Objective To evaluate the diagnosis of proximal vas deferens and epididymis with contrast-enhanced ultrasonography (CEUS) in healthy adult canines. Methods Six healthy male adult dogs were dissected to expose unilateral vas deferens. CEUS was performed after injecting ultrasound contrast agents into vas deferens by catheter. X ray angiography was taken after injecting compound meglumine diatrizoate. The image features of CEUS and X ray angiography were displayed by CEUS. Results Under CEUS, inguinal segment of proximal vas deferens were straight and symmetrical in diameter, scrotal segment of vas deferens were tortuous, and the tail of epididymis were speckled which enhanced shaped as a comma. The inguinal and scrotal segment of vas deferens section was display well by X-ray angiography. Conclusions CEUS is a new method in proximal vas deferens and tail of epididymis examination. It can provide guidance for the disease diagnosis of seminal vesicle and ductal system.

Specific antibodies purified from sera of rabbit infected with Toxoplasma gondii were used to immun-oscreen the 12-mer and 7-mer phage random peptide libraries. By 3 rounds of screening, 70 clones were picked out ra-ndomly to test the specificity and 43 were found positive by ELISA. 23 of the 43 positive clones showing strong reaction in ELISA were picked out for sequencing. Clone A7 was selected as mimic antigen in ELISA test after analysis of the 23 short peptide sequences. 23 out of 47 sera from rabbits infected with Toxoplasma gondii were ELISA positive by A7 mimic antigen with a positive rate of 68.1%, and 10 of 155 sera from healthy persons showed false positive with a sp-ecificity of 93.5%. The primary result suggests that mimic antigen may have a potential use in diagnose of toxoplasmosis.

ObjectiveTo investigate the haemostatic effect of combining pulsed therapeutic ultrasound (TUS) and microbubble (MB) on splenic trauma.MethodsForteen healthy dogs were divided into 3 groups.Six animals were treated by microbubble-enhanced therapeutic ultrasound (MEUS),the other eight animals were treated with TUS only ( n =4) and the MB only ( n =4) served aS the controls. The spleens of all animals were surgically exposed and a 20 mm long,5 mm deep incision was created on the spleens using scalpel.Human prothrombin complex (HPC) was slowly injected prior to the treatment.During the procedure, the initial bleeding rate and the bleeding rate after the treatment were calculated using gauze weighing.Contrast enhanced ultrasound (CEUS) was performed to assess the blocking effects of splenic circulation. The targeted spleens were harvested for pathological examination.ResultsThere were no differences in initial bleeding rate before treatment. After treatment,in the MEUS group oozed blood was found and the bleeding rate were lower than that of the TUS and the MB groups ( P <0.05).A nonenhanced or perfusion defect region was formed within the treated area. The histological results showed splenic sinus hyperemia, platelet aggregation and intravascular thrombosis. Conclusions Microbubbles enhanced pulsed therapeutic ultrasound can arrest the regional blood circulation of spleen by disrupting the microvasculature and this can be used in splenic heamostasis for treating spleen trauma.

This study is to explore new lead compounds by inhibition of Pin1 for anticancer therapy using temperature sensitive mutants. As Pin1 is conserved from yeast to human, we established a high-throughput screening method for Pin1 inhibitors, which employed yeast assay. This method led to the identification of one potent hits, 8-11. In vitro, 8-11 inhibited purified Pin1 enzyme activity with IC50 of (10.40 +/- 1.68) micromol x L(-1), induced G1 phase arrest and apoptosis, showed inhibitory effects on a series of cancer cell proliferation, reduced Cyclin D1 expression, was defined as reciprocally matched for protein-ligand complex in virtual docking analysis and reduced cell migration ability. In vivo, we could observe reduction of tumor volume after treatment with 8-11 in xenograft mice compared with vehicle DMSO treatment. Altogether, these results provide for the first time the involvement of 8-11 in the anticancer activity against Pin1.

Objective To explore the effects of all-trans-retinoic acid (ATRA) on the proliferation of human lung adenocarcinoma cell line A549 and the expression of APLNR (apelin receptor) gene. Methods The inhibition of proliferation of human lung adenocarcinoma cell lines A549 cultured in vitro with or without ATRA was measured by MTT (methyl thiazolyl tetrazolium,MTT) method. The morphological changes in the cells were observed by light microscopy. The cell cycle and apoptosis were analyzed by flow cytometry. The levels of APLNR,cyclin D1 and p16 proteins were detected by western blot. Results After treatment of ATRA,the proliferation of A549 cells was obviously inhibited in dose-and time-independent manner (P < 0.01).The cell morphology was significantly changed. The cycle of A549 cells was blocked at G0/G1 phase and the apoptosis rate was increased. With the increasing concentration of ATRA,the expressions of cyclin D1 and APLNR were down-regulated but the expression of p16 was up-regulated (P < 0.01).Conclusion ATRA could inhibit the proliferation of A549 cells by retardant cell cycle of A549 cells at G0/G1 phase and inducing the apoptosis, and down-regulate the expression of APLNR gene.

Objective:To understand the results of serum protein electrophoresis in patients with acute brucellosis, and to provide objective indicators for clinical diagnosis and treatment of acute brucellosis patients. Methods:Thirty acute brucellosis patients diagnosed by the Center for Disease Prevention and Control of Donghe District, Baotou City from January to June 2021 were selected as the case group, and 30 local healthy persons who took physical examination during the same period were selected as the control group. Blood samples were collected from the two groups of people, serum was separated and serum protein electrophoresis was performed. Independent sample t-tests were used to compare total protein concentrations, protein fractions (albumin, α1-globulin, α2-globulin, β-globulin and γ-globulin) and albumin/globulin (A/G) ratio. Results:The age of patients in the case group was (52.35 ± 6.74) years old, with 25 males and 5 females. The course of the disease ranged from 0.3 to 2.6 months, with an average of 1.78 months. The age of people in the control group was (52.74 ± 5.47) years old, with 25 males and 5 females. There were no statistically significant difference in age and sex distuibution between the two groups ( P > 0.05). The concentrations of total protein and α2-globulin fractions were not significantly different between the two groups ( t = 0.78, 2.04, P > 0.05, respectively). Compared with the control group, the differences in the percentage of albumin, α1-globulin, β-globulin, γ-globulin in total protein and A/G ratio were statistically significant ( t = 4.78, 4.76, 2.89, 2.20, 3.65, P < 0.05). Conclusions:The percentage of serum albumin in total protein and A/G ratio decrease in patients with acute brucellosis, while the percentage of α1-globulin, β-globulin and γ-globulin in total protein increase. The percentage of albumin, α1-globulin, β-globulin, γ-globulin in total protein and A/G ratio can be uesed as objective indicators for clinical treatment of acute brucellosis.
