2.Preparation and clinical application of cDNA microarray for combined detection of hepatitis virus

Objective To develop the cDNA microarray for the combined detection of hepatitis virus, and to study the feasibility of applying the microarray in clinical setting. Methods For the small and simple genome of HBV and HDV, the specific primers of PCR were designed with Primer Premier 5.0 program according to the conserved region of HBV and HDV, and 10 and 4 gene fragments were obtained respectively, which could be used as the probes of gene chip. As for the complex genome of HCV, the technique of restriction display PCR (RD-PCR) was employed. Some of gene fragments were selected which were comparatively more specific and sensitive as microarray probes. In order to explore the experimental conditions of microarray in vitro detection, three types of gene chip were prepared successively including HBV and HDV simultaneous detection, HCV detection and modified HCV detection. Results The hybridized signals on the gene chip showed that the effect in detection was satisfactory. Through the prepared gene chips mentioned above, some probes with good quality were selected and the microarray was prepared for HBV, HCV and HDV simultaneous detection. The diagnostic capability of the microarray was evaluated following the washing and scanning steps. Linearity: Serial dilutions of the target DNA or cDNA showed that a strong linear relationship existed between the various concentrations of target DNA or cDNA and the fluorescence intensities obtained from microarray assay (r=0.990 2, r=0.992 1, r=0.981 9), and that the detection range for the microarray was from 104 to 1011 copies/ml. Specificity: Samples from other viruses such as YFV, JET and DV were also subjected to the test and the results were all negative. Reproducibility: The reproducibility of this assay system was evaluated by repeated measurements, and the within-run coefficient of validation of HBV, HCV and HDV were 7.1%, 7.2% and 6.6%, respectively, while the between-run coefficient of validation was 7.9%, 8.2% and 7.6%, respectively. Accuracy: By using the BLAST and the GenBank database, the identity of the obtained sequences including 16 fragments of PCR, 24 RD fragments of HCV and some positive serum samples were verified, each sequenced product was confirmed to be a genome fragment of expected size. In order to fulfill the request of clinical diagnosis, a modified protocol for microarray detection was established. This new protocol consisted of two hours of hybridization, omitting the steps of prehybridization and purification of samples, and the hybridization temperature was elevated from 42℃ to 52℃, et al. The whole protocol could be completed in less than 8 hours. 98, 42 and 5 serum samples from hepatitis B, C and D patients and 130 samples from healthy people were analyzed, respectively, by microarray assay and real-time PCR (Taqmam method). 3.Effects of He-Ne irradiation on matrix metalloproteinases- 9 expression during experimental tooth movement in rabbits

Objective To explore the feasibility of He-Ne laser irradiation in promoting orthodontic tooth movement and periodontal remodeling through observing the effect of low power laser on the expression and distribution of matrix metalloproteinases- 9(MMP- 9) during orthodontic tooth movement in rabbit. Methods Thirty rabbits were divided into 1d,3d,5d,7d,14d and 21d groups, each group were given a coil spring connected to the maxillary upper first molar to the upper incisor and fixed bilaterally by a ligature wire and set force to 80g. He-Ne laser irradiation was performed on the right side of the orthodontic tooth, and the left side served as control. Radiation time ranged from 1 day to 5 days, once a day, 15min each time. All of the tissue sections were proceeded with MMP- 9 immunohistochemical staining. MMP- 9 average gray scale in periodontal tissue of the orthodontic teeth was analyzed by a Computer Image Analyzing System. Results The expression level of MMP- 9 increased earlier and lasted for a longer time in the compression and tension area on the side of irradiation by He-Ne laser as compared to those on the control side. There was significant difference between the irradiation side and the control side in terms of the MMP- 9 expression gray scale in the tension zones at 5 and 7 days post-operation, and in the pressure zones at 3,5,7 day post-operation. Conclusion During the movement of orthodontic teeth, local irradiation with He-Ne laser could increase the expression of MMP-9. It was suggested that the irradiation with low power laser accelerate blood vessel and bone remodeling.
4.Effect of basic fibroblast growth factor on the proliferation and differentiation of human preadipocytes

BACKGROUND:Previous studies have shown that basic fibroblast growth factor (bFGF) can enhance fat survival rate by promoting vessel regeneration of transplanted fat.However,there is no clear conclusion addressing its effects on adipocytes.OBJECTIVE:To investigate the effect of bFGF on the proliferation and differentiation of human preadipocytes.DESIGN,TIME AND SETTING:Cytological in vitro comparison observation was conducted at the Medical College of Chinese People's Armed Police Force from March 2006 to December 2007.MATERIALS:Totally 10 samples of adipose tissue from patients undergoing abdominal liposuction were used in this study,including 1 male and 9 females,aged 21-45 years.METHODS:The human preadipocytes were obtained from human abdominal fat particles by isolation,culture and natural purity.The experiment was divided into bFGF and control groups.Cells in the control group were incubated in DMEM.Cells in the bFGF group were incubated in DMEM supplemented with 100 ng/L bFGF.MAIN OUTCOME MEASURES:Morphology of cultured cells was studied to determine the growth curve at 0,3,6,9,12,15 days following culture by inverted phase contrast microscope.Fat aggregation was observed in cells using oil red O staining.Absorbance value was examined using enzyme-linked immunosorbent assay.RESULTS:The preadipocytes represented the spindle-like character with strong proliferation.During 1 to 15 days,the preadipocytes proliferated into monolayers.Some cells became single bubble cells with lipid droplets deposited,close to maturity.The cells in bFGF group had no significant differences with the control group in cell morphology,but the amount of cells in bFGF group was 44% more than the control group.Following oil red O staining,lipid droplet was found in the preadipocytes,showing dark red.Following 15 days of incubation,the concentration of lipid droplet reached a peak.The absorbanca of bFGF group was increased 300% compared with the control group (P < 0.05).CONCLUSION:The bFGF not only promote the proliferation of human preadipocytes,but also induce the differentiation of preadipocytes into mature adipocytes.

