1.Clinical study on the relationship between plasma level of homocysteine and progressive ischemic stroke of carotid artery system as well as carotid artery atherosclerosis

Objective To explore the role of plasma level of homocysteine ( Hey ) in progressive ischemic stroke (PIS)of carotid artery system and to investigate the relationship between Hcy and carotid artery atherosclerosis(AS).Methods Plasma Hcy concentrations were measured in 68 patients with progressive ischemic stroke of carotid artery system (PIS group)and 72 patients with non-progressive ischemic stroke of carotid artery system ( non-PIS group ).Plasma Hcy level and carotid artery intima-media thickness ( IMT ) were compared among PIS group and non-PIS group as well as normal control group.The linear correlation between plasma Hcy level and IMT in PIS group was analyzed.Results Plasma Hcy concentrations was respectively (22.52 ± 5.26) μmol/L,( 16.38 ± 4.23) μmol/L and( 10.13 ± 2.16) μmol/L in PIS group,non-PIS group and normal control group( F =56.87,P < 0.01 ).Compared PIS group and non-PIS group with normal control group,the differences were statistically significant ( P < 0.01 ).And there was also significant difference on plasma Hcy levels between PIS group and non-PIS group( P < 0.01 ).The thickness of carotid artery IMT was respectively(1.12 ± 0.34)mm,(0.96 ± 0.27)mm and(0.43 ± 0.21 )mm in PIS group,non-PIS group and normal control group( F =28.47,P < 0.01 ).Compared PIS group and non-PIS group with normal control group,the differences were statistically significant( P < 0.01 ).And there was also significant difference on IMT between PIS group and non-PIS group ( P < 0.01 ).In PIS group plasma Hcy level was positively correlated with the carotid artery IMT ( r =0.687,P < 0.01 ).Conclusion The plasma Hcy level is closely related to the occurrence and development of carotid artery atherosclerosis and PIS of carotid artery system,detection of plasma Hcy level helps to predict the occurrence of PIS of carotid artery system and provide an evidence for the therapy decision.
2.Treatment of 30 Cases of Post-herpetic Neuralgia by Acupuncture Combined with Point Injection

Objective: To observe the effect of acupuncture around the diseased region combined with point injection on post-herpetic neuralgia. Methods: Point injection on 30 cases in the related segments of spinal cord after needling around the diseased region. Results: Twenty-seven cases got effect and the total effect rate was 90%. Conclusion: Acupuncture combined with point injection has remarkable curative effect on post-herpetic neuralgia.
3.Correlation of smokers in catenin wnt/β signal transduction pathways in expression and research on the incidence of squamous cell carcinoma of the lung

Objective To investigate the correlation of smokers in wnt/β-catenin signaling pathway activation as well as their correlation with lung squamous cell carcinoma incidence.Methods In the Department of Respiratory Medicine of the People's Hospital of Wuhan University,30 cases lung squamous cell carcinoma patients with a history of smoking,30 cases lung squamous cell carcinoma patients with no history of smoking,30 cases bronchiectasis patients with a history of smoking,30 cases bronchiectasis patients with no history of smoking were chose respectively as A group,B group,C group and D group.After the patients were treated by surgery,the removal of diseased tissue used immunohistochemical staining for CD68 + testing,TNF-α,wnt-2,β-catenin and the expression of TCF-4 were measured by using Western blot hybridization method,then wnt-2,β-catenin,TCF-4 transcription of mRNA concentrations were detected by using RT-PCR method.Results Immunohistochemical studies showed that cells in TNF-α,CD68 + expression of A group,B group,C group and D group were respectively 0.895±0.036 and 0.931±0.237,0.125±0.018 and 0.631±0.064,0.083±0.042 and 0.311±0.062,0.053±0.004 and 0.196±0.020,the differences were statistically significant(P<0.05).Westblot test Results showed that wnt-2,β-catenin,TCF-4 expression level showed a gradually decreasing trend,the difference was statistically significant(P<0.05),testing of RT-PCR showed that wnt-2,β-catenin,TCF-4 translation level showed a gradually decreasing trend,the difference was statistically significant(P<0.05).Conclusion Smoking for body wnt/β-catenin pathway has a cell activation,after cigarette activate wnt/β-catenin signaling pathway in the incidence of lung squamous cell carcinoma of the body will greatly enhance the smoking crowd wnt/β-catenin signaling transduction circumstances.The detection of wnt/beta-catenin signal transduction in smokers can be used to indicate the occurrence of lung squamous cell carcinoma.
4.Clinical efficacy of transurethral drugs perfusion combined with ultrashort wave for treatment of chronic bacterial prostatitis

