Objective To explore the feasibility of He-Ne laser irradiation in promoting orthodontic tooth movement and periodontal remodeling through observing the effect of low power laser on the expression and distribution of matrix metalloproteinases- 9(MMP- 9) during orthodontic tooth movement in rabbit. Methods Thirty rabbits were divided into 1d,3d,5d,7d,14d and 21d groups, each group were given a coil spring connected to the maxillary upper first molar to the upper incisor and fixed bilaterally by a ligature wire and set force to 80g. He-Ne laser irradiation was performed on the right side of the orthodontic tooth, and the left side served as control. Radiation time ranged from 1 day to 5 days, once a day, 15min each time. All of the tissue sections were proceeded with MMP- 9 immunohistochemical staining. MMP- 9 average gray scale in periodontal tissue of the orthodontic teeth was analyzed by a Computer Image Analyzing System. Results The expression level of MMP- 9 increased earlier and lasted for a longer time in the compression and tension area on the side of irradiation by He-Ne laser as compared to those on the control side. There was significant difference between the irradiation side and the control side in terms of the MMP- 9 expression gray scale in the tension zones at 5 and 7 days post-operation, and in the pressure zones at 3,5,7 day post-operation. Conclusion During the movement of orthodontic teeth, local irradiation with He-Ne laser could increase the expression of MMP-9. It was suggested that the irradiation with low power laser accelerate blood vessel and bone remodeling.

BACKGROUND:Bone mesenchymal stem cells(BMSCs) can accelerate vascularization of myocardial cell, relieve myocardial remodeling, and improve heart function. However, whether its paracrine action can facilitate the differentiation of CD117+ cardiac stem cells(CSCs) by effecting transforming growth factor-? Ⅲ(TGF-? Ⅲ) receptor expression is still unknown.OBJECTIVE:To study the influence of BMSCs on the expression of TGF-? Ⅲ receptor in the process of differentiation of CD117+ CSCs.DESIGN,TIME AND SETTING:In vitro cytology contract observation. The experiment was performed at the Xinhua Hospital from February 2008 to February 2009.MATERIALS:Neonatal rats, weighing 5-8 g,were used to prepare CSCs.Sprague Dawley rats,4 weeks old, weighing 200-250 g,were prepared for BMSCs.METHODS:CD117+CSCs were obtained from neonatal rats by explant cultures and magnetic bead cell sorting.Immunohistochemical experiment was performed to identify specificity of CSCs.CD117+CSCs suspension were synchronization without serum for 24 hours, washed by PBS for 3 times, and then cultured with DMEM/F12,followed by inoculation into 6-well plate. The cells were divided into the control and co-culture groups. In the control group,CSCs were induced differentiation by cardiac spheres, and Transwell chamber were used in the co-culture group.MAIN OUTCOME MEASURES:The growth of CSCs was observed by inverted microscope. The changes of cardiac Troponin T(cTnT),connexin43,TGF-? Ⅲ receptor,smad2 and phospho-smad2 expressions were detected by western blot at days 1,3,5 and 7 after culture.RESULTS:①CSCs isolated by explants cultures and MACS expressed CD117,which grew in disperse with smaller body.② The expression of cTnT and connexin43 were significantly higher in the co-culture group than that of the control group at days 5 and 7(P

Objective To investigate the anti-inflammatory/immune modulatory effects of high-expressing membrane bound M-CSF-hematopoietic malignant cells on macrophages. Methods After coculturing RAW264.7 and murine macrophage cell line with Namalwa-M, a cell line stably expressing mM-CSF, and companing with Nainalwa-V, a cell line stably transfected with the empty vector as the control, flow cytometry was used to detect the expression of the marker of alternatively activated macrophages, CD206, and intracellular expression of IL-10, IL-12, IL-6 and TNF-α to study the immunophenotype of RAW264.7; phagocytic assays to investigate their functional activity in vitro. Results RAW264.7 cocultured with Namalwa-M consistently showed high-level expression of CD206, which indicated activities of these macrophage cells were increased. Furthermore, these RAW264.7 expressing high levels of IL-10, TNF-a and low levels of IL-12, IL-6, as determined by intracellular staining were suggested that the phagocytic activity was increased. Functionally, RAW264.7 cocultured with Namalwa-M showed a higher level of phagocytic activity. Conclusion Macrophage generated in vitro after cocultured with mM-CSF-expressing hematopoietic malignant cell line could be transformed into abnormal macrophage with an immunophenotype defined as IL-10-high, IL-12-low, IL-6-low and TNF-α-high.
