Objective To observe the cognitive fuction difference between post-stroke depression(PSD) and non-post-stroke depression(NPSD) patients.To evaluate the clinical value of exploratory eye movement(EEM) and event-related potentials(ERP) on PSD.Methods EEM and ERP test were used for 50 PSD,50 NPSD patients and 48 normal controls.The results were compared.Results The abnormal rate of EEM and ERP test in PSD group were 96%(48/50) and 92%(46/50)respectively,and the abnormal rate of EEM and ERP test in NPSD group were 86%(43/50)and 82%(41/50)respectively.The number of eye fixation(NEF)and the responsive search score(RSS)of PSD and NPSD group were significant lower and the scores of discriminant(D) were significant higher than those in the normal control group(all P
2.Influence of Acupuncture on Cephalic Acupuncture Points Region Over the Brain-stem Evoked Potential of Patients Suffering Insufficient Blood-Supply of Vertebral Basilar Artery

Objective From the angle of electrophysiology, go further into the mechanism of action of acupuncture treatment to the insufficient blood-supply of vertebral basilar artery and at the same time, make a comparison of the effects between the acupuncture on cephalic meridian points and zonation cephalic acupuncture. Methods Through the contrast before and after adopting the self-acupuncture on the testees of 28 cases (including healthy persons of seven cases and TIA patients of 21 cases), observe the change of immediate effect of all waves latent period values of BAEP. Results The BAEP wave latent period of healthy persons (exclusive of I wave of auditory fainting region) assumes a relative delay within the physiological range and by comparison between points and region, the delay of V-wave Baihui point is evidently higher than that of auditory fainting region. However, the latent period of all waves of TIA patients is relatively moved up, especially there are evident differences before and after the I-wave acupuncture is carried out. Viewing from the comparison between points and region, there is no significant difference. Conclusion The function of acupuncture lies in the adjustment of antagonistic body state and it is most effective under the pathological state. Acupuncture will play the main role of excitation to the auditory nerve conduction of BAEP of TIA patients (from cochlea to midbrain) and the influence over the peripheral nerves is more sensitive than over the center ones. Although points and region have their own relative specificities, there is no significant difference between them.
3.Changes in behavior and spinal cord dorsal horn CGRP following chronic compression of DRG in rats

Objective To develop a reliable and reproducible modei of hyperalgesia arising from chronic compression of dorsal root ganglion (DRG) and investigate the contribution of spinal cord dorsal horn calcitonin gene-related peptide (CGRP) to the clinical manifestation of sciatica.Methods Sixty healthy Wistar rats of either sex weighing 200-300 g were randomly divided into three groups: group A control ( n = 12) ; group B sham operation (n = 24) and group C chronic compression of DRG ( n = 24) . In group B and C the animals were anesthetized with intraperitoneal 1.25% thiopental sodium (50mg?kg-1) . The animals were then placed in prone position and the left L4 and L5 intervertebral foramina were exposed. In group C a 4 mm long 1-0 chromic catgut was inserted into the intervertebral foramen. Motor function of the hind limbs was assessed by gait disturbance scores (GDS) , and paw-withdrawal threshold (PWT) was measured with modified Randall algometer 3, 5, 7, 10, 14, 17, 21 and 28 days after operation. In group B and C 12 animals were decapitated on the 5th and 28th days after operation respectively and lumbar section of spinal cord was removed for determination of dorsal horn CGRP content (by radio-immunoassay) and microscopic examination of DRG. Results In chronic compression group (C) significant hyperalgesia occurred after operation and reached a peak one week after operation then gradually decreased but didn't return to normai on the 28th day, while in sham operation group (B) postoperative hyperalgesia quickly decreased and returned to normai on the 7th day. In group C dorsal horn CGRP content on the compressed side significantly increased on the 5th and 28th day after operation as compared with that on the contralateral side in the sham operation and control group. Microscopic examination of DRG showed significant edema of endoneurium and neurons which persisted on the 28th day but significantly attenuated. Conclusion This animal model reliably produces a disorder resembling low back pain. It is easy to perform less traumatic and produces significant hyperalgesia. The dorsal horn CGRP content on the compressed side significantly increases, which is consistent with chronic pain.
4.Study on P300 potentials in patients with first cerebral stroke

