1.Differentiation of mesenchymal stem cells from rat bone marrow into cardiomyocyte-like cells in vitro

Objective To explore the optimal condition for the differentiation of rat bone marrow mesenchymal stem cells (MSCs) into cardiomyocyte-like cells in vitro. Methods The MSCs were purified by percoll mediated density gradient centrifugation, and then randomly divided into A, B and C groups treated with 5, 10, 20 ?mol/L 5-azacytidine, respectively. MSCs in each group were treated with 5-azacytidine for 12, 24 and 48 h, and then cultured for 28 d before examination. The morphological changes in MSCs were observed with inverted microscope. The expression of ?-actin and cTnT was determined by immunohistochemistry and the protein expression of cTnT was determined by Western blotting. Results At day 28 after induction with 5-azacytidine, scattered myogenic structure was observed in the plasma of MSC under electronic microscope. Positive expression of ?-actin and cTnT were detected with immunohistochemistry. As compared with 10 ?mol/L (48 h) and 20 ?mol/L (12, 24 and 48 h) subgroups, there was no significant difference in the expression of cardiac troponin T among 5 ?mol/L (48 h) and 10 ?mol/L (24 h) groups without effect on the biological characteristics. Conclusion MSCs could be differentiated into cardiomyocyte-like cells by 5-azacytidine in vitro. Induction with 5 ?mol/L 5-azacytidine for 48 h or 10 ?mol/L of 5-azacytidine for 24 h is the optimal condition for the differentiation of MSCs into cardiomyocyte-like cells in vitro.

The Chinese and English published literature regarding the effects of acupuncture on microcirculation in recent years were analyzed, and it was found out that the theory of TCM meridian had subtle connection with microcirculation on structure and function, which could provide a possible breakthrough for the research of meridian essence. Acupuncture, through multiple links, multiple paths and multiple levels, could regulate microcirculation under physiological or pathological condition, which was a bidirectional benign adjustment function, and reflected the characteristic of acupoint specificity. Due to many shortcomings in the current studies, studies with rigorous design, large sample size and multicenter are expected in the further to make a standard systematized research.

3.Health care follow-up after chronic heart failture can reduce cardiovascular events and improve quality of life

Objective To observe the impatcts of health care follow-up after chronic heart failture on cardiovascular events and qualigy of life.Methods One hundred and sixty-six patients who were confirmed to have chronic heart failture in our hospital during February 2007 and December 2008 were enrolled in this investigation.The patients were then assigned to the health management group (n=65) or the non-health management group (n=101) and followed up for 5 years.All-cause mortality,readmission for heart failture and quality of life (SF-36 score) were evaluated.Results There was no significant difference of all-cause mortalitv between the two groups (health management group:12.30% (8/65); non-health management group:14.85% (15/101; x2=0.22,P>0.05),although a statistically significant difference of readmission rate was found between the two groups (health management group:12.30% (9/65); non-health management group:35.64% (36/101; x2=9.51,P<0.05).In health management group,the scores of SF-36 were significantly increased at 3 (114.11 ±2.76) or 5 years (116.07± 15.43) when compared with baseline (91.37 ± 2.62) (x2 =102.39,P<0.05).For the non-health management group,the scores of SF-36 were significantly decreased from 92.65±3.65 at baseline to 90.09±5.12 at 3 years or 89.08±5.71 at 5 years (x2=102.39,P<0.05).Conclusion Health care follow-up after chronic heart failture may not decrease all-cause mortality,although can decrease readmission rate and improve quality of life of the patients.
4.Effect of rosiglitazone on SREBP-1 and TGF-β1 expressions and accumulation of ECM in renal tubular cells of Wistar rats treated with high fat diet

