1.Histocompatibility evaluation of nano hydroxyapatite-40% zirconia composite bioceramic

[Objective] To evaluate the histocompatibility of nano hydroxyapatite-zirconia composite bioceramic.[Methods]According to the standard of ISO 10993-1,cytotoxicity experiment,acute toxicity test,hemolysis test and in vivo implantation(90 days)test were conducted to evaluate the histocompatibility of nano hydroxyapatite-zirconia composite bioceramic.[Results]The score of cytotoxicity experiment was lower than grade I,and there was no significant inhibition of cell growth,no acute toxic reaction or hemolytic reaction.And the in vivo implantation met the requirements of the biological evaluation of implant materials.[Conclusion]Nano hydroxyzpatie-zirconia composite bioceramic showed a good histocompatibility.It has a broad prospect as a biomaterial scaffold in the bone tissue engineering.
2.Analysis of somatic mutations in membrane proteins of tumor cells

Objective To screen specific mutations on extracellular regions of membrane proteins ( extracellular membrane protein mutations ) in tumor cells and provide the reference information for target searching in cancer precision medicine .Methods Somatic mutations on extracellular regions of membrane proteins of 7042 tumor samples were collected to screen all specific extracellular membrane protein mutations, and the overall distribution of these mutations were obtained by statistical analysis.Genes, gene site and cancer types occured high frequency of extracellular membrane protein mutations were identified.Results 97193 specific extracellular membrane protein mutations were obtained from 4938362 somatic mutations in 7042 tumor samples (30 cancer types), the statistical analysis showed that 4347 genes and 65532 sites were involved in these specific mutations.The study further analyzed five genes (MUC16、LRP1B、CSMD3、RYR2、USH2A), one site (17:37868208) and six cancer types (including colorectal cancer, melanoma, uterine cancer, brain lower grade glioma, lung adenocarcinoma and stomach adenocarcinoma) which occured high frequency of extracellular membrane protein mutations.Conclusion An information library of specific mutations on extracellular regions of membrane proteins was established and the distribution of these specific mutations was obtained which can provide reference information for target detection in targeted cancer therapy and immunological therapy.
3.Bibliometric analysis of antibody-drug conjugates

Objective To retrieve and analyze domestic and international literatures about antibody-drug conjugates, and understand the recent progress and current situation.Methods PubMed, Web of Science and CNKI were searched to collect all the literatures connected with ADCs from the beginning to January, 2016.Endnote X7 was used to sort out and summarize.The type of literature, published year, first author, research institution, published journal, cited frequency, research contents and patent situation were analyzed with bibliometric methods.Results A total of 645 literatures were included, among which 495 were foreign articles and 150 were Chinese articles.The literatures greatly increased after the 21st century.The top one nation and journal which published the most articles were America and Clinical Cancer Research, respectively.Krop IE and Younes A published the most articles.Among them, the most frequently cited paper was cited up to 686 times.Selection of the targets, site-specific drug conjugation to antibodies and cytotoxic agents were frequently involved.Conclusion ADCs, which have made breakthrough progress, are the focus in the field of cancer research.However, there is still room for improvement, and we still need further exploration.
4.Antimicrobial resistance of bacteria strains producing extend spectrum β-lactamase in clinic

ObjectiveTo investigate the resistance character and trend of bacteria strains producing extend spectrum β-lactamase (ESBLs) in clinic.MethodsThe ESBLs-producting K. pneumoniae and E.coli producing ESBLs isolated from Apr 2001 to Mar 2004 in our hospital were screened and confirmed according to NCCLs method.ResultsK. pneumoniae and E.coli with the ESBLs phenotype were increased in recent 3 years with the resistance rate and Gm(MIC) also higher than that not producing ESBLs.ConclusionThe dection of bacteria strains producing ESBLs will be enhanced in order to provide reasonable treatment information.
5.Analysis of nosocomial infection in patients with spinal cord injury

ObjectiveTo investigate the risk factors of nosocomial infection(NI) of patients with spinal cord injury(SCI),strengthen the prevention and control measures and reduce the NI rate.MethodsClinical data of 967 patients hospitalized in our hospital from Jan 2002 to Dec 2004 were analyzed retrospectively.ResultsThe nosocomial infection rate of SCI patients was 30.3%(293 cases),the most common infection sites were urinary tract,respiratory tract,gastrointestinal tract and wound.The major pathogen was gram-negative bacteria.The infection rate of patients with cervical SCI was significantly higher than patients with thoracic and lumbar SCI(P<0.01).ConclusionSCI patients are susceptible to NI;rehabilitation at early stage should performance to improve living ability of patients and decrease the NI rate.
6.Clinical research on effects of fosamax on the healing of radius distal osteoporotic fracture in postmenopausal women

