The education,as well as the fostering environment,of the basic medical college postgraduates has its strong "local features".Our college exerts the subject advantages,empha-sizes on the feature-fostering mode,and implements level-management(That is: college—teaching office—scientific research office).Management is centered on the daily fostering of the depart-ment,teaching office and scientific research office.With the target management as the guidance,we strengthen process management of different stages including the study of basic knowledge,topic-selection,topic opening,experiment research and thesis-writing while attaching importance to the behavioral management,consciousness of teamwork,spirit of commitment and devotion,foster and improve the comprehensive quality of communication ability,pursuit of truth etc.

Recommending postgraduate without tests is the path to improve the quality of postgraduate and choose talents with news ideas.In order to consummate the recommendation in local key medical college,the school of postgraduate in Hebei medical university formulated the selecting quota and standard scientifically and reasonably,selected the morally and academically excellent graduates,reasonably distributed quota of recommended exam-free postgraduates,reinforced cooperation between different colleges,explored the linkage among learning,studying and producing and attempted orderform talent-training model.
4.Effect of increased serum TNF-αon lipid accumulation in kidney of diabetes

Aim Todeterminetherelationshipbe-tween serum TNF-α and renal abnormal lipid metabo-lismindiabeticmice.Methods CD1micewerein-jected intraperitoneally with STZ (150 mg·kg-1 ) and the type 1 diabetic mice were determined with high fasting blood glucose ( >16. 7 mmol · L-1 ) 72 hours after injection. After fed for one month, normal control mice and diabetic mice were sacrificed. The serum TNF-α level was detected by the method of ELISA. Immunohistochemistry and Western blot were used to explore the expression of SREBP-1 and ADRP. Re-sults TheresultsofELISAshowedthatserumTNF-αwas increased by 7 . 73 times in diabetic mice compared with normal mice. SREBP-1 and ADRP expressions were located in renal tubular cells. Again, it was con-firmed by Western blot that SREBP-1 precursor seg-ment, SREBP-1 mature segment and ADRP were re-spectively enhanced by 2. 31 times, 1. 74 times and 1. 72 times in diabetic mice in comparison with normal control mice . In addition , the data analysis revealed a
positive correlation ( correlation coefficient 0. 914 ) be-tween serum TNF-αand renal ADRP expression. Con-clusion TheincreasedserumTNF-αmaybeoneof factors involved in renal lipid accumulation of diabe-tes.
5.The time-dependent effect of insulin on the expression of SREBP-1,FAS and lipid droplet formation in HKC cells

Aim To investigate the time-dependent effect of insulin on the expression of SREBP-1(sterol regulatory element binding protein-1),FAS(fat acid synthase)and lipid droplet formation in HKC cells(human renal proximal tubular epithelial cells line).MethodsHKC cells were respectively treated with 100 nmol·L~(-1) insulin for 0,2,4,6,12 h and 24 h.The analysis of SREBP-1 and FAS mRNA was performed by RT-PCR and the expression of SREBP-1 protein was detected by Western blot and immunocytochemistry.Furthermore,Oil Red O staining was used to determine cellular lipid droplet formation.ResultsCompared with HKC cells of 0 h group,there was no difference of SREBP-1 and FAS mRNA in HKC cells of 2 h group.However,the expression of SREBP-1 and FAS mRNA was significantly increased in HKC cells of 4,6 h and 12 h group.Further,the most expression of SREBP-1 and FAS mRNA was at 6 h group and was respectively increased by 3.578 and 4.272 times compared with 0 h group.The results of Western blot showed that the precursor and mature segments of SREBP-1 protein in 4,6 h and 12 h group HKC cells were increased and those of 6 h group HKC cells were the highest and about 4.106 and 5.167 times than those of 0 h group HKC cells.Immunocytochemistry presented the result that SREBP-1 protein was located in the plasma and the expression of 4,6 h and 12 h group HKC cells was significantly higher than that of 0,2 h and 24 h group HKC cells.The result of Oil Red O staining showed that lipid droplet markedly deposited in 6 h group HKC cells,contrarily,no lipid droplet was found in HKC cells of other groups.ConclusionAbove results suggested that insulin up-regulated SREBP-1 and FAS in time-dependent manner that led to cellular lipid droplet deposit,which may play an important role in the pathogenesis of renal lipid accumulation in metabolism syndrome.
6.Expression of spliced XBP-1 s and ADRP in kidney of diabetic rats

