2.Determination of Vitamin D_2, Vitamin A and Vitamin E in Cernevit-12 for Injection by HPLC

OBJECTIVE: To establish a HPLC method for the simultaneously determination of Vitamin D2, Vitamin A and Vitamin E in Cernevit-12 for injection. METHODS: An Inertsil(ODS-2) C18 column was selected as separation column at 30℃. The mobile phase consisted of acetonitrile-methanol-methylene chloride (80∶10∶10) at a flow rate of 1.0mL?min-1. The detection wavelength was set at 265nm and the injection volume was 10?L. RESULTS: The linear ranges of Vitamin D2, Vitamin E acetic ester and Vitamin A palmitate were 0.61~1.43?g?mL-1, 222.6~519.4?g?mL-1 and 1.21~2.82mg?mL-1 respectively. The average recoveries were 99.79%, 99.53% and 101.63%, with RSD of 1.66%, 0.184 6%, 0.113 4%. CONCLUSION: The method is simple, rapid and accurate, and suitable for the determination of Cernevit-12.
3.Micro-pulse laser therapy for acute central serous chorioretinopathy

Objective To observe the effect of micro-pulse laser in the treatment of acute central serous chorioretinopathy (CSC).Methods 105 cases (114 eyes) with clinically diagnosed acute CSC were included in the study,including 78 males (86 eyes) and 27 females (28 eyes) with an average age of (40.40±7.80) years,and mean duration of 26 days.All patients were examined for best corrected visual acuity (BCVA),slit lamp microscope and pre-lens,direct ophthalmoscopy,color fundus photography,fluorescein angiography combined with indocyanine green angiography (ICGA),and micro-perimetry.Patients were randomly divided into micro-pulse laser treatment group (treatment group,61 eyes) and control group (51 eyes).The age (Z =-0.374),gender ratio (x2=0.010),disease duration (Z=-0.525),BCVA (t=1.885),foveal thickness (CFT) (t=-1.754) and macular light sensitivity (t=1.255) were similar between the two groups.The micro-pluse laser treatment was performed with an 810 nm infrared diode laser at the active leakage site on retinal pigment epithelium guided by ICGA.The exposure time was 0.2 s,effective working time was 15%,the laser spot diameter was 100 μm,and the distance between 2 spots was 100 μm.The control group received pseudo-treatment using the same laser parameters.After 2 weeks,1month,3 months and 6 months of treatment,all patients were examined with BCVA,ocular fundus,optical coherence tomography and micro-perimetry.These parameters were compared between the 2 groups.Results The subretinal fluid of 39 eyes in the micro-pulse laser group and that of 3 eyes in the control group were absorbed completely.The cure rates in the micro-pulse laser group (61.9%) was higher than that in the control group (x2 =38.015,P<0.01).In the micro-pulse laser group,the mean BCVA was 67.81±11.70 at baseline,which increased significantly to 75±9.91,76.78± 9.43,78.56 ± 8.57 and 78.52 ± 8.60 at 2 weeks,1 month,3 months and 6 months after treatment respectively.In the control group,that was 63.86±10.35,64.20±11.43,63.90±10.88,64.55±11.04,64.10±11.12 at baseline,2 weeks,1 month,3 months,6 months post-treatment respectively.The mean post treatment BCVA at each time point were significantly higher in the micro-pulse laser group than that in the control group (P< 0.01).The mean CFT was (380.94 ± 50.73) μm at baseline,which reduced to (268.44±44.20),(242.78±41.31),(235.46±38.44),(235.56±38.71) μm at 2 weeks,1 month,3 months and 6 months post-treatment respectively in the nmicro-pulse laser group.In the control group,that was (397.98±52.61),(334.55±59.15),(316.16±55.25),(314.47±53.27),(321.51±55.74) μm at baseline,2 weeks,1 month,3 months,6 months post-treatment respectively.The mean post-treatment CFT at each time point in the micro-pulse laser group were decreased significantly compared to that in the control group (P<0.01).The mean central retinal sensitivity was (15.03 ± 2.00) dB at baseline,which enhanced to (17.06 ± 1.71),(17.37 ± 1.61),(17.56 ± 1.58),(17.48 ± 1.53) dB at 2 weeks,1 month,3 months and 6 months post-treatment respectively in the micro-pulse laser group.In the control group,that was (14.54±2.22),(14.80±2.16),(14.88±2.09),(14.82±2.07),(14.69±2.11) dB at baseline,2 weeks,1 month,3 months and 6 months post-treatment respectively.The mean central retinal sensitivity at each time point of post-treatment in the micro-pulse laser group were enhanced significantly compared to that in the controlled group(P<0.01).Conclusion In the treatment of acute CSC,micro-pulse laser can improve BCVA,reduce the mean CFT and improve the mean central retinal sensitivity.It is an effective and safe method to treat acute CSC.
4. Effects of abstinence on locomotor activity and contextual fear conditioning in morphine-dependent mice

