1.The discovery of the interaction between augmenter of liver regeneration and Na~+, K~+-ATPase with yeast two-hybrid system

AIM: To screen the proteins interacting with human augmenter of liver regeneration (hALR) by yeast two-hybrid system and to study the mechanism of hALR action. METHODS: hALR bait plasmid was constructed by ligating the gene of hALR into pGBKT7, then transformed into yeast AH109. The yeast strain AH109 containing pGBKT7-hALR was mated with yeast Y187 containing human liver cDNA library plasmid. Diploid yeast was plated on SD/-trp-leu-his-ade (QDO) for screening and on QDO containing X-?-gal for further selection.The AD/library inserts were amplified by PCR and the PCR products were characterized by digesting with Sau3AⅠ and HaeⅢ restriction enzyme to eliminate the duplicates. After sequencing, the positive clones were analysed by bioinformatics. RESULTS: Several positive clones were obtaind. The sequencing and analysis shown that one of them is 669 bp DNA fragment encoding ? subunit of Na +,K +-ATPase. The 224 bp 3′terminal DNA fragment is non-encoder region, and the 445 bp 5′terminal DNA encodes C-terminal 147 amino acid residues of Na +, K +-ATPase ? subunit. CONCLUSION: The results of screening proteins using yeast two-hybrid system showed that hALR could interact directly with Na +, K +-ATPase in the yeast cell.
2.Clinical effect and mechanism of combination of ozone,ozonated water and improved vacuum sealing drainage in healing complex wounds

Objective To investigate the clinical effect of combination of ozone ,ozonated water and improved Vacuum Sealing Drainage(VSD) in healing complex wound ,and to observe the expression of fibroblast growth factors(FGF) and interleukin-1(IL-1) in its surface .Methods 40 patients with complex extremity trauma were selected and divided into 4 groups randomly .These 4 groups were Ozone+VSD group and Ozonated Water+ VSD group ,VSD group and Control group ,and received ozone combined VSD treatment ,ozonated water combined VSD treatment ,pure VSD treatment ,and traditional treatment .Each group contained 10 patients .Comparison of clinical effect ,bacteria loads and the expression of FGF and IL-1 in wound surface was made in 4 groups . Results The average cure time ,average replacing frequency and infection rate in Control group were (31 .5 ± 6 .6)days ,(15 .4 ± 3 .9) times and 40% respectively .The average cure time ,twice VSD cases and infection rate in VSD group were (19 .2 ± 2 .3)days , 5 cases and 10% respectively .There were no case of infection or received twice VSD treatment in Ozone + VSD group and Ozonated Water+VSD group .The average cure time were (14 .9 ± 2 .7) days and (14 .1 ± 3 .0) days .The Ozone+ VSD group and Ozonated Water+VSD group had best clinical effect ,and the Ozonated Water+ VSD group had least bacteria loads in wound surface (P<0 .05) .The expression of FGF and IL-1 in 4 group had obvious differences ,and it had statistical significance(P<0 .05) .Conclusion Both ozone or ozonated water combined VSD treatment could decrease IL-1 and increase FGF expression ,accelerate recovery time ,inhibit bacterial infection ,especially ozonated water combined VSD treatment had better effect .
3.Clinical Efficacy of Integrated Traditional Chinese and Western Medicines in the Treatment of Patients with Ocular Herpes Zoster

Objective:To observe the clinical curative effect of integrated traditional Chinese and western medicine in the treat-ment of patients with ocular herpes zoster. Methods:Seventy patients with ocular herpes zoster in our hospital were selected during De-cember 2011 to December 2013. According to random number table,the patients were divided into two groups. The patients in the con-trol group were treated with ganciclovir 5. 0 mg·kg-1 ·h-1 , ivd, qd and ganciclovir ophthalmic gel, qid. The patients in the observa-tion group were treated with Banlangen granules and acupuncture additionally. The clinical efficacy, the time of blistering, pain and crusting and adverse reactions in the two groups were observed and compared. Results:After the treatment, the effective rate of the ob-servation group was 94. 29% , while that of the control group was 77. 14% (P<0. 05). The time of blistering, pain and crusting in the observation group was significantly shorter than that in the control group (P<0. 05). The incidence of adverse reactions of the ob-servation group was 2. 86%, while that of the control group was 11. 43%(P>0. 05). Conclusion:The clinical efficacy of integrated traditional Chinese and western medicine in the treatment of ocular herpes zoster is obviously better than that of western medicine treat-ment alone with fewer adverse reactions and shorter time of blistering, pain and crusting.
4.The Relationship between Preoperative MRI Characteristics and The Perioperative Outcomes of Microvas- cular Decompression in Primary Trigeminal Neuralgia

