2.Essential role of thyroid hormones in brain developm ent
Chinese Journal of Endocrinology and Metabolism 2001;0(05):-
Thyroid hormones (TH) play an imp or tant role in the development and maturity of the mammalian brain. Maternal TH de ficiency, even subclinical hypothyroidism, leads to poor neurodevelopment of the fetus. Screening of pregnant women for hypothyroidism is recommended. Identific ation of the TH-responsive genes in the developing brain and investigation on t heir functions are helpful to provide insights into the molecular mechanisms by which TH influence brain development.

Objective To investigate the effect of continuous renal replacement therapy (CRRT) on the function of monocyte in patients with severe acute renal failure (ARF).Methods The function of monocyte in 42 patients with ARF accepting CRRT was dynamically observed and compared with that of healthy people. Results Cytokine secreting function of monocyte in patients with ARF was stronger than that in healthy people[TNF-α:(1 212.61 ±107.43) vs (857.34 ±83.45) ng/L,IL-6:(2 518.22 ±228.22) vs (1 948.13 ±218.23) ng/L,IL-8:(416.18 ±54.46) vs (229.31 ± 39.83) ng/L] (t =6.47,7.09,6.45,all P < 0.05) 48 h after CRRT,cytokine secreting function declined to normal. Conclusion CRRT corrects the disorder of cytokine secreting function of monocyte in patients with ARF,decreases the inflammation response.

Abstract Objective:To observe and compare the effects of immune responses of HBV DNA caccine by insertion IL-2 gene.Try to ex-Plore the efficient way to enhance the immune effect of DNA vaccines. Methods:The recombinant expression plasmid vector peDNA3.1-S/IL-2with fusion gene of HBsAg and IL-2 cloned was constructed by PCR method and DNA recombinant technique. Then, it was transfered into Cos-7 cells by cation lyposomes and was detected its transient expressing product in vitro. At last, observed and compared the cellular and humoralimmune responses to HBsAg in C57BL/6 mice by intramuscular injection. Results:The pcDNA3. 1-S/IL-2 was constructed successfully by themethod of restriction endonucleases digestion, RCR and DNA sequencing. It can increase the titers of anti-HBs antibody. The bioactivity of IL-2and lymphocyte proliferation responses to HBsAg induced by the mouse spleen immunized with psDNA3.1-S/IL-2 were improved also. Conclu-sion:These results showed the expression of HBsAg-IL-2 fused gene can enhance the immune responses and suggested the insertion of IL-2 maybe a practicable way to enhance the DNA vaccine efficacy.

Chinese Journal of Endocrinology and Metabolism 1986;0(03):-
The effect of 17?-estradiol ( 17?-E2 ) on expression of estrogen receptor ?( ER?) was observed in cultured MC63 cells and human osteoblast-like (HOB) cells. The results show that the expression of ER* is in a dose-dependent manner with 17p-E2 (0-1?10-6 mol/L) in MC63 cells and has an optimal 17?-E2 concentration (1?10-8 mol/L) in HOB cells resulting in maximum expression of ERp protein.

Chinese Journal of Schistosomiasis Control 2014;(1):103-105
Objective To evaluate the long-term therapeutic effect of Ruangan pills on advanced schistosomiasis combined with ascites. Methods The data of 54 advanced schistosomiasis patients with ascites were collected, and the patients were divid-ed into two groups namely a treatment group and a control group according to whether taking Ruangan pills. The effective rates,im-provement status of symptoms and levels of serum albumin(ALB)and hyaluronic acid(HA)of the two groups were compared. Re-sults The effective rates of the treatment group and control group were 92.59%and 44.44%,respectively,and the difference be-tween them was statistically significant(P<0.05). After the treatment for 9 and 12 months, the percentages of patients without as-cites and patients with symptom improvement in the treatment group were 77.78%and 92.59%,92.59%and 96.30%,respective-ly,while those in the control group were 29.63%and 37.00%,48.15%and 51.85%,respectively,and the differences between the two groups by different time were all statistical significant(all P<0.05). After the treatment for 9 months, the percentages of patients with the normal levels of ALB and HA were 88.89% and 59.26%,while those in the control group were 40.74% and 14.81%,respectively,and the differences between the two groups were statistically significant(both P < 0.05). Conclusion The long-term treatment of Ruangan pills can not only improve clinical symptoms but also control the ascites recurrence, however, the therapeutic effect and the recurrence rate of ascites in longer-term still need further observation.

Journal of Chinese Physician 2001;0(10):-
Objective The recombinant fluorescent eukaryotic expressing vector containing hTERT cDNA was transfected into human embryonic fibroblasts (hEFs) to explore the effects of exogenous hTERT on the type I and III collagens expression in hEFs. Methods p IRES2-EGFP-hTERT plasmid and pIRES2-EGFP plasmids were transfected into primary hEFs respectively by Lipofectin reagent. Expression of type I and III collagen was determined by Western blotting and the content of type I and III collagens in the cellular medium at 3 days after transfection were examined by radio-immunoassay. Results The expression levels of type I and III collagens in hTERT gene transfected hEFs(hEF-hTERT) were obviously higher than those in untransfected hEFs and vacant vector transfected hEFs(hEF-EGFP). The content of type I and III collagens in the cellular medium in hEF-hTERT cells at 3 days after transfection was also higher than that in untransfected hEFs and hEF-EGFP cells. Conclusions The synthesis ability of type I and III collagens in hEFs could be promoted by exogenous hTERT gene transfection.

OBJECTIVE To investigate and elucidate the role of histamine on the regulation of tight junctions(TJs) gene expression of the airway epithelial cells. METHODS We cultured primary human bronchial epithelial cells from healthy and asthmatic individuals in the presence of histamine for 96 hours in air-liquid interface (ALI) culture system. The TJs expression on the level of mRNA and protein was determined by RT-qPCR and immunofluorescence (confocal microscopy), respectively. RESULTS A RT-qPCR analysis revealed that the mRNA expression of several TJs were up- or down-regulated by histamine. However, examination by confocal microscope revealed no detectable alterations in the morphology of the tight junctions after stimulating with histamine. CONCLUSION Our findings that histamine could regulate the mRNA expression of TJs of bronchial epithelial cells. That might contribute to the pathogenesis of chronic airway inflammatory diseases.
