1.STUDIES ON ORIGIN AND HEMOPOIETIC FUNCTION OF PERITONEAL CELLS OF MICE

In this paper we established a method of preparing chromosomes of peritoneal cells and made use of the method for observing the Y-chromosome of marrow and peritoneal cells of female mice which had been irradiated with 850 rad ~(60)Co-?, rays and then transplanted with marrow cells of male mice. The results showed that the chromosome karyotypes of the marrow cells of the female mice were of the XY type of male mice. It means that the marrow cells of the recipients were substituted by those of the donors. At the same time, the chromosome types of the peritoneal cells of the recipients were also of the XY type. It is suggested that the peritoneal cells were derived from the marrow cells.By the use of the method for assaying CFU-C and CFU-S of mouse marrow cells, CFU-C and CFU-S of peritoneal cells of mice were assayed. The result demonstrated that there were a number of CFU-C and CFU-S among the peritoneal cells.On the observation of the cell morphology of CFU-C and CFU-S, it was discovered that CFU-C differentiated mainly into mononuclear phagocytes, while CFUS had the ability to differentiate into erythrocytie series and granulocytic series, which implies that there were some multipotential stem ceils among peritoneal cells of mice.When more than 10~6 peritoneal cells per mouse were transplanted into the mice which had received 850 rad of ~(60)Co-? rays, their life span could be prolonged.It is still a question unsetteled that whether the stem cells in the peritoneal cavity differentiate diroctly into the mononuclear phagocytes or the mononuclear cells in the blood penetrate into the peritoneal cavity and then differentiate into the macrophages.
2.Correction of inverted nipple with bilateral areolar rhomboid dermal flaps by crossed filling and supporting

Objective To explore a new methodology with bilateral rhomboid dermal flaps for surgical treatment of inverted nipple. Methods 36 cases with 64 inverted nipples were involved in this study. Among these cases, 8 of them were evaluated as mild degree, while 19 moderate and 9 severe. In this procedure, the bilateral rhomboid incisions were designed. The rhomboid skin was deepithelialized and two dermal flaps were created. Then these two dermal flaps were filled crosswise through the tunnel beneath the nipple and fixed as supporting. After the operation, the continuous traction of the nipple lasted for one week, and sutured out 10 days later. Results None of these 36 cases appeared nipple blood circulation disorder after operation. Postoperative follow-up for 6 months-3 years showed that the appearance of the nipple was satisfactory. There were no recurrence and sensory disturbance. Conclusions The method introduced in this study is simple, the appearance of nipple is natural and no complication has been found. The incision is larvaceous while the scar is invisible. Meanwhile this method is also benefit for reserving the function of breast-feeding. It is proved to be an ideal method for correcting the inverted nipple.
3.MORHPOLOGICAL AND HISTOCHEMICAL OBSERVATION OF MYOCARDIUM OF KESHAN DISEASE

Keshan disease was an endemic cardiomyopathy in China. The very low selenium intake of local people was considered to be an important causal factor. The main pathological characteristics of this disease was multifo-cal necrosis and fibrous replacement of myocardium that was scattered throughout the wall of all chambers.Two patterns of myocardial necrosis, myofibrillar pattern and mitochondrial pattern were distinguished in electron microscopy. The myofibrillar pattern was characterized by myofibril segmentation. It agreed well with the contraction band necrosis described in light microscopy. It was mainly seen in acute Keshan heart and might be related to circulatory disorders. Mitochondrial pattern was identical with myocytolysis of conventional pathology. It represented the typical lesion of Keshan disease.Mitochondria showed early and conspicuous changes in involved myo-cytes. Myofibrillar damage seemed to be secondary to the mitochondrial injury in the development of myocytolysis.Histochemical studies revealed that the acid phosphatase activity was obviously increased in muscle fibers surrounding the necrotic foci, and the succinic dehydrogenase activity was greatly reduced in damaged myocardio-cytes.
4.Development of a Forensic Multiplex Amplification STR Kit for 15 Autosomal STR Loci and 10 Y-STR Loci

Objective To establisha multiplex STR genotyping m ethod for autosom al STR and Y-STR loci in forensic biological practice. Methods W idely used autosom al STR loci and Y-STR loci w ere se-lected. A set of PC R prim ers w as designed, and a 5-dye fluorescent labeled STR multiplex PC R reagent kit w as developed. Results A kit w as developed w hich can sim ultaneously detect 15 autosom al STR loci, 10 Y-STR loci, and an Amelogenin. Conclusion The 15 autosom al STR plus 10 Y-STR kit in com bination w ith capillary electrophoresis m ethod w as used to STR genotyping w ith accurate and reli-able results. The new one-step testing kit can potentially be w idely used in forensic cases and D N A databank in the future.
