1.Development of Hyperpurificatory Biosafety Cabinet

Objective To create a sterilized condition in field operation and the daily grind avoiding the intercross contamination between the experimentation and outside. Methods The air in the experimentation was filtrated by primary and advanced filter. High intense ultraviolet was used to sterilize the air, and negative compression was kept in the experiment condition. Results Hyperpurificatory biosafety cabinet guaranteed the sterilization condition in field operation and the daily grind, avoiding the intercross contamination. Conclusion Hyperpurificatory biosafety cabinet can be used in medical treatment, pharmacy, food industry, electrical industry, instrumentation, bioengineering, scientific tests, especially biology species identification in biological warfare.
2.Clinical Study of Treatment with Nourishing Qi and Activating Blood Circulation Chinese Medicine Combined with Conventional Western Medicine for Coronary Heart Disease Angina Pectoris

Objective To observe the therapeutic effects of nourishing qi and activating blood circulation Chinese medicine combined with conventional western medicine on coronary heart disease(CHD) angina pectoris.Methods Eight-three patients with CHD angina pectoris were randomized into treatment group(53 cases) and control group(30 cases).The treatment group was treated with Chinese medicine of nourishing qi and activating blood circulation combined with conventional western medicine,and control group was treated with conventional western medicine only.The changes of clinical symptoms and electrocardiogram were observed,and serum IL-6 was detected before and after treatment.Results The clinical symptoms of angina pectoris and electrocardiogram were obviously improved,and IL-6 was decreased.The treatment group was better than the control group(P

DQA1*0501.TSHR peptides of 183~198、195~210、248~263、301~320、343~362 and 357~376 could bind with high affinity to both HLA-DR3 and HLA-DQA1*0501,all of their IC50 values less than(1 ?mol/L),but they could bind to neither HLA-DR7 nor HLA-DQA1*0201 with high affinity.Conclusion Epitopes of TSHR 183~198,195~210,248~263,301~320,343~362 and 357~376 on the TSHR extracellular domain might be the auto-antigens to cause GD.
4.Expressions of interleukin-27,-35 and Epstein-Barr virus-induced gene 3 mRNA in peripheral blood of patients with psoriasis vulgaris

Objective To detect the expression of interleukin (IL)-27,-35 and their common subunit Epstein-Barr virus-induced gene 3 (EBI3) mRNA in peripheral blood from patients with psoriasis vulgaris (PV).Methods Peripheral blood was collected from patients with psoriasis vulgaris (PV) in progressive (n =22),stationary (n =20) and regressive (n =20) stage as well as 30 healthy human controls.A competitive enzymelinked immunosorbent assay (ELISA) was performed to determine the serum level of IL-27 and IL-35.SYBR Green-based real-time reverse transcription (RT)-PCR was carried out to measure the mRNA expression of EBI3 in peripheral blood mononuclear cells (PBMCs).Results The patients with psoriasis vulgaris showed a higher level of IL-27 and IL-35 compared with the healthy controls.The serum level of IL-27 was significantly higher in the patients with progressive,stationary and regressive psoriasis vulgaris than in the healthy human controls ((226.72 ±71.32) ng/L,(160.80 ± 20.02) ng/L and (181.93 ± 27.36) ng/L vs.(138.66 ± 33.93) ng/L,P< 0.01,0.05,0.01,respectively),and higher in the patients with progressive and regressive psoriasis vulgaris than in those with stationary psoriasis vulgaris (P < 0.01,0.05,respectively).Increased serum level of IL-35 was observed in the patients with regressive psoriasis compared with those with progressive and stationary psoriasis and healthy controls ((355.78 ± 49.82) ng/L vs.(309.36 ± 36.10) ng/L,(299.14 ± 31.98) ng/L and (292.43 ± 36.26) ng/L,all P < 0.01),while no statistical difference was observed between the patients with progressive and stationary psoriasos and healthy controls.The mRNA expression level of EBI3 in the patients with progressive,stationary and regressive psoriasis was 20.15,16.17 and 19.74 times,respectively,higher than that in the healthy controls.Conclusion IL-27,IL-35 and EBI3 may function as anti-inflammatory factors in the development,as well as the reversal of progression,of psoriasis.