Objective: To study the impact of different abstinence periods on locomotor activity and contextual fear conditioning in morphine-dependent mice. Methods: Morphine hydrochloride was administrated (sc) to mice at a gradually increasing dose for 7 d to establish morphine-dependent model. After the last injection, animals were divided into MW1 d (Morphine Withdrawal for 1 d), MW7 d, MW21 d, SW1 d (Saline Withdrawal for 1 d), SW7 d and SW21 d groups randomly; the spontaneous activities of mice, including the numbers of crossing and rearing, grooming times, the number of center entries and time spent in center area, and the time spent in peripheral area, were observed in the open field box after different periods of abstinence. After the observation, the mice were trained using eight unconditioned stimulus (0.5 s; 0.5 mA foot shock) and contextual fear conditioning was tested 24 h later. Results: The times of crossing and center entries decreased in MW1 d and MW7 d mice (W1 d: P<0.001, P<0.001; W7 d: P<0.001, P<0.05); the time of rearing in MW1 d group was also decreased significantly (P<0.001); mice in MW1 d group had more grooming behaviors (P<0.05). The periods spent in the center and peripheral areas were not significantly different between all the 3 morphine-dependent groups and their corresponding saline withdrawal groups. In addition, a history of morphine injection selectively impaired the contextual fear conditioning in mice of MW1 d group (P <0.05). Conclusion: During the early period of morphine abstinence, mice have a decreased spontaneous activity and an impaired contextual fear conditioning.

Objective To study the regulating roles of PAI 2 in the differentiation mechanism of the human epidermis. Methods Human skins were take from the early, middle and late human embryos respectively and observed with immunocytochemistry and in situ hybridization techniques. The cell culture and dot blot were also used in the observation of the materials from late embryo. Results 1 PAI 2 exhibits a very high experssion in the development of embryonic period, with the highest level in the middle embryonic phase while the transcripts of PAI 2 still keep a rather high level in the late human embryonic stage. 2 PAI 2 is mainly localized in the superficial, more differentiatied layers of the epidermis.3 PAI 2 is localizated in peripheral cytoplasm of the vitro or vivo keratinocyte.Conclusion PAI 2 is involved in the regulation of the keratinocyte differentiation. lschemic postconditioning(IPo)is a way that after long ischemia on liver graft,animals are given one or several brief reperfusion-ischemia before persistent reperfusion to improve the hepatic tolerance and relieve the ischemie reperfusion injury.It has been proved an effective and controlled method to attenuate the ischemic reperfusion injury.Protective mechanism of ischemic postconditioning on hepatic graft is related with protecting sinus hepaticus endotheliocyte and hepatic microcirculation,relieving hepatic cells injury and inflammatory reaction induced by oxygen free radicals,relieving calcium ovedoad in hepatic cells and mitochondria,regulating apoptosis genes,transforming ion channels condition in mitochondria.This article will makes a brief review on protective mechanism ofhepatic ischemic posteanditioning.
8.Anti-inflammatory, Analgesia and Antipyretic Effects of Chaige Qingre Granules

To investigate the anti-inflammatory, analgesia and antipyretic effects of Chaige Qingre granules. Meth-ods:Kunming mice were used to make auricle swelling model by dimthylphenanthrene, and the auricle swelling degree was measured to test the anti-inflammatory effect of Chaige Qingre granules. Mice were used to make pain models by acetic acid, and the body twis-ting of the mice was counted to observe the analgesia effect of the granules. The fever model in rats was induced by yeast, and the body temperature of the rats was measured to investigate the antipyretic effects of the granules. Results:Chaige Qingre granules could relieve the auricle swelling degree significantly, and the inhibition rate was 46. 3% in the high dose group. High and middle dose groups could reduce the body twisting frequency with the inhibition rate of 51. 1% and 61. 8%, respectively. Every Chaige Qingre granules group could lower the body temperature after one hour of the administration, and the temperature could be reduced 0. 56 degrees Celsius in the high dose group. Conclusion:Chaige Qingre granules can effectively inhibit auricle swelling degree, relieve pain and reduce heat.
9.Pharmacokinetics and Serum Fingerprints of Decoction for Puerperal Blood Stasis

OBJECTIVE:To study the pharmacokinetics and serum fingerprints of Decoction for Puerperal Blood Stasis at different time.METHODS:The content of Ferulaic acid(FA) was determined by HPLC.The pharmacokinetic study was carried out.The active constituent in the Decoction for Puerperal Blood Stasis was analyzed based on the serum fingerprints of FA.RESULTS:The distribution and metabolism of FA in rabbits after oral administration of Decoction for Puerperal Blood Stasis assummed two compartment model.The fingerprints between serum at different time and blank serum showed significant differences.CONCLUSION:The content change of FA monitored with serum fingerprints was in line with the rule of pharmacokinetics of FA in Decoction for Puerperal Blood Stasis.The fingerprints can be applied to clarify the pharmacokinetic characteristics,the pharmacological action and the action mechanism of Decoction for Puerperal Blood Stasis.

Multiple organ dysfunction syndrome(MODS),a hot topic worldwide,has made rapid progress with nigh mortality.MODS in pediatrics versus MODS in adults are similar but different.Due to special age-related physiological characteristics.It is difficult to carry out randomized controlled clinical study compared with adults.Diagnosis and treatment of pediatric MODS can only be obtained with reference to adult MODS.This study reviews on the epidemiology,clinical scoring system,pathogenesis,clinical features and treatment of MODS in pediatrics.