Objective To explore the change of P300 potentials in first cerebral stroke patients,and to find the objective index in judging the cognition in these patients.Methods P300 potentials test was used for sixty patients with their first episode of single loci stroke and sixty-two healthy people.The results were compared in the two groups.Results Latency periods of N 2 and P 3 of P300 potentials in cerebral stroke group were longer than those in the controls,the amplitude of P 3 of P300 potentials in cerebral stroke group was lower than that in the controls.There was significant difference between the two groups(P0.05).Conclusion P300 potentials can reflect the cognition in cerebral stroke patients,which has remarkable clinical value in making reasonable treatment plan.
5.Effects of fluoxetine hydrochloride on depressive symptoms and P300 after cerebral stroke

BACKGROUND: The depressive emotion and cognition impairment after cerebral stroke are closely connected with the evolution and curative effect and prognosis of the disease, so how to improve depressive emotion and cognition impairment with reasonable drug has important significance to enhance clinical rehabilitation for the patients.OBJECTIVE: To treat patients with post-stroke depression with fluoxetine hydrochloride, andevaluate the changes of their depressive emotion and cognitive function with self-rating depression scale (SDS) and P300 potential.DESIGN: A randomized controlled trial based on patients. SETTING: Henan Provincial Psychiatric Hospital. PARTICIPANTS: Eighty-two inpatients with the first-attack of poststroke depression, who were selected from the Department of Neurology,Henan Provincial Psychiatric Hospital between December 1999 and June 2003, were divided randomly into treatment group (n=41) and control group (n=41).METHODS: All the patients were given neurological routine treatment;besides, those in the treatment group were treated with fluoxetine hyhad taken other psychotropic drugs before entering the group, fluoxetine oxetine was 20 mg, which was taken orally by 1 tablet every morning. All the patients were evaluated with SDS and P300 potentials test at 1 week (the first evaluation) and 6 weeks (reevaluation) after treatment.latencies of N1, P2, N2 and P3 waves and amplitude of P3 wave in P300 potentials.RESULTS: All the patients in the treatment group (n=41) and control group fore treatment, the scores of SDS and the results of P300 potentials were not Reevaluation: After treatment, the score of SDS in the treatment group was significantly lower than that in the control group (40.32±7.2, 48.31±8.02,t=3.89, P < 0.01); the latencies of N2 and P3 waves in P300 potentials test in the treatment group were obviously lower than those in the control group [(235.5±22.9), (248.3±23.4) ms; (339.1±25.3), (348.6±25.5) ms, P < 0.05],and the amplitude of P3 wave was obviously higher than that in the controlgroup [(6.3±1.9), (4.9±2.0) μV, P < 0.05]. 6.Interventional effect of radix bupleuri and radix scutellariae at different ratios on the changes in the indexes of liver, spleen, thymus gland and suprarenal gland and vitamin C in suprarenal gland of rats with acute alcoholic liver injury