Objective To evaluate the relationship between preoperative MRI characteristics and the perioperative outcomes of microvascular decompression in primary trigeminal neuralgia. Methods To analyze the relationship between preoperative MRI characteristics and the perioperative outcomes in 103 primary trigeminal neuralgia patients with micro-vascular decompression in Nanfang Hospital. The MRI features such as the ratio of CPA area, TGN cross-sectional area and TGN length was evaluated together with the TGN oppression distance, the position of TGN, the position of basilar ar-tery and the type of offending vessel as well as the outocmes including complete disappearance, temporary remission and ineffectiveness. Results Univariate analysis showed that the oppression orientation (P=0.017), oppression distance (P<0.001), offending vascular type (P=0.016), TGN cross-sectional area ratio (P<0.001) were the influencing factors of periop-erative outcomes. Logistic regression analysis showed that the offending vascular type (P=0.002)and TGN cross-sectional area ratio (P=0.020) were the main predictive factors of perioperative outcomes of microvascular decompression. Conclu-sions Preoperative thin slice MRI scanning showed that the offending artery, non-atrophy nerve roots, far distance from op-pression point to brainstem may be the favorable factors of perioperative outcomes of microvascular decompression.
5.Effect of sodium butyrate on apoptosis and stromal interaction molecule and Orai1 activity in human colon cancer HCT-116 cells in vitro.

OBJECTIVETo investigate the mechanism underlying sodium butyrate (NaB)-induced apoptosis of a human colon cancer cell line HCT-116.
METHODSThe apoptosis of HCT-116 cells induced by NaB was confirmed by hoechst33342 staining and AnnexinV+ PI assay. The changes in the intracellular localization of stromal interaction molecule (STIM1) and Orai1 following NaB treatment were detected by immunofluorescence technique. Western blotting was used to investigate the protein expression levels of STIM1 and Orai1.
RESULTSNaB induced apoptosis and caused translocation and colocalization of STIM1 and Orai1 in HCT-116 cells.
CONCLUSIONThe apoptosis of human colon cancer cells induced by NaB is correlated to the redistribution of STIM1 and Orai1.

6.Research on quality specification of Herba Justiciae.

OBJECTIVETo provide scientific basis for the utilization and development of Herba Justiciae by setting up the quality control specification of Herba Justiciae.
METHODMoisture and ash were determined by aquametry and method of ash determination. And the bioactive constituents were analyzed by HPLC.
RESULTThe contents of total ash, acid-insoluble ash, and moisture of 28 samples from different origins were determined. The quantitative analysis of chinensinaphthol methyl ether by HPLC were preformed, respectively.
CONCLUSIONThe established method can be used for the quality control of Herba Justiciae.

7.Epidemiological and clinical characteristics of human brucellosis in Enshi Tujia and Miao Autonomous Prefecture of Hubei Province from 2012 to 2019

Objective:To analyze the epidemiological and clinical characteristics of human brucellosis in Enshi Tujia and Miao Autonomous Prefecture (Enshi Prefecture) of Hubei Province, and to provide a basis for prevention and control of local brucellosis.Methods:Using descriptive epidemiological methods, the data of reported cases of brucellosis in Enshi Prefecture from 2012 to 2019 (from the "Enshi Infectious Disease Reporting Information Management System" and the medical record system of hospitals in counties and cities within the jurisdiction of Enshi Prefecture) were collected, and the three distributions (population, time, region distributions) and clinical manifestations of human brucellosis cases were statistically described and analyzed.Results:A total of 78 brucellosis cases were reported in Enshi Prefecture from 2012 to 2019, with an average annual incidence rate of 0.193 5/100 000. Among the 78 cases of brucellosis reported, the sex ratio of men to women was 2.12∶1.00 (53∶25); the age of onset was mainly from 30 to 59 years old, accounting for 75.64% (59/78); the occupation was mainly farmer, accounting for 88.46% (69/78); the main contact animal was sheep, accounting for 80.77% (63/78). There were reported cases of brucellosis throughout the year, and the main onset months were May, June, September, November, and December, accounting for 55.13% (43/78) of the total number. From 2012 to 2019, all counties and cities in Enshi Prefecture except Xianfeng County and Hefeng County had reported cases. Among them, Lichuan City had the most cases, with 55 cases (70.51%). The main clinical manifestations of reported cases of brucellosis were fever, fatigue, hyperhidrosis, and muscle and joint pain, which accounted for 98.72% (77 cases), 89.74% (70 cases), 79.49% (62 cases), and 69.23% (54 cases), respectively.Conclusions:The majority of patients with brucellosis in Enshi Prefecture are young and middle-aged male farmers, with the highest incidence in Lichuan City. Relevant departments should increase the propaganda of brucellosis, increase the people's awareness of disease prevention, and strengthen the prevention and control measures of high-risk groups and regions to reduce the incidence of brucellosis.
