1.Molecular mechanisms of Taishan Ganoderma Lucidum amylose in prevention of rat atherosclerosis

AIM: To study the change of blood lipid and difference of expressed gene in chest aorta cells of atherosclerosis model rats with or without the treatment of Taishan Ganoderma Lucidum amylose.METHODS: The concentrations of triglyceride (TG), total cholesterol (TC) and high-density lipoprotein cholesterol (HDL-C) in serum were measured by automatic serum biochemical assay. Differential-display reverse transcription-PCR (DDRT-PCR) was used to analyze the differential expressed gene in chest aorta cells of atherosclerosis model rat with or without the treatment of Taishan Ganoderma Lucidum amylose. The differential expressed cDNA was confirmed by reverse Northern blotting.RESULTS: Up-regulated and down-regulated cDNA were isolated in chest aorta cells of atherosclerosis model rats with or without Taishan Ganoderma Lucidum amylose treatment. The known up-regulated genes included cytB561, ERP72, nitric oxide synthase 2 and Gpx1. The known down-regulated genes included thymosin β4, FGFRI oncogene, ICAM-1, FK506 binding protein, c-myc and TNF-α. CONCLUSION: Taishan Ganoderma Lucidum amylose decreases the level of blood lipid.
2.Role of Kupffer Cells in High Fat Diet Induced Hepatic Insulin Resistance

Objective To explore the role of Kupffer cells in high fat diet induced hepatic insulin resistance. Meth-ods Eighty-four C57BL/6J mice were divided into two groups: normal chow (NC) group (7 subgroups) and high fat diet (HFD) group (7 subgroups). Glucose tolerance tests were performed at feeding time of 1,2,4,8,12 and 16 weeks in one NC group and one HFD group. The mice livers at the same feeding time were obtained in other 6 NC groups and 6 HFD groups respectively. The expression levels of IRS1-ser307, Akt-ser473, S6K1-thr389 and p-JNK were detected by Western blot as-say. The values of MCP-1,TNF-α,IL-1βand IL-10 mRNA were examined by qRT-PCR. Results Compared with NC group, the impaired glucose tolerance was found from the first week in HFD group (P < 0.05). The hepatic expressions of IRS1-ser307, S6K1-thr389 (4-16 weeks ) and p-JNK(2-16 weeks ) increased and Akt-ser473(8-16 weeks )decreased in HFD group than those of NC group. The same results were gained by analysis of protein relative expression (all P<0.05). The hepatic pro-inflammatory factor MCP-1,TNF-αand IL-1βmRNA expressions were higher in HFD group than those in NC group during 2-16 weeks (all P < 0.01). The anti-inflammatory factor IL-10 mRNA was significantly lower in HFD group than that in NC group during 4-16 weeks (all P<0.01). Conclusion High fat diet maybe play a role in the hepatic insulin resistance by stimulating M1 Kupffer cells to secrete pro-inflammatory factor and inhibiting M2 Kupffer cells to se-crete anti-inflammatory factor.

Objective To observe the protective effect of?-asarone,the active components from Rhizoma Acori Tarinowii,on human umbilical venous endothelial cell strain ECV304 injury induced by oxidized low-density lipoprotein(ox-LDL)and to investigate its influence on proliferation of vascular smooth muscle cells(VSMC). Methods The well-grown ECV304 cells were randomized into 5 groups:normal group,control group,high-, middle-and low-dose?-asarone groups.Except the normal group,ECV in the other groups were co-cultured with ox-LDL 25?g/mL,and the?-asarone groups were additionally with?-asarone 30,15 and 7.5?g/mL added respectively.Flow cytometry was used to detect the cell apoptotic rate and the mitochondrial membrane potential (MMP)of ECV304 as well as the proliferation index and the DNA content in different cell phases of VSMC. 4.Identification of the regulation elements in heat-inducible Lehsp23.8 promoter.

The promoter of mitochondria-localized small heat shock protein gene in Lycopersicon esculentum (Lehsp23.8) is characterized as strongly heat-inducible. In this study, to determine how the expression of Lehsp23.8 is regulated, we conducted five expression vectors carrying the gus gene driven by the 5' deletion products of the Lehsp23.8 promoter. The corresponding transgenic tobacco plants were generated via Agrobacterium tumefaciens-mediated transformation. Transgenic plants were identified by PCR and Southern blotting analysis. GUS activities under heat-shock conditions were characterized in transgenic tobacco plants. After heat shock, obvious GUS staining was detected in the leaves, shoots, roots, flowers and fruits of the transgenic tobacco plants. The result of fluorometric GUS assays in leaves showed that the heat-induced GUS activity of the 565 bp promoter was the strongest, while that of the 255 bp promoter was the lowest. Deletion analysis shows that the smallest promoter fragment (-255 bp to -23 bp) is sufficient for heat induction. It also indicates that the sequences between -255 bp and -565 bp serve as enhancers, while the sequences between -565 bp and -871 bp can repress the heat-induced activity of the Lehsp23.8 promoter.

5.Comparison of bacterioplankton communities in three heavily polluted streams in China.

OBJECTIVETo compare the bacterioplankton communities in streams exposed to pollution of different types.
METHODSThe bacterioplankton communities in three selected heavily polluted streams were investigated by using terminal-restriction fragment length polymorphism (T-RFLP) analysis in combination with 16S rRNA gene clone library analysis.
RESULTSBoth T-RFLP and 16S rRNA gene clone library revealed a great difference in bacterioplankton community composition in the different streams.
CONCLUSIONThis work might provide some new insights into bioremediation of heavily polluted streams.