BACKGROUND:. Until now modern science cannot explain clearly aboutthe mechanism of acute alcoholic liver injury. Besides such methods as givingup drinking, or symptomatic treatment, there is no specific therapy and remedy. Traditional Chinese Medicine (TCM) has obvious advantages for this: Itcannot only improve the clinical symptoms, but also adjust organism immunefunction, and has very good development future. OBJECTIVE: To study the effects of radix bupleuri and radix scutellariae incompatibility with the ratio of different dosage (2:1, 1:1, 1:2) on acute alcoholicliver injury by regulating the system of endocrine-immunity network in rats. DESIGN: Groups are divided randomly and experiments are made underblank conditions. SETTING: TCM Clinical Laboratory of Pharmaceutical School, Henan University of TCM. MATERIALS: From September to November 2002 in TCM Clinical Laboratory of Pharmaceutical School, Henan University of TCM, the general condi tion of the animals were as follows: 96 healthy Wistar rats were divided into 8groups: blank group, model group, radix bupleuri group, radix scutellariaegroup, xiao-chaihu group, radix bupleuri-radix scutellariae 2:1 group, radix bupleuri-radix scutellariae 1:1 group, radix bupleuri-radix scutellariae 1:2 group. METHODS: ① According to the ratio of 2:1, 1:1, 1:2, radix bupleuri andradix scutellariae were taken respectively (480 g, 240 g; 360 g, 360 g; 240 g, 480 g) to produce the water decocting nedicine. Except the blank group(duplicated by saline of the same volume through consecutive gastric infusion), the other groups are duplicated through consecutive gastric infusionby 56 degrees of Er-Guotou White Spirit (7 mL/kg) two times a day for tendays until the model-making is finished. From the first day, the medicine wasgiven two times a day. The blank group and the model group were given thedistilled water of the same volume. The other groups were given the properwater decocting medicine. The dosage in xiao-chaihu group was 10 g/kg andin the other was all 12 g/kg. ② 16 hours later after the last time when thewhite spirit was given, blood was taken from stomach aorta to produceserum. Liver, spleen, chest gland, adrenal gland were quickly removed. Thecalculating formula of an organ index: An organ index= The weight of an organ (g)/The weight of a body (g) ×100%. The calculating formula of vitamin C content in adrenal gland (mg/g): Content of vitamin C= [(The Avalue of the testing tube-The A value of the blank tube)/ (The A value ofthe standard tube-The A value of the blank tube)] ×density of the standardtube (6 mg/L)×dilution times of the sample before tested/protein content(g/L). ③ Comparison between the groups was shown by using variancesimilar test and single-factor variance analysis.MAIN OUTCOME MEASURES: Index of rats' liver, spleen, chest gland, and adrenal gland (on the right side); the determination of vitamin C content in rats' right adrenal gland.RESULTS: During the experiment, 3 rats in the model group died. Tworats died in radix scutellariae group and radix bupleuri-radix scutellariae group respectively, In each of the other groups only one rat died. The condition of the number of the rats which were used for analyzing results:There were 12 in blank group; 9 in model group; 10 in radix bupleuri group, radix bupleuri-radix scutellariae 1:1 group respectively; 11 in each of the other groups. ① Comparison of index of rats in liver, spleen, chest gland, adrenal gland (right side) between different groups: There was no significant difference between the indexes of rats' liver in different groups.The index of spleen, chest gland, adrenal gland in the model group were all greatly less than in blank group (P < 0.05-0.01). The index of spleen in the radix bupleuri group wcre greatly higher than in the model group, it could make the reduced spleen index return to normal (P < 0.05); The index of chest in radix scutellariae group, radix bupleuri-radix scutellariae 1:1 group, radix bupleuri-radix scutellariae 1:2 group and xiao-chaihu group were greatly higher than in model group (P < 0.05); The index of adrenal gland in radix bupleuri-radix scutellariae 1:2 group and xiao-chaihu group were greatly higher than in model group (P < 0.05). ② Comparison of vitamin C content in rats' right adrenal gland between the groups: Compared with blank group, the vitamin C content of adrenal gland in the model group had no significant difference. While that of radix bupleuri group, xiao-chaihu group, radix bupleuri-radix scutellariae groups were greatly less than that of model group (P < 0.05-0.01), especially radix bupleuri-radix scutellariae 1:1 group had the most significant influence (P < 0.01).CONCLUSION: Interfering functional injuries of liver, spleen and suprarenal gland is one of the mechanisms of acute alcoholic liver injury.Generally speaking, bupleuri-radix scutellariae 1:2 group has the greatest effect on the testing index.
7.Effects of repetitive transcranial magnetic stimulation on depression and cognition in the treatment of post-stroke depression