Objective To observe the effects of fosamax on the fracture healing and the bone mineral density(BMD)in postmenopausal women with radius distal osteoporotic fracture(RDOF).Methods All the 62 patients with RDOF were randomly divided into 2 groups after the fracture was fixed manually.Thirty-two patients in treated group took fosamax 1 tablet(70 mg)per week for 12 weeks and caltrate D 600 mg per day,while the other 30 patients in control group took caltrate D 600 mg per day only.The BMD and the fracture healing time were detected after 12 weeks' treatment.Results The fracture healing time was(9.31±2.50)weeks in treated group and was(13.0±2.8)weeks in control group(t=5.54,P<0.01).BMD was significantly increased after treatment in treated group[(0.615±0.075)g/cm2 vs.(0.665±0.085)g/cm2,t=2.50,P<0.05],while there was no obvious change of BMD in control group[(0.620±0.085)g/cm2 vs.(0.617±0.075)g/cm2,P>0.05].BMD was higher in treated group than in control group after treatment(t=2.46,P<0.05). Conclusions Fosamax can promote formation of bony callus,increase BMD and shorten external fixation time of radius distal osteoporotic fracture in postmenopausal women.
7.Clinical efficacy of alendronate treatment of early-stage adult nontraumatic avascular necrosis of femoral head

Objective To evaluate the influence of the alendronate treatment in early-stage adult nontraumatic avascular necrosis of femoral head. Methods The 83 patients with nontraumatic avascular necrosis of femoral head were enrolled in this study. They were given oral alendronate 70 mg weekly, and evaluated with Harris criteria before and after treatment. Results In the patients with ARCO Ⅰ necrosis, the scores of pain and function were higher after treatment than before [(41.45±3.55) scores vs. (38. 48± 5.55) scores, t = 3. 70, P = 0. 001; (45.06 ± 1.50) scores vs. (43.97 ±2.31) scores, t= 3.76, P= 0. 001]. In the patients with ARCO Ⅱ necrosis, the scores of pain,function and activity were also higher after treatment than before [(40. 40±4.31 ) scores vs. (37.32±6. 65) scores, t=4.06, P=0.00; (42.90±2.70) scores vs. (41.66±3.35) scores, t=3.15, P=0.003; (4.76±0.47) scores vs. (4.42±0.70) scores, t=3.35, P=0.002]. Conclusions Alendronate is effective in treatment of early-stage adult nontraumatic avascular necrosis of femoral head, in particular for ARCO Ⅱ patients. But its long-term effect is worth researching in future.
8.Quality Analysis of Compound Yinqiao and Paracetamol and Chlorphenamine Maleate Capsules

Objective: Combining with national drug sampling program,to evaluate the current quality situation and problems of compound Yinqiao and paracetamol and chlorphenamine maleate capsules by testing and analyzing 78 batches of samples collected from the realm of drug production and circulation all over the country in 2015.Methods: As the current standard could not control the product quality, and combined the prescribed examination with exploratory research, the method of HPLC was used to determine the contents.GC and TLC was respectively used to identify peppermint oil and study on forsythin.Near infrared spectrum database was established by near-infrared spectroscopy (NIRS), which provided the basis for rapid testing.The results of the prescribed examination and exploratory research were statistically analyzed.Results: There were significant differences in the results of the prescribed examination and exploratory research.Conclusion: The results of exploratory research show that there are many defects in the statutory standards.The product quality of different manufactures was divers, and it is necessary to guide them to improve the preparation process and the quality.More exclusive, accurate and sensitive methods should be used to comprehensively control the quality.
9.Identification and differentiation of breast cancer stem cells under tumor microenvironment

BACKGROUND:Breast cancer stem cel s have a greater impact on the occurrence and metastasis of breast cancer. Under simulated tumor microenvironment, we can better analyze the proliferation and differentiation of breast cancer stem cel s. OBJECTIVE:To explore the tumor microenvironment effect on the differentiation of breast cancer stem cel s. METHODS:Breast cancer cel s and MCF-7 cel s were primarily cultured in fibroblast supernatant and serum-free PCM-2 medium, and formation of breast cancer cel s microspheres was observed. Proliferative ability of breast cancer cel s was detected using MTT colorimetry, and the surface markers of breast cancer stem cel s and epithelial-mesenchymal transition markers were measured using immunocytochemistry and RT-PCR methods. RESULTS AND CONCLUSION:The diameter of primary cel microspheres was larger in the serum-free PCM-2 medium than in the fibroblast supernatant, but the culture speed was faster in the fibroblast supernatant than the serum-free PCM-2 medium. At 3 days of primary culture, the expression of ALDH1 in primary cel s was greatly higher in the serum-free PCM-2 medium than in the fibroblast supernatant. However, the expressions of E-cadherin and vimentin were up-regulated in the fibroblast supernatant than in the serum-free PCM-2 medium. In addition, the expressions of E-cadherin and vimentin in MCF-7 cel s cultured in the fibroblast supernatant were up-regulated, while the expressions of ALDH1 and Oct-4 were downregulated. These findings indicate that the tumor environment has some certain effects on the growth and differentiation of breast cancer stem cel s, and some cytokines secreted from fibroblast supernatant can promote the proliferation and differentiation of breast cancer stem cel microspheres to some extent.