Objective To assess the efficacy and safety of mifepristone combined with oral or vaginal misoprostol for termination of pregnancy between 8 and 16 weeks of gestation. Methods This was a randomized, multi-center, open clinical trial. A total of 625 women at 8-16 weeks of gestation were randomized to receive 200 mg oral mifepristone followed by either oral misoprostol 400 μg every 3 hours or vaginal misoprostol 400μg every 6 hours for a maximum of 4 doses 36-48 hours later. There were 417 women in oral group with 198 at 8-9 weeks and 219 at 10-16 weeks, while 208 women in vaginal group with 99 at 8-9 weeks and 109 at 10-16 weeks. The outcome measures were the success abortion rate, induction to abortion interval, the amount of bleeding, reoccurrence of menstruation and adverse events. Results Abortion rate was significantly higher in vaginal group [98.1% (202/206)] than that in oral group [94.0%(390/415), P=0.023]; concerning termination of pregnancy at 8-9 weeks and 10-16 weeks respectively, there were no significant differences between oral and vaginal groups (P=0.156, P=0.073). The induction to abortion interval was no significant difference in oral and vaginal group in different gestational weeks ( P=0.238, P=0.273). The average induction to abortion interval was (4.1 ± 6.6) hours and (6.0 ± 4.5) hours respectively in terminating 8-9 weeks and 10-16 weeks of gestation. Concerning the amount of bleeding within 2 hours of placenta expulsion, there was significant difference between oral group [(63±46) ml] and vaginal group [(55 ± 45) ml] in terminating 8-9 weeks of gestation (P=0.047), while there was no significant difference between groups in terminating 10-16 weeks of gestation [oral group (76 ± 52) ml versus vaginal group (76 ± 61) ml, P=0.507]. The reoccurrence of menstruation was about 37 days in both oral and vaginal groups. Two cases of incomplete abortion were serious adverse events (SAE) relating to treatment. The common adverse events (AE) of nausea and vomiting were significantly higher in oral group [57.2% (239/417), 36.3% (151/417)] than those in vaginal group [45.4% (94/208), 26.1% (54/208); P=0.005, 0.011]. Conclusion Oral or vaginal misoprostol combined with mifepristone, is effective and safe for termination of pregnancy between 8 and 16 weeks of gestation.
8.Safety and short-term effect of irreversible electroporation ablation of hepatic neoplasms

Objective To explore the safety and short?term efficacy of irreversible electroporation (IRE)ablation which is a novel ablation technology in unresectable hepatic neoplasms. Methods Patients with pathologically diagnosed as liver cancer or liver metastases were prospectively enrolled. The patients were not suitable for surgery with PS score ≤ 2. Exclusion criteria included who was not tolerate general anesthesia, severe liver and kidney dysfunction, and with cardiac pacemaker. A total of 16 patients were included in this study. There was 12 males and 4 females, aged 40 to 86 years with mean age (60 ± 10)y. Ultrasound and CT guided percutaneous IRE ablation was performed. Perioperative hemodynamic changes were reviewed. Liver and kindey function before and 7 d after ablation was compare by t test. The adverse reactions within 30 d after ablation treatment were recorded. CT and MR scans within 1 month were performed and the 30 d curative effect was evaluated by the modified RECIST criteria. Results All patients received IRE treatment successfully, and some patients experienced adverse reactions within 30 days after ablation, including abdominal pain in 7 cases, peritoneal effusion in 5 cases, hydrothorax in 4 cases, fever in 3 cases, cough, nausea and vomiting in 2 cases, biliary tract infection and thrombocytopenia in 1 case. After symptomatic treatment, these symptoms were improved. Severe complications, such as massive haemorrhage and bile leakage didn't occur. At 30 days after ablation, the curative effects were evaluated. Complete response (CR) was achieved in 1 patient , partial response (PR) was achieved in 12 patients, stable disease (SD) was in 2 patients , and progressive disease(PD) was 1 patients . The tumor relief rate (complete response+partial response) was 81.3%. Conclusions IRE ablation in the treatment of unresectable hepatic malignant tumor could have many advantages, including high safety, mild adverse reactions, and short?term efficacy. However, its long?term effect still need further observation.
9.Theoretical construction and practical research on the curriculum system of children's rehabilitation specialty in rehabilitation therapy

This article takes the setting of children's rehabilitation specialty in the rehabilitation therapy of Xiamen Medical College as an example, and expounds that in the process of cultivating talents in children's rehabilitation, theoretically, a training model of "2.5+0.5+1" has been built closely around the training goals of "solid foundation, wide caliber and strong skills", highlighting basic pediatric courses, core skills courses for children's rehabilitation therapy and special courses for children's rehabilitation. In practice, the training of "early clinical practice and early probation" for children's rehabilitation skills is stressed. The results show that students have greatly improved their professional knowledge and practical ability, and the effect of "early clinical practice and early probation" is obvious. Among the students who choose the direction of children's rehabilitation, 95% of the students want to engage in the career of children's rehabilitation and believe that the curriculum system will be of great help to their future employment.
10.Analysis of related substances in montelukast sodium granules by HPLC-MS/MS

The aim of this paper was to study the species and sources of related substances in montelukast sodium granules based on HPLC-MS/MS. The related substances of the already marketed montelukast sodium granules(Singulair, 4 mg)were firstly separated and identified by HPLC-MS/MS method, and their chemical structures and sources were confirmed according to the pulished synthesis process and degradation studies. The results revealed that there were 7 related substances, whose chemical structures and sources were confirmed based on the MS/MS technique and the related literature. Impurity 7, montelukast quinoline ring oxynitride, had not been reported. This study systematically analyzed the related substances in the commercical product, which provides useful information for the development and quality control of the product.