Objective To investigate the effect of urinary kallidinogenase on serum concentrations of hydrogen sulfide (H2S),neuron-specific enolase (NSE),and S100β in patients with cerebral infarction (CI).Methods From June 2011 to June 2014,80 patients with CI were chosen as study objectives.All patients were divided into two groups:40 patients in study group (urinary kallidinogenase group),and 40 patients in control group.The death rate,the rate of complication and National Institute of Health Stroke Scale (NIHSS) were compared between two groups.The concentrations of H2S,NSE,and S100βwas compared between two groups.Results In study group,the death rate was 5.00% (2/40),the rate of complication was 22.50% (9/40);in control group,the death rate was 12.50% (5/40),the rate of complication was 15.00% (6/40);and no significant significance was found between two groups (P > 0.05).The NIHSS was (11.2 ± 3.2) in the study group,and (15.7 ± 2.7) in the control group,with statistically significant difference between two groups (P < 0.05).After treatment,the concentrations of H2 S,NSE,and S100β of two groups were decreased significantly (P <0.05).At 1w,2w,and 3w,the concentrations of H2S,NSE and S100βhad statistically significant difference between two groups (P < 0.05).Conclusions Urinary kallidinogenase has a cerebral protective effect,which can decrease the concentration of H2S,and increase the concentrations of NSE and S100βin CI patients.

7.Effects of focal vibration on the physical properties of the muscles of patients with neck-shoulder pain

Objective To evaluate the effects of focal vibration on the physical properties and surface electroyograms of persons with neck-shoulder pain.Methods Thirty-four patients with neck-shoulder pain received 3 to 5 applications of vibratory stimulation of the inner side of the trapezius and scapula for 30 s each time.They reported their pain level using a visual analogue scale.Muscle tone and the elasticity and stiffness of the trapezius muscle were measured bilaterally using a MyotonPRO Muscular Function Evaluation System.Surface electromyography (sEMG) was performed bilaterally during arm extension while standing.The testing was repeated four times:before treatment (baseline),immediately after treatment (IAT),2 hours after treatment (2AT) and 6 hours after treatment (6AT).Results There was a significant reduction in reported pain at IAT and 2AT compared to baseline,but no significant difference was found between the perceived pain at baseline and at 6AT.The values of oscillation frequency,logarithmic decrement and dynamic stiffness had decreased significantly at IAT and 2AT compared to baseline.The average AEMG during upper-arm flexion of the trapezius on the painful side had increased significantly at IAT comparcd to that before the treatment,while the average IEMG and MF were both significantly higher than before the treatment.At 2AT the average AEMG and IEMG were both still significantly higher than at the baseline,but there was no longer a significant difference in the average MF.By 6AT none of the three indicators differed significantly from those before the treatment.Conclusions A single session of focal vibration can relieve pain,increase muscle activation as well as decrease muscle stiffness for people with neck-shoulder pain within 2 hours.

9.Study on Dsg3 induced activation of T cells in first degree relatives of patients with pemphigus vulparis

Objective To investigate The Th1/Th2 and Tc1/Tc2 polarization in the peripheral blood of first degree relatives of pemphigus vulgaris(PV) and healthy control individuals, and to approach the mechanism of the Dsg3-specific autoimmunity in PV. Methods The peripheral blood mononuclear cells (PBMC) from first degree relatives and healthy control was stimulated for72 h with Dsg3 and without Dsg3. Th1/Th2, Tc1/Tc2 was assessed by four-color flow cytometry. Results The mean frequency of Dsg3-spe-cific Th2 cells for PV antibody positive first degree relatives was 10.13%±3.72%, compared with stimula-tion without antigen 7.28%±3.58%, the difference was significant (P<0.05). The percentage Dsg3-spe-cific Th2 was markedly higher in the PV antibody positive first degree relatives group than that in the control group(10.13%±3.72% vs 6.10%±2.82%, P<0.05) , Tc2 was markedly higher also (20.01%± 10.43% v514.91%±8.06%, 20.01%±10.43% vs 9.58%±5.49%, P<0.05). Conclusion When Dsg3 stimulated PBMC were used to stimulate autologous T cells an increased amount of Th2 and Tc2 was observed, it is implied that the imbalance of Th1/Th2, Tc1/Tc2 might play an important role in the initia-tion of PV.
10.Expression of Ki-67 in moderate and high risk gastrointestinal stromal tumors with CD117 immunoreactive

Objective To study the expression and significance of Ki-67 in gastrointestinal stromal tumors(GIST) with CD117 immunoreactive. Methods The expression of Ki-67 was detected by SP method in 25 cases of high risk GISTS, 18 cases of moderate GISTS and 33 cases of extremely low and low risk GISTS,which were compared with the follow-up results. The relationship between Ki-67 index (LI) and risk degree was analyzed. Results Forty patients with moderate and high risk GIST were followed-up, including 26 alive,12 die of GIST and 2 die of other causes. Compared with patients of extremely low and low risk GIST, Ki-67 LI>5 % was correlated with moderate and high risk cases (P <0.01), meanwhile Ki-67 LI was positively correlated with tumor sizes of >5 cm and tumor mitotic cell count >5/50 HPF, but was not with locations(P >0.05). Conclusion Ki-67 LI>5 %, tumor size>5 cm and tumor mitotic cell count>5/50 HPF are risk indicator for GIST with CD117 immunoreactive.
