1.Gelatinized starch is more feasible for starch tolerance test

Aim To study the effect of gelatinized starch on starch tolerance test.Methods Acarbose with gelatinized starch or ungelatinized starch was administered to ICR mice by gavage,and blood samples were collected at 30,60 and 120 minutes after treatment to measure blood glucose levels,whose rising percentage calculated at 30 min and AUC in every group.Results Compared with the ungelatinizedstarch group,blood glucose levels at 30 and 60 minutes after starch loading were significantly elevated in the gelatinized-starch group.Acarbose drastically flatted the elevation of blood glucose after gelatinized-starch load,but it had little effect on the ungelatinized-starch test.Conclusion It is more feasible to use gelatinized starch to carry out starch tolerance test than to use ungelatinized starch.
2.Evaluation of the pharmacoeconomics of alprostadil in pharmacotherapeutic schemes for cerebral infarction

Objective:To investigate the curative effects and the economic effects of Alprostadil in Pharmacotherapeutic Schemes for Cerebral Infarction. Methods:118 cases of cerebral infarction, admitted from March 2001 to March 2003, were collected and divided into two groups: Alprostadil was given to group Ⅰ, Alprostadil was not given to group Ⅱ. Data was evaluated using the pharmacoeconomics cost effectiveness analysis. Results: The costs of two groups were 13 826.13 and 7 884.55 yuan respectively. The effective rates of two groups were 92.89% and 80.33% respectively. The cost-effective rates of two groups were 148.70 and 98.15 yuan / per unit of effective. It cost 469.69 yuan for group Ⅰ to increase per unit of effective above group Ⅱ. Conclusion: GroupⅡ had lower cost- effective ratio, and groupⅠ had higher effective rate. If patients care about effectiveness more than cost, it maybe useful to take groupⅠ.
3.Magnetic resonance diffusion tensor imaging of cervical cord in healthy children

Objective To investigate the development of the cervical cord in children by using diffusion tensor imaging.Methods Ninety healthy children were undergone with diffusion tensor imaging of the cervical cord by using single-shot spin-echo echo planar image sequence.The values of apparent diffusion coefficients (ADC), fractional anisotropy (FA), average length(Ltract) and volume of tracts(Vtract) were measured in the cervical regions.Results The measurements of each group were as follow:ADC value:0.9747 ±0.2777,0.8493 ±0.2236,0.8210 ±0.1432,0.9198 ± 0.1444,0.9048 ±0.1676;FA value:0.4117 ±0.0391,0.4712 ±0.0199,0.4944 ±0.0439,0.5608 ±0.0443,0.6169 ± 0.0551;Ltract:25.61 ±8.63,24.66 ±7.14,27.03 ±7.23,34.93 ±10.99,37.63 ±10.22;Vtract:3.07 ±1.49,3.00 ± 1.52,3.81 ±1.33,5.41 ±2.35,6.64 ±2.84.FA value, Ltract and Vtract showed significance in different age groups , while ADC value was found no difference ( P<0.001) .Post-Hoc test revealed that FA value was significantly different between age group I and Ⅱ.FA value, Ltractand Vtract presented significantly different between group ⅢandⅣ.FA value difference was also found between group Ⅳand V.FA value, Ltract and Vtract were all positively corelative with age (F=1.758, P=0.145 ) .Conclusion Development of the cervical cord shows periodicity with periodic features .Diffusion Tensor Imaging can be used as a tool to observe and evaluate development of the cervical cord in children .
4.Distribution and Speciation of Lead in Moss Collected from a Lead-Zinc Mining Area by Micro-X-Ray Fluorescence and X-Ray Absorption Near Edge Structure Analysis