Objeetive To explore a new therapeutic way of transurethral drugs perfusion combined with ultrashort wave on treating chronic bacteria prostatitis.Methods 241 cases with chronic bacteria prostatltis were randomly divided into two groups.the treatment group were treated with transurethral perfusion combined with ultrashort wave.the control group were treated with transurcthral drugs perfusion.Results Total effective rate in the treatment group was 85.9%.that of the control group was 36.7%.It had a significant difference between two groups(P<0.01).Conclusion Transurethral drugs perfusion combined with ultrashort wave could promote the curative effect of antibiotic maximumly,so it is an effective method to treat chronic bacterial prostatitis.
5.Clinical study of serum C-reactive protein,supplement C3 and C4 in the patients with systemic inflammatory response syndrome induced by acute cerebral infarction

Objective To study the role of serum C-reactive protein (CRP) ,supplement C3 and CA in sys-temic inflammatory response syndrome (SIRS) induced by acute cerebral infarction(ACI).Methods The levels of CRP,C3 and CA were measured by automatic immunoturbidimetry in 52 ACI cases accompanied with SIRS (SIRS group) ,50 cases of acute simple cerebral infarction(SACI group) and 46 normal cases (control group).Results The serum CRP, C3 ,and CA levels were significantly different among the three groups( F=720.6,580.8,94.1 ,P <0.01) ,the serum CRP levels in SIRS group were significantly higher than those in SACI group(P <0.01 ).The ser-um C3 and CA levels in SIRS were significantly lower than those in SACI group(P <0.01).With the diagnosis crite-rion items increasing, the serum CRP levels tended to be higher (t = 7.69 ,P < 0.01 ), and the serum C3, CA levels tended to be lower meeting SIRS diagnosis criterion numbers increasing respectively (t=10.68 and 10.53, P <0.01).Conclusion The serum ClIP levels are higher significantly and the serum levels of C3 and C4 are lower sig-nificantly when ACI progresses to SIRS.The serum CRP,C3,CA level changes can be used as an index for judging the progress of ACI progressed to SIRS.
6.The experimental study of the effect of medicine-induced posterior vitreous detachment on proliferative vitreoretinopathy

Objective To observe the effect of medicine-induced posterior vitreous detachment (PVD) on proliferative vitreoretinopathy (PVR).Methods.PVR was induced in the left eyes of 24 pigmented rabbits by intravitreal injection with platelet rich plasma.The rabbits were randomly divided into two experimental groups (group A and B) and one control group with 8 eyes in each group.Three hours later,the eyes in group A and B and the control group underwent intravireal injection with 1 U plasmin 0.05 ml+ 20 U hyaluronidase 0.05 ml,plasmin 0.1 ml,and balance salt solution 0.1 ml,respectively.The grade of PVR was recorded 1,7,and 28 days after the intravitreal injection,and the eyes were examined by flash electroretinogram (FERG),B-scan,and retinal histopathological examination.Results The PVR models of rabbit eyes were induced successfully.On the 7th day after injection,complete and partial PVD was found in 5 and 3 eyes respectively in group A;partial PVD in 5 eyes and no complete PVD was observed in group B;there was no PVD in the other 3 eyes in group B and also in the eyes in the control group.On the 28th day after intravitreal injection,PVR grade of group A and B were both obviously lower than that of the control group(D= 75.6,98.9;P = 0.003,P = 0.011) ;On the 7th and 28th day after injection,the b-wave amplitude in group A and B was significantly higher than that in the control group;PVR grade of the PVD eyes was lower than that of non-PVD eyes;PVR grade of the complete PVD eyes was only 0 ~ 1.Conclusions Three hours after the PVR models of rabbit eyes were induced,complete PVD induced by intravitreal injection of plasmin combined with hyaluronidase could prevent the development of PVR of rabbit eyes in some degree;partial PVD induced by plasmin alone or combined with hyaluronidase could relieve the development of PVR.
7.Role of CD4 + CD25 + regulatory T cells in the patients with ulcerative colitis