AIM: To study the effect of high fat diet on the expression of sterol regulatory element biding protein-1 (SREBP-1) and transforming growth factor β_1 (TGF-β_1) in renal tubular cells and rosiglitazone intervention. METHODS: Wistar rats were treated with high fat diet and rosiglitazone for 3 months. The serum glucose, serum insulin and serum triglyceride were detected. Oil Red O staining was used to observe the renal lipid deposit and Masson staining was for the detection of ECM accumulation. SREBP-1, TGF-β_1 and FN protein were determined by the methods of immunohistochemistry and Western blotting. SREBP-1 mRNA was detected by in situ hybridization. RESULTS: Rosiglitazone prevented effectively the increase in serum glucose, serum insulin and serum triglyceride resulted from high fat diet. High fat diet led to lipid droplet formation in renal tubular cells and interstitial ECM accumulation, which was decreased by rosiglitazone treatment. Compared to normal rats, SREBP-1 protein and SREBP-1 mRNA showed high expressions in high fat diet rats that were lowered by rosiglitazone. The precursor segment and mature segment of SREBP-1 protein were decreased by 27.39% and 27.32%. Similarly, the high expressions of TGF-β_1 and FN protein in kidney of high fat diet rats were also prevented by rosiglitazone intervention. Compared to high fat diet rats, the expression of TGF-β_1 in rosiglitazone treatment rats was lowered by 19.14%. CONCLUSION: Rosiglitazone prevents effectively the over-expression of SREBP-1 and TGF-β_1 in renal tubular cells, and decreases lipid accumulation and ECM production in rats fed with high fat diet.
5.Effect of Electroacupuncture at Point Zusanli (ST36) on Gastric HSP70 mRNA in Rats with Stress Gastric Ulcer

Objective To investigate the effect of electroacupuncture at point Zusanli (ST36) on gastric expression of heat shock protein 70 (HSP70) mRNA in rats with stress gastric ulcer and explore the molecular mechanism of its prevention and treatment of gastric ulcer. Method Fifty-one SD rats were randomly allocated to normal control (normal), stress gastric ulcer (model) and electroacupuncture at point Zusanli (electroacupuncture) groups. A model of stress gastric ulcer was made by cold restraint. From the second day after model making, the electroacupuncture group received electroacupuncture at point Zusanli 20 min once daily for three consecutive days, with left and right points Zusanli alternately needled; the model group, the same restraint treatment as in the electroacupuncture group; the normal group, no treatment. Gastric mucosal ulcer index (UI) was calculated using the GUTH’s method. The pathological morphology of gastric tissue was observed by HE staining. Gastric HSP70 mRNA expression was determined by RT-PCR. Result Different numbers and sizes of obvious gastric mucosal ulcers were observed, UI increased significantly (P<0.001) and HSP70 mRNA expression decreased significantly (P<0.05) in the model group compared with the normal group. The pathological change in gastric tissue improved somewhat, gastric mucosal UI decreased significantly (P<0.05) and HSP70 mRNA expression increased significantly (P<0.01) in the electroacupuncture group compared with the model group. Conclusion Electroacupuncture at point Zusanli can upregulate gastric HSP70 mRNA expression and promote the repair of gastric mucosa injuries in rats with stress gastric ulcer.
6.Hemolytic anemia as the first manifestation in hepatolenticular degeneration combined with ichthyosis:a case report

Objective To explore the clinical features of hepatolenticular degeneration (HLD) with hemolytic anemia as the first manifestation, and its relationship with ichthyosis.Method The clinical manifestations, course, diagnosis, and treatment of HLD combined with ichthyosis in one child who presented with hemolytic anemia as the ifrst manifestation, were retrospectively analyzed.Results In ten-year-old girl, the ifrst symptom was fatigue and jaundice, and then progressed to abdominal distension 5 days after admission. The reticulocyte percentage was 8.5%. Coombs test was negative. The K-F rings appeared on the cornea. The CT scan of Abdomen showed pleural and peritoneal effusion, abnormal substance deposition in liver and splenomegaly. There were mutations inATP7B andTGM1 genes, both of which were located on fourteenth of chromosomes. The diagnosis of HLD combined with ichthyosis was conifrmed. After symptomatic and supportive treatments such as removal of copper, prevention of the copper absorption and supplement of vitamin A, the disease was brought to remission.Conclusion HLD with hemolytic anemia as the ifrst manifestation, combined with ichthyosis was extremely rare.
7.Nursing about the patch test for 122 patients with chronic eczema

Objective To discuss the key points of nursing the patch test for patients with chronic eczema. Method Analyze the clinical data and the concreted nursing measures which obtained from 122 chronic eczema patients. Result The top 5 chemical allergic resources of these patients were 0.1% thiomersalate, 5% nickel sulfate,7% aromatic compound,1% formaldehyde(water solution),1% styrene. Conclusion\ The key points of getting successful patch test were following the attention issues before,during and after patch test,and to pay attention to the living nursing and explanation of patch test for patients.
8.Factors affecting blood lipid control among residents at a high risk of atherosclerotic cardiovascular disease

