2.Effect of SGK1 on the Protection of Ischemia Reperfusion Model Rats

Objective To seek the effect of SGK1 on the protection and prognosis of cerebral ischemia reperfusion. Methods Animal model of rats was used to construct them into experimental group(A)and control group(B). The impact of cerebral ischemia reperfusion on the hippocampus neuron cell apoptosis was simulated by making the overexpression of SGK1 and using PI3K inhibitors LY294002 to deal with animal models. Results The overexpression of SGK1 could reduce the neuron cell apoptosis caused by cerebral ischemia reperfusion to a certain extent,reverse the expressions of Cleaved aspas 3 protein,pro-apoptotic protein Bax and inhibitor of apoptoasis protein Bcl-2 through the PI3K/Akt-mediated signaling pathway. Conclusion SGK1 can protect the tissues with ischemia-reperfusion,which may provide the biological evidence for future clinical applications.
3.A clinical research about predicting the changing of malignant tumor patients serum glucose after chemotherapy by SELDI technology

Objective By surface-enhanced laser desorption / ionization time-of-flight mass spectrometry(SELDI-TOF-MS), the serum pmteomic fingerprints related with the changing of malignant tumor patients' serum glucose after chemotherapy was selected and constructed as an predictive model. Methods By SELDI-TOF-MS, the serum of 182 malignant tumor patients who had received chemotherapy were tested, and the pmteomic fingerprints were received. After 2 years follow-up, all the patients were divided into 3 groups: the euglycemia group(136 people), the carbohydrate tolerance abnormality group(27 people), and the diabetes mellitus group (19 people). The proteomic fingerprints were analyzed by Biomarker Wizard Software and the idio-proteomic fingerprint of protective models were constructed by BPS (biomarker pattern software). Results The diagnosis model composed with 2 proteins (M/Z values were 5298 and 9608) could classify the carbohydrate tolerance abnormality group, and the diabetes mellitus group correctly. In the test model, the sensitivity and specificity were 81.48 %(22/27) and 100.00 %(17/17) respectively, the accuracy was 88.64 % (39/44). The diagnosis model composed with 3 proteins (M/Z values were 10324, 2761 and 4084) could classify the diabetes mellitus group and the euglycemia group correctly. In the test model, the sensitivity and specificity were 62.35 %(53/85) and 88.24 %(15/17) respectively, the accuracy was 66.67 %(68/102). The diagnosis model composed with 6 proteins (M/Z values were 5895,6010,6099,3930,5430 and 2495) could classify the diabetes mellitus group and the the carbohydrate tolerance abnormality group correctly. In the test model, the sensitivity and specificity were 77.65 %(66/85) and 96.30 %(26/27) respectively, the accuracy was 82.14 %(92/112). Conclusion SELDI-TOF-MS could be utilized to analyze protein profiling in screening serum glucose changing-related biomarkers and developing diagnostic and predictive patterns, and the developed patterns may be used to predict the changing of serum glucose after chemotherapy in malignant tumor patients.
4.Relations between increasing of blood sugar and cytorines after chemotherapy

Objective To explore the correlation between the increasing of blood glucose and cytokines in malignant tumor patients after chemotherapy.Methods According to the blood glucose after chemotherapy,155 cases of malignant tumor patient were divided into 3 groups:euglycemia group,IGT(impaired glucose tolerance)group and DM(diabetes mellitus)group.The variance of cytokines in the 3 groups were analyzed.Results Compared with pre-chemotherapy,IL-6,TNF-αincreased greatly in IGT group and DM group,while IL-10 decreased greatly.The statistical difference were obvious compared with euglycemia group.Conclusion There are close correlation between the increasing of blood glucose and cytokines in malignant tumor patients after chemotherapy.
5.Efficiency Evaluation of Iodized Salt Supply in Prevention of Iodine Deficiency Disease in Tianjin in 1995-2005

Objective To evaluate the efficiency of ten years of iodized salt supply in prevention and control of iodine deficiency disease (IDD) in Tianjin. Methods 1 200 pupils aged 8-10 years with 1:1 of sex ratio were chosen in the regions where iodized salt supply had been conducted for 10 years (1995-2005), the prevalence rate of goiter, the level of urine iodine and the iodized salt consumption were investigated. Results The prevalence rate of goiter of children aged 8-10 years in Tianjin decreased from 32.6% to 1.8% after 10 years of iodized salt supply, the median of urinary iodine increased from 96 ?g/L to 228 ?g/L. In the high water iodine regions, the prevalence rate of goiter was 8.1%, the median of urinary iodine was 576 ?g/L. Conclusion 10 years of iodized salt supply can significantly prevent and control IDD in the regions where there is iodine deficiency in drinking water, but the problem should not be neglected that iodine excess in the regions where the concentration of iodine in drinking water is higher. Reasonable iodine supplement is the key to prevent and control IDD as a whole.