Purpose To investigate the expression of XBP-1s and ADRP in kidney of diabetic rats. Methods Diabetic rat models were successfully established by intraperitoneal injection of STZ. After two months rats were sacrificed and XBP-1s and ADRP were de-tected by immunohistochemistry and Western blot. Results XBP-1s and ADRP were located in renal tubular cells and increased by a-bout 2. 017 times and 1. 544 times in comparison with normal control rats (P<0. 05). Moreover, it was shown that high expression of XBP-1s was commonly accompanied with increased ADRP by Pearson correlation analysis and the correlation coefficient was 0. 723 (P<0. 05). Conclusion The increased XBP-1s may cause the up-regulation of ADRP in the kidney of diabetic rats.
7.Screening of Active Compounds Against HIV-1 Reverse Transcriptase Based on Molecular Docking

Objective To screen the non-nucleoside compounds against HIV-1 reverse transcriptase by molecular modeling and bioactivity assay. Methods Surflex-Dock module of Tripos SYBYL software was used to simulate the binding pattern of 22 000 compounds in SPECS database with the active pocket of HIV-1 reverse transcriptase. Based on the simulation results, the interaction mode between the above compounds and the crystal structure of HIV-1 reverse transcriptase was analyzed. The compounds with higher docking scores and better binding pattern were determined by anti-HIV-1 ac tivities test in vitro. Results The virtual screening results showed that the docking conformation of 1- (4-fluorophenyl) -3- [2- (1H-indol-3-yl) ethyl] thiourea was similar to the embedded ligand in Rilpivirine crystal structure. 1- ( 4-fluorophenyl) -3- [ 2- ( 1H-indol-3-yl) ethyl] thiourea was held together with the key residue Lys101 in docking pocket of HIV-1 reverse transcriptase by hydrogen bonds, and hadπ-πstacking action together with the conservative residue Trp229 and the aromatic residue Tyr181 respectively. The bioassay in vitro results showed that when the proliferation rate of C8166 lymphocyte syncytium infected by HIV-1ⅢB arrived 50% ( EC50) , the concentration of 1- ( 4-fluorophenyl) -3- [ 2- ( 1H-indol-3-yl) ethyl] thiourea was 5.45μg/mL. Conclusion Molecule docking technology is an effective approach to reducing the screening of candidate compounds with micromolecular activity, and can be used to predict the interaction mode between the compound and the target receptor. In the study, active compound 1- (4-fluorophenyl) -3- [2- (1H-indol-3-yl) ethyl] thiourea has been screened out by molecule docking technology.
8.A randomized trial on the efficacy and safety of Advagraf vs tacrolimus in prevention of acute liver allograft rejection

Objective To evaluate the efficacy and safety of tacrolimus exposure in stable liver transplant recipients converted from FK506 twice a day to Advagraf (tacrolimus extended-release capsules) once daily. Methods This was an open-label, random, control and multi-center study.Eligible patients were 19 to 70 years of age, 6 months post-transplant with stable renal and hepatic function and receiving stable doses of tacrolimus twice a day for 2 weeks prior to enrollment. There were 86 patients in the experimental group and the control group, separately. The average age of experimental group and control group was 46 ± 10 and 49 ± 9, respectively. Patients in experimental group received Advagraf, once daily, and the dose was adjusted according to the drug concentration,and the drug concentration was between 2 to 10 μg/L. The control group given tacrolimus, twice daily, and the drug concentration was between 2 to 10 μg/L. Results The incidence of acute rejection reaction was 1.20 % and 1.18 % respectively in experimental group and control group, and the 95 %confidence interval was -3.25% ~3.31 % and -3.26% ~ 3.34 %, individually. There was 1 case of acute rejection reaction in experimental group and control group, respectively. The patient and organ survival rate was 100%. Sixteen adverse events occurred in 15 patients (17.65 %) of the experimental group, and 10 adverse events occurred in 10 patients (11.63 %) of control group. Severe adverse events relating to the test drug in experimental group occurred in 4 patients (4. 71 %). and 2 patients (2. 33) in control group.Conclision Clinical trials indicated that Advagraf has efficacy and safety profiles similar to those of tacrolimus. The drug is safe and may improve patient compliance.
