Based on the superior optical properties and the advantage of surface-modifiable capacity,the novel fluorescent nanoparticle quantum dots can conjugate with many kinds of biomolecules or chemical materials to form target quantum dots,which play an important role in biomedicine.The target ability this probe possesses offers a more accurate tumor localization,and minimizes the side effect to normal cells while strengthens the curative effect.At present,the probe makes a great progress in many areas,such as cell and molecule recognition,imaging in vivo,biosensing and diseases treating.

OBJECTIVE:To establish the quality standard of Fufang jinqin tablet. METHODS:TLC was conducted for the qual-itative identification of Cortex phellodendri and Lonicera japonica in the preparation. And HPLC was conducted for the content de-termination of baicalin;the column was Zorbax Eclipse XDB-C18 with mobile phase of methanol-0.2% phosphoric acid water(40:60,V/V)at a flow rate of 1.0 ml/min,the detection wavelength was 280 nm,column temperature was 35 ℃,and the injection vol-ume of 10 μl. RESULTS:C. phellodendri and L. japonica of TLC showed clear spots and good separation. The linear range of ba-icalin was 0.533-4.264 μg(r=0.999 9);RSDs of precision,stability and reproducibility tests were lower than 2%;recovery was 96.95%-99.70%(RSD=1.18%,n=6). CONCLUSIONS:The standard can be used for the quality control of Fufang jinqin tablet.

[Objective] To retrospectively analyze the treatment of tailor's bunion with minimal incision osteotomy,and to investigate the indications and effects of this procedure.[Method]Thirty-seven patients(69 feet)underwent the procedure from July 2002 to August 2007.The axial and lateral films of all feet with loading were taken before and after operation.AOFAS were recorded and analyzed preoperatively and postoperatively.[Result]The forth intermetatarsal angle was 14.36??7.32? before operation and 9.36??2.92?after operation.The unguis aduncus angle of digitus quintus pedis was 20.44??7.36? before operation and 4.36??1.35?after operation.The forth reforming intermetatarsal angle was 10.36??.2.81? before operation and 7.83??2.37?after operation.The exstrophy angle of the fifth metatarsal was 5.46??1.70? before operation and 2.13??0.38?after operation.The score of AOFAS was 45.7?5.6 before operation and 85.3?5.1 after operation.[Conclusion]The treatment of tailor's bunion with minimal incision osteotomy is easy to operate and its therapeutic effect is convincing.

AIM: To investigate the role of K + channels in the decreased hypoxic pulmonary vasoconstriction(HPV) in chronic hypoxic rats. METHODS: Blockers of three kinds of K + channels, 4-AP(voltage dependent K + channel blocker), TEA(Ca 2+ activated K + channel blocker)\, GLIB(ATP sensitive K + channel blocker) were used in isolated perfused rat lungs to detect the role of K + channels in HPV. RESULTS: In normal rats, 4-AP and TEA, but not GLIB, both elicited a significant increase in pulmonary artery baseline pressure, and also potentiated the acute hypoxic pulmonary vasoconstriction. In chronic hypoxic rats, the HPV is significantly decreased, while 4-AP, TEA, GLIB all elicited a significant but smaller increase in pulmonary artery baseline pressure. Additionally, all these three blockers potentiated the HPV stronger in chronic hypoxic rats than in control rats. CONCLUSION: The opening of Kv, K Ca , K ATP might modulate the hypoxic pulmonary vasoconstriction in isolated rat lungs, and the increase in this modulation by potassium channel in chronic hypoxic rats might play a role in its decrease in HPV.

Objective:To simultaneously determine the contents of asarinin, prim-O-glucosylcimifugin and 5-O-methylvisammio-side in Xinqin granules. Methods:An HPLC method was used. The determination was performed on a ZORBAX Eclipse XDB-C18 col-umn(150 mm ×4.6mm,5 μm) with the mobile phase consisting of menthol (A)-water (B) with gradient elution. The flow rate was 1. 0 ml·min-1 . The column temperature was 30℃. The detection wavelength was set at 254 nm from 0 to 30 min and 287nm from 30 to 55 min. The injection volume was 10μl. Results:The linear range of prim-O-glucosylcimifugin, asarinin and 5-O-methylvisammio-side was 10.210-163.400 μg·ml-1(r=0.999 7),10.160-162.600 μg·ml-1(r=0.999 8) and 5.015-80.240 μg·ml-1(r=0. 999 8), respectively. The average recovery was 100. 30%(RSD=1. 6%, n=6),101. 53%(RSD=1. 1%,n=6) and 101. 12%(RSD=1. 2%, n=6), respectively. Conclusion: The method is simple and accurate, which can be used in the quality control of Xinqin granules.