Objective To explore the effects of repetitive transcranial magnetic stimulation (rTMS) and fluoxetine on depression and cognition in the treatment of post-stroke depression (PSD).Methods A total of 82 PSD patients were divided into a treatment group and a control group using a random number table.Besides conventional neurological therapy,the treatment group was treated with rTMS combined with fluoxetine,while the control group was treated only with fluoxetine.Forty healthy persons acted as normal controls.The Hamilton depression rating scale (HAMD) was used to evaluate depressed emotions,and event-related potential (ERP) P300 and exploratory eye movement (EEM) were used to evaluate cognitive function.The three groups were tested before treatment and after 8 weeks of treatment.Results After 8 weeks of treatment the HAMD scores in both the treatment and control groups had decreased significantly compared with before treatment.The HAMD scores decreased significantly more in the treatment group than in the control group.Before treatment,the N2 and P3 iatencies of P300 in the treatment and control groups were significant longer than those in the normal group,and the average amplitude of P3 in the treatment and control groups was significantly lower than among the normal controls.Before treatment,the number of eye fixations (NEF) and the average responsive search score (RSS) in the treatment group and control groups were significantly lower than in the normal group.After 8 weeks after treatment,the N2 and P3 latencies were significantly shorter and the amplitude of P3 was significantly higher in the treatment and control groups than before treatment.The NEF and the average RSS in the treatment and control groups had increased significantly compared with before treatment.All of these indexes improved significantly more in the treatment group than in the control group.Conclusion rTMS combined with fluoxetine can improve depression and cognitive function among PSD patients better than antidepressant treatment alone.
8.Clinical study on Yinzhi-huang combined with probiotics in the treatment of neonatal hyperbilirubinemia

Objective To investigate the therapeutic efficacy of Yinzhi-huang combined with probiotics in the treatment of neonatal hyperbilirubinemia.Methods 80 neonates of hyperbilirubinemia were recruited into a treatment group (40 cases) and a control group (40 cases).The control group was treated with the gap blue light irradiation,besides,patients with rapid increase or high level of bilirubin were additionally treated with acid-correcting agents,Liver enzyme inducer,and albumin infusion; while the treatment group was additionally treated with Yinzhi-huang and probiotics on the basis of the control group.The values of serum total bilirubin before the treatment Was determined in both groups.During treatment,the changes of daily bilirubin,were monitored with percutaneous bilirubin monitor,and the time for serum bilirubin values decreased to normal level was studied in both groups.Results The two groups showed no significant difference in examination of the serum bilirubin values before treatment(P>0.05).The daily bilirubin decreased value of treatment group(53.07± 17.80) μmol/L showed statistically significant compared with the control group(30.56± 13.43)μmol/L(P<0.05); the time for serum bilirubin values decreased to normal level in the treatment group (4.87± 2.06) d was significantly different compared with the control group (7.12± 2.33) d,(P< 0.05).The apparent effective rate in the treatment group 75% (30/40) was higher than that in the control group 35%(14/40)with statistical significance(x2=5.89,P<0.05).The total effective rate in the treatment group 92.5% (37/40)was higher than the control group 75% (30/40)with statistical significance (x2=4.50,P<0.05).Conclusion Yinzhi-huang combined with probiotics has better effects than conventional treatment in decreasing the daily bilirubin values,restoring normal serum bilirubin time of neonatal hyperbilirubinemia and it is worthy of clinical application.

10.In vitro differentiation of islet-like cells from mouse bone-marrow stem cells on a PLGA scaffold

Objective To investigated the potential influences of poly(lactic-co-glycolicacid)(PLGA)scaffold as a platform on the differentiation of mouse bone-marrow stem cells to islet-like cells.Methods Mouse bonemarrow stem cells were grown and differentiated in culture with or without PLGA scaffold,and cell morphology and functions were compared within these groups.Results The PLGA scaffold showed fine biological compatibility.Differentiated islet-like cells were dithizone (DTZ) positive,insulin and C-peptide double positive,glucagon positive and somatostatin positive in both groups.Under electron microscope there were ultrastructures similar to that of islet β cells in cells of both groups.Cells with PLGA scaffold secreted more insulin under high level glucose stimulation (P<0.01).Conclusions PLGA scaffold was biologically compatible and improved function of the differentiated islet-like cells.