Moss has high tolerance and accumulating capacity to heavy metal.In this study, the distribution of heavy metal elements in moss sampled from lead-zinc mine was analyzed by X-ray fluorescence spectrometry.The speciation of lead was analyzed by X-ray absorption near-edge spectroscopy.Research showed that the contents of Pb, Zn, Cd and As in the moss of the mining area were extremely high, and their maximum concentration were 1.06 mg/g , 1.23 mg/g, 30.5 μg/g, 13.2 μg/g, respectively.Besides, the shoots especially the new tissue of the moss were the major sites for accumulation and storage of heavy metals.The micro-distribution characteristics varied among Hypnum plumaeforme and Brachytheciumprocumbens, indicating the difference of different species of moss in absorption pathway, accumulation and tolerance mechanisms for heavy metal.Linear combination fitting results indicated that the main lead speciation in moss was lead phosphate (78%) and lead oxide (22%), which suggested that the precipitation of lead phosphate might be the main mechanism of tolerance for moss.
5.Construction of autocatalytic caspase-3 and its effects of inducing apoptosis in human ovarian carcinoma

Objective To construct the autocatalytic caspase-3 and investigate its apoptosis- inducing effect in ovarian cancer in vitro and in vivo.Methods PCR recombination technique was used to construct autocatalytic caspase-3 which is named as rev-caspase-3,and Ad-Max system was used to prepare recombinant adenovirus containing rev-caspase-3,which is named as Ad-rev-easp3.Immunohistochemistry was used to detect active caspase-3 expression.Cell counting kit,flow cytometry and western blot were used to measure cell survival rate,apoptotic rate,cell cycle distribution and the expressions of plT,active subunit of caspase-3,and p85,the poly(adenosine diphosphate-ribose)polymerase(PARP)cleavage segment,respectively.Transmission electron microscope was used to detect cell ultrastrueture,and real time PCR was used to detect apoptosis-related gene expression.Subcutaneous tumor models and abdominally spread tumor models of human ovarian carcinoma were established using AO cells in BALB/c nude mice. The mouse survival rates were measured for abdominally spread tumor models,and the volume of tumor nodules were determined for subcutaneous tumor models following the treatments of rev-caspase-3.Results Active caspase-3 protein was significantly expressed,and the expression levels of active subunit of caspase- 3,p17,and the PARP cleavage segment,p85,were significantly elevated in cells treated with rev-caspase- 3.The decrease of cell survival rate and the increase of cell apoptotic rate were detected following Ad-rev- casp3 treatment.Treatments with Ad-rev-casp3 [ multiplicity of infection(MOI)was 70 ] resulted in survival rate of 30.3% and apoptotic rate of 40.2%.There was a significant increase in cell number of S- phase(56.5%),while there was no significant apoptosis(3.4%)following treatments with Ad-rev-casp3 at a low dosage of MOI=10.Cells treated with rev-caspase-3 displayed significant apoptotic morphology. The levels of active caspase-3 gene expressions(9.44)significantly increased.Rev-caspase-3 treatment significantly prolonged survival,the mean survival duration was(213?16)days,and suppressed tumor growth(tumor growth suppression rate was 70%),when compared with treatment with phosphate buffered saline(PBS).Conclusion Recombinant adenovirus containing rev-caspase-3 can significantly induce apoptosis of ovarian carcinoma cells,suppress tumor growth and prolong the mouse survival duration.
7.Mechanism of resistance of Pseudomonas aeruginosa to carbapenems

Objective To investigate the mechanism of resistance of clinical isolates of Pseudomonas aeruginosa to carbapenems.Methods Fifty-eight strains of imipenem-and meropenem-resistant Pseudomonas aeruginosa were isolated.Modified K-B technique was adopted in the susceptibility test by adding ?-lactamases inhibitors and efflux pumps inhibitors to M-H agar plates.The refined three-dimensional extract test was used to detect ?-lactamases.The genes of metallo-enzyme IMP,VIM,as well as outer membrane porin D2(OprD2) were analyzed with polymerase chain reaction.Results Among the 58 strains of carbapenems-resistant Pseudomonas aeruginosa,53 produced over-expressed active efflux and continuously produced large amont of AmpC enzyme,15 of which were accompanied by the loss of OprD2,and 1 of which were accompanied by extended spectrum ?-lactamases(ESBLs).Among the 5 strains which neither produced over-expressed active efflux nor ?-lactamases,only 1 was found with OprD2 gene deletion.Metallo-enzyme was not detected in any of the 58 strains.Conclusion The mechanism of resistance of Pseudomonas aeruginosa to carbapenems was mainly the production of the over-expressed active efflux combined with the continuous production of large amount of AmpC enzyme.Sixteen of the strains were accompanied by the loss of OprD2 gene.
8.Evaluation of the effect of human biological stimulation feedback instrument for screening of metabolic syndrome

