 ms interval is the most efficient .
4.Clinical application of purse?string suture on the treatment of patients with peptic ulcer bleeding

Objective To study the clinical efficacy and safety of purse-string suture on the treatment of peptic ulcer bleeding. Methods Data of 42 patients with peptic ulcer bleeding treated by purse-string suture with hexiejia combined with endoloop in Dongguan Tung Wah Hospital from January 2016 to January 2017 were retrospectively analyzed. The immediate hemostasis rate, effective hemostasis rate, additional surgery rate, recurrence rate in two weeks and mortality were analyzed. Results Among 20 cases with active bleeding during operation, 19 were immediately arrested, and 1 case of failure was diagnosed as marginal ulcer after gastric resection. The overall effective hemostasis rate was 97. 6%(41/42), additional surgery rate was only 2. 4%( 1/42). The vital signs of 41 patients with successful endoscopic hemostasis were stable after operation, and no fever, hematemesis, black stool or other symptoms occurred. No recurrence of bleeding or death occurred within two weeks. The ulcers were healed well in follow-up of one month. Conclusion Endoscopic purse-string suture is a safe and feasible method for treatment of patients with peptic ulcer bleeding.
5.Study on status and influencing factors of disease knowledge among patients with ulcerative colitis

Objective To investigate the knowledge level among patients with ulcerative colitis, and analyze its influencing factors. Methods A total of 147 patients with ulcerative colitis were recruited by using Chinese version of inflammatory bowel disease knowledge questionnaire to assess their knowledge level. The influencing factors were explored by using general information questionnaire and self-efficacy questionnaire of inflammatory bowel disease. Results The total score of disease knowledge among patients with ulcerative colitis was (6.58 ± 1.57) points, Correct rate of disease knowledge was 28.6%; the total score of self-efficacy was (227.45 ± 32.25) points, the score index was 78.28%. The educational level, duration of disease, patients′ age and management of medical and nursing were the influencing factors of disease knowledge, which could explain 44.3% of total variation. Conclusions The knowledge level among patients with ulcerative colitis needs to be improved.Nursing staff should be based on influencing factors, pay attention to improve the knowledge level of patients with nursing intervention or health education,through a variety of ways to meet the patients need of disease knowledge, improve theirquality of life.
6. Mediating effect of self-efficacy and colleague solidarity on innovative behavior and career success of nurses

Objective:
To investigate effects of personal innovation behaviors, self-efficacy and colleague solidarity of nurses on career success, and to provide the basis for career planning and management of clinical nurses.
Methods:
A total of 441 registered nurses from eight grade three and first-class hospitals in Tianjin were measured with personal innovation behaviors scale, self-efficacy scale, colleague solidarity of nurses' scale and career success scale.
Results:
The total score of personal innovation behavior was (21.58±4.81). The total score of self-efficacy was (27.68±6.53). The total score of colleague solidarity of nurses was (95.12±9.42). The total score of career success was (35.75±9.27). The personal innovation behaviors, self-efficacy, colleague solidarity of nurses career success and each dimension showed positive correlation(
7.Curcumin attenuates IL-1β-induced chondrocyte damage by modulating the DUSP1/p38 MAPK pathway

Objective:
To investigate the inhibitory effect of curcumin(Cur) on IL-1β-induced cartilage damage and to study the relationship between the regulatory mechanisms of the DUSP1/p38 MAPK signalling pathway in the above process.
Methods:
Chondrocytes(C28/I2) and postoperative primary chondrocytes from osteoarthritis patients were divided into control and experimental groups, and the experimental group was treated with different concentrations of Cur(0, 10, 20, 40, 60, 80 μmol/L) after applying the inflammatory induction treatment with IL-1β(10 μg/L). The cell proliferation inhibition rate was determined by cell viability assay(CCK-8), the apoptosis rate was detected by flow cytometry assay. Real-time fluorescence quantitative PCR(qRT-PCR), Western blot, and immunofluorescence assay were used to detect type II collagen α1 chain(Collagen Ⅱ), matrix metallopeptidase 13(MMP13), interleukin-1β(IL-1β), BCL2-related X protein(Bax), B lymphocytoma-2(Bcl-2), dual-specificity phosphatase 1(DUSP1), p38 mitogen-activated protein kinase(p38), and phosphorylated p38 mitogen-activated protein kinase(p-p38) RNA and protein expression levels. The role of the DUSP1/p38 MAPK axis in the inhibition of chondrocyte oxidative stress, apoptosis and inflammation by Cur was further validated using DUSP1 interfering RNA and p38 MAPK pathway inhibitor(SB).
Results:
Cur significantly inhibited the IL-1β-induced decrease in chondrocyte viability and significantly reduced the levels of oxidative stress, apoptosis, and inflammation in chondrocytes; Cur inhibited the expression of MMP13, IL-1β, Bax, and p-p38 proteins, while the expression of Collagen II, Bcl-2, and DUSP1 proteins significantly increased; IL-1β and interfering RNA silencing DUSP1 activated the p38 pathway, while Cur inhibited the activation of the p38 pathway; the use of p38 MAPK pathway inhibitors reduced cellular inflammation.
Conclusion
Cur attenuates IL-1β-induced oxidative stress, apoptosis and inflammation in chondrocytes by promoting the expression of DUSP1 protein and inhibiting the activation of p38 MAPK pathway.