Objective:
To investigate the proportion of achieving the blood lipid control target and its influencing factors among residents at a high risk of atherosclerotic cardiovascular disease (ASCVD), so as to provide insights into management of blood lipid among residents at a high risk of ASCVD.
Methods:
Residents at a high risk of ASCVD and at ages of 35 to 70 years were sampled using a multi-stage cluster sampling method from 6 counties (districts) in Shaoxing City from May to July 2021. The residents' demographics, smoking, alcohol consumption and medical history of chronic diseases were collected using questionnaires, the height, weight, waist circumference (WC) and blood pressure were measured, and the total cholesterol (TC), triacylglycerol (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C) and fasting blood glucose were detected. The proportion of blood lipids achieving the control target was analyzed, and factors affecting the proportion of blood lipids achieving the control target were identified using a multivariable logistic regression model.
Results:
A total of 1 695 individuals at a high risk of ASCVD were enrolled, including 940 men (55.46%) and 755 women (44.54%), with a mean age of (62.56±6.08) years. There were 285 participants that achieved the target of blood lipid control (16.81%). Multivariable logistic regression analysis identified gender (male, OR=1.962, 95%CI: 1.396-2.758), age (OR=1.037, 95%CI: 1.013-1.061), WC (OR=0.979, 95%CI: 0.964-0.995), diastolic blood pressure (OR=0.981, 95%CI: 0.967-0.994), smoking (OR=1.485, 95%CI: 1.034-2.133), alcohol consumption (OR=0.684, 95%CI: 0.498-0.941), hypertension (OR=1.428, 95%CI: 1.006-2.207), administration of hypoglycemic drugs (OR=2.326, 95%CI: 1.720-3.144) as factors affecting the achievement of the target for blood lipid control among residents at a high risk of ASCVD.
Conclusions
Individuals at a high risk of ASCVD with higher WC, higher diastolic blood pressure and alcohol consumption are less likely to achieve the target for blood lipid control, while male individuals with older age, hypertension and administration of hypogcemic drugs are more likely to achieve the target for blood lipid control.
9.Molecular Identification and Phylogenetic Analyses of Coxsackievirus A24v Causing an Outbreak of Acute Hemorrhagic Conjunctivitis in Jiangxi, China, in 2010.

To identify the cause of an outbreak of acute hemorrhagic conjunctivitis (AHC) in Jiangxi (China) in 2010, 20 eye conjunctival swabs were first collected from AHC patients. Then, viruses were isola- ted and tested for human enterovirus 70, coxsackievirus A24 variant (CV-A24v) and adenovirus using the polymerase chain reaction. All CV-A24v isolates underwent sequencing of 3C and VP1 coding regions. Then, a phylogenetic tree was constructed for Jiangxi CV-A24v and worldwide CV-A24v based on,3C and VP1 regions, respectively. Ten out of 20 specimens were positive for CV-A24v, implying that the outbreak was caused by CV-A24v. The phylogenetic tree based on the 3C region showed that Jiangxi CV- A24v belonged to cluster 5 in genotype IV (GIV-C5) with strains isolated throughout the world after 2010, and were divided further into A and B lineages. Phylogenetic analyses of the VP1 region showed that all of the worldwide CV-A24v strains isolated after 2000 could be divided into five groups (1-5). Jiangxi CV-A24v was classified into group 5 and also divided further into A and B lineages upon analyses of the 3C region. These data suggested that CV-A24v causing AHC outbreaks in China in 2010 belonged to GIV-C3 and GIV-C5. At least two transmission lineages were circulated in Jiangxi in 2010. The classification of CV-A24v isolated after 2010 worldwide using the phylogenetic tree based on the VP1 region was almost consistent with that based on the 3C region and also had significant chronological clustering.

Objective To obtain a better understaning of the clinical features of Castleman tumor associated paraneoplastic pemphigus. Methods The clinical features and therapy of 10 cases of this disease, diagnosed in the Department of Dermatology of Peking University First Hospital were analyzed. Results Castleman tumor was shown to be the most common neoplasm associated with paraneoplastic pemphigus in China. The clinical presentations, histopathologic characteristics, CT scan findings, and immunologic features were all unique. The early diagnosis and removal of the Castleman tumor are crucial for the treatment of this tumor-associated autoimmune disease. Conclusions Because Castleman tumor is directly related to the induction of autoimmunity, early diagnosis and prompt removal of the tumor are essential to the management of this disease.