Objective To investigate the levels of CD4 + CD25 + regulatory T cells and the Foxp3 in the peripheral blood of patients with ulcerative colitis ( UC), and analyze its role in the pathogenesis of UC. Methods From February 2007 to December 2008, 40 UC patients (23 active and 17 remissive), 33 irritable bowel syndrome (IBS) patients, and 32 normal controls entered our study. CD4 + CD25 + T cells were detected with flow cytometric assay. The expression of Foxp3 mRNA in peripheral blood mononuclear cell (PBMC) was detected by RTPCR. Interleukin-10 (IL-10) and transforming growth factor-β (TGF-β) in the serum from the peripheral blood of UC patients, IBS patients, and normal controls were determined with ELISA. Results There were no significant differences of the peripheral CD4 +T cell numbers among the active, remissive UC and IBS patients, and normal control groups (P=0. 126). The positive rate of CD4+ CD25+ T cells in patients with active and remissive UC were significantly lower than those in IBS patients and normal controls ( both P < 0. 01 ) it was more lower in the active UC patients than the remissive UC patients ( P < 0. 001 ). However, the positive rate of CD4 + CD25 + T cells showed no significant difference between IBS patients and normal control groups ( P = 0. 343 ). The percentage of CD4 + CD25 + T cells was negatively correlated with UC disease activity index ( r = - 0. 660, P < 0. 001 ) and with erythrocyte sedimentation rate (r = -0. 572, P =0. 001 ). The expressions of Foxp3 mRNA in PBMC from active and remissive UC patients were significantly lower than those in both IBS patients and normal controls ( both P <0. 001 ). There was no significant difference of the plasma levels of IL-10 or TGF-β among the active/remissive UC patients, IBS patients, and normal controls ( all P > 0. 05 ). Conclusions CD4+ CD25 + regulatory T cells remarkably decline in the active UC and show certain increase in remissive UC, indicating that these cells may be involved in the pathogenesis of UC. The decreased expression of Foxp3 mRNA may be an important factor of the aberrant developmental disorder of CD4 + CD25 + regulatory T cells.

Inflammatory reaction plays an important role in the occurrence and development of ischemic stroke. Monocyte chemotactic protein-1 (MCP-1) is one of the subfamily members of the CC chemokine class. It can attract and activate a variety of cells, such as monocytes and T lymphocytes, promote the expression of cytokines, and participate in the occurrence of ischemic brain injury. This article reviews the progresses on the MCP-1 and ischemic stroke-related research.
9.Study of genetic variation of the structural region of hepatitis C virus

Objective To study the role of the structural gene variation of hepatitis C virus(HCV) in the chronicity of HCV infection. Methods Serum samples from four HCV infection acquired during six-month follow up and four patients with chronic hepatitis C type were obtained. A 1-kb fragment containing the 3' half of core ,completed E1 and 5' half of E2/NS1 was amplified by nested reverse transcription polymerase chain reaction (RT-PCR). For each,30 cloned cDNAs were examined by a method that combined heteroduplex analysis (HAD) and a single-stranded conformational polymorphisim (SSCP)assay to optimize selection of clones with unique gel shift patterns (clonotypes )as a measure of the quasispecies complexity. Results There was obvious difference between the SSCP bands of cloned cDNAs representing core-E1 and E2 region,but more distinct difference between heteroduplex and the homoduplex of clones spanning E2 region which accurately reflected the degree of heterogeneity. Quasispecies complexity was higher in those with persistent viremia than those with acute viremia. Conclusion HCV persistence is associated with complex quasispecies in viral structural region.
10.Pulse wave: the bridge connecting traditional Chinese medicine with Western medicine

Pulse wave, a research focus in both traditional Chinese medicine and Western medicine, which contains a wealth of human physiological and pathological information, has always been the concern of medical practitioners. On the basis of the review on the development of the pulse wave theory and its applications, the existing problems in this field are discussed in this paper. On the basis of the status quo of pulse wave information extraction methods and the utilization of pulse wave in traditional Chinese and Western medicine, a more in-depth study on pulse wave is proposed to make it a bridge connecting traditional Chinese medicine with Western medicine.