Objective To assess the effect of a noninvasive equipment (the human biological stimulation feedback instrument) for the screening of metabolic syndrome (MS). Methods The samples were selected from the International Health Care Center of the Second Affiliated Hospital of Zhejiang University College of Medicine. From January to December 2012, according to the inclusion criteria, a total of 5674 consecutive participants (3 437 men, 2 237 women) without medical history of diabetes mellitus underwent general health screening in this hospital, which included questionnaire investigation, all MS components and the risk assessment of the human biological stimulation feedback instrument. MS was defined according to 2004 the Chinese Diabetes Society (CDS) criteria. Results The 5 674 subjects were divided into three groups in accordance with the human biological stimulation feedback instrument risk score: the normal group, metabolic abnormalities low risk group, metabolic abnormalities high risk group. The detection rate of MS corresponding to the three groups was 8.73% (251/2 876), 29.32% (612/2 087), 32.77% (233/711), respectively (trend χ2=372.14,P<0.001). With the increase of risk score, the detection rate of MS increased. A total of 1 096 (male 879, female 217) were diagnosed MS by CDS criteria. The area under the curve (AUC) of the test was 0.674 (95%CI 0.658-0.691, P<0.000 1)for predicting MS. The corresponding cutoff value was 25.00%, the sensitivity, specificity, positive predictive value and negative predictive value of diagnosis of MS was 77.80%, 56.00%, 30.20%and 91.30%, respectively;409 subjects who had no history of MS and diabetes mellitus follow up for two years, the AUC of the test was 0.719 (95%CI 0.644-0.794)for predicting MS. The corresponding cutoff value was 25.00%, too. And the sensitivity and specificity of diagnosis of MS was 78.00%and 59.80%, respectively. The detection rate of MS was positively correlated with the risk score. Conclusion We found that the human biological stimulation feedback instrument is a potential screening approach for early predicting MS in health checkup with simple and noninvasive procedure and high negative predictive value, although its diagnostic efficiency is not so good. And it might have a warning effect on the progress of MS.

Objective:To investigate the clinical manifestations and causative factors of adverse drug reactions following cinepazide mahate injection and provide reference for the safe use of drugs.Method:503 cases treated with cine- pazide maleate in our hospital were retrospectively studied and the results were statistically analyzed by SPSS(versionl 1.0).Result:Of 503 patients,27 cases presented some adverse events with an incidence of 5.4%,and adverse drug reac- tions were found in 11 cases with an incidence of 2.2%.The main adverse drug reactions were nervous,gastrointestinal and dermal reactions.The adverse drug reactions had no relation with sex,but with age of patients and combination use of drugs(P
10.Investigation of clinical application of the self-made curved laryngoscope

OBJECTIVE To discuss the clinical utility of the self-made curved laryngoscope.METHODS Clinical data of 198 patients with vocal cord polyps from September 2012 to October 2014 was analyzed retrospectively in which 99 cases were treated under the self-made curved larngoscopic surgeries (curved laryngoscope group) and 99 cases were treated by microlaryngoscopic surgeries (microlaryngoscope group).The duration of follow-up was 3 months in all patients.The clinical efficacy, times of inserting laryngoscope, glottic exposure and operation complications of two kinds of operations were observed and compared.RESULTS The total clinical efficacy in curved laryngoscope group showed no significant difference as compared with that in microlaryngoscope group. But for patients with difficult laryngeal exposure,the efficacy of curved laryngoscopic surgery was significantly better compared with that in the microlaryngoscopic surgery group. The results of times of inserting laryngoscope, glottic exposure and operation complications in the self-made curved larngoscope group were all better than those in microlaryngoscope group (P <0.05). CONCLUSION The operation under the self-made curved laryngoscope is a minimal invasive procedure to the laryngeal mucosa, with such more advantages like clear view of operation field and accurate management to the lesion tissue.Furthermore, the self-made curved laryngoscopic surgery is the first choice for the treatment of patients with difficult laryngeal exposure.
