1.Effect of ketamine on the cGMP level in different brain areas in a rat model of CFA-induced arthritis

Objective To investigate the effect of ketamine on the cGMP content in cerebral cortex, hypothalamus and brain stem in a rat model of CFA-induced arthritis. Methods Twenty-four SD rats weighing 200-220 g were randomly divided into three groups with 8 animals in each group, group Ⅰ: normal rat + intraperitoneal normal saline (ip NS);group Ⅱ : arthritic rat+ ip NS;group Ⅲ: arthritic rat + ip ketamine 100mg?kg-1 . Arthritis was produced by injecting CFA 0.05 ml into left ankle joint which developed inflammatory response within 24h and became hyperalgesic. The hyperalgesia persisted for more than 6 weeks. Three days after the arthritis model was established, paw withdrawal latency (PWL) to radiant heat was measured. 15 min after ip ketamine the animals were decapitated and cerebral cortex, hypothalamus and brain stem were separately isolated for determination of cGMP content using radioimmunoassay. 2.Clinical observation on the treatment of 69 patients with diabetic neurogenic bladder with acupoint injection therapy

Objective To investigate an effective therapy to cure Diabetic Neurogenic Bladder (DNB).Methods By adopting multi-center,random,comparative and single-blind clinical experiments,138 patients with DNB from three centers were divided equally into two groups,namely a control group and a therapy group.Patients in the therapy group were treated with acupoint injection therapy with Astragalus injection,while others were treated by western medical based therapy.After four weeks,the effectiveness was collected.Results After the treatment,the TCM syndrome integral of the treatment group and the control group was (18.47± 1.67) and (23.19±2.82) respectively,both reduced than that before the treatment[(29.25±2.12) and (29.13 ± 1.69) respectively]; the difference between the two groups after treatment was statistically significant (P<0.05) ; bladder residual urine volume in the treatment group and the control group was (88.47± 16.7) ml and (143.19±28.2)ml respectively,both reduced than that before the therapy [(308.90±22.6)ml and (305.90± 20.8)ml respectively],the difference between the two groups after treatment was statistically significant (P<0.05) ;The total effective rate was 88.4% and 72.5% in the treatment group and the control group respectively,showing statistical difference(P<0.05).Conclusion It is confirmed that acupoint injection therapy with Astragalus injection is an effective curative method for DNB.
3.Investigation of Wnt/β-catenin signaling pathway on regulation of Runx2 in cementoblasts under mechanical stress in vitro.

OBJECTIVEPeriodontal tissue remodeling includes remodeling of alveolar bone, periodontal ligament, and cementum. Cementoblast plays a main role in repairing root resorption. Canonical Wnt/β-catenin signaling can promote the odontogenic differentiation in osteoblast. However, the mechanism on how the orthodontic force influences the function of cementoblast and the relationship between the canonical Wnt/β-catenin signaling and Runx2 of cementoblast are not yet known. The aim of this study is focus on this relationship.
METHODSOCCM30 cementoblasts were subjected to mechanical strain by four-point bending system with tension stress for 0, 3, 6, and 12 h. They were pretreated with different concentrations of Dikkopf-1 (DKK1) for 48 h. Western blot analysis was performed to detect the β-catenin levels in the nucleus. Runx2 mRNA was observed by real-time quantitative polymerase chain reaction (RT-PCR). OCCM30 cementoblasts were then pretreated with 150 ng · mL(-1) DKK1 for 48 h and subjected to mechanical strain by FX4000T system with tension stress for 12 h. Western blot analysis was conducted to detect the β-catenin levels in the nucleus, and Runx2 mRNA was observed by RT-PCR.
RESULTSOCCM30 cementoblasts had significantly higher Runx2 mRNA and β-catenin levels after being loaded with mechanical stress. The amount of Runx2 mRNA in OCCM30 cementoblasts was significantly decreased by DKK1. When OCCM30 cemento-blasts were pretreated with DKK1 without stress, their β-catenin level was significantly decreased by DKK1 and Wnt signaling was blocked. When they were not pretreated with stress, the β-catenin level with DKK1 was lower than that without DKK1. Without DKK1, the β-catenin level in OCCM30 cemento- blasts increased afterbeing loaded with mechanical stress. With DKK1, the β-catenin level in OCCM30 cementoblasts, which were loaded with mechanical stress, was higher than that without mechanical stress.
CONCLUSIONCementoblasts had higher Runx2 mRNA expression under mechanical stress because of the Wnt/β-catenin signaling pathway effect.

4.Investigation of correlative factors of non-operative treatment in the senile cataract and countermeasure of health education

ObjectiveTo investigate correlative factors of non-operative treatment in the senile cataract, and provide gist for the health education measures.Methods1513 retiring old people had a whole medical examination, and the data was analyzed.ResultsThe proportion of binoculus cataract was 45.9%. Predilection age was 70~79 years old. Cognition information and iatrogenic effects were major risk factors in the non-operative therapeutic agents. Cataract was the third in the elder chronic disease.ConclusionMany senile patients with cataract have not enough knowledge about cataract operation although having better medical treatment and economy conditions. So the closed attention to the health education must be paid in the future.
5.SNP microarray analysis of retention abortion chorionic villus.