7.Comparison of endoscopic and open McKeown esophagectomy

Objective To evaluate the feasibility of endooscopic minimally invasive McKeown esophagectomy in the treatment of esophageal carcinoma. Methods From June 2012 to August 2015, the data of 180 patients with esophageal carcinoma was retrospectively analyzed. The patients were divided into endoscopy McKeown esophagectomy group (EME group) and open McKeown esophagectomy group (OME group), each group had 90 patients. The clinical pathological data, perioperative data and postoperative complications between the two groups were analyzed. Results The operation time in EME group was longer than that in OME group [(289 ± 30) min vs. (252 ± 28) min, t= 8.063, P< 0.05]. The intraoperative blood loss and postoperative blood loss on the first day after operation in EME group were less than those in OME group [(178.94 ± 47.84) ml vs. (323.44 ± 59.51) ml, t=17.952, P<0.05; (220.93 ± 49.07) ml vs. (362.59 ± 68.37) ml, t= -15.968, P< 0.05]. There was no significant difference between the two groups in the number of lymph node dissection (t= -1.913), postoperative hospitalization days (χ2= 4.858) and postoperative complications (exact probability method) (all P> 0.05). Conclusions Endoscopic minimally invasive McKeown esophagectomy has the same effect as open surgery, and trauma is small. Therefore, for the patients who are suitable for the minimally invasive surgery, it can be preferred.
8.The characteristics of attentional bias in high anxiety sensitivity individuals

Objective To investigate the characteristics and mechanisms of attentional bias in anxiety sensitivity individuals.Methods By using Anxiety Sensitivity Index (ASI),23 participants were included in the high score group and 20 participants were included in the low score group.Then,2 (between-subject factor:the high and low score group) ×2 (within-subject factor:the positive and negative picture) mixed design experiments were adopted.Emotional faces picture pairs were chosen as stimuli.Picture pairs were presented 100 ms in experiment 1 and 1 250 ms in experiment 2.Dot-probe task was adopted to inspect the attentional bias and the response time and correct rate were recorded.Results Experiment 1 implied the main effect of type of pictures was found in mixed design experiments(F(1,41)=4.40,P<0.05).The reaction time of two groups in positive pictures was greater than zero((12.22±30.24) ms vs (10.07±21.55) ms).It showed input effect to positive pictures.An input effect due to the reaction time of the high score group was greater than zero to negative pictures((4.81± 17.88)ms),while the low score group tended to avoid the negative pictures ((-6.81 ±21.33) ms).Experiment 2 implied positive score was not significant between two groups (F(1,41) =0.29,P>0.05).And positive score showed the attentional bias to some certain extent.Significant outcome was found by negative score between two groups (F(1,41) =6.41,P<0.05).It implied that the high score group tended to avoid the negative pictures and the low score group had the tendency of input effect.Conclusion At the initial stage of attention,anxiety sensitivity individuals had the attentional bias to negative emotional faces and avoidance in the late stage of attention.It suggests that the attentional bias of anxiety sensitivity individuals may have an important effect on the development of the mental disorders.
9.Study on the expression of LGT proteinic fingerprint in the change of TSH,FT3,FT4

Objective Exploration of TSH, FT3, FT4 changes in critical condition prior to death.Methods 84 patients that were anti-tumor therapy acceptable and not associated with the treatment of thyroid disease in patients with terminal tumors. Age from 24 to 85-year-old, male 56, female 28. The use of SELDI technology (surface-enhanced laser description / ionization time-of-flight mass spectrometry protein chip technology) checks to M/Z for the 11 100 + H~11 900 + H peak of the cluster, divided into the positive group for critically ill patients with early warning protein (LGT) and the negative group for it. All patients were given TSH, FT3, FT4 inspection. The results were statistically analyzed. Results TSH, FT3, FT4 in the LGT positive group was significantly lower than in the negative LGT group. There was no correlation between TSH,FT3 and FT4 in the LGT positive group;In the negative LGT group FT3 and FT4 had a positive correlation,and FT3 and TSH no relevance. Conclusion Thyroid function is normal in the stage of cancer patients stable condition. Possible pituitary-hypothyroidism may occur as the disease advances to the end or before death.This may be an important factor causing the acceleration to the death of the patients. However, the cause and effect between this phenomenon and the early warning of protein fingerprint group(LGT) is still not clear. In critical condition checking thyroid function and render supplementary amount of thyroid hormone in accordance with the checking results may be beneficial.
10.Whole-genome sequencing: a new approach for understanding of pathogenesis and individualized treatment of cancer.

With the development of sequencing technology, the cost of whole-genome sequencing was significantly declined.Meanwhile, with the application of combined whole-genome sequencing with epigenetic analysis on methylation and histone acetylation, the comprehensive and systematic analysis of numerous samples became a reality and we are able to re-understand the genesis and development of cancer. New ideas are emerging in comparative genomics research methods, from comparison of genomes among different individuals to horizontal self-comparison of different tissues and vertical self-comparison of genomes recently.Individualized diagnosis and treatment of cancer has shown a bright future.