Objective To investigate the effects of different concentrations of crocin on receptor activator of nuclear factor kappa B ligand (RANKL)-induced osteoclastogenesis using the monocyte-macrophage cell line RAW264.7. Methods The monocyte-macrophage cell line RAW264.7 was cultured routinely.After treatment with 0,6.25,12.5,25, 50,100,200 and 400 μmol/L crocin,cell counting kit-8(CCK-8)assay was used to analyze the viability of RAW264.7 cells to screen out the appropriate experimental concentration. RAW264.7 cells were induced by RANKL (100 ng/L) to form osteoclasts. After treated with 0, 12.5, 50 and 100 μmol/L crocin respectively, the number of osteolasts was counted by tatrate resistant acid phosphatasec (TRAP) staining. Real-time PCR was used to detect the mRNA expression levels of calcitonin receptor(CTR),nuclear factor of active T cells 1(NFATC1),C-fos and TRAP.Results No significant effects of crocin (within 0-100 μmol/L) were found on the viability of RAW264.7 cells (P>0.05). However, When crocin concentration was over 100 μmol/L,the cell proliferation was decreased,and which showed a significant inhibitory effect on proliferation (P<0.05). Thus, 0-100 μmol/L crocin was selected as the experiment concentration. The amount of differentiated osteolasts and the expression levels of CTR,NFATC1,C-fos and TRAP mRNA were decreased significantly with the increased concentrations of crocin(P<0.05).Conclusion At a certain concentration(0-100 μmol/L),the higher levels of crocin could inhibit RANKL-induced osteoclastogenesis.

Background and purpose:Somatostatin receptors have been found in a variety of tumors and are therefore amenable to treatment with somatostatin analogs, like octreotide. However, the study of SSTRs expression has been rarely studied in nasopharyngeal cancer.We investigated the expression of somatostatin receptors gene subtypes in human nasopharyngeal cancer cell line CNE_ 2 . Methods:We have harvested cultured human nasopharyngeal cancer cell line CNE_ 2 . Using both techniques, reverse transcriptase-polymerase chain reaction (RT-PCR) and immunohistochemical assay, we analysed mRNA of different subtypes of somatostatin receptors in human nasopharyngeal cancer cell line CNE_ 2 .Results:The positive rate of somatostatin receptors subtype SSTR_ 1 SSTR_ 2 SSTR_ 4 was manifested in the human nasopharyngeal carcinoma cell line CNE_ 2 by RT-PCR and immunohistochemical assay. According to immunohistochemical assay, SSTR_ 1 and SSTR_ 2A showed strongly positive expression and SSTR_ 3 and SSTR_ 5 negative expression,respectively.Conclusions:There are more than one SSTR subtypes expressed in the human nasopharyndeal carcinoma cell line CNE_ 2 . This study demonstrated the presence of SSTR_1,SSTR_ 2 in the human nasopharyndeal carcinoma cell line CNE_ 2 .

Objective To summarize the experiences of diagnosis and treatment of dermatofibrosarcoma protuberans (DFSP). Methods The clinical data of 16 patients with DFSP were analyzed retrospectively. All the patients with DFSP underwent extensive tumor resection (3～5cm margin from the tumors).Free full-thickness skin flap transplantations was performed on 6 cases , three cases received skin flap shifting operations, and 1 patient with abdominal wall deficiency was repaired by artificial mesh . Results All the 16 patients presented as painless cutaneous nodules which enlarged slowly, but the majority of the tumors grew quickly in recent shorter time. Ten cases were primary tumors, and six were recurrence. The time of recurrence after surgery ranged from 3 months to 10.5 years with 1～3 times recurrence. There was no operative complications. Eleven patients were followed-up for 1～11 years and all survived,only one patient developed tumor local recurrence seven years after surgery. Conclusions A slowly enlarged painless cutaneous or subcutaneous nodules quickly growing in recent time is the clinical characteristic feature of DFSP. Extensive tumor resection is the treatment of primary DFSP.

Objective To compare the effect of a carbohydrate-electrolyte-protein solution (CEPS,2％ protein plus 4％ carbohydrate),carbohydrate-electrolyte solution (CES,6％ carbohydrate),and non-caloric sweetened placebo (PLA)on 21 km running performance of female recreational marathon runners.Methods In a randomized cross-over design,12 female recreational marathon runners (age:33.4 ± 2.1 years,body mass index:21.10 ± 0.6 kg/m2,and maximal oxygen consumption:48.5 ± 1.9 mL/kg/min)performed a 21 km time trial (TT)run on three occasions separated by at least 28 days.During the first 5 km,participants ran on the treadmill with 70％VO2max.Thereafter,the participants ran at whatever speed they wished for the remaining 16 km of the performance run.In each main trial,participants ingested the CEPS,CES,or PLA at a rate of 150 mL every 2.5 km.The blood samples,gas samples,heart rate,core temperature,and subjective measures were conducted every 5 km throughout the exercise.Results Time of TT run completion was slightly shorter (P ＜ 0.05)in the CES trial (97.8 ±1.6 min)than in the PLA trial (102.4 ± 2.4 min),with no significant differences between the CEPS trial (100.7 ± 2.9 min)and the other two trials (P＞0.05).There were no differences in core temperature,blood lactate,heart rate,CHO oxidation rate,fat oxidation rate,CHO oxidation amount,and fat oxidation amount among the different trials (all P＞0.05).The average blood glucose was higher in CES trial than PLA trial (P＜0.05).The average respiratory exchange rate was higher in CEPS trim than PLA trial (P＜0.05).Conclusion Compared with ingesting the PLA,ingesting the 6％ CES improves the 21 km TT run performance in recreational female runners.However,no additional benefit of CEPS was observed on endurance performance.
