Journal of Chongqing Medical University 2007;0(08):-
Objective:To explore the mechanisms of ERK in affecting estrogen receptor signaling pathway by investigating the changes of the expression levels of nuclear receptor co-activatorPCAF protein and wild-type P53 protein and their gene transcription in the process of estrogen promoting transformation of cell cycle and resisting apoptosis of breast cancer cell MCF-7 after inhibitting ERK. Methods:Estrogen receptor-positive breast cancer cells MCF-7 were divided into 17?-estradiol treatment group,17 ?-E2 + ERK inhibitor PD98059 treatment group, and the control group. The apoptosis of cell and cycle were analyzed by flow cytometry. The expression of phosphorylated ERK1/2 and PCAF protein and wild-type p53 were detected by Western blot. The expression of pcaf mRNA and wild-type p53 mRNA were detected by RT-PCR. Results:With phosphorylation inhibitor of ERK PD98059,the effect of 17?-estradiol in resisting apoptosis and prmoting transformation of MCF-7 cell cycle was reversed, and the rate of early apoptosis of MCF-7 cell was raised(P0.05).The protein expression level and gene transcription of wild-type P53 were reinforced (P

Chinese Journal of Tissue Engineering Research 2016;20(21):3150-3155
BACKGROUND:Because of poor biocompatibility, conventional intraocular lenses (IOLs) used to improve visual acuity can result in a higher incidence of inflammatory foreign body reactions.
OBJECTIVE:To study the biocompatibility and therapeutic effects of hirudin peptide-modified acrylic IOL on eye diseases.
METHODS:By immersion coating method, we prepared a polymethylmethacrylate IOL by the modification with hirudin polypeptides and observed its biocompatibility through the cytological observation. In this study, six New Zealand white rabbits were equivalently randomized into conventional and hirudin peptide grouppolypeptide hirudin group. Rabbits in the conventional group were given conventional lens implantation; while those in the hirudin peptide group underwent hirudin peptide-modified polymethylmethacrylate IOL.
RESULTS AND CONCLUSION:Three days after implantation of hirudin peptide-modified polymethylmethacrylate IOL, cels proliferated and spread wel on the acrylic IOL, but on the cel membrane, there were a few cels most of which were gathered sphericaly with no spreading. In the hirudin peptide group, some cels attached to the border zone migrated to the IOL surface, but did not impede the central visual field; in the conventional group, the IOL surface was stil covered by fibrotic cel membrane.Comparedwith the conventional group, the score in the SchirmerItest was increased but the breakup time of tear film decreased in the hirudin peptide group. To conclude, the polymethylmethacrylate IOL modified by hirudin peptides has good physicochemical properties and biocompatibility, increases visual acuity, and yields satisfactory outcomes in the cataract treatment, which provides a new insight into the treatment of cataract.

Objective:To investigate the role of extracellular signal-regulated protein kinase(ERK) in estrogen-induced proliferation and cell cycle transformation of breast cancer cell line MCF-7 and the related mechanisms. Methods: Estrogen receptor-positive breast cancer cell line MCF-7 was used in our study. The effects of 17?-E2 on the proliferation of MCF-7 cells was investigated by MTT assay to determine the optimal concentration of 17?-E2 for the following experiment. The effect of PD98059 on 17?-E2-induced proliferation of MCF-7 cells was measured by MTT assay to determine the intermediate concentration of PD98059.The cell cycle was analyzed by flow cytometry and telomerase activity was determined by Telomerase repeat amplification protocol PCR(TRAP-PCR) silver staining. The expression of wild-type p53 and phosphorylated ERK1/2 protein was determined by Western blotting and the expression of wild-type p53 mRNA was detected by RT-PCR.Results: ERK phosphorylation inhibitor PD98059 inhibited the proliferation of MCF-7 cells treated with 17?-E2 in a time-and dose-dependent manner(P

Objective To analyze and evaluate the 48 selected highly polymorphic SNP loci in Chinese Han population on genetic and forensic aspects. Methods Samples from 200 unrelated Han individuals in East China were collected.48 X-SNP polymorphic genetic markers were selected according to the information providedby NCBI and HapMap,and then were typed by SNPlex~(TM) System for establishing genetic data. Results The 48 X-SNP loci were highly polymorphic markers in East China Han population except rs6527549,and their polymorphism information contents(PIC)were all above 0.32,the discrimination power(DP)in females and males were above 0.56 and 0.40 respectively, the probability of exclusion(PE)in duos and trios were above 0.20 and 0.32 respectively. In addition, linkage disequilibriums were observed among some loci. Conclusion The 48 X-SNP loci union is suitable for developing high-throughout automated research and also useful for special parentage testing.

Cancer Research and Clinic 2010;22(z1):32-35
Objective To study the expression of POKemon and pl4ARF genes in colorectal cancer cell lines. Methods The expressions of POKemon and pl4ARF were detected in 5 human colorectal cancer cell lines by RT-PCR and immucytochemistry method. Results The expressions of POKemon were positive in cell lines of SW480, SW480/M5, SW620 and LOVO, and were negative in HCT116. The expression of pl4ARF was positive in HCT116 and negative in other cell lines. Conclusion POkemon is expressed positively in colorectal cancer cell lines and its function is negatively correlated with pl4ARF.

Objective To find the best cut point of FPG to predict diabetes(DM) and impaired glucose tolerance(IGT) and to study its effects on the metabolism status. Methods The ROC analysis of FPG in 3189 citizens from some area of Chongqing without a previous history of diabetes was done by OGTT in a cross-section study. The metabolic features of impaired glucose regulation(IGR) and its subcategories were analyzed according to 5.6 mmol/L and 6.1 mmol/L as the FPG threshold for IFG.Results The areas under the ROC curve were 0.899 for diabetes and 0.728 for IGT.The cut point of FPG with the best equilibrium between sensitivity and specificity was 5.6 mmol/L for diabetes and 5.2 mmol/L for IGT.Among people whose 2 hPG

Objective To investigate the effects of chloroquine on metabolism of insulin, glucose, lipids and expression of insulinase gene (EIG) in patients with type 2 diabetes mellitus. Methods Fasting plasma insulin (FINS),fasting plasma glucose (FPG),glycoslated hemoglobin (HbA1c),plasma lipids and lipoprotein, insulinase activity of erythrocytes (EIA) and EIG were determined in 30 patients with type 2 diabetes mellitus laking chloroquine for 14 day in dosage of 250mg twice daily,27 diabetic patients laking placebo and 20 normal subjects.Insulin sensitive index (ISI) were also calculated. Results Chloroquine caused a decrease in total plasma cholesterol, low density lipoprotein,EIA and EIG,and an increase in ISI,plasma high density lipoprotein and subclass 2.No change of these indices was observed in diabetic patients with placebo. Conclusion Chloroquine can ameliorate the dyslipidemia and insulin sensitivity of patients with type 2 diabetes mellitus. This may be due to a decrease in EIG,which may result in lowered degradation of insulin.