This study aimed to assess the efficacy of intraoperative neural monitoring (IONM) in preventing recurrent laryngeal nerve palsy during thyroid cancer surgery, analyze and elaborate the theory, operation standard, application, benefits and disadvantage based on clinical practice.
6.Investigation of Agars Culture Mediums Sterilized by Ethylene Epoxide

Objective To develop a new method for sterilization in agars culture mediums,i.e.Agars culture mediums is sealed to be sterilized and preserved after resolution.Methods The effect of rudimental Ethylene Epoxide was investigated,and the germiculture results by different preserving methods were analyzed.Air sedimentation bacterium was sampled by different methods to analyze the bacterium capture rate.Results Ethylene Epoxide sterilized agars culture mediums bacterium capture rate was similar with regular agars.Conclusion Ethylene Epoxide sterilized agars culture mediums can be used in sampling of clinical treatment and other outburst incident,reducing the preparation time.
7.Clinical value of dual-source CT in evaluating coronary artery disease

Objective To explore the clinical value of dual-source CT(DSCT) in the diagnosis of coronary artery disease by comparing the result of DSCT and coronary angiography(CAG).Methods From July 2009 to August 2010,36 patients with suspected coronary hear disease underwent both DSCT coronary angiography(DSCTCA) and selective GAG examination.The results of DSCT coronary angiography were compared with CAG which was the gold standard of diagnosing coronary disease.The sensitivity,specificity,positive and negative predictive value,and accuracy of DSCT were evaluated.Results 144 coronary arteries were well evaluated of 36 cases.The sensitivity,specificity,positive and negative predictive value,and accuracy of DSCT in the diagnosis of coronary hear disease were 100.00%,66.67%,93.75%,100.00% and 97.20% respectively by the number of patients;The McNemar test showed no significant difference between two methods( x2 =0.500,P >0.05 ) with a higher diagnosis concordance (Kapp =0.768,P <0.01).By detecting moderate to severe coronary artery stenosis,the sensitivity,specificity,positive and negative predictive value,accuracy were 95.24%,96.08%,90.91%,98.00%,95.83%,respectively.The McNemar test showed no significant difference between two methods(x2 =0.167,P >0.05) with a higher diagnosis concordance ( Kapp =0.899,P < 0.01 ).Conclusion DSCT has a diagnostic accuracy of coronary heart disease close to that CAG,especially in evaluating patients with moderate to severe coronary artery stenosis,it has high compliance and consistency with CAG.DSCT is a noninvasive,safe and reliable radiographic method.

Objective To investigate the pathophysiological mechanism of proliferation and apoptosis in spermatogenic cell of idiopathic oligospermia. Methods Forty one patients with oligospermia were included in the trial.Testicular tissues,which were obtained with Tru Cut needle biopsy,were observed under light microscope.Expressions of p16 and PCNA,incidence of apoptosis in spermatogenic cells were detected by immunohistochemistry and in situ 3′ DNA end labeling technique. 10.The role of stanniocalcin 2 in oral squamous cells carcinoma

Background and purpose:About 80%patients with oral and maxillofacial malignant tumor are oral squamous cell carcinoma (OSCC). OSCC is one of the most common cancers in the world with less than 50%survival rate over 5 years. This experiment aimed to explore the effect of stanniocalcin 2 (STC2) on apoptosis, proliferation, migration and invasion of OSCC cell. Methods:RNA interference (RNAi) vector pLKO.1-shSTC2 was constructed and transfected into KB cells. Cell proliferation and cell apoptosis were then assessed by CCK8, APC Annexin V/7-AAD and lfow cytometry. Differences of migration and invasion between KB scr and KB STC2i were examined by cell scratch and transwell tests. Finally, this study detected the apoptosis-associated proteins and metastasis-associated proteins by Western blot. Results:STC2 down-regulation plasmid was constructed and transfected into KB cells. CCK8 prolifera-tion assay revealed that the STC2 down-regulation inhibited KB cells proliferation. By treating with cisplatin, this study found that STC2 silence could facilitate cell apoptosis signiifcantly. With the knock down of STC2 gene, the expressions of Bcl-2, Caveolin-1 andβ-catenin were decreased but the expression of bax was obviously increased. Conclusion:These data suggest that STC2 may be involved in the apoptosis, proliferation, migration and invasion of OSCC KB cells. Simultaneously, it can signiifcantly weaken the sensitivity of KB cells to chemotherapeutic drug cisplatin.