OBJECTIVETo compare villus cell culture and karyotype analysis with single nucleotide polymorphism (SNP) microarray technology for the detection of chorionic villus chromosome in patients with retention of abortion.
METHODSForty cases were analyzed with the two methods.
RESULTSChorionic villus culturing was successful in 29 cases, among which 10 were found to have an abnormal karyotypes. For the SNP microarray analysis, all 40 cases were successful, among which 16 were shown to have an abnormal molecular karyotype.
CONCLUSIONSNP microarray technology is highly accurate and specific, which is particularly suitable for the detection of chromosomal deletions or duplications, uniparental disomy, low-percentage mosaicism and other chromosomal abnormalities. It has provided an effective supplement to the conventional chorionic villus culture and karyotype analysis.

6.Donations after cardiac death kidney transplantation in northwest China.

OBJECTIVETo explore the effect of donations after cardiac death (DCD) kidney transplant performed in northwest China and the measures for management of delayed graft function (DGF).
METHODSIn the period of 2011-2013, a total of 51 families of DCD donor gave their consent to organ donation by signing the informed consent with the help by a Red Cross Organization (ROC) coordinator, and 102 kidneys were retrieved by organ procurement organization (OPO) teams. Ninety-four operations of renal transplantation were carried out in our hospital. All the patients were followed-up and based on the occurrence of DGF after transplantation, they were divided into DGF group and non-DGF group for comparative studies.
RESULTSThe success rate of donation after cardiac death was 29.3%, and the incidence of post-transplantation DGF was 27.7%. The 1-year human/kidney survival rate was 98.9%/95.7%. Within six months after the transplant, the values of eGFR in DGF group were significantly lower and serum creatinine significantly higher than those in non-DGF group (P<0.05), but no significant differences were found between the two groups thereafter (P>0.05). The occurrence of DGF in LifePort mechanical perfusion cohorts was significantly lower than that in the simple cold preservation group (21.5% vs. 41.4%, P<0.05).
CONCLUSIONThe overall effect of DCD kidney transplant is good despite a high incidence of early DGF, and we recommend the use of low-temperature mechanical perfusion for storage and transportation of DCD donor kidney.

7.Changes in CD40 expression in the pallium and hippocampus in epileptic rats.

OBJECTIVE:
To investigate the expression of inflammatory molecule CD40 in the pallium and hippocampus of rats after status epilepticus (SE).
METHODS:
The expression of CD40 in the pallium, the different areas of hippocampus and the different cells from the lithium-pilocarpine epileptic rats at different time points were examined by immunohistochemistry and double-immunofluorescent labeling.
RESULTS:
After SE, CD40 expression was obviously inhibited, especially in hippocampus. CD40 was mainly expressed in the activated microglia. CD40 positive cells reached a peak at the 3rd day and returned to a slightly higher level at the 7th day after SE compared with the level before SE.
CONCLUSION
Elevation of CD40 expression in the activated microglia can promote inflammatory injury of rat's hippocampus, suggesting that CD40 induced-signal pathway is involved in inflammatory injury in the hippocampus after SE.

8.Status risk factors and prevention and control strategies of familial aggregation of Helicobacter pylori infection

Objective To investigate the status of familial aggregation of Helicobacter pylori (Hp) infection in Jinniu District, Chengdu, and analyze its risk factors so as to provide a basis for developing prevention and control strategies of family aggregation of Hp infection. Methods A total of 172 subjects in the Second Affiliated Hospital of Chengdu Medical College · 416 Hospital of Nuclear Industry from January 2022 to January 2023 were selected as the research subjects. All subjects underwent 13C-urea breath test (13C-UBT) to diagnose whether there was Hp infection. Analyze the current situation of family aggregation of Hp infection in the region, collect general data of survey subjects, analyze the relevant factors affecting Hp family aggregation infection, and develop prevention and control strategies based on this. Results A total of 242 people from 97 households were surveyed, and the Hp family aggregation rate was 29.33%. Univariate analysis showed that there were statistically significant differences in family aggregation of Hp infection in terms of different age groups (χ2=9.719, P=0.008), marital status (χ2=8.496, P=0.014), occupations (χ2=19.462, P<0.001), frequencies of dining out (χ2=5.457, P=0.019), previous Hp test results (χ2 =4.131, P=0.042) and test results after treatment (χ2=12.000, P=0.001), with statistical significance (P<0.05). Multivariate logistic regression analysis showed that the frequency of dining out 2 days or more per week and a positive Hp test results in the past were risk factors for family aggregation of Hp infection, while the occupation of teachers/medical staff/management/technology personnel and a negative Hp results after treatment were protective factors (P<0.05). Conclusion Family aggregation of Hp infection is related to family members' occupation, frequency of dining out, previous Hp test results and Hp test results after eradication, which deserves attention in clinical practice.
